발효 차가버섯 추출물이 인체 종양세포주 증식에 미치는 영향 (Effects of Water Extract from fermented Chaga Mushroom(Inonotus obliquus) on the Proliferation of Human Cancer Cell Lines.)

  • 발효 및 비발효 차가버섯 수용성 추출물이 정상 세포주 NIH3T3 mouse normal fibroblast cell 및 인체 종양 세포주 AGS human gastric cancer cell(위암), HCT-15 human colon cancer cell(대장암), Hep3B human hepatoma cancer cell(간암), MCF-7 human breast cancer cell(유방암), HeLa human cervical cancer cell(자궁경부암)에서 MTT(3-[4,5-dimethylthiazol-2-yl]-2,5-diphenyl tetrazolium bromide) assay 방법에 의한 세포 증식 억제와 암세포 증식억제의 기전 연구의 일환으로 apoptosis가 일어날 때 나타나는 DNA fragmentation을 agarose gel electrophoresis 방법으로 검토하였다. 인체 종양 세포주의 생육저해 효과가 발효 차가버섯 추출물이 비발효 차가버섯 추출물보다 강한 것으로 나타났다. 그러나 동일한 실험조건하에서 마우스 정상 세포주 NIH3T3은 80% 이상의 생존율을 나타내어 정상 세포주에는 큰 영향을 미치지 않는 것으로 나타났다. 특히, 발효 및 비발효 차가버섯 추출물에서 본 실험에 사용한 세포주 중에서 대장암 세포주 HCT-15에 대해 가장 세포 증식 억제효과가 뛰어났으며, 이러한 효과는 첨가 농도 의존적 이였다. 발효 및 비발효 차가버섯 추출물에 의한 암세포 증식억제가 기전 연구로 apoptosis가 일어날 때 나타나는 DNA fragmentation을 세포로부터 genomic DNA를 분리하여 agarose gel electrophoresis 방법으로 조사한 결과, 정상세포인 NIH3T3 세포는 DNA fragmentation이 거의 일어나지 않아 세포 생존율 결과와 유사한 경향을 보였으나, 특히 대장암 세포주인 HCT-15에서는 발효 차가버섯뿐만 아니라 비발효 차가버섯 추출물에서도 DNA fragmentation이 많이 일어나는 것이 관찰되어 암세포 증식억제 효과가 높다는 결과를 뒷받침 해주고 있다.

  • 인삼과 계피 추출물의 인체 직장암 세포(HT-29), 인체 간암 세포(HepG2)및 인체 결장암 세포(HRT-18)의 증식에 미치는 영향을 in vitro에서 확인하였다. 암세포의 배양액에서 인삼 추출물 또는 계피 추출물의 효과는 농도에 비례하여 암세포의 증식을 억제하였다. 세포증식의 억제 정도는 인삼과 계피 추출물을 단독으로 첨가한 것보다 병용하여 첨가한 경우 현저한 상승 효과를 나타내어, 낮은 인삼 농도에서도 HT-29, HepG2및 HRT-18암세포의 증식을 효과적으로 억제하였으며 심지어는 사멸시켰다. 인삼과 계피의 혼합물은 세포주기 중 G1 단계에서 S단계로의 진행을 지체시킴으로써 암세포의 증식을 억제하는 것으로 나타났다.

고추 부위별추출물에 의한 종양세포의 세포사유도 - Hepatoma 세포와 MCE-7 세포 - (Induction of Cancer Cell Apoptosis by the Extract of Capsicum annuum L. var. angulosum Mill Sorted According to the Parts in Hepatoma Cells and MCF-7 Cells)

  • Under the active search for biologically active novel agents for cancer prevention and treatment, some agents have been found from plants which are easily available. Our previous research on them revealed that C. annuum L. var. angulosum Mill have high antiproliferating effect on cancer cells. However, it has not been known whether the anticancer efficacy is different according to each part of C. annuum L. var. angulosum Mill or whether it can be changed by timing of harvest or solvent for extraction. Thus we compared the efficacy of each part of C. annuum L. var. angulosum Mill and assessed how much difference in the efficacy can be made according to the time of harvest or solvents for extraction. We observed the morphologic change and apoptosis 48 hr after treatment with the extract of each part of C. annuum L. var. angulosum Mill in MCF-7 mammary gland adenocarcinoma cells and human hepatoma cells. We also counted cancer cells by trypan blue method and MTT method to check the cytotoxicity. The leaf extract showed the highest anticancer effect among all the parts of C. annuum L. var. angulosum Mill; 50% and 70% reduction in the number of cancer cells was observed at 25 $\mu\textrm{g}$/mι and 50 $\mu\textrm{g}$/mι, respectively. It was more than 2 times as potent as 5-fluorouracil (5-FU). We found chromosomal fragmentation, clumping, and destuction by PI staining, and DNA fragmentation by electrophoresis. In conclusion, this study suggests that leaf extraction using water as solvent has the highest antiproliferative and apoptotic activity in cancer cells compared with other parts of extraction.

  • A coumestan derivative, psoralidin (1) was found to be a cytotoxic principle of the seeds of Psoralea corylifolia L (Leguminosae) with the IC_{50}$ values of 0.3 and 0.4 ug/ml against the HT-29 (colon) and MCF-7 (breast) human cancer cell lines, respectively. A coumarin, angelicin (2) was also isolated as a marginally cytotoxic agent along with an inactive compound, psoralene (3) from the plant. The isolates 1-3 were not active against the A54l(lung) and HepG2 (liver hepatoma) cancer cell lines.

Intracellular $Ca^{2+}$ release mediates apoptosis induced by ascorbic acid (vitamin C) in HepG2 human hepatoma cells

  • Ascorbic acid (vitamin C) has been shown to have anti-cancer actions. However, the exact mechanism of this action is not fully understood. In this study we investigated the possible mechanism of anti-cancer action of ascorbic acid in HepG2 human hepatoblastoma cells. Ascorbic acid induced apoptotic cell death in a dose-dependent manner in the HepG2 cells, assessed by the flow cytometric analysis of hypodiploid nuclei stained with propodium iodide. In addition, ascorbic acid increased intracellular Ca$\^$2+/ concentration, whereas the level of reactive oxygen species was not significantly changed, suggesting that ascorbic acid may not alter cellular redox potential in the cells. PU.1 Is Identified as a Novel Metastasis Suppressor in Hepatocellular Carcinoma Regulating the miR-615-5p/IGF2 Axis

  • Invasion and metastasis is the major cause of tumor recurrence, difficulty for cure and low survival rate. Excavating key transcription factors, which can regulate tumor invasion and metastasis, are crucial to the development of therapeutic strategies for cancers. PU.1 is a master hematopoietic transcription factor and a vital regulator in life. Here, we report that, compared to adjacent non-cancerous tissues, expression of PU.1 mRNA in metastatic hepatocellular carcinoma (HCC), but not primary HCC, was significantly down-regulated. In addition, levels of PU.1 mRNA in metastatic hepatoma cell lines MHCC97L and MHCC97H were much lower than in non-metastatic Hep3B cells. Transwell invasion assays after PU.1 siRNA transfection showed that the invasion of hepatoma cell lines was increased markedly by PU.1 knockdown. Oppositely, overexpression of PU.1 suppressed the invasion of these cells. However, knockdown and overexpression of PU.1 did not influence proliferation. Finally, we tried to explore the potential mechanism of PU.1 suppressing hepatoma cell invasion. ChIP-qPCR analysis showed that PU.1 exhibited a high binding capacity with miR-615-5p promoter sequence. Overexpression of PU.1 caused a dramatic increase of pri-, pre- and mature miR-615-5p, as well as a marked decrease of miR-615-5p target gene IGF2. These data indicate that PU.1 inhibits invasion of human HCC through promoting miR-615-5p and suppressing IGF2. These findings improve our understanding of PU.1 regulatory roles and provided a potential target for metastatic HCC diagnosis and therapy.

Inhibition of SMP30 Gene Expression Influences the Biological Characteristics of Human Hep G2 Cells

  • Senescence marker protein 30 (SMP30), a hepatocellular carcinoma (HCe) associated antigen had been identified by our research group. To study its mechanisms of regulation and associations with the occurrence and development of HCe, we inhibited expression by RNAi technique, and observed effects on the biological characteristics of Hep G2 cells. In cell viability assays, cell growth in the experimental group (with siRNA transfection) was elevated. In Transwell invasion assays, compared with blank and control groups, numbers of invading cells in the experimental group were significantly increased, whereas in apoptosis assays, the percentage apoptosis demonstrated no differences, but after UV irradiation, that in the experimental group was higher than the other two groups. In a word, SMP30 can inhibit the proliferation and invasion of human hepatoma cells and thus can be regarded as a cancer suppressive factor.

온청음(溫淸飮)이 인체 간암세포의 세포주기 G1 Arrest에 미치는 영향 (G1 Arrest of the Cell Cycle by Onchungeum in Human Hepatocarcinoma Cells)

  • Onchungeum, a herbal formula, which has been used for treatment of anemia due to bleeding, discharging blood and skin disease. In the present study, it was examined the effects of extract of Onchungeum (OCE) on the growth of human hepatocarcinoma cell lines Hep3B (p53 null type) and HepG2 (p53 wild type) in order to investigate the anti-proliferative mechanism by OCE. Treatment of Hep3B and HepG2 cells to OCE resulted in the growth inhibition in a dose-dependent manner, however Hep3B cell line exhibited a relatively strong anti-proliferative activity to OEC. Flow cytometric analysis revealed that OCE treatment in Hep3B cells caused G1 phase arrest of the cell cycle, which was associated with various morphological changes in a dose-dependent fashion. RT-PCR and immunoblotting data revealed that treatment of OCE caused the down-regulation of cyclin D1 expression, however the levels of cyclin E expression were not changed by OCE. The G1 arrest of the cell cycle was also associated with the induction of Cdk inhibitor p27 by OCE. Because the p53 gene is null in Hep3B cells, it is most likely that the induction of p21 is mediated through a p53-independent pathway. Moreover, p27 detected in anti-Cdk4 and anti-Cdk2 immunoprecipitates from the OCE-treated cells, suggesting that OCE-induced p27 protein blocks Cdk kinase activities by directing binding to the cyclin/Cdk complexes. Furthermore, OCE treatment potently suppresses the phosphorylation of retinoblastoma proteins and the levels of the transcription factor E2F-1 expression. Taken together, these results indicated that the growth inhibitory effect of OCE in Hep3B hepatoma cells was associated with the induction of G1 arrest of the cell cycle through regulation of several major growth regulatory gene products.

매생이 추출분획의 암 예방 및 면역증진 효과 (Effects of Cancer Prevention and Immune Stimulation of Fractions from Capsosiphon fulvescens)

  • The fractions of Capsosiphon fulvescens were studied to verify the anticancer and immunostimulating activity. The fractions from the ethanol extract of C. fulvescens were prepared by the systematic extraction procedure with the solvents such as hexane, ethyl ether, methanol, butanol and H$_2$O. The cytotoxic effects of C. fulvescens fractions against human leukemia cell line U937, mouse neuroblastoma cell line (NB41A3), human hepatoma cell line (HepG2)and rat glioma cell line (C6) were investigated. Ethyl ether fraction of C. fulvescens showed the highest cytotoxicity against all four cell lines tested. In addition, H$_2$O fraction also showed relatively high cytotoxicity. Dose dependent patterns were observed on all four cell lines. The immune-stimulating effects of C. fulvescens fractions on rat macrophage cell line (RAW 264.7) were also investigated. All five fractions of C. fulvescens extract stimulated NO production with concentration dependant manner. These results suggest that C. fulvescens may be a useful candidate for a natural antitumor and immune-stimulating agent.

Liposome-mediated Induction of Apoptosis of Human Hepatoma Cells by C-Myc Antisense Phosphorothioate Oligodeoxynucleotide and 5-Fluorouracil

  • Background: The aim of this study was to investigate the effect of a c-myc antisense oligodeoxynucleotide and 5-fluorouracil on the expression of c-myc, invasion and proliferation of HEPG-2 liver cancer cells. Materials and Methods: HEPG-2 cells were treated with lipiosome-mediated c-myc ADSON and 5-fluorouracil. The proliferation inhibition rate and invasion were measured by MTT and invasion assay, respectively. Cell apoptosis was detected by flow cytometry and expression of c-myc by RT-PCR and immunohistochemistry. Results: The proliferation inhibition rate was significantly higher in the antisense oligodeoxynucleotide added-5-fluorouracil group than single antisense oligodeoxynucleotide or 5-fluorouracil group (p<0.05). G0/G1 cells in the antisense oligodeoxynucleotide group and S cells in the 5-fluorouracil groups were significantly increased than that in the control group, respectively (P<0.01). The amplification strips of PCR products in 5-FU, ASODN and combination groups were significantly weaker than that in the control group (P<0.01). The percentage of c-myc-protein-positive cells were significantly lower in antisense oligodeoxynucleotide, 5-fluorouracil and combination groups than that in the control group (P<0.01). Conclusions: A liposome-mediated c-myc antisense oligodeoxynucleotide and 5-fluorouracil can inhibit the proliferation and invasion of liver cancer cells by reducing the expression of c-myc. A c-myc antisense oligodeoxynucleotide can increase the sensitivity of liver cancer cells to 5-fluorouracil and decrease the dosage of the agent necessary for efficacy, providing an experimental basis for the clinical therapy of liver cancer.
