Scutellaria baicalensis $G_{EORGI}$ (Scutellariae Radix) has been used to clear heat and to dry dampness in the stomach or intestines, which manifests as diarrhea or dysenteric disorder. In this study, we investigated the effective preparation of active components in Scutellariae Radix using the methods of solvent extraction and absorption fractionation for the development of new functional food or pharmaceuticals. The marker substances, baicalin, baicalein, wogonoside, and wogonin were directly isolated from the Scutellariae Radix. There chemical structures were elucidated by spectroscopic analysis. The Scutellariae Radix was extracted with hot water. To enhance yield of flavonoids in Scutellariae Radix, the hot water extract was dissolved in ethanol with concentration dependent manner. The precipitates were separated using centrifugal techniques at 10,000 rpm. Supernatant liquid was applied to the HPLC for quantification of major compounds. Separately, the hot water extract was absorbed on Diaion HP-20 resin. And then, the absorbed fraction was eluted with methanol for HPLC. The contents of baicalin, baicalein, wogonoside and wogonin in different treatment methods were analyzed by HPLC. Total amount of four major components were 16.9% in 50% ethanol extract, 21.7% in 70% ethanol extract, 20.5% in 90% ethanol extract, and 39.3% in absorbed fraction of Diaion HP-20 resin. In these results, we found that resin absorption method is suitable for the extraction of enriched flavonoids from Scutellariae Radix.
The present study, antioxidant and anti-inflammatory effects were measured of 6 endemic plants in Indonesian extracted by hot water or ethanol. The Nipa Fruticans Wurmb ethanol extract (NFWE) and Orthosiphon aristatus ethanol extract (OAE) showed the highest polyphenol and flavonoid contents of 203.70 and 33.70 ㎍/mg, respectively. Antioxidant activity of OAE was highest in DPPH radical scavenging activity (77.49% at 10 ㎍/mL) and ABTS+ radical scavenging activity (93.36% at 10 ㎍/mL). FRAP activity was significantly higher in NFWE than other extracts. Anti-inflammatory effects of 6 endemic plants in Indonesian extracted by hot water or ethanol were examined using nitric oxide (NO) inhibition assays. In the LPS-induced BV2 cells, OAE showed the highest inhibition of NO production without toxicity. The results of this study, suggest that OAE is a potential functional raw material for antioxidant and anti-inflammatory.

To determinate the content of acanthoic acid and kaurenoic acid in 70% EtOH and hot water extracts of Eleutherococcus gracilistylus, quantitative analysis of each compound in samples was carried out by a HPLC-UVD. Also, the identification of each acompound in samples was successfully assigned by LC-MS analysis. In result, the contents of acanthoic acid and kaurenoic acid in 70% ethanoic extracts were 28.84±0.21 mg/g (2.88%), 26.38±1.63 mg/g (2.64%), respectively. However, the content of two compounds in hot-water extracts was not observed. In conclusion, it shows that 70% ethanol as a best extraction solvent to extract the acanthoic acid and its metabolite from Eleutherococcus gracilistylus was better than hot-water solvent. The 70% ethanol complex extract of Allium Hookeri and Eleutherococcus gracilistylus showed better effectiveness. In addition, the 70% ethanol extract complex of Allium Hookeri and Eleutherococcus gracilistylus showed better effects than the hot water solvent of DPPH radical scavenging ability, total polyphenols, and flavonoids content. The anti-inflammatory activity were significantly or partially reduced by treatment with ethanol extract complex(SEC) by Allium Hookeri and Eleutherococcus gracilistylus.
This study was conducted to gather basic data on the physiological activities of flower extracts from 4 cultivars of white lotus to promote the increase of public consumption. Four cultivars of white lotus: c.v. 'Garam', 'Choeue', 'Baekhwageonryeon', and 'Seungdal' were harvested on 14, August, 2007, and the physiological activities of flower extracts obtained by hot water extraction and ethanol extraction were examined. Total phenol content was highest in the 'Garam' flower hot water extracts, $144.2{\mu}g{\cdot}mL^{-1}$, and lowest in the 'Seungdal' flower ethanol extracts, $63.4{\mu}g{\cdot}mL^{-1}$. DPPH radical scavenging activity was highest at 2,000ppm flower extract of 'Garam' obtained by hot water extraction and ethanol extraction of 90.9% and 83.0%, respectively. Total flavonoid content in the flower extracts of 'Garam' obtained by hot water extraction and ethanol extraction showed the highest levels at $34.1{\mu}g{\cdot}mL^{-1}\;and\;33.9^{\mu}g{\cdot}mL^{-1}$, respectively. Nitrite scavenging activity at 1,000ppm flower extract was highest in 'Garam'. Extracts obtain by hot water and ethanol showed 91.3% and 80.4% activity, respectively. No significant difference in tyrosinase inhibition activity was observed among the cultivars or as a result of extraction method. Overall anti-microbial activity of the flower extracts was slightly higher in 'Garam' than in the other cultivars.
The inhibitory effect of cacao bean husk (CBH) extract on glucosyltransferase(GTasc) from Streptococcus mutans B13 was investigated. Water solube extract from CBH showeda sarong inhibitory effect (88-89%) on GTase from Streptococcus mutans Bl3. GTase inhibitors from sequential extraction by hot water or water-methanol had the strongest inhibition. Sources, fermentation, and types of solvents and fumigation processes did not influence the effect. These active compounds proved to be polyphones through acid hydrolytic analysis of the precipitates by ammonium sulfate or ethanol and proteinase K. It was also confirmed by additional column chromatography of Sephadex G-50, Sephadex LH-20 and DEAE-Sephdex A-50.
The purpose of this study is to investigate antioxidant properties in microbial fermentation products of Lonicera japonica Thunb extract. The bacterium Lactobacillus plantarum NHP1 was isolated from conventional fermented foods. Modern pharmacological studies show that Lonicera japonica Thunb and its active principles of wide pharmacological actions. For instance, they show a strong efficacy in antibacterial, anti-inflammatory, antiviral, anti-endotoxin, blood fat reducing, antipyretic, and antioxidant activities. The extract of Lonicera japonica Thunb was obtained by extracting dried Lonicera japonica Thunb using either hot water or 70% ethanol as a solvent. Fermentation was performed in a 2L fermentor containing 1.2 L of extractat conditions of $30^{\circ}C$ and 100 rpm for 48 hr. The amount of cholorogenic acid was $2.65{\mu}g/g$ in hot water extract. The total phenolic content (GAE, gallic acid equivalent) in hot water and 70% ethanol were $56.5{\pm}4.9$ GAE mg/g and $72.7{\pm}5.3$ GAE mg/g, respectively. After fermentation, the phenolic content increased to 30.2% in hot water and 12.9% in ethanol extract. In the same manner, flavonoid content increased to more than 75% regardless of extract solvent. ABTS (2,2-azino-bis-3-ethylbenzthiazoline-6-sulfonic acid) value noticeably increased to 50% after fermentation.

Ethanol concentration in blood, brain and liver of rats was shown to be effectively lowered by arrowroot flower extract. The lowering effect for ethanol concentration in blood was maximum when measured after 1 hour from ethanol feeding. Hot water extract was more effective than 80% ethanol extract. The treatment of extract at 10 min. before ethanol feeding gave a better result than that at 10 min after or 1 hour before ethanol feeding. The ethanol concentration in brain and liver was lowered as found in the blood ethanol concentration. Acetaldehyde was not detected either in blood or the tissues. The optimal amount of the Puerariae flos was 55.7 mg/kg body weight. The newly developed analytical method using dichloromethane as extracting solvent was proven to be very effective in terms of speed and simplicity.
Ethanol and hot water extracts were prepared from Lindera obtusiloba Blume (LO) and Zanthoxylum piperitum DC (ZP) and used to evaluate their antimicrobial activities and thermal stability against six foodborne pathogens (3 gram-positive and 3 gram-negative bacteria). The antimicrobial activities were assessed using the agar diffusion method, and the thermal stabilities of extracts were examined after heat treatment at 60, 70, 80, and $100^{\circ}C$ for 10 min. The zones of inhibition by the LO extract or the ZP extract of the tested microorganisms were in the range of 21-30 mm and 19-25 mm, respectively, at 100 mg/mL concentrations. The 60% ethanol extract and the hot water extracts from LO showed the strongest antimicrobial effects against MRSA and Staphylococcus aurues, respectively. For the extract from ZP, the strongest antimicrobial effect was shown against S. aurues by 60% ethanol, and the weakest antimicrobial effect was shown against E. coli by the hot water extracts. The ZP extracts showed that the gram-positive bacteria were more sensitive than gram-negative bacteria. For the thermal stability of the extracts, the antimicrobial effects stabilized after heat treatment. Overall, the data suggest that the extracts have a potential for application in various food products for which a natural antimicrobial additive is desired.
Flavonoids in pine needles are known to be effective scavengers of free radical. Especially, proanthocyanidin, a kind of flavonoids possesses cardiovascular protection and antioxidative activities. Here, we evaluated proanthocyanidin contents in total polyphenolic compounds of pine needle extracts prepared by using hot water, ethanol, hexane or sub-supercritical $CO_2$. Analyses of total polyphenolic compounds and proanthocyanidin in each extracts indicated that hot water extract contained the highest concentrations, but sub-supercritical extract contained the lowest concentrations. On the other hand, evaluation of proanthocyanidin contents in total polyphenolic compounds in each extracts showed that sub-supercritical extract possessed the highest content, but hot water extract possessed the lowest content. These results indicate that extracts containing high concentrations of both total polyphenolic compounds and proanthocyanidin could be obtained by using hot water or ethanol extractions. Furthermore, extract containing high content of proanthocyanidin out of total polyphenolic compounds could be achieved by using sub-supercritical extraction. Measurement of antioxidative activities of extracts showed that hot water extract possessed the highest activity. In this study, we prepared extracts from pine needles by four different methods and evaluated the antioxidative compounds in extracts that could be used for effective components of functional food products.
Objectives : Acori Rhizoma is one of the common widely used herbal medicines with diverse bioactive effects. However, little evidence has been reported about the potential anti-platelet activity of Acori Rhizoma. The present study examined the effects on platelet aggregation by Acori Rhizoma. Methods : In this study, we tested the in vitro effect of 16 kinds of Acori Rhizoma extracts by hot water or 70% ethanol on collagen-induced platelet aggregation in human whole blood using the impedance method of aggregometry. Results : Among them, 2 kinds of 70% ethanol extract and 1 kind of hot water extract showed the significant inhibiting effect on whole blood aggregation. In particular, Acorus gramineus extracts were selected as the most effective candidate. Conclusiions : The results from this experiment provide pharmacological evidence for the traditional medicine, suggesting that Acorus gramineus could be help problems of blood circulation more than Acorus tatarinowii.
