Objectives: Diabetic nephropathy is the most common cause of end stage renal disease. Transforming growth factor (TGF)-${\beta}1$, type IV collagen, advanced glycation end-products (AGEs), and angiotensin II type 1 receptor (AT1) are the main factors of diabetic nephropathy. We investigated the effects of Prunus on renal function and histopathological changes of diabetic nephropathy rat model induced by unilateral nephrectomy and streptozotocin. Methods: Diabetes was induced in male Sprague-Dawley rats ($290{\pm}10g$) by injecting streptozotocin (55 mg/kg) into the tail vein after unilateral nephrectomy. Rats were divided into 3 groups (n=6): normal, control, and Prunus. After 8 weeks of oral administration of Prunus extract on the Prunus group from 3 days after streptozotocin injection, we checked weight, 24 hrs urine, blood biochemistry and renal tissue to evaluate renal function and histopathological changes by examining parameters including albuminuria, BUN, creatinine, cholesterol, low density lipoprotein (LDL), triglyceride, TGF-${\beta}1$, type IV collagen, AGEs, and AT1. We also measured mRNA expression of TGF-${\beta}1$, type IV collagen, AGEs, and AT1 by Real Time polymerase chain reaction (RT-PCR). Results: Prunus decreased the amount of 24 hrs proteinuria, and inhibited histopathological changes of diabetic nephropathy including the expression and accumulation of TGF-${\beta}1$, type IV collagen and AGEs which could promote development of diabetic nephropathy. Prunus also inhibited mRNA expression of TGF-${\beta}1$, type IV collagen. Conclusions: These findings suggest that Prunus might protect the renal function and inhibit the development of renal injury by regulating factors including TGF-${\beta}1$, type IV collagen, AGEs, except AT1, so Prunus can be used for diabetic patients to prevent the progression of diabetic nephropathy.
This study compared the effects of diets consisting of experimental extruded pellets (EP), commercial soft extruded pellets (SEP), and raw-fish-based moist pellets (MP) on the growth, flesh quality, and histology of the Korean rockfish Sebastes schlegeli. Three replicated groups of 60 fish each (initial mean weight 152 g) were fed one of two experimental EPs (EP1 and EP2), two commercial SEPs (SEP1 and SEP2), or MP for 11 weeks. The survival, weight gain, and final mean weight of the fish did not differ significantly among the groups. The highest feed efficiency was in fish fed MP (P<0.05), and the protein efficiency ratio of fish fed MP was significantly higher than that of fish fed EP1, EP2, or SEP1, but not significantly different from that of fish fed EP2. The daily feed intake of the fish fed the EPs and SEPs was significantly higher than that of the fish fed MP (P<0.05). The condition factor, Hepatosomatic Index, and Viscerasomatic Index of the fish did not differ significantly among the groups. No notable differences in the textural properties of the dorsal muscle, plasma biochemical parameters, or histological features of the fish were observed in fish fed any diet. This suggests that extruded pellets can be used to feed growing Korean rockfish without compromising growth, flesh quality, or histopathological change in comparison to raw fish-based moist pellets.
Effects of Sardine Oil Feeding and Vitamin E Supplementation on Histopathological Changes and $\alpha$-L-fucosidase activity in experimental hepatocarcinogenesis. Sprague-Dawley rats weighing 80~90 g were fed the diet containing either 15% corn oil (CO) or sardine oil (SO) with or without vitamin E supplements (dl-$\alpha$-tocopherol acetate 800 IU/kg diet) for 8 weeks. After 2 weeks of feeding, the rats were given a single intraperitoneal injectin of diethylnitrosamine (DEN, 200 mg/kg BW). From the fifth week, rats were given 0.02% acetylaminofluorene (AAF) in diet for 4 weeks. At the seventh week, 0.05% phenobarbital in liver and hepatic glutathione S-transferase palcental form positive (GST-P+) foci were examined by Hematoxylin& Eosin (H&E) staining and immunohistochemical method, respectively. Serum $\alpha$-L-fucosidase activity was determined. The livers fromt he carcinogen treated rats showed significantly increased formation of GST-P+ foci at sacrifice points while the livers fromthe non-carcinogen treated groups showed almost no foci. Although GST-P+ foci formation was not affected by dietary oil, it was increased unexpectedly by vitamin E supplementation. Histopathological changes were similar to patterns of GST-P+ foci formation in almost all groups. Serum $\alpha$-L-fucosidase activities were increased by carcinogen treatment in all dietary groups. $\alpha$-L-fucosidase activities were positively correlated with GST-P+ foci formation. There results suggest that excessive vitamin E supplementation can enhance hepatocarcinogenesis although the mechanisms involved are not clearly understood.
Objectives: Burn is a severe dermal injury caused by heat. We were to investigated the effects of acupuncture or pharmacopunture treatment for the histologic and morphologic recovery on the mouse with the 3rd grade burn skin. Methods : We divided into 3 groups. One was a control group(n=3) that was not treated any treatments. Another was a acupuncture group(n=3) that was treated only acupuncture. The other was a pharmacopuncture group(n=3) that was treated only pharmacopuncture. We made a 3rd grade burned skin with the stainless steel heating apparatus. We made a treatment for the mice for a week(2 times for a day, totally 14 times treatments). We observed a dermal morphologic recovery on the mice and a histopathological photographs of the burn skin and subcutaneous tissue with H&E stain, Masson's trichrome stain, and VEGF, FGF and c-kit immunohistochemical stain. Results : The pharmacopuncture group were a better morphologic recovery than control group and acupuncture group. And the pharmacopuncture group were a better histopathological recovery than control group and acupuncture group on the burn skin and subcutaneous tissue with H&E stain, Masson's trichrome stain. And the pharmacopuncture group were a better histopathological recovery than control group and acupuncture group on the burn skin and subcutaneous tissue with VEGF, FGF and c-kit immunohistochemical stain. Conclusions : We suggest that the pharmacopuncture treatment is a better histologic and morphologic recovery than the no treatment or the acupuncture group with the burned mouse skin.
Objectives : This study was to evaluate Sibjeondaebo-tang’s(Shiquan dabutang) effect to rat hypothyroidism which induced by 6-n-propyl-2-thiouracil(PTU)Methods : Aqueous extracts of Sibjeondaebo-tang(Shiquan dabutang)(21.29%) were administered by oral dose of 300 and 150 mg/kg(rat’s body weight) for 42 days from 2 weeks before start of PTU treatment. During this moment the body weight, thyroid gland weights, liver weight, serum thyroid hormone, lipid content and liver antioxidant defense system were measured with histopathological changes of thyroid glands, Liver and these result were compared with LevoT4 0.5mg/kg treated ratsResults : As a result of PTU treatment, marked decreases of body weight, liver weight, serum thyroid hormone levels of T3, T4, triglyceride content and liver CAT activation were observed with marked increases of thyroid gland weight, thyroid gland hormone TSH, activate liver H2O2 and SOD, serum Total cholesterol, HDL and AST amount were admitted. In addition, marked hyperplasia of follicular cells with reduce of follicular colloid contents and sizes were showed at histopathological inspections. These PTU-induces histopathological changes related to hypothyroidism were significantly reduced by treatment of both different dosages of Sibjeondaebo-tang(Shiquan dabutang).Conclusions: This study suggest that Sibjeondaebo-tang(Shiquan dabutang) extracts have favorable effect on the thyroid hormone productions with beneficial effect on hypothyroidism medicated by modulatory effects on antioxidant defense system.
Acetaminophen (APAP) and gentamicin are widely used for many patients, but little in-formation is available regarding the combined effects of APAP and gentamicin. This study was aimed to investigate the potent nephrotoxicity following combined-treatment with APAP and gentamicin. Serum biochemical parameters and histopathological changes in the kidney were observed in female SD rats after continuous daily treatment with either 600 mg/kg/day APAP, and/or 300 mg/kg/day gentamicin for 3 days, and compared with saline sham-treated control animals. APAP and gentamicin combination-treated rats exhibited inconsistent increasing tendency in blood urea nitrogen (BUN) by 96 hours after the last treatment, compared to control or the animals treated with each drug. The relative kidney weights were also increased. Histopathological findings of kidneys revealed that necrosis of proximal convoluted tubules were higher in rats treated with APAP and gentamicin combination than the rats treated with each drug alone. These results suggest that combination use of both drugs have more severe nephrotoxicity than treating each drug alone.
Objective: The root bark of Ulmus davidiana var. Japonica (Ulmi Radicis cortex, URC) is a medicinal herb used for promoting diuresis and treating dampness. In Korea, URC has long been used as an efficacious therapy for inflammation, burns, frostbite and skin diseases such as eczema and psoriasis. Methods: In the present study, we used 1-fluoro-2,4-dinitrofluorobenzene (DNFB)-induced contact dermatitis (CD) mouse model to investigate the anti-allergic and the anti-inflammatory effects of URC on skin lesion, histopathological changes and specific antibody production. Results: URC treatment, 10 mg/mL, effectively inhibited skin lesions induced by repeated paintings with DNFB. In the histopathological observation, topical application of URC inhibited spongiosis. In addition, URC lowered the production levels of total immunoglobulin and IgG2a in serum. Conclusion: These data indicate that URC has an anti-inflammatory effect that produces an improvement of skin lesions in CD mice.
The purpose of this study was to determine the gingival response to the various restorations for 3 weeks, 5 weeks and 8 weeks respectively after they had been inserted in 42 tooth of 5dogs. The histopathological observation was also performed to evaluate the effect of the variuos restorations on gingival tissue. They inclued gold, copper and nickel-chrome alloy. The following findings were obtained.
1. The gingivae adjacent to the well adapted and polished restorations and their margins with a level of gingival crest were grossly and histopathologically found no specific changes.
2. The gingive adjacent to the ill fitting and unpolished restorations and their margins with subgingival extension of 1 to 1.5mm were not grossly found any changes but hitopathologically, the inflammatory changes.
3. Thee wee no obvious difference in gingival response among the various alloys in histopathological observation.
Estrous and non-estrous rabbits were inoculated with E. coli or Streptococcus pyogenes, and the mixture of the two organisms, and bacterial count and histopathological studies of uterine horns were made to observe the effects of estrogen on the resistance of the uterus to bacterial infection. The results obtained were summarized as followings; 1. Four hours after inoculation of bactoria into uterine horn, the number of organisms was significantly lower in estrous rabbits than in non-estrous regardless of the kind of organisms inoculated. 2. The highest reduction rate of the organisms among the three bacterial inoculation groups was found in estrous rabbits inoculated with E. coli, and the lowest reduction rate was with Streptococcus pyogenes. 3. Histopathological changes of uterine horns induced five days after bacterial inoculation were observed. In estrous rabbits, a mild inflammatory reaction was found in Streptococcus pyogenes group, but a slight inflammatory reaction and only a negligible inflammatory reaction were observed in mixed bacteria group, and in E. coli group respectively. In non-estrous group, however, a marked inflammatory reaction was observed in Streptococcus pyogenes group, a moderate inflammatory reaction and a slight inflammatory reaction were observed in the mixed bacterial group and E. coli group, respectively.
Hepatoprotective effects of the water extract of Protaetia brevitarsis larva (PB) was investigated in carbon tetrachloride ($CCI_{4}$) treated male Sprague-Dawley rats. PB administration protected rats against ALT, AST and LDH elevations induced by $CCI_{4}$, as well as the severity of liver damage. PB recovered the decrease in serum level of high-density lipoprotein-cholesterol and the increase in serum level of low-density lipoprotein-cholesterol induced by $CCI_{4}$. In histopathological observation, massive fatty change and necrosis in the centrilobular area, degenerative change including pyknosis of nucleus and swelling of parenchymal cell induced by $CCI_{4}$ were clearly protected by PB. These histopathological findings paralleled with the serum biochemical results. The present results demonstrated that the water extract of PB may have the hepatoprotective effect against $CCI_{4}$-induced liver damage in vivo.
