Development of the 3rd Generation Anticancer Platinum Complex as New Drug

  • Life Science Research Center of SK Chemicals has developed a 3rd-generation anticancer platinum drug for the first time in the nation′s 100-year-old pharmaceutical industry. The Korea Food and Drug Administration (KFDA) approved the sale of "Sunpla" (code name SKI 2053R, general name : Heptaplatin) on July 14, 1999 for the treatment of advance, metastatic gastric cancer. Cisplatin, the 1 st-generation anticancer drug, which was developed by Bristol-Myers of the United States in 1976, is one of the most potent anticancer drugs and is a major component of combination chemotherapy for a variety of human cancers. However its clinical usefulness has frequently been limited not only by undesirable side effects such as severe renal toxicity, nausea, vomiting, ototoxicity, and neurotoxicity but also by the development of resistance. Carboplatin, the 2nd-generation anticancer platinum drug, which was also developed by Bristol-Myers in 1986, has modified the problems of the renal and gastrointestinal toxicities of cisplatin. Carboplatin, however, has no enhanced therapeutic efficacy over cisplatin and does not possess the property to overcome cross-resistance to cisplatin.

  • Recently, people are gradually concerned about environmental issue, bionics, environmental-friendly or biocompatible materials. Poly(vinyl alcohol) (PVA) is suitable for these materials, because it is typically water-soluble polymer that have linear-flexible chains, a material of no toxicity for human, and biodegradable polymer[1]. Antimicrobial Activity of Biosurfactants

  • Biosurfactant has antibiological activities. Due to its low toxicity, biosurfactant can be applied to human health care products. Posseing the antibiological activities as well as the surfactant properties implies that biosurfactant can be widely used in the cosmetic industry. In this presentation, applications of biosurfactant have been reviewed including the antimicrobial activities of sophorolipid. Sophorolipid, a biosurfactant produced from Candida bombicola ATCC 22214, showed antimicrobial activity against Bacillus subtilis, Staphylococcus xylosus, Streptococcus mutans, and Propionibacterium acne at 4, 1, 1, 0.5 ppm as MIC (minimum inhibitory concentration). Also 100 ppm of sophorolipid inhibited 50% of cell growth of plant pathogenic fungus, B. cinera. However, sophorolipid showed no effect on the Escherichia coli., indicating its selective antimicrobial activity depending on the cell wall structure. Treatment of B. subtilis with sophorolipid increased the leakage of intracellular enzyme, malate dehydrogenase, indicating the possible interaction of sophorolipid with cellular membrane. Between lactone-type and acid-type sophorolipid, the former showed higher antimicrobial activity.

  • There shoul be high possibility of preventive and immune eficacy aqainst cancer when the anti-cancer plants are properly utilized in human dietary life as it is evident that considerable part of wild plantd or both medicinal use and ood are growing naturally in the mountainous area in korea, some of which have ben proved to be anti-cancer plants. this study, at this initial stage, has been done to acquire the fundamentals of 235 kinds of ante-cancer plants growing naturally in korea. from the results o thisresearch, the anti-cancer plants have been sorted out by amily and its number. and also the status of regional distribution of those plants and the kinds of anti-cancer plants used for both medicine and food were studied and experimented to see the medical efficacy against anti-tumor. this experiments were conduced in accodance with the total packed cell volume method and cyto toxicity method.

  • Coating ceramic balls activate water molecular in water as radiate far-infrared radiation of high efficiency, and then prevent formation of scale and corrosion of pipe. Therefore, but only keep away drop of heat exchange efficiency of boiler, not also remove formed scale. As a result, pipe of boiler has clean and has form thin protection film in inner of pipe. Also, the water treated by rust preventing system using the ceramic balls, that is harmless, tasteless, odorless, and non-toxicity in the human body, and it can use drinking water. This rust preventing system can save energy and protect environment.

  • Objectives : We review a result of studies until the present and suggest the method of medical treatment for the clinical treatment of Scolopendrid Herbalacupuncture. Methods : We analysis the paper of the bibliographic studies, the experiment studies and the clinical studies from 2001 developed Scolopendrid Herbalacupuncture and grope for the course of studies. Results : 1. Scolopendrid Herbalacupuncture is proved the clinical safety by the aninmal and human tests. 2. The pharmacological action of Scolopendra subspinipes mutilans L. Koch is anti-convulsive action, analgesic action, lowering blood pressure, anti-inflammatory action, anti-tumor action and microbe inhibition 3. Scolopendrid Herbalacupuncture has been a fine effect to the entrapment neuropathy and inflammatory. 4. Scolopendrid Herbalacupuncture was thought effective on a acute phase and to the excessive symptoms. The Sub-chronic toxicity experiment observing the response after hypodermic medication over 90 days, The Genetic-mutagenic toxity experiment and the clinical effect studies are necessary.

Biodistribution and Hemolysis Study of Terplex Gene Delivery System in Mice

  • Polymeric gene delivery system attracts profound attention as it shows less toxicity, versatility, and reasonable gene expression efficiency. Terplex system, a synthetic biopolymeric gene delivery system consisting of stearyl poly-L-lysine (stearyl-PLL) and low density lipoprotein (LDL) was evaluated for its body distribution of gene expression of exogenously administered pDNA after tail-vein injection in mice. Kidney and spleen are two major organs with highest gene expression, whereas liver and heart showed marginal gene expression among the organs examined. Hemolytic effect of the terplex system was evaluated using human red blood cells, where terplex system did not cause significant hemolysis at the concentrations above the experimental ranges, although unmodified PLL or stearyl-PLL without LDL did. Serum stability of terplex system against enzymatic degradation was also significantly enhanced, presumably due to the steric stabilization from the polymers. Based on these findings and along with its high in vitro transfection efficiency, terplex system could serve as a safe and efficient polymeric gene delivery system with many applications for the in vivo gene therapy.

  • Methylmercury (MeHg) is a well-known neurotoxicant that causes severe damage to the central nervous system in humans. Many reports have shown that MeHg is poisonous to human body through contaminated foods and has released into the environment. Despite many studies on the pathogenesis of MeHg-induced central neuropathy, no useful mechanism of toxicity has been established so far.(omitted)

  • Cadmium is an environmental pollutant and exhibits nephrotoxicity, hepatotoxicity and immunotoxicity. Recently, cadmium was found to induce DNA fragmentation, a biochemical hallmark of apoptosis, in cultured renal cells, hepatocytes and neuroblastoma cell. Therefore, the various toxicities of cadmium are thought to be caused by the induction of apoptosis. Lipids-derived pro-apoptotic ceramide has emerged as an important intracellular signaling molecule that mediates diverse cellular effects, of which programmed cell death, or apoptosis, has attracted significant interest.
