Insect Resistance and Horticultural Trait Genetic Values of Potato Families

  • This work aimed to evaluate 11 potato families for insect resistance horticultural traits. The families were derived from crosses between introduced insect resistant and adapted genotypes. A randomized complete block design, with three replications was used. The plot consisted of 25 genotypes of a family, summing up to 75 genotypes tested per family. Two susceptible cultivars were included in the experiment as checks. The genotypes were evaluated for insect resistance, tuber yield traits, tuber appearance and tuber skin smoothness. The genetic value for the intensity of insect attack in the leaves and in the tubers was lower(higher resistance) in the 11 families than in the checks. For yield traits, there was no predominant effect among the families regarding the origin of the resistant donor species(Solanum berthaultii and/or S. chacoense). However, there was a large contribution of the C-1485-16-87 recurrent parent in crosses with resistant genotypes for tuber yield and tuber number. In relation to tuber appearance, only the family derived form the C-1485-16-87/ ND140(S. berthaultii) cross did not differ from the checks. had genetic value tuber skin smoothness, it was identified that families derived from S. berthaultii resistant parents showed higher genetic values than the check cultivars. For tuber skin smoothness, the genetic values of the families neither differed significantly from each other nor from the checks.

Identification of Drought Tolerant Genotypes by Evaluating Morpho-physiological Traits in Pepper

  • The fast-changing climatic conditions make plants to be vulnerable to many abiotic stresses. Drought stress is one of the limiting factors that affect pepper production in water deficient regions. It affects plant growth and development by altering physiological, morphological, and metabolic processes. Breeding drought tolerant varieties is one of the mitigation strategies to overcome the ever increasing drought disaster. Hence, screening of new drought tolerant pepper genotypes is essential. The current study was aimed to identify new drought tolerant genotypes among the collection of pepper genetic resources. In total, 70 pepper genotypes were screened for drought tolerance after exposure to drought stress condition. The pepper genotypes were classified as highly tolerant, intermediate, or severely sensitive to drought stress based on the phenotypic analysis. Consequently, 13 genotypes significantly exhibited higher recovery rate after drought stress and were classified as highly tolerant. Comparative analysis of morphological and physiological parameters and expression of drought responsive genes between tolerant and susceptible pepper genotypes will be presented and discussed. The identified tolerant genotypes will be useful resources for breeding drought tolerant pepper cultivars.

  • 유전체 전장 연관분석 (GWAS)은 단일염기다형성(SNP)의 유전자형과 표현형 간의 통계적인 연관성을 분석함으로써 품종 선발용 SNP Marker 개발에 응용되고 있다. 본 연구에서는 Tano Red와 Ruby seedless 교배실생 278 계통을 대상으로 여러 과실 특성에 따른 관련 SNP를 동정함으로써 육종 선발에 필요한 DNA marker 개발에 필요한 기초 유전 자료를 얻고자 하였다. 한 계통 당 5~10개의 포도알을 선택하여 과립중, 과육탄성, 과피탄성, 과육경도, 과피경도, 과립당 종자갯수, 과립당 종자무게 및 인장강도를 측정하였다. 각 개체는 Genotyping by sequencing (GBS) 방법으로 Sequencing하여 Reference genome (Vitis vinifera PN40024 12X v2.)과 mapping 하였다. MAF (Minor allele frequency) >5%, Missing Data <30% 의 조건을 가진 SNPs 만 1차 선발하여 TASSEL과 GAPIT 프로그램으로 GWAS 분석을 하였다. Manhattan plot 결과 과립중 형질에서는 33개, 과립당 종자무게 25개와 인장강도에서는 20개의 통계학적으로 유의한 SNPs 가 선발되었고, 특이적으로 이들 모두 18번 염색체에서 발견되었다. 그러나 나머지 형질에서는 유의한 차이를 보이는 SNPs를 선발하지 못하였다. 과실의 인장강도는 수확 후 저장성과 유통과정에 영향을 미치기 때문에 Marker 개발을 통한 품종선별이 중요하다. 향후 이러한 특성과 본 연구를 통해 동정된 SNPs 의 상관관계를 구체적으로 연구하여 Marker 개발에 활용하고자 한다.

Cultivation and Characteristics of Licorice F1 (Glycyrrhiza glabra × G. uralensis) Lines

  • Licorice, a traditional Korean medicinal plant, is recognized for its main active ingredient, glycyrrhizin. The level of glycyrrhizin in Chinese licorice (Glycyrrhiza uralensis) is lower than the reference level (2.5%) set by the Korean Pharmacopoeia, reducing its value as a medicinal herb. In this study, we aimed to overcome this problem by generating an interspecific licorice hybrid by crossing European licorice (G. glabra) with Chinese licorice, resulting in the production of 32 $F_1$ lines. A comparison of genetic traits revealed variations in glycyrrhizin content among lines, ranging from 1.5 to 5.6%, with a mean value of 3.2%; these values are higher than that of the parental plants. Additionally, 25 lines (78.1%) had a glycyrrhizin content greater than 2.5%, which is higher than the reference level set by the Korean Pharmacopoeia. Four of these lines had glycyrrhizin levels higher than the WHO recommended level of 4.0%. A comparison of phenotypic characteristics showed that the leaves of the hybrids possessed all of the characteristics of European and Chinese licorice; however, the stems of most hybrids had characteristics of European licorice. Finally, we determined the genetic distances of 34 samples of Glycyrrhiza plants (parents, 32 $F_1$ lines) by random amplified polymorphic DNA (RAPD); the $F_1$ lines showed a close genetic distance. We plan to develop to a cultivar using five of these lines (glycyrrhizin content < 4.0%).

Early Autumn Maturing Pear Cultivar 'Sinhwa' with Fascinating Very Soft Flesh (부드러운 육질이 매력적인 중생종 배 '신화')

  • Pear cultivar 'Sinhwa' (Pyrus pyrifolia var. culta Nakai) was originated from a cross between 'Niitaka' and Whasan' with the aims of improving the fruit quality and the traits of cultivation and of early maturing more than 'Whasan' cultivar at Pear Research Station of National Institute of Horticultural & Herbal Science, Rural Development Administration in 1995. 'Sinhwa' was preliminarily selected in 2004 and named in 2009. The tree shows vigorous growth habit and semi-spread characters like 'Niitaka'. Furthermore, it has a sufficient and well upkeep of the flower bud, so it can be more easily cultivated in orchards. In the flower characteristics, flowering time of 'Sinhwa' is $11^{th}$ April like as maternal parent 'Niitaka'. Also 'Sinhwa' has short of pollen grains, so it is need above two pollinizer cultivars. 'Sinhwa' is highly resistant to black leaf spot (Alternaria kikuchiana) and relatively strong to pear scab (Venturia nashicola) in field condition. The optimum harvest time is around Sep. $15^{th}$ in Naju, which is ahead of 'Whasan' about 10 days in the harvest period. The fruit shape is oblate and fruit skin color is yellowish-brown during harvesting time. The average weight of fruit is 627 g, and the soluble solids content is $13.0^{\circ}Brix$. The flesh is very soft and juicy, and renders good eating quality. Shelf life is about 30 days under the room temperature condition.

Genomics and Molecular Markers for Major Cucurbitaceae Crops (주요 박과작물의 유전체 및 분자마커 연구 현황)

  • Watermelon and melon are economically important Cucurbitaceae crops. Recently, the development of molecular markers based on the construction of genetic linkage maps and detection of DNA sequence variants through next generation sequencing are essential as molecular breeding strategies for crop improvement that uses marker-assisted selection and backcrossing. In this paper, we intended to provide useful information for molecular breeding of watermelon and melon by analyzing the current status of international and domestic research efforts on genomics and molecular markers. Due to diverse genetic maps constructed and the reference genome sequencing completed in the past, DNA markers that are useful for selecting important traits including yield, fruit quality, and disease resistances have been reported and publicly available. To date, more than 16 genetic maps and loci and linked markers for more than 40 traits have reported for each watermelon and melon. Furthermore, the functional genes that are responsible for those traits are being continuously discovered by high-density genetic map and map-based cloning. In addition, whole genome resequencing of various germplasm is under progress based on the reference genome. Not only by the efforts for developing novel molecular markers, but application of public marker information currently available will greatly facilitate breeding process through genomics-assisted breeding.

Floral Analysis in the Kimpo Landfills and Its Peripheral Region (김포 매립지와 그 근린 지역의 식물상 분석)

  • Floral analysis about vegetation of Kimpo landfills and its periphery region was carried out. The study area was defined to a square $(81km^2)$ of which center was located at 250m in front of Andongpo, Komdan-myon, Kimpo-gun in the northwest part of the Kyunggi Province. This study was accomplished by analyzing five qualitative traits such as ecological strategy, reproductive strategy, distribution type, native/foreign division and life-form throughout actual investigation of the flora. The flora was composed of 536 taxa which comprise 105 families, 343 genera, 458 species, 1 subaspecies, 70 varieties and 7 forms (including 92 species of horticultural plant). Lythrum salicaria community, Spiraea salicifolia community, ottelia alismoides community and woods of Alnus japonica were recorded ? 새 retrictive distribution according to habitat characteristics. Owing to severe anthropogenic interferences such as construction of the Kimpo landfills in the study areas, a high proportion of ruderal plants and neophytes in the floral composition was recognized. The proportion of individual distribution type and therophyte was very high throughout whole study areas, and plant communities in the reclaimed areas were characterized by high proportion of phalanx plant species. Analysis of the correlations between meshes with ecological traits of plant species showed that Kimpo landfills and its periphery region was divided into two vegetation types, coastal type and inland type, as a result of possible saline effects.

Cultural and Morphological Characteristics of a New White Button Mushroom Cultivar 'Saedo'

  • Since development of the first Korean button mushroom cultivar 'Sae-Ah' in 2010, mushroom farmers have demanded an improved cultivar. Although a new cultivar, 'Saejeong', was developed in 2011, this cultivar has thus far been difficult to cultivate in typical mushroom farms. To solve this problem, another new cultivar, 'Saedo', was developed in 2012. This new cultivar was selected by genetic analysis from crossing the heterokaryon line A175 and the homokaryon line ASI1346-15. The heterokaryon A175 was a crossed line between strains ASI1038-211 and ASI1346-20. The mycelium of 'Saedo' showed good growth on compost dextrose agar at $25^{\circ}C$. The morphological traits such as the pileus (cap) and stipe of the 'Saedo' cultivar were thicker than those of the reference control 'Saejeong', and the production yield was increased by 1.89-fold compared to that of the reference control. A field experiment of the 'Saedo' cultivar has been ongoing since 2013. The yield of the 'Saedo' cultivar was found to be substantially higher than that of the reference control during the $2^{nd}$ flush.

Effect of GA3 and BA on Plant Growth of Ranunculus Cultivars

  • Ranunculus asiaticus characterizes colorful and attractive flower shapes that are related with the ornamental value of bulbous plants. Improving ornamental value of bulbous flowers has been the general goal of floricultural market. Gibberellic acid ($GA_3$) and benzyladenine (BA) play an important role in growth and developmental processes in floriculture. Combinational treatments of these two hormones have been used in floriculture to improve flower quality. We assessed the effects of combined $GA_3$ and BA, as well as the individual effects of each hormone, on growth characteristics using soil drench application to eight R. asiaticus cultivars, 'Giallo Millepetali', 'Bianco Millepetali', 'Arancio Millepetali', 'Rosa SC', 'Arancio Pratolino', 'Giallo Pratolino', 'Bianco Pratolino', and 'Rosa Ch Pratolino'. $GA_3$ treatments increased plant height and first flower size of R. asiaticus cultivars. Moreover, about 5 to 9 days to flowering were averagely shortened by $GA_3$ treatments compared to controls. On the other hand, the opposites, including first flower size and days to flowering, were observed for cultivars treated with BA, compared with controls. Treatments of $GA_3$ + BA generally affected growth traits, such as plant height, flower size, and the timing of flowering on some R. asiaticus cultivars. In particular, about 5 to 6 days to flowering were reduced on average by Treatments of $GA_3$ + BA. Our results showed positive growth effects, including plant height, days to flowering, first flower height, number of flowers from the application of individual and combined hormones to R. asiaticus cultivars and demonstrate a role for these hormones in future bulbous floriculture.

'Kowon', a New Korean Ginseng Cultivars with High Yield and Alternaria Blight Resistance

  • Recently, there has been increased attention to the development of new plant cultivars with enhanced resistance to biotic and abiotic stress. To develop ginseng cultivars with such traits, systematic breeding programs and comprehensive field studies are prerequisites. In this study, we applied a pure-line selection method to identify a ginseng cultivar with enhanced stress resistance. Phenotypic and agronomic characteristics, seed yield, and physiological responses to biotic and abiotic stresses were investigated according to the guidelines of the International Union for the Protection of New Varieties of Plants (UPOV). In the newly developed 'Kowon' cultivar, the time of emergence, flowering, and berry maturity were intermediate between those of the controls, 'Yunpoong' and 'Chunpoong'. The stem length of 'Kowon' was intermediate, whereas the root length was shorter and the main root diameter was greater than those of 'Chunpoong'. In local adaptability tests conducted in three regions, the yield of 'Kowon' was $666kg{\cdot}10a^{-1}$; 27% and 4% higher than that of 'Chunpoong' and 'Yunpoong'. Diseases such as Alternaria blight, Phytophthora blight, mulberry mealybug, and nematode infestation did not occur in 'Kowon'; and it also exhibited moderate resistance to damping-off and anthracnose. In these cases, yellow spots occurred on aerial parts and the rusty skin of the root, and it exhibited moderate resistance at high temperatures. Our study demonstrates that 'Kowon', which has a high root weight and enhanced biotic/abiotic stress resistance, is a superior cultivar that could increase farmers' income.
