형질전환 식물을 이용한 phytoremediation

  • Tobacco plants were transformed by A. tumefaciens harboring human ferritin gene and they were subjected to investigate for the expression of transformed gene as well as heavy metal accumulation. Seed from self-fertilized transgenic plants was germinated on media containing toxic level of Cd, Cu, Zn, Fe, Mn and scored for tolerance to this heavy metals. There is difference in growth rate between transgenic and control plants, especially Cd, Cu. And transgenic plants accumulated more heavy metals than control plants.

The Effect of Dietary Phytate Content on Iron Absorption and Status in Young Korean Women

  • This study investigated the effects of dietary phyate reduction on the apparent absorption and biochemical parameters of iron status in young Korean women. Fourteen healthy, young women consumed low and high phytate diets for ten days of each experimental period. Duplicate diet samples, a fasting blood sample on day 9, and complete fecal samples for five consecutive days starting from day 5 of each diet period were collected. The iron content of diet and fecal samples were analyzed to calculate apparent absorption. Serum samples were analyzed for iron, ferritin, transferrin receptor and TIBC; transferrin saturation was also calculated. The apparent absorption of iron tended to increase in the low phytate period (32.51%) compared to the high phytate period (17.91%), but the difference was not significant (p=0.06). Serum ferritin decreased and serum transferrin receptor increased significantly during the low phytate diet although the mean values were within the normal range. Serum iron and transferrin saturation did not change significantly. In conclusion, the results indicated that reducing dietary phytate for ten days negatively affected iron nutritional parameters, but it moderately and positively affected apparent iron absorption in young Korean women. Further research on the long-term effects of a low phytate diet with an adequate iron content for vows Korean women is necessary.

미숙아 빈혈에 대한 Recombinant Human Erythropoietin의 효과 : 예비연구 (Effect of Recombinant Human Erythropoietin in the Anemia of Prematurity : a Pilot Study)

  • 1991년 1월부터 1993년 2월까지 영남대학교 의과대학 부속병원에서 출생하여 신생아 중환자실에서 치료 받았던 12명의 미숙아를 대상으로 무작위로 대조군과 rHuEPO투여군으로 나누어 체중 kg당 100unit의 rHuEPO을 1주간 3회씩 4주동안 피하주사하고 rHuEPO투여 시작과 같이 체중 kg당 3mg의 elemental iron을 매일 경구투여 하였고 출생 후 수유가 가능할 때 부터 25unit씩의 비타민 E를 매일 경구투여 하였다. 망상적혈구, 혈색소치, 백혈구수, 혈소판수, 혈청 철, ferritin, 총 철결합능, erythropoietin농도, 활력징후, 체중, 간기능, 신장기능 및 피하주사부 위의 국소 부작용을 비교해 본 결과 치료 시작전 erythropoietin의 농도는 대조군과 rHuEPO투여군 사이에 차이는 없었으며 모두 정상 범위에 속했고 rHuEPO투여군에서는 치료 1주에서 3주사이에 급격한 망상적혈구의 경향이 관찰되었고 전반적으로 혈색소치, 백혈구수, 혈소판수, 혈청 철, ferritin, TIBC등에서는 차이가 없었다. 그리고 체중증가 및 활력증상, 간기능, 신장기능에서도 대조군과 rHuEPO투여군 사이에서 차이가 없었고 rHuEPO 투여 기간 동안 계속 정상적으로 유지되었다. 결론적으로 우리의 연구에서는 rHuEPO치료로 뚜렷한 혈색소치 및 헤마토크리의 증가를 볼 수 없었지만 망상적혈구가 치료 초기에 증가되는 경향이 관찰되었고 치료로 인한 특이할 만한 부작용이 없었으므로 첫째, 치료 기간 중의 예견할 수 없이 발생하는 패혈증, 무시할 수 없는 채혈량과 spontaneous endogenous erythropoietin생산이 rHuEPO의 반응에 미치는 영향, 둘째. rHuEPO의 가장 높은 효과를 얻기 위한 보다 적절한 투여용량, 기간 및 rHuEPO의 투여를 출생 후부터 바로 예방 목적으로 할 것인지 또는 출생 몇 주 후부터 rescue therapy로 할 것인지 투여 시작 시기, 셋째, 용량의 증가 및 감소에 동반되는 부작용의 정도등을 고려해서 앞으로 더 많은 미숙아를 대상으로 지속적인 연구를 한다면 rHuEPO을 이용해 더욱 더 활발하고 효과적으로 미숙아 빈혈을 치료할 수 있으리라 기대한다.

사람 혈청 트란스페린수용체의 단클론 항체를 이용한 방사면역측정과 철영양상태의 진단 (Immunoradiometric Assay using Monoclonal Antibody Against Human Serum Transferrin Receptor for Diagnosis of Iron Deficiency)

  • The soluble transferrin receptor(TfR) in human serum has been shown recently to be a truncated form of intact membrane bound receptor containing most of the extracellular domain. We purfied the transferin-free TfR from human serum by immounoaffinity chromatography which produced the single protein identity in high resolution gel chormatography. The monoclonal antibodies(MAb) against purifed serum TfR were produced by fusion of spleen cells o fimmunized Balb/c mice and SP2 cells. Ten hybrids producing MAb specific for serum TfR were identifed and determine their iostypes. A immunoraddiometric assay (IRMA) for serum TfR was established using two monoclonal IgG1 antibodies as the coating and indicator antibodies on the bosis of their suitability in sandwich IRMA of serum TfR. The mean serum TfR levels in the 15 normal male, 15 normal female, and 19 iron-deficient subjects were 5.4$\pm$0.98, 4.6$\pm$0/76, and 18.0$\pm$12.8mg/1, respectively, and the difference in mean values between normal and iron deficient subjects was significant(p=0.0005). There existed the inverse logarithmic relationship(r=-0.9336, p<0.0001) between the serum TfR and ferritin levels.

Distinctive clinical features of HPeV-3 infection in 2 neonates with a sepsis-like illness

  • We report a human parechovirus-3 (HPeV-3) infection in 2 neonates who had prolonged fever (>5 days) with palmar-plantar erythema. This distinctive rash was observed 4-5 days after fever onset, just before defervescence. Elevated aspartate aminotransferase, lactate dehydrogenase, and ferritin levels were characteristic laboratory findings in the 2 cases, suggesting tissue damage caused by hypercytokinemia. Case 1 was treated with intravenous immunoglobulin, considering the possibility of severe systemic inflammatory responses. The initial ferritin level was 385 ng/mL (range, 0-400 ng/mL); however, the level increased to 2,581 ng/dL on day 5 after fever onset. Case 2 presented with milder clinical symptoms, and the patient recovered spontaneously. HPeV-3 was detected in cerebrospinal fluid and/or blood samples, but no other causative agents were detected. The findings from our cases, in accordance with recent studies, suggest that clinical features such as palmar-plantar erythema and/or hyperferritinemia might be indicators of HPeV-3 infection in neonates with sepsis-like illness. In clinical practice, where virology testing is not easily accessible, clinical features such as palmar-plantar erythema and/or hyperferritinemia might be helpful to diagnose HPeV-3 infection.

운동선수들의 혈액분석을 통한 Etrythropoietin 간접도핑검사 (Blood Analysis for Indirect Doping Control of Erythropoietin in Sports)

  • The use of recombinant human erythropoietin (rhEPO), a stimulator of erythropoiesis, banned in sports because of the medical risk associated with thrombosis. Due to analytical difficulties to differentiate between natural human EPO (hEPO) and rhEPO, blood parameters of erythropoiesis such as contents of hemoglobin (cut-off value <17.5 g/d l for man, and < 16.0 g/dl for women), hematocrit and reticulocytes (cut-off value <2.0%) were measured to focus the misuse of rhEPO. We conducted anti-doping test for 122 blood samples of the World Cup athletes. The mean values of key parameters are as follows; 14.5$\pm$1.0 g/dl for hemoglobin, 41.7$\pm$2.8% for hematocrit, and 1.3$\pm$0.4% for reticulocyte. Blood sample was found to be stable up to 8 hours for the reticulocyte measurement. In addition, the soluble transferrin receptor and ferritin levels were measured by immunoassay methods using plasma samples (n=28) in which the mean value was 0.8$\pm$0.5 $\mu\textrm{g}$/$m\ell$ and 54.6$\pm$33.7 ng/$m\ell$, respectively. The results indicate that all samples tested were negative for the blood parameters of indirect anti-doping test for hEPO misuse. The statistical evaluation suggest that several other parameters such as red blood cell, mean corpuscular hemoglobin concentration, mean corpuscular volume, mean corpuscular hemoglobin and white blood cell could be considered as factors influencing hEPO function in addition to five parameters mentioned.

CD Gene Microarray Profiles of Bambusae Caulis in Liquamen in Human Mast Cell

  • Bambusae Caulis in Liquamen(BCL) has been used to relieve the cough and asthma, and remove the phlegm in traditional Oriental medicine. In recent years, it was studied for its antiinflammatory, antiallergenic, immune-modulating, and anticarcinogenic capabilities. This experiment was performed to evaluate the microarray profiles of CD genes in human mast cells before and after BCL treatment. The results are as follows: The expression of 51 of the genes studied was up-regulated in the Bel-treated group; they include the genes coding L apoferritin, beta-2-microglobulin, ferritin light polypeptide, CD63, monocyte chemotactic and activating fact, heme oxygenase 1, CD140a, integrin alpha M, colony stimulating factor 2 receptor, eukaryotic translation elongation factor, CD37, interleukin 18, NADH dehydrogenase 1 beta, CD48, 5-lipoxygenase activating protein, interleukin 4, ribosomal protein L5, GABA(A) receptor-associated protein, beta-tubulin, integrin beta 1, CD162, CD32, lymphotoxin beta, alpha-tublin, integrin alpha L, CD2, CD151, CD331, 90 kDa heat shock protein, CD59, CD3Z, microsomal glutathione S-transferase 2, CD33, CD162R, cyclophilinA, CD84, interleukin 9 receptor, interleukin 11, CD117, CD39-Like 2, and so forth. The expression of 7 of the genes studied was down-regulated in the BCL-treated group; they include the genes coding con, CD238, SCF, CD160, CD231, CD24, and CD130. Consequently, the treatment of BCL on the human mast cells increased the expression of 51 genes and decreased the expression of 7 genes. These data would provide a fundamental basis to the traditional applications of Bambusae Caulis in Liquamen.

유근피로부터 분리한 hederagenin 3-O-b-D-glucopyranosyl(1→3)-a-L-rhamnopyranosyl(1→2)-a-L-arabinopyranoside (HDL)의 항산화 효과 (Antioxidant Effect of Hederagenin 3-O-b-D-Glucopyranosyl(1→3)-a-L-Rhamnopyranosyl(1→2)-a-L-Arabinopyranoside (HDL) Isolated from Root Bark of Ulmus davidiana)

  • 본 연구는 유근피(root bark of Ulmus davidiana)에서 분리한 화합물인 hederagenin 3-O-b-D-glucopyranosyl($1{\rightarrow}3$)-a-L-rhamnopyranosyl($1{\rightarrow}2$)-a-L-arabinopyranoside (HDL)을 이용하여 $CoCl_2$에 의해 생성된 ROS에 대한 항산화 기전을 밝히기 위하여, ROS 생성과 관련된 산화 효소 및 항산화 효소에 대한 저해효과를 조사하였다. 또한 HDL이 $CoCl_2$에 의해 생성된 ROS의 조절을 통해 산화적 스트레스와 관련된 단백질 발현 및 세포주기에 미치는 영향을 조사하였다. 그 결과 HDL은 $CoCl_2$에 의해 유발된 xanthine oxidase와 $H_2O_2$ 생성 증가를 억제하였고, 산화와 관련된 SOD, CAT의 활성을 증가시켜 $H_2O_2$의 가수분해를 촉진하였다. 그리고 HDL은 $CoCl_2$에 의해 유발된 ferritin의 손상과 ferritin iron의 방출을 억제하 였으며, 지질과산화의 증가를 억제하였다. 뿐만 아니라 HDL은 $CoCl_2$에 의해 증가된 G1 phase의 세포를 감소시켰으며, 세포주기와 관련된 p53 및 $p21^{CIP1/WAF1}$의 발현을 감소시켰다. 이러한 연구결과들은 HDL이 천연물로부터 유래한 독성이 없는 항산화제로서의 가능성을 제시한다.

결핵균 감염에 의한 THP-1 세포에서의 Prothymosin alpha 유전자 발현증가 (Up-regulation of Prothymosin alpha in THP-1 Cells Infected with Mycobacterium tuberculosis)

  • Mycobacterium tuberculosis is capable of growing and survival within macrophage. The purpose of this study was to identify the genes regulated by infection of mycobacteria in human monocytic THP-1 cells. We used the differential display reverse transcriptase polymerase chain reaction (DD RT-PCR) and nothern blot analysis to confirm the differentially expressed genes from THP-1 cells infected with live Mycobacterium tuberculosis H37Rv, heat-killed Mycobacterium tuberculosis H37Rv and live Mycobacterium bovis BCG. Among many up or down-regulated clones, 27 clones were sequenced and compared with known genes on GenBank. Thirteen of over-expressed clones from THP-1 cells infected with live Mycobacterium tuberculosis H37Rv were identical to human prothymosin alpha, eight were novel clones and six clones showed homology with Human ferritin H chain, Esherichia coli bgl, Mouse RNA-dependent EIF-2 alpha kinase, E. coli htrL, Hyaluronan receptor and T cell receptor. Our result suggests that Mycobacterium tuberculosis might regulate prothymosin alpha gene transcription in monocytic THP-1 cell.
