MicroRNA (miRNA) dysregulations are associated with various types of human cancers, and miRNAs can function as tumor suppressors and oncogenes. Emerging evidence has shown that miRNA pathway is also altered during colorectal tumorigenesis. The detection of cancer-related miRNAs in stool samples may become useful diagnostic marker for colorectal cancer, because miRNAs in stool samples has high stability, and maintains a high portion of its original level. Recent studies reported that stool-based miRNAs can offer more sensitivity and specificity than currently used stool-based screening methods for CRC. In addition, unlike fecal occult blood test, sampling on consecutive dates and special dietary restrictions are not required. In this review, the authors discuss stool-based miRNA for the early diagnosis of CRC and perspectives on future application.
Twenty healthy adult volunteers (divided into two groups) were given with 100 mL of acidophilus milk, twice a day, containing $1{\times}10^8\;cfu/mL$ of Lactobacillus acidophilus SNUL 01 or SNUL 02 originated from healthy Korean adults. The floral changes of lactobacilli, Escherichia coli, and Clostridium perfringens surviving in the feces as well as the pH of the fecal material have been studied for three weeks. Initial pH of the fecal samples before taking acidophilus milk were about pH 7 and it has been decreased to about pH 5 when the acidophilus milk was given. Population of fecal lactobacilli increased from about $10^7\;cfu/g\;to\;10^8\;cfu/g$ of fecal material after 4 days of acidophilus milk intake and the high population of lactobacilli was maintained up to 21 days. Viable cells of the fecal E. coli and C. perfringens have been decreased from about $10^7{\sim}10^8\;cfu/g\;and\;10^5{\sim}10^6\;cfu/g\;to\;10^6\;cfu/g\;and\;10^3\;cfu/g$ of fecal material after 21 days, respectively. In conclusion, continuous intake of acidophilus milk made with L. acidophilus SNUL 01 and SNUL 02 helped maintaining normal intestinal microflora, suppress harmful microorganisms.
We compared the efficacy and applicability of a modified formalin-ether concentration technique (M-FECT) to the conventional FECT (C-FECT) and the agar plate culture (APC) method for the detection of Strongyloides stercoralis larvae. For this purpose, we used 600 human fecal specimens collected in an endemic area of southern Thailand. In the M-FECT, we used 2 layers of wire meshes, instead of gauze, to avoid the loss by absorption/adhesion of larvae to the gauze during filtration, and we reduced the exposure time of S. stercoralis larvae in stool samples to formalin. By such simple modifications, the efficacy of M-FECT has become comparable to APC and was much better than that of C-FECT for the diagnosis of strongyloidiasis.
Quantitative fecal egg counts represented as the number of eggs per gram of feces (EPG) are generally a reliable parameter to estimate the worm burden of intestinal and hepatic parasitoses. Although Haplorchis taichui (Digenea: Heterophyidae) is one of the most common minute human intestinal flukes, little is known about the relationship between EPG and the actual worm burden in patients or the severity of the disease. In the present study, fecal samples were collected from 25 villagers in northern Thailand before and after praziquantel treatment. The EPG values of each participant were determined by the modified cellophane thick smear method, and adult worms were collected from the whole stool after the treatment. Eggs per day per worm (EPDPW) of H. taichui were estimated 82 from egg counts and expelled worms. The EPG was not well correlated with the worm burden, and a reverse correlation was observed between the EPDPW and the worm burden.
In order to investigate the distribution of dominant Bifidobacterium species in intestinal microflora of Korean adults and seniors, SDS-PAGE profiles of whole cell proteins were used for the identification of bifidobacteria. To confirm the reliability of SDS-PAGE, the Bifidobacterium species identified by SDS-PAGE of whole cell proteins were validated by using 16S rDNA sequencing analysis. The results of SDSPAGE corresponded well with those determined by the analysis of 16S rDNA sequencing. Based on the analysis of SDS-PAGE patterns on unidentified fecal strains which showed positive in fructose-6-phosphate phosphoketolase activity, B. adolescentis, B. longum, and B. bifidum were identified in the feces of adults, and B. adolescentis, B. longum, B. bifidum, B. breve, and B. dentium were identified in those of seniors. In most of the fecal samples tested, the predominant Bifidobacterium species consisted of only a few species, and differences in the distribution and numbers of Bifidobacterium species were observed between adults and seniors. B. adolescentis and B. longum were found to be the most common species in feces of adults, but not in seniors. Accordingly, the distribution and abundance of bifidobacteria in the human intestinal microflora varied depending on the age of hosts.
The study examined the effect of urbanization on the water quality of Nwaorie and Otamiri Rivers in Owerri metropolis, Imo State, South-East Nigeria. Water samples were collected from Nwaorie and Otamiri Rivers from four sampling stations up and down stream. Water parameters analyzed using standard procedures were: color, turbidity, temperature, pH, total hardness, total solids, metals (Iron and magnesium), anions (nitrate and ammonia) and Fecal coliform. Results showed increased levels of coloration, iron, ammonia, turbidity and fecal coliform which exceeded the World Health Organization (WHO) permissible limits for drinking water. Increase in these parameters indicated influx of industrial effluent from the nearby industries and indiscriminate disposal of wastes at the bank of the rivers. Application of ANOVA showed various degree of variation in pollutants levels between the two rivers and at different sampling points. River Nwaorie was observed to be more impacted than River Otamiri. High values of iron observed from the study could be deleterious to human health if the river water is consumed without treatment. The study, therefore, recommended proper waste management and disposal as well as effluent treatments in Owerri municipal against pollution of surface water.
Blastocystis is one of the most commonly detected genera of protozoan parasites in the human intestines as well as the intestines of many other species such as pigs in several geographical regions worldwide. However, no studies have examined Blastocystis in pigs in Korea. In this study, PCR and nucleotide sequencing were performed to evaluate the genetic diversity and zoonotic potential of Blastocystis using pig fecal samples. We obtained 646 stool samples from groups of piglets, weaners, growers, finishers, and sows in Korea. A total of 390 Blastocystis-positive samples were identified, and the infection rate was 60.4%. The infection rates were significantly related to age and region. The 4 subtypes (STs) of Blastocystis confirmed by phylogenetic analysis were ST1, ST2, ST3, and ST5, indicating the high genetic diversity of Blastocystis in Korean pigs. ST5 was highly distributed in Korean pigs among detected STs in this study. Some sequences were closely related to those of Blastocystis isolated from humans. This is the first study of Blastocystis in pigs in Korea. Based on the results, Blastocystis is prevalent in Korean pigs. Although a small number of samples were obtained in some areas, the clinical development of Blastocystis infection in pigs and potential for human transmission should be further examined.
As a series of basic study for the prevention of zoonotic parasites, fecal samples and internal organs of cats in Iri and its vicinity were examined for helminth and their ova or protozoan oocysts from November 1994 to March 1995. The results obtained were summarized as follows; Out of the 133 samples, 101(75.9%) were positive and 135(101.5%) were cumulative positive for parasites. The following were identified in the samples and positive numbers of each species detected were ; Toxocura cari, 51;Isospora spp., 27;Spirometra erinacei, 26;Tasnka taentaeformis, 24;Diphyllobothrium latum, 4;and Capillaria spp., 3, respectively. In analyzing the state of infection, 64.9%(61 heads) of all the cats inspected had a single infection, while the rest(excluding the 32 uninfected cats) had mixed infections : 27.6%(26 heads), 6.4%(6 heads) and 1.1%(1 heads) had double, triple, quadruple Infections, respectively. From these results, it can be concluded that since the helminth, zoonotic parasites, in Korean autochthonal cats cause not only various disorders, but also can be very harmful to human beings, they are considered to be of great significance to public health.
The increasing prevalence of Toxoplasma gondii infection in the human population in the Republic of Korea (= Korea) is due to various reasons such as an increase in meat consumption. However, the importance of cats in transmitting T. gondii infection through oocysts to humans has seldom been assessed. A total of 300 fecal samples of stray cats captured around Seoul from June to August 2013 were examined for T. gondii B1 gene (indicating the presence of oocysts) using nested-PCR. Fourteen (4.7%) of 300 cats examined were positive for B1 gene. Female cats (7.5%) showed a higher prevalence than male cats (1.4%). Cats younger than 3 months (5.5%) showed a higher prevalence than cats (1.5%) older than 3 months. For laboratory passage of the positive samples, the fecal suspension (0.2 ml) of B1 gene positive cats was orally inoculated into experimental mice. Brain tissues of the mice were obtained after 40 days and examined for the presence of tissue cysts. Two isolates were successfully passaged (designated KNIH-1 and KNIH-2) and were molecularly analyzed using the SAG5D and SAG5E gene sequences. The SAG5D and SAG5E gene sequences showed high homologies with the ME49 strain (less virulent strain). The results indicated the importance of stray cats in transmitting T. gondii to humans in Korea, as revealed by detection of B1 gene in fecal samples. T. gondii isolates from cats were successfully passaged in the laboratory for the first time in Korea.
Current reports suggest that obesity is a serious global health issue. Emerging evidence has predicted strong links between obesity and the human gut microbiota. However, only a few such studies have been conducted in Asia, and the gut microbiota of lean and obese adult Asians remains largely unexplored. Here, we investigated the potential relationship between gut microbiota, body massindex (BMI), and metabolic parameters in adults from Thailand, where obesity is increasing rapidly. Fecal and blood samples were collected from 42 volunteers who were allocated into lean, overweight, and obese groups. The fecal microbiota was examined by quantitative PCR analysis. Bacteroidetes, Firmicutes, and Staphylococcus spp. and methanogens were most abundant in lean volunteers. Overweight volunteers majorly harbored Christensenella minuta and Akkermansia muciniphila, ${\gamma}-Proteobacteria$, and bacteria belonging to the genus Ruminococcus. Methanogens and bacteria belonging to the phylum Bacteroidetes were negatively correlated with adiposity markers (BMI and waist circumference), but positive correlated with high-density lipoprotein, suggesting that they can be used as leanness markers. While some of our results agree with those of previous reports, results regarding the contributions of specific taxa to obesity were inconsistent. This is the first study to report the adult gut microbiota in Southeast Asian populations using molecular techniques and biochemical markers and provides a foundation for future studies in this field.
