The global spread of flaviviruses has triggered major outbreaks worldwide, significantly impacting public health, society, and economies. This has intensified research efforts to understand how flaviviruses interact with their hosts and manipulate the immune system, underscoring the need for advanced research tools. RNA-sequencing (RNA-seq) technologies have revolutionized our understanding of flavivirus infections by offering transcriptome analysis to dissect the intricate dynamics of virus-host interactions. Bulk RNA-seq provides a macroscopic overview of gene expression changes in virus-infected cells, offering insights into infection mechanisms and host responses at the molecular level. Single-cell RNA sequencing (scRNA-seq) provides unprecedented resolution by analyzing individual infected cells, revealing remarkable cellular heterogeneity within the host response. A particularly innovative advancement, virus-inclusive single-cell RNA sequencing (viscRNA-seq), addresses the challenges posed by non-polyadenylated flavivirus genomes, unveiling intricate details of virus-host interactions. In this review, we discuss the contributions of bulk RNA-seq, scRNA-seq, and viscRNA-seq to the field, exploring their implications in cell line experiments and studies on patients infected with various flavivirus species. Comprehensive transcriptome analyses from RNA-seq technologies are pivotal in accelerating the development of effective diagnostics and therapeutics, paving the way for innovative treatments and enhancing our preparedness for future outbreaks.
When olfactory placodes are transplanted at stages 23/24 from Xenopus laevis to Xenopus borealis hosts of the same age, it is possible to distinguish the cell populations of the host and donor due to the peculiar nuclear Q bands specific to X. borealis. I have replaced the eye anlage in each of a number of X. borealis with the transplanted olfactory placode of an individual X. laevis, or vice versa. In most instances, the placode of the donor fuses with that of the host. When fusion occurs, but not when the host and donor orqans grow separately, the cells of the donor were replaced gradually and according to a characteristic pattern by cells of the host. The basal cells of the donor were the first to be replaced, followed by the more matured cells of the sensory epithelium. This cellular substitution, proceeding in an orderly fashion from bottom to upper layers of the epithelium, depends on the fusion of the two organs. This observation suggests intercellular contacts in the mitotic zone of the two organs favor the host's cells over those of the donor.
Many important horticultural and field crops are susceptible to virus infections or may possess a degree of resistance to some viruses, but become infected by others. Plant viruses enter cells through the presence of wounds, and replicate intracellularly small genomes that encode genes required for replication, cell-to-cell movement and encapsidation. There are numerous evidences from specific virus-host interactions to require the involvement of host factors and steps during viral replication cycle. However, viruses should deal with host defense responses either by general or specific mechanisms, targeting viral components or genome itself. On the other hand, the host plants have also adapted to defend themselves against viral attack by operating different lines of resistance responses. The defense-related interactions provide new insights into the complex molecular strategies for hosts for defense and counter-defense employed by viruses.
The portion of Cuscuta japonica haustorium which lies external to the host tissues, the upper haustorium, was investigated at the light- and electron-microscopic levels. The haustorium lightly contacted with the host was formed by the expansion of the epidermis and cortex of the stem at the contact side, and to have a group of meristematic cells within the haustorial cortex. When such a haustorium was closely contacted with the host, the meristematic region transformed into a primordial structure of the endophyte (endophyte primordium, EP) which may penetrate into the host tissues. EP consisted of the three kinds of cell group: dividing cells at the adaxial or proximal side; large, elongate cells (idioblasts) at the middle portion,; compressed cells at the abaxial or basal side. the idioblasts were characterized by the presence of large nucleus, dense cytoplasm, several small vacuoles, and abundant cell organelles including the multilamellar structures and cytosegresomes, and thus suggested to have a high metabolic activity. The features of the EP were discussed in relation to the possibility of the penetrating into host tissues.

Adaptive responses to diverse microbial pathogens might be limited in relatively few types. Host cell responses to pathogens are believed to be patterned or stereotyped along with species or class. We tried to compose the host response to Mycoplasma in terms of cellular gene expression. Although gene expression profile of two host HeLa and 293 cells were quite different each other, 30 genes were differentially expressed by mycoplasma infection in both of HeLa and 293 cells. Six of them (PR48, MADH4, MKPX, CRK, RBM7, NEK3) were related to cell cycle or proliferation. Another category of genes like IL1 HY1, KLRF1, TNFSF14, GBP1 were host defense to elicit immune responses. With this set of genes, we establish the prediction model for mycoplasma contamination.
5G goes beyond people to serve indoor and outdoor companies and industries, as well as campuses such as halls, industrial complexes, educational institutions, stadiums, dense urban areas, rural areas, and government institutions. Therefore, a new approach to small cells is needed. Accordingly, 3GPP and Small Cell Forum are researching 5G small cell architecture; 3GPP, Small Cell Forum, and 5G Alliance for Connected Industries and Automation are also researching private networks tailored to meet the specific requirements of various companies and local governments. In particular, in the UK, a small cell-based technology is required for realizing the Joint Operator Technical Specifications-Neutral Host In-Building specification to cost-effectively secure indoor coverage. Further, the research on the SON(Self-Organizing Network) technology for small cells in 5G, where commercialization has begun, is required. The 5G-based small cell structure, private network, and Neutral Host In-Building and SON reviewed in this study are at the initial research stages; therefore, additional research is needed to secure the competitiveness of the small cell technology in 5G and Beyond 5G.
The glycoprotein of novirhabdoviruses is known to play a critical role in the determination of host specificity. Viral hemorrhagic septicemia viruses (VHSVs) in different genotypes have different glycoprotein sequences and show different preferences for specific cell lines. In this study, to know whether the glycoprotein is solely responsible for the host cell preference of VHSV, a recombinant VHSV expressing vesicular stomatitis virus (VSV) glycoprotein instead of VHSV IVa glycoprotein (rVHSV-VSV-G) was generated by reverse genetics and inoculated into several fish cell lines, then, cytopathic effect (CPE) and viral growth caused by rVHSV-VSV-G infection were compared with those caused by rVHSV-wild that was previously generated and has the same genomic sequence with wild-type VHSV except a few nucleotides. The plaque numbers of rVHSV-VSV-G were significantly higher in EPC, BF-2 and GF cells than those of rVHSV-wild. However, in HINAE cells (originated from olive flounder), rVHSV-VSV-G titer was significantly lower than rVHSV-wild titer, and both recombinant VHSVs were not grown well in CHSE-214 cells. Although statistical significances were detected in the titers between rVHSV-wild and rVHSV-VSV-G in several cell lines, the cell line-preference order of rVHSV-VSV-G was not different from that of rVHSV-wild. These results suggest that the replacement of VHSV glycoprotein may not completely change host cell preference, and other regions of VHSV might also involve in the determination of host cell preference.
Foot-and-Mouth Disease (FMD) is a highly contagious trans-boundary viral disease caused by FMD virus, which causes huge economic losses. FMDV infects cloven hoofed (two-toed) mammals such as cattle, sheep, goats, pigs and various wildlife species. To control the FMDV, it is necessary to understand the life cycle and the pathogenesis of FMDV in host. Especially, the protein-protein interaction between FMDV and host will help to understand the survival cycle of viruses in host cell and establish new therapeutic strategies. However, the computational approach for protein-protein interaction between FMDV and pig hosts have not been applied to studies of the onset mechanism of FMDV. In the present work, we have performed the prediction of the pig's proteins which interact with FMDV based on RNA-Seq data, protein sequence, and structure information. After identifying the virus-host interaction, we looked for meaningful pathways and anticipated changes in the host caused by infection with FMDV. A total of 78 proteins of pig were predicted as interacting with FMDV. The 156 interactions include 94 interactions predicted by sequence-based method and the 62 interactions predicted by structure-based method using domain information. The protein interaction network contained integrin as well as STYK1, VTCN1, IDO1, CDH3, SLA-DQB1, FER, and FGFR2 which were related to the up-regulation of inflammation and the down-regulation of cell adhesion and host defense systems such as macrophage and leukocytes. These results provide clues to the knowledge and mechanism of how FMDV affects the host cell.
Plant pathogenic oomycetes, such as Phytophthora spp., are the causal agent of the most devastating plant diseases. During infection, these pathogens accomplish parasitic colonization of plants by modulating host defenses through an array of disease effector proteins. These effectors are classified in two classes based on their target sites in the host plant. Apoplastic effectors are secreted into the plant extracellular space, and cytoplasmic effectors are translocated inside the plant cell, through the haustoria that enter inside living host cell. Recent characterization of some oomycete Avr genes showed that they encode effector protein with general modular structure including N-terminal conserved RXLR-DEER motif. More detailed evidences suggest that these AVR effectors are secreted by the pathogenic oomycetes and then translocated into the host plant cell during infection. Recent findings indicated that one of the P. infestans effector, Avrblb2, specifically induces hypersensitive response (HR) in the presence of Solanum bulbocastanum late blight resistance genes Rpi-blb2. On the other hand, another secreted RXLR protein PexRD8 originated from P. infestans suppressed the HCD triggered by the elicitin INF1. In this review, we described recent progress in characterized RXLR effectors in Phytophthora spp. and their dual functions as modulators of host plant immunity.
The induced resistance of cucumber leaves treated with oligochitosan to the infection of the cucumber powdery mildew, Sphaerotheca fuliginea, was investigated using transmission electron microscopy. The results showed that when the plants were treated with oligochitosan and challenged with inoculum, a significant decrease of the disease occurred. The mycelial development in the treated leaves was markedly inhibited. The cytoplasm of the powdery mildew mycelium was aggregated, with its organelles disintegrated and the cytoplasm collapsed. The protoplasm in haustoria became electron-dense. Haustoria became malformed, their organelles disintegrated, the hausterial wall thickened and eventually the whole complex necrotized. The host cells produced defence structures and materials associated with infection and a hypersensitive response. The host cell wall was thickened and deeply stained; several layers of papilla structure were produced under the cell wall; dark materials were deposited between the cell wall and plasmalemma; extrahaustorial plasmalemma was deeply stained and extrahaustorial matrix appositions had large deposits of electron-dense material; the cytoplasm was disordered, host organelles disintegrated and eventually the whole host cell disintegrated and necrotized.
