Human Papillomavirus Genotypes Associated with Mucopurulent Cervicitis and Cervical Cancer in Hangzhou, China

  • Background: To investigate the infection status and predominant genotype distribution of human papillomavirus (HPV) infection among Chinese patients with mucopurulent cervicitis (MPC) or cervical cancer (CC) in Hangzhou. Methods: Initially, 217 cases of healthy cervix controls (n=50), acute MPC (n=89), and CC (n=78) were included; samples were collected between January 1, 2010, and January 30, 2013. Cervical specimens were screened for HPV using a nested polymerase chain reaction assay and DNA sequencing. Results: Overall prevalence of HPV infection was 16.7% in the control group, 51.9% in the MPC group, and 84.4% in the CC group. The predominant genotype detected in all 3 groups was the oncogenic variant HPV 16 (55.8%, 17.3%, and 6.3% in the CC, MPC and control specimens, respectively), HPV58 was the second most predominant HPV type in CC (9.1%), MPC (8.6%), and control group (4.2%). Most or all of the genotypes were oncogenic in the three groups. Conclusions: Infection with HPV was found to be prevalent among Chinese women with MPC or CC and oncogenic variants were in the majority. Therefore, peoples who suffered MPC with HPV DNA positive should be regularly followed-up, for prevention and early treatment of cervical cancer.

개흉술 후 발생한 종격동염의 대흉근-복직근 양경근피판을 이용한 치료 (Pectoralis Major-Rectus Abdominis Bipedicle Muscle Flap in Treatment of Postoperative Mediastinitis)

  • Although the incidence of mediastinal wound infection in patient undergoing median sternotomy for cardiovascular surgery is relatively low(less than 1%), it is not only a devastating and potentially life-threatening complication but also associated morbidity, mortality and cost are unacceptably high. During the past few decades various methods had been applied for the treatment of postoperative mediastinitis. Currently, chest wall reconstruction by using muscle flaps-especially pectoralis major muscle and rectus abdominis muscle are commonly selected for the reconstruction after wide debridement has become widely accepted. We performed bilateral pectoralis major-rectus abdominis muscles in-continuity bipedicle flap to overcome the limit of each flap for reconstruction of sternal defects in 17 patients. We analyzed the results of the surgery. Recurrent infection developed in 17.6% of cases and abdominal herniation was observed in one patient. There was no postoperative hematoma or death. We conclude that this flap is very valuable in reconstruction of the anterior chest wall defect caused by post-sternotomy infection because it provides sufficient volume to fill the entire mediastinum, and the complication rate compares favorably to that of other methods.

Mitochondria-Targeted Apoptosis in Human Cytomegalovirus-Infected Cells

  • Mitochondria often play central roles in apoptotic pathways, and disruption of the mitochondrial transmembrane potential (${\Delta}{\psi}m$) has been observed in various cells undergoing apoptosis. Human cytomegalovirus (HCMV) infection induces apoptosis in permissive cells; however, investigations of mitochondria-targeted apoptosis in HCMV-infected human foreskin fibroblast (HFF) cells have been limited. Here, we investigated the mitochondrial apoptosis pathway in HCMV-infected HFF cells. Flow cytometry analysis using JC-1 revealed that HCMV infection induces disruption of ${\Delta}{\psi}m$ in HFF cells when administered 24 h post-infection (hpi), and this disruption was maximized at 48 hpi. Moreover, cytochrome c, normally a mitochondrial inner membrane protein, was detected in cytoplasmic extracts of HCMV-infected cells, but not mock-infected cells, by western blot analysis at 24 hpi. A caspase activity assay based on fluorescence spectrophotometry using a fluorogenic substrate revealed an increase in caspase-3 activity at 48 hpi in HCMV-infected cells. Caspase-8 activity was increased at 72 hpi in HCMV-infected cells. These results imply that HCMV infection induces mitochondria-mediated apoptosis in HFF cells.

긴이형흡충과 여러 종의 장흡충에 혼합감염된 인체감염 1례 (One human case of natural infection by Heterophyopsis continua and three other species of intestinal trematodes)

  • 긴이형흡충(Heterophyopsis continua)에 의한 인체감염 1례를 보고한다. 이 사람은 53세 남자로 경상남도 진주시에 거주하였으며 수년 동안 반염수어와 민물고기를 즐겨 생식했다고 하였다. 대변 겁사에서 간흡충 요코가와흡충. 극구흡충의 충란이 검출되어 이 사람에게 praziquantel을 투여하였다. 치료 후 설사변에서 긴이형흡충 2마리, 유해이형흡충 12마리, 요코가와흡충 2,329마리와 호르텐스극구흡충 2따리를 수집하였다. 이 사람의 임상증상은 소화불량 상복부불쾌감, 식욕부진, 피로감 등이었다. 이 증상은 소수의 긴이형흡충, 유해이형흡충과 호르텐스극구흡충 감염보다는 많은 수의 요코가와흡충 감염에 의한 것으로 해석되었다. 이 증례는 문헌상 긴이형흡충 인체감염례의 국내 제3례에 해당된다.

간호대학생의 자궁경부암 및 인유두종 바이러스감염 예방행위의도 영향요인 (The Factor of Influencing Cervical Cancer and Human Papilloma Virus(HPV) Infection Preventive Behavioral Intention of Nursing Students)

  • The purpose of this study was to identify factors of influencing cervical cancer and Human papilloma virus(HPV) infection preventive behavioral intention of nursing students. The research design for this study was a descriptive survey design. Data collection was done using self-report questionnaires with 218 nursing students in B city from November 30 to 15 December, 2015. Data were analyzed using percentage, mean, standard deviation, t-test, ANOVA, Pearson's correlation analysis and multiple regression with SPSS Win 21.0 program. The significant factors of cervical cancer and HPV infection preventive behavioral intention was cervical cancer knowledge(${\beta}=.246$, p<.001), health belief about HPV vaccination(${\beta}=-.223$, p<.001), HPV vaccination(${\beta}=.173$, p=.008), and Gynecological Disease History(${\beta}=.145$, p=.026). And these factors explained 15.4% of the variance in the cervical cancer and HPV Infection preventive behavioral intention. In conclusion, the results of this study indicated that it's a need to develop the effective education program in addition to knowledge improvement on HPV and cervical cancer for nursing students.

  • Coronavirus disease 2019 (COVID-19) is caused by severe acute respiratory syndrome coronavirus 2 (SARS-CoV-2). Since the emergence of SARS-CoV-2 in the human population in late 2019, it has spread on an unprecedented scale worldwide leading to the first coronavirus pandemic. SARS-CoV-2 infection results in a wide range of clinical manifestations from asymptomatic to fatal cases. Although intensive research has been undertaken to increase understanding of the complex biology of SARS-CoV-2 infection, the detailed mechanisms underpinning the severe pathogenesis and interactions between the virus and the host immune response are not well understood. Thus, the development of appropriate animal models that recapitulate human clinical manifestations and immune responses against SARS-CoV-2 is crucial. Although many animal models are currently available for the study of SARS-CoV-2 infection, each has distinct advantages and disadvantages, and some models show variable results between and within species. Thus, we aim to discuss the different animal models, including mice, hamsters, ferrets, and non-human primates, employed for SARS-CoV-2 infection studies and outline their individual strengths and limitations for use in studies aimed at increasing understanding of coronavirus pathogenesis. Moreover, a significant advantage of these animal models is that they can be tailored, providing unique options specific to the scientific goals of each researcher.

Regulation of Innate Immune Response to Fungal Infection in Caenorhabditis elegans by SHN-1/SHANK

  • In Caenorhabditis elegans, SHN-1 is the homologue of SHANK, a scaffolding protein. In this study, we determined the molecular basis for SHN-1/SHANK in the regulation of innate immune response to fungal infection. Mutation of shn-1 increased the susceptibility to Candida albicans infection and suppressed the innate immune response. After C. albicans infection for 6, 12, or 24 h, both transcriptional expression of shn-1 and SHN-1::GFP expression were increased, implying that the activated SHN-1 may mediate a protection mechanism for C. elegans against the adverse effects from fungal infection. SHN-1 acted in both the neurons and the intestine to regulate the innate immune response to fungal infection. In the neurons, GLR-1, an AMPA ionotropic glutamate receptor, was identified as the downstream target in the regulation of innate immune response to fungal infection. GLR-1 further positively affected the function of SER-7-mediated serotonin signaling and antagonized the function of DAT-1-mediated dopamine signaling in the regulation of innate immune response to fungal infection. Our study suggests the novel function of SHN-1/SHANK in the regulation of innate immune response to fungal infection. Moreover, our results also denote the crucial role of neurotransmitter signals in mediating the function of SHN-1/SHANK in regulating innate immune response to fungal infection.

Human Papilloma Virus Detection by INNOLiPA HPV in Prostate Tissue from Men of Northeast Mexico

  • Background: Prostatic adenocarcinoma by Prosate cancer (PCa) is the most prevalent cancer and the second cause of cancer-related death among men in the Western world. Human papilloma virus (HPV) may be considered as a preventable risk factor. In this study, we assessed the frequencies of HPV infection in prostatic adenocarcinoma and benign prostatic hyperplasia (BPH) cases in Northeast Mexico. Materials and Methods: A total of 87 paraffin-embedded blocks (from 25 and 62 patients with definite diagnoses of BPH and adenocarcinoma, respectively) were selected and subjected to INNOLiPA HPV Genotyping to detect 28 high- and low-risk HPV types. The rates of infection were compared in the two studied groups. Results: INNOLiPA HPV demonstrated great sensitivity for HPV detection on paraffin-embedded tissue. Global prevalence was 14.9% (13/87). HPV infection was positive in 19.4% (12/62) of patients with adenocarcinoma and 4.0% (1/25) of patients with BPH. HPV-11, which is considered to be low risk, was more prevalent. Interestingly, one patient with BPH and six with prostate cancer showed examples considered to be high risk (HPV-18, -51, -52, and -66). Conclusion: A higher rate of HPV infection among Mexican patients with prostatic carcinoma than among those with BPH was observed. HPV infections may thus contribute to the risk of prostate cancer. Further studies are required to elucidate any roles of HPV infection in prostate disease in Mexico and the effect of prevention and treatment of HPV infection on prostatic adenocarcinoma.

Kefir Grain의 Polysaccharide에 의한 HRV S-2의 MA-104 세포 감염억제 (Inhibitory Effect of Polysaccharide from Kefir Grain on the Infection of MA-104 Cell by Human Rotavirus)

  • 본 연구에서는kefir중의 polysaccharide분획이 HRV의 MA-104세포 감염에 미치는 억제효과를 조사하였다. (1) 추출된 polysaccharide는 DEAE 이온교환 chromatography에서 fraction I의 미결합 분획과 fraction II, III, IV의 결합 분획으로 나뉘어졌다. (2) Polysaccharide fraction II, III, IV는 각각 4.8, 5.3, 1.4 mg/mL의 농도로 첨가했을 때 HRV의 MA-104 세포감염에 대하여 약 97%, 99%, 100%의 억제효과를 나타냈다. 본 연구의 결과를 종합하면 kefir의 다당체가 유아설사변에서 분리한 rotavirus S-2가 MA-104 cell 감염에 97% 이상 억제효과가 있어 동절기 유아의 바이러스성 급성설사 예방에 kefir를 급식하는 것이 바람직하다고 사료된다.

Four Human Cases of Acanthotrema felis (Digenea: Heterophyidae) Infection in Korea

  • Acanthotrema felis is an intestinal trematode of cats originally reported from the Republic of Korea. Only 1 human case infected with a single adult worm has been previously recorded. In the present study, we report 4 human cases infected with a total of 10 worms recovered after anthelmintic treatment and purging. All 4 patients reside in coastal areas of Jeollanam-do, Korea, and have consumed brackish water fish including the gobies, Acanthogobius flavimanus. The worms averaged 0.47 mm in length and 0.27 mm in width, and had 3 sclerites on the ventrogenital sac; 1 was short and thumb-like, another was long and blunt-ended, and the 3rd was long and broad-tipped. They were identified as A. felis Sohn, Han, & Chai, 2003. Surveys on coastal areas to detect further human cases infected with A. felis are required.
