  • The features of special brand eggs are growing more and more diverse and it is hard to draw a clear distinction between these and ordinary (no-brand) eggs. Special brand eggs range from those with objectively recognizable characteristics to those given only an unsubstantial product image and price differentials are among them great, too. The relation between product features and prices is unclear. Special brand eggs are the commodity whose characteristics are the vaguest of all livestock products. Farm's brand eggs produce a high profitability to producers is they are directly sold to consumers, by, for example, home-delivery service. But if they are sold to supermarkets, etc., producer's (poultry farmer's) income becomes lower by the amount of distributors' margin, reducing the profitability substantially. Thus how to increase the ration of retailing is important for farmers to secure a high profit. The sales strategies of poultry are the combination of two elements, that is, new product development (product differentiation) and creation of new market. But it is difficult for special brand egg producers to develop products with clear characteristics (use value) distinct form ordinary eggs and so these producers depend on the factors of appearance, such as the color of egg shells and package. Special brand eggs manage to keep their marketable value by the combination of the few product features and product image. Thus NB eggs from feed producers have a great market-ability since they can take advantage of the power of patents and TV commercials. However, market differentiation affects profitability much more than product features and price gaps are very wide between directly sold and wholesaled eggs. The producers of special brand eggs have come to the turning point where they have to decide whether they will content with being the subcontractors for NB and PB eggs or they will continue to keep their independence in production and marketing.

Studies on Cultural Characteristics for High Density Fermentation of Phellinus linteus WI-001 (Phellinus linteus WI-001 균사체의 고밀도 배양을 위한 배양학적 특성 연구)

  • Various environmental factors such as pH, temperature and initial glucose concentration were investigated for enhancing cell growth in fermentations of Phellinus linteus WI-OOl, a producer of polysaccarides with potent anticancer activities. Optimal pH and temperature were around 5.5 and $28^{\circ}C$, respectively. Relatively little variation of pH was observed ranging between 5.5 and 6.5 during the whole fermentation period. Maximum cell concentration and specific growth rate were investigated in the media containing initial glucose concentrations of 0.5%, 1 %, 2%, 3% and 4%. High initial glucose concentration enhanced biomass production but showed negative effect on specific growth rate. In bioreactor experiments with various feeding strategies, increases of 28% and 42% in final cell concentration were obtaind as compared to conventional batch process, by adopting pulse and continuous supplement of 2% glucose solution, respectively.

  • Chinese cabbage is a staple food to Korean, which has the high degree of self-sufficiency and worldwide breeding technique in the aspect of seed. However, the producers' competitiveness has been decreasing after the agricultural product market was open. In order to cope with this problem, the government introduced some policies for promoting seed industry in 2007 to reflect producer's needs for high quality seeds of Chinese cabbage. These policies will be a good opportunity for producers to secure and promote the producers' competitiveness against low-price importing Chinese cabbage. In this aspect it is very important to know how well these policies are established and what Chinese cabbage farmers want in regard to a new variety of Chinese cabbage seed. This study was carried out to look over the Chinese cabbage producers' using pattern about a new variety of seed and show some directions for efficient way of diffusion of a new variety of seed to producers using a survey research. The main results are as follows. The producers thought the characteristics of a new variety of seed most important factor compared to other factors such as the seed price, and easiness of cabbage sales when they choose a seed. Also, the 65% of respondents were willing to accept a new variety of seed and thought the government support for an exhibition field and diffusing public information about a new variety of Chinese cabbage seed are important in accepting a new variety of seed.

  • In recent years, research at the Environmental Microbial and Food Safety Laboratory (EMFSL), Agricultural Research Service (ARS) has focused on the development of novel image-based sensing technologies to address agro-food safety concerns, and transformation of these novel technologies into practical instrumentation for industrial implementations. The line-scan-based hyperspectral imaging techniques have often served as a research tool to develop rapid multispectral methods based on only a few spectral bands for rapid online applications. We developed a newer line-scan hyperspectral imaging platform for high-speed inspection on high-throughput processing lines, capable of simultaneous multiple inspection algorithms for different agro-food safety problems such as poultry carcass inspection for wholesomeness and apple inspection for fecal contamination and defect detection. In addition, portable imaging devices were developed for in situ identification of contamination sites and for use by agrofood producer and processor operations for cleaning and sanitation inspection of food processing surfaces. The aim of this presentation is to illustrate recent advances in the above agro.food safety sensing technologies.

Characterization of the Fecal Microbial Communities of Duroc Pigs Using 16S rRNA Gene Pyrosequencing

  • This study characterized the fecal bacterial community structure and inter-individual variation in 30-week-old Duroc pigs, which are known for their excellent meat quality. Pyrosequencing of the V1-V3 hypervariable regions of the 16S rRNA genes generated 108,254 valid reads and 508 operational taxonomic units at a 95% identity cut-off (genus level). Bacterial diversity and species richness as measured by the Shannon diversity index were significantly greater than those reported previously using denaturation gradient gel electrophoresis; thus, this study provides substantial information related to both known bacteria and the untapped portion of unclassified bacteria in the population. The bacterial composition of Duroc pig fecal samples was investigated at the phylum, class, family, and genus levels. Firmicutes and Bacteroidetes predominated at the phylum level, while Clostridia and Bacteroidia were most abundant at the class level. This study also detected prominent inter-individual variation starting at the family level. Among the core microbiome, which was observed at the genus level, Prevotella was consistently dominant, as well as a bacterial phylotype related to Oscillibacter valericigenes, a valerate producer. This study found high bacterial diversity and compositional variation among individuals of the same breed line, as well as high abundance of unclassified bacterial phylotypes that may have important functions in the growth performance of Duroc pigs.

Genetic and biochemical evidence for redundant pathways leading to mycosporine-like amino acid biosynthesis in the cyanobacterium Sphaerospermopsis torques-reginae ITEP-024

  • Cyanobacteria have been widely reported to produce a variety of UV-absorbing mycosporine-like amino acids (MAAs). Herein, we reported production of the unusual MAA, mycosporine-glycine-alanine (MGA) in the cyanobacterium Sphaerospermopsis torques-reginae ITEP-024 using a newly developed UHPLC-DAD-MS/HRMS (ultra-high performance liquid chromatography-diode array detection-high resolution tandem mass spectrometry) method. MGA had previously been first identified in a red-algae, but S. torques-reginae strain ITEP-024 is the first cyanobacteria to be reported as an MGA producer. Herein, the chemical structure of MGA is fully elucidated from one-dimensional / two-dimensional nuclear magnetic resonance and HRMS data analyses. MAAs are unusually produced constitutively in S. torques-reginae ITEP-024, and this production was further enhanced following UV-irradiance. It has been proposed that MAA biosynthesis proceeds in cyanobacteria from the pentose phosphate pathway intermediate sedoheptulose 7-phosphate. Annotation of a gene cluster encoded in the genome sequence of S. torques-reginae ITEP-024 supports these gene products could catalyse the biosynthesis of MAAs. However, addition of glyphosate to cultures of S. torques-reginae ITEP-024 abolished constitutive and ultra-violet radiation induced production of MGA, shinorine and porphyra-334. This finding supports involvement of the shikimic acid pathway in the biosynthesis of MAAs by this species.

Properties of ${\beta}$-Galactosidase from Bacillus licheniformis Isolated from Cheongkookjang (청국장 유래 Bacillus licheniformis의 ${\beta}$-Galactosidase 특성)

  • A bacterial strain was isolated from homemade Cheongkookjang as a producer of the ${\beta}$-galactosidase, capable of hydrolyzing lactose to liberate galactose and glucose residues. The isolate YB-1105 has been identified as Bacillus licheniformis on the basis of its 16S rDNA sequence, morphology and biochemical properties. ${\beta}$-Galactosidase activity was detected in both the culture supernatant and the cell extract of B. licheniformis YB-1105. The enzymes of both fractions demonstrated maximum activity for hydrolysis of para-nitrophenyl-${\beta}$-D-galactopyranoside (pNP-${\beta}Gal$) under identical reaction conditions of pH 6.5 and $50^{\circ}C$. However, ${\beta}$-galactosidase activity from the culture filtrate was affected more than that from the cell free extract at acidic pHs and high temperatures. The hydrolyzing activity of both ${\beta}$-galactosidases for pNP-${\beta}Gal$ was dramatically decreased by the addition of low concentrations of galactose, but was only marginally decreased by high concentrations of glucose or mannose.

  • As basic study for purpose bioremedation in oil-contaminated environment, Primarily, we isolated biosurfactant producer- strains utilized of oil-agar plate, and measured surface tension and emulsifying activity. We investigated in oil-contaminated soil and sea water. In this laboratory, Pseudomonas sp. EL-012S strain isolated from oil-contaminated soil was able to product novel biosurfactant under the optimal culture condition. Its condition was n-hexadecane 2.0%, NH$_4$NO$_3$0.4%, Na$_2$HPO$_4$0.6%, KH$_2$PO$_4$0.4%, MgSO$_4$.7$H_2O$ 0.02%, CaCl$_2$.2$H_2O$ 0.001%, FeSO.7$H_2O$ 0.001%, initial pH 7.0 and aeration at 3$0^{\circ}C$, respectively. This biosurfactant was produced in both late-exponential and early-stationary phase. The biosurfactant from Pseudomonas sp. EL-012S was composed of carbohydrate, lipid and protein. The purified-biosurfactant was examined two (biosurfactant type I, II) with the silica gel G60 column chromatography and the purified biosurfactant confirmed thin layer chromatography, high performed liquid chromatography and gas chromatography. The biosurfactant type I involved in carbohydrate-lipid-protein characteristics lowered surface tension of water to 27dyne/cm and interfacial tension 4.5dyne/cm aginst to n-hexadecane and the biosurfactant type B involved in carbohydrate lipid characteristics lowered surface tension of water to 30dyne/cm and interfacial tension 8dyne/cm against to n-hexadecane. Specially type I had the properties such as strong emulsifying activity, emulsion stability, pH-stability, thermo-stability, high cleaning activity and forming ability.

Isolation and Characterization of Indigenous Marine Diatom Achnanthidium sp. BS-001 Producing a high Content of Omega-3 Fatty Acid and Fucoxanthin Production (오메가-3 지방산 및 푸코잔틴 고함량 토착 규조류 아크난티디움 균주의 분리 및 특성)

  • Omega-3 fatty acids and carotenoids, which are known as representative high-value substances derived from microalgae, are being studied from various diatoms. Most of the diatoms contain fucoxanthin and omega-3 fatty acid. Fucoxanthin produced by diatom has been reported as bioactive compounds exhibiting strong antioxidant, anticancer and anti-inflammatory activities. However, the low growth rate and fucoxanthin content of diatoms are one of the big obstacles to the industrial application. In this study, indigenous marine diatom Achnanthidium sp. BS-001 was isolated for a candidate of fucoxanthin producer. Light intensity and temperature for the culture of Achnanthidium sp. BS-001 were optimized on PhotoBiobox. Optimization of silicate concentration for increasing BS-001 biomass productivity was confirmed in F/2 medium with various concentration of sodium silicate. As a result, condition of light intensity, temperature, and silicate concentration for optimal cultivation were $150{\mu}mol{\cdot}m^{-2}{\cdot}s^{-1}$, $18^{\circ}C$ and 0.106 mM, respectively. Maximum biomass productivity reaches to $154.3mg{\cdot}L^{-1}{\cdot}day^{-1}$, and then the content of omega-3 fatty acids and fucoxanthin were $19.4mg{\cdot}g^{-1}$, $9.05mg{\cdot}g^{-1}$, respectively. These results indicate that Achnanthidium sp. BS-001 has the potential to be used as a source of omega-3 fatty acids and fucoxanthin.

  • Smart Grids are intelligent next generating Electric Power System (EPS) that provide environment-friendliness, high-efficiency, and high-trustworthiness by integrating information and communication technology with electric power technology. Smart grids help to supply power more efficiently and safely than past systems by bilaterally exchanging information between the user and power producer. In addition, it alleviates environmental problems by using renewable energy resources. However, smart grids have many cyber security risks because of the bilateral service, the increase of small and medium-sized energy resources, and the installation of multi-sensors or control devices. These cyber risks can cause critical problems within a national grid through even small errors. Therefore, in order to reduce these risks, it is necessary to establish a cyber security strategy and apply it from the developmental stage to the implementation stage. This thesis analyzes and recommends security strategy in order to resolve the security risks. By applying cyber security strategy to a smart grid, it will provide a stepping-stone to creating a safe and dependable smart grid.
