Production of Reuterin by Immobilized Lactobacillus reuteri (Lactobacillus reuteri의 고정화 세포를 이용한 루테린 생산)

  • Lactobacillus reuteri residing in human and animal intestines converts glycerol into reuterin (antimicrobial substance) in anaerobic condition. Attempt was made to increase production efficiency of L. reuteri by employing immobilized cells. L. reuteri was immobilized in agarose beads, which were then reacted with 250 mM glycerol solution. Batch-type production of reuterin with immobilized cells (0.5% agarose beads) lasted for about 36 h, although reuterin production decreased with passage of time. In continuous-type production, period of reuterin production with immobilized cells was extended about twofold and production ratio increased 1.5-fold (502 mM) compared with suspended cells (315 mM). Maximum concentration of reuterin reached 47 mM at 80 min after reaction with glycerol solution. Results of this study indicate that immobilization of Lactobacillus reuteri in agarose beads increased reuterin production.

A Study on the Properties and Functions of the Twelve Main Meridians: Focusing on the Relationships between the Internal Organs and the Meridians (12 경락의 속성과 기능에 대한 연구: 경락과 장부의 관계를 중심으로)

  • Objectives and Methods : This study is to understand the properties and functions of twelve meridians. It aims to understand the meaning of the meridian names, and to study the properties of the six qi and the method of movement of the meridians by analyzing the six qi of the three yin and three yang from a meteorological perspective. The functions of twelve meridians based on the relationship between the operation of the internal organs and the meridians are then examined. Results and Conclusions : (1) The three yin and three yang includes the properties of six qi at the meridians. (2) The fundamental qi at reverting yin meridian lesser yin meridian and greater yin meridian has the properties of wind, heat and dampness, respectively, which are yang qi, while the fundamental qi at lesser yang meridian, yang brightness meridian and greater yang meridian has the properties of ministerial fire, dryness and cold, respectively, which are yin qi. (3) In the circulation of meridian qi, yang qi ascends at yin meridians while yin qi descends at yang meridians. (4) Three yin meridians belonging to the viscera help the function of intestines, which saves essence qi and then makes it go upward, while three yang meridians belonging to the bowels help the function of converting foodstuff and making it go downward. (5) In the human body, the internal organs play the primary role, and the meridians serve as helpers of the functioning of the internal organs by controlling the six qi.

Identification of Anti-Angiogenic and Anti-Cell Adhesion Materials from Halophilic Enterobacteria of the Trachurus japonicus

  • The halophilic enterobacteria, Enterobacteria cancerogenus, was isolated from the intestines of the fusiform fish (Trachurus japonicus) to yield a protein-like material termed PLM-f74. PLM-f74 was characterized by strong inhibition ratios to angiogenesis (82.8% at the concentration of $18.5{\mu}g/ml$) and elevated antioxidative capacities with low toxicity. The PLM-f74 is a glycoprotein comprised of saccharides and amino acids. PLM-f74 inhibited cell adhesion that non-activated U937 monocytic cell adhesion to HUVECs activated with $IL-1{\beta}$ by 78.0%, and the adherence of U937 cells treated with the PLM-f74 and stimulated with $IL-1{\beta}$ to unstimulated HUVECs decreased by 102%. When both cell types were pretreated with PLM-f74, the adhesion of U937 cells to $IL-1{\beta}$-stimulated HUVECs was completely suppressed by 121% at a concentration of $18.5{\mu}g/ml$. PLM-f74 blocked signal pathways from VEGFR2, PI3K, ${\beta}$-catenin, and VE-cadherin to NF-kB, based on western bolt analysis. It also inhibited IL-l-stimulated HUVEC expression of the adhesion molecules, ICAM-l by 40%, VCAM-l by 60%, and E-selectin by 70% at the same concentration noted above. New anti-angiogenic and anti-cell adhesion materials showing elevated antioxidative capacities, and non-toxicity may be expected from these results.

  • "Uihakdokseogi(醫學讀書記)" is a casual work which mentions Ujaekyeong(尤在涇)'s thoughts on various problems found through extensive studying or in practice. The book does not focus on a single topic, thus the spectrum of the work is broad and is filled with problems which would interest most medical professionals. The major contents of the first volume of "Uihakdokseogi(醫學讀書記)" include differences of view on health preservation, the Five Circuit Phases[五運] and Six Atmospheric Influences[六氣], diseases, and the "Naegyeong(內經)", mentions on errors in transcription , and the gap between certain contents in "Yeongchu(靈樞)" and "Somun(素問)". U[尤在涇] asserts that Eum and Yang(陰陽) in a human should be in harmony as is the Gi(氣) of the sky and earth[天地] is, and that one should live according to the Gi(氣) of the four seasons to live a healthy life. He does not vary largely from the context of other writers on the matter of the Five Circuit Phases [五運] and Six Atmospheric Influences[六氣], and focuses on the concepts of predominant Gi[主氣], guest Gi[客氣], corresponding years[天符] , correlating years[歲會], and Taeeul corresponding years[太乙天符]. He mentions causes, symptoms, and treatments of various diseases such as cough due to asthenia of the viscera, stagnation of Yang(陽), stagnation of Eum(陰), abscess of the stomach, hard abscess of the intestines, upper emaciation, edema of the limbs, inability to raise the limbs, broken thigh, turbid fluid, inversion of Gi(氣) flow, sudden onset of fainting with cold extremities, diarrhea due to disorder of Gi(氣), and malaria. U[尤在涇] also points out faults of "Classic of Acupuncture and Moxibustion[甲乙經]" in the understanding of "Naegyeong(內經)".

Effects of added chlorides and their concentrations on growth and nitrate content in leaf lettuce(Lactuca sativa L.) in hydroponics.

  • Nitrate in edible vegetables is converted to nitrite by nitrate reductase(NR) and/or bacteria in intestines. Nitrite and amino, in the intestine of some animals and human, bind to form nitrosamine, which is toxic and known as carcinogen. This study was carried out to examine the effect of added chlorides and their concentrations on growth, yield and nitrate content in leaf lettuce plants in hydroponics. Seeds of lettuce cv, "Samsunjokchukmyon" were planted on April 29, and seedlings were planted on June 2, and were cultured until July 5 in 1998. KCI and CaCl₂ were used as chloride source and their concentrations were 1, 2 and 4 me/L, respectively, in the lettuce standard nutrient solution for National Horticultural Research Institute(NHRI). Completely randomized design with 3 replications was used. Nitrate content and NR activity were measured 2 and 5 weeks after planting(WAP). The obtained results were summarized as the follows : Leaf weight per plant was difference from harvest dates and treatments, but total leaf weight was not significantly different among treatments. Number of leaves was higher in KCI 2 me/L, CaCl₂1 me/L and control at 2 WAP than the others, and was higher in KCI 1 me/L, and control at 3 WAP than others, and was higher in control at 5 WAP. Total number of harvested leaves was the highest in control with 14, which followed by KCI 2 me/L and CaCl₂1 me/L. Nitrate content was decreased by addition of chloride in nutrient solution. Nitrate content in the 3rd and 9th leaves was significantly decreased. NR activity was higher in control and CaCl₂ addition treatments, while KCI addition treatments reduced NR activity. However, no direct relationship with nitrate was observed. Growth characteristics such leaf length and leaf width were not significantly influenced by chloride addition.

  • In the past, antibiotics and antimicrobial substances have been used for the purpose of promoting the growth of livestock or treating livestock, but various problems such as the presence of livestock products or resistant bacteria have emerged. Recently, regulations on the use of antibiotics have been strengthened worldwide, and probiotics are attracting attention as an alternative. Probiotic microorganisms have already been used for human use, such as intestinal abnormal fermentation, diarrhea, and indigestion. In livestock, beneficial microorganisms are increasing in use for the purpose of improving productivity, such as promoting livestock development and preventing diarrhea. Therefore, it is advisable to understand livestock probiotics in deeper and think about effective uses. The role of probiotics in the livestock sector is made with microorganisms themselves, so it is a substance that promotes livestock growth and improves feed efficiency by settling in the intestines of livestock, suppressing the growth of other harmful microorganisms, helping digestion and absorption of ingested feed, and helping to synthesize other nutrients. There is a need for a probiotic that suppresses intestinal bacteria by supplying probiotics used as a means to minimize the effects of stress in livestock management, thereby suppressing disease outbreaks by maintaining beneficial microorganisms and suppressing pathogenic microorganisms. The purpose of this paper is to develop a brand of feed additive probiotics to improve health conditions due to increased feed intake, improve the efficiency of use of feed nutrients, inhibit the decomposition and production of toxic substances, increase immunity, reduce odor in livestock, and improve the environment. We investigated and analyzed feed additive probiotics already on the market, and developed the naming and logo of suitable feed additive probiotic brands in livestock. We hoped that the newly developed product will be used in the field and help solve problems in the livestock field.

A Study in the Daqi- Theories by Yu Chang and Zhang Xi Chun (천담유창여장석순적 「대기」론)

  • In the Far-Eastern traditional medicine, Oi[Energy] implies a wide range of meaning and is emphasized. There is nothing that is not related to this Qi, as seen in physiology, pathology, the relationship between human body and nature, the movements of intestines and gyeongnak[energy networks], the process of outbreak and change of illness, remedial laws, the features and effects of drugs, and so on. Accordingly, Nei Jing also says, 'every sickness arises from OL' The Qi has multi-meanings, and each of lots Of past physicians researched and analyzed it in different perspective, thus making the Qi-theories much richer. Still. there were not so many physicians who discussed the theme of Daqi. The denomination of Daqi is seen in Nei Jing and Jin Gui Yao Lue, and the physicians like Yu Chang in Ming dynasty and Zhang Xi Chun in Cheng dynasty, etc. applied and utilized Daqi by exploring its functional actions for human life and associating it with clinical practices. Yu Chang said that Daqi is Xiong Zong Yangqi[Positive Energy in Breast] governing every Qi, and that if this Qi is full, it spreads through body and protects the body from sickness, and vice versa. Summarizing his researches on the Daqi in Jin Gui Yao Lue and on the opinions of Yu Chang as well as his experiences, Chang Xi Chun maintained that the Qi accumulated in breast must be named Daqi, which constitutes the contents of Zongqi[Chief Energy] mentioned in Nei Jing. Once the Qi is vacant, breathing is not smooth, whole body is enervated, spirit becomes dim, thinking ability falls drastically. Furthermore, if the Qi is extremely vacant or more worsens, breathing stops. And he prescribed the medicines including Sheng Xian Tang as remedies against the symptom of Daqi XiaXian[Fall in Great Energy]. The recognitions of Daqi by Yu Chang and Zhang Xi Chun are consistent with each other. At any rate, their theories and prescriptions may be high in practical value in contemporary clinics.

Probiotic Effects of Lactobacillus plantarum and Leuconostoc mesenteroides Isolated from Kimchi (김치에서 분리한 Lactobacillus plantarum과 Leuconostoc mesenteroides의 프로바이오틱 효과)

  • The probiotic effects of kimchi lactic acid bacteria (LAB), Lactobacillus plantarum (Lab. plantarum) and Leuconostoc mesenteroides (Leu. mesenteroides), were studied. Lab. plantarum KCCM 11352P (LPpnu) and Leu. mesenteroides KCCM 11353P (LMpnu) were isolated from kimchi and were the predominant LAB. We compared their probiotic effects with Lactobacillus rhamnosus GG (LRgg), a well-known probiotic LAB. LPpnu showed better probiotic activities than LRgg. LMpnu also exhibited almost equal activities as LRgg. These two kimchi LAB strains exhibited resistance to gastric and bile acid, adhesion to intestines, and thermal stability. In particular, LPpnu showed excellent probiotic properties. In addition, LPpnu showed greater antioxidant activity by scavenging DPPH radicals or hydroxyl radicals than LMpnu or LRgg. LPpnu also inhibited growth of HT-29 human colon cancer cells by inducing apoptosis, increasing Bax and suppressing Bcl-2 expression compared to LMpnu or LRgg. Taken together, LPpnu and LMpnu could be used as probiotics, and LPpnu exhibited the most beneficial probiotic activities with anti-oxidant and anti-cancer properties.

Effect of Lactobacillus brevis HY7401 Intake on the Serum Ethanol Concentration in Rats (Lactobacillus brevis HY7401 섭취가 쥐의 혈중 알코올 수준에 미치는 영향)

  • Possibility of Lactobacillus strains able to metabolize ethanol and acetaldehyde in vitro and in vitro was studied. Lactobacillus brevis strains showed higher alcohol dehydrogenase (ADH) and aldehyde dehydrogenase (ALDH) activities than those of other lactic acid bacteria strains. L. brevis HY7401 exhibited the highest ADH and ALDH activities and decreased considerable amounts of ethanol and acetaldehyde in vitro. L. brevis HY7401 cell intake significantly decreased serum ethanol levels in rats fed ethanol (4g/kg BW) compared to control groups. Ethanol level in small intestines of rats fed L. brevis HY7401 was about 50%, and their acetic acid concentration was twofold higher than control. Results reveal L. brevis HY7401, isolated from human, metabolizes ethanol and acetaldehyde in vitro and in vivo.
