This study was performed to investigate the effects of liquid culture of Agaricus blazei on the lipid metabolism and enzyme activities in growing male rats. Sprague-Dawley rats were given four different types of diets for a succeeding period of 5 weeks, respectively: a normal diet group (7% corn oil), a high fat diet group (7% com oil + 15% lard), a 20 or 30% Agaricus diet groups (high fat diet + 20 or 30% Agaricus in water) according to the levels of Agaricus supplementation. The body weight gains, food intake, food efficiency ratios, and hepatic, kidney, spleen and pancreas weights of the rats fed 20 or 30% Agaricus diets were similar to those of the rats fed high fat diet. The epididymal fat pad weight of the rats fed high fat diet and 20 or 30% Agaricus diets were significantly higher than that of the rats fed normal diet. The concentrations of serum triglyceride, total cholesterol, LDL-cholesterol and HDL-cholesterol, and the activity of glutamic pyruvic transaminase in the rats fed 30% Agaricus diet were significantly lower than those in the rats 114 high fat diet. But the concentrations of hepatic total cholesterol and triglyceride of rats fed the 20 or 30% Agaricus diets were similar to those of rats fed the high fat diet. The HDL-oholesterol/total-cholesterol ratio of the rat fed 30% Agaricus diet was significantly higher than that of the rats fed high fat diet, The activity of glutamic oxaloacetic transaminase in the rats fed 20 or 30% Agaricus diets were similar to those in the rats fed high fat diet. There were no differences in the concentrations of total protein, hemoglobin and glucose, and the activities of alkaline phosphatase, and the atherogenic index in the serum among the experimental groups. These results showed that the 30% Agaricus diet feeding decreased the total cholesterol, the triglyceride and the LDL-cholesterol, and increased the HDL-oholesterol/total cholesterol ratio in serum of rats.
This study was conducted to investigate whether dietary factors, normal fat and genistein leads to beneficial improvement of lipid metabolism and oxidative stress in adult hyperlipidemic male rats. Seven wk-old male SD rats were fed high fat diet (15% fat, 1% cholesterol) for 4 wks for induction of hyperlipidemic model rat. Weight-matched rats were then assigned to four groups according to dietary fat level (7% or 15% fat) and genistein contents (0 or 320 mg/kg diet). Food intake was significantly decreased by both high fat intake and genistein supplementation compared with normal fat intake and genistein no supplementaion. But weight gain was significantly decreased by genistein supplementation in normal fat intake compared with the other groups. Total lipid, total cholesterol and triglyceride in serum and liver were significantly decreased by normal fat intake compared with high fat intake. But total cholesterol in liver was significantly increased by genistein supplementation in both high fat and normal fat intake. TBARS in serum and liver was less produced by normal fat intake compared with high fat intake but TBARS in liver was significantly increased by genistein supplementation compared with genistein no supplementation in normal fat intake. Glutathione reductase activity in erythrocytes was significantly reduced by genistein supplementation in normal fat intake compared with the other groups. Glutathione peroxidase and glutathione reductase activities in liver were significantly inhibited by normal fat intake compared with high fat intake. Catalase activity in liver was significantly increased by genistein supplementation compared with genistein no supplementation in high fat intake. Nitrite was significantly decreased by normal fat intake compared with high fat intake. These results suggest that normal fat intake has the treatment effect against risk factors related with cardiovascular disease by reducing lipid profiles, lipid peroxidation. And genistein shows action as a antioxidant replacing antioxidant enzymes but also may act as prooxidant causing the production of TBARS.

The effects of Pleurotus (P.) eryngii on the weight gains, food efficiency ratios, serum and hepatic lipid concentrations were investigated in male rats fed the high fat diets. Twenty one week-old Sprague-Dawley rats were given three different types of diet for 10 weeks, respectively: a control diet (high fat; $20\%$ lard), two kinds of P. eryngii diet supplemented with $3\%\;or\;5\%$ of dry P. eryngii powder in substitutes for the same amounts of cellulose in the control diets. The body weight gains and food efficiency ratios of the rats fed $3\%\;and\;5\%$ P. eryngii diets were significantly lower than those of the rats fed the control diet. The concentrations of hepatic total lipid and triglyceride in the rats fed the P. eryngii diets were significantly lower than those in the rats fed the control diet. But the hepatic cholesterol contents of the rats fed the P. eryngii diets were similar to those of the rats fed the control diet. The concentrations in serum total cholesterol, LDL-cholesterol, and atherogenic index ratios were significantly lower in the rats fed the P. eryngii diets compared to those fed the control diet. The HDL-cholesterol/total- cholesterol ratio was significantly higher in the rats fed the P. eryngii diets compared to those fed the control diet. There were no significant difference found in the serum triglyceride, phospholipid and HDL-cholesterol concentrations among the experimental groups. These results showed that feeding of P. eryngii powder feeding decreased the total cholesterol, LDL-cholesterol and atherogenic index and increased the HDL-cholesterol/total-cholesterol ratio in serum of the rats fed high fat diet.
Objectives: To investigate the effects of Chrysanthemum indicum L. pharmacopuncture on lipids, antioxidant capacity and anti-inflammation in rats fed high-fat diet. Methods: Hyperlipidemic rats induced by high-fat diet were divided into 5 groups: no treatment control (normal, n=8), high-fat diet only control (control, n=8), high-fat diet and Chrysanthemum indicum L. pharmacopuncture at CV4 group (TI, n=8), high-fat diet and Chrysanthemum indicum L. pharmacopuncture at CV17 group (TII, n=8), and high-fat diet and Chrysanthemum indicum L. pharmacopuncture at EX-HN3 group (TIII, n=8). They were given pharmacopuncture accordingly every other day for two weeks followed by analyses of lowering lipids effects, oxidative capacity and anti-inflammatory effects. Results: Compared with the control, pharmacopuncture groups showed significantly decreased plasma total cholesterol (TC), liver thiobarbituric acid reactive substance (TBARS), catalase, glutathione peroxidase, neutrophils, monocytes, plasma and liver IL-$1{\beta}$, and plasma and liver IL-6. In other parameters including plasma and liver triglyceride, liver TC, LDL-cholesterol, HDL-cholesterol, liver TBARS, supraoxide dismutase, total protein, albumin, blood cell analysis, plasma and liver TNF-$\alpha$, and IL-10, there was no significant difference between control and pharmacopuncture groups. No clear acupoint-specificity was observed. Conclusions: Chrysanthemum indicum L. pharmacopuncture may improve control of hyperlipidemia.
We found previously that dietary high fat caused obesity, and levan supplementation to the regular diet reduced adiposity and serum lipids. In the present study, we examined the effects of levan [high-molecular-mass $\beta$-(2,6)-linked fructose polymer] supplement on the development of obesity and lipid metabolism in rats fed with high-fat diet. Thus, to determine whether the dietary levan may have the anti-obesity and hypolipidemic effects, 4-wk-old Sprague Dawley male rats were fed with high-fat diet for 6 wk to induce obesity, and subsequently fed with 0, 1, 5, or 10% levan supplemented high-fat diets (w/w) for another 4 wk. For the comparison, a normal control group was fed with AIN-76A diet. Supplementation with levan resulted in a significant reduction of high-fat-induced body weight gain, white fat (i.e., epididymal, visceral, and peritoneal fat) development, adipocyte hypertrophy, and the development of hyperinsulinemia and hyperlipidemia in a dose-dependent manner. Serum triglyceride and free fatty acid levels were greatly reduced by levan supplementation. Serum total cholesterol level was reduced, whereas the HDL cholesterol level was increased by dietary levan. The expression of uncoupling protein (UCP) was increased by dietary high fat, and was further induced by levan supplementation. The mRNA level of UCP1, 2, and 3 in brown adipose tissue (BAT) and UCP3 in skeletal muscle was upregulated in rats fed with dietary levan. In conclusion, upregulated UCP mRNA expression may contribute to suppression of development of obesity through increased energy expenditure. The present results suggest that levan supplementation to the diet is beneficial in suppressing diet-induced obesity and hyperlipidemia.
As the obesity has been known to be related with the hyperlipidemia, cardiovascular disease, cerebral apoplexy, fatty liver, and other chronic diseases, recent researches have focused on the functional food materials and their anti-obesity activities. This study was performed to study the effects of vanilloid family capsaicin, major pungent ingredient of hot chillies and peppers, on anti-obesity activities. ICR male mice were fed one of the pellet diet, basal diet, and high fat diet with capsaicin (45 $\mu\textrm{g}$/day) solution for 5 days. Mice in the corresponding control groups were given water for 5 days. In results, capsaicin reduced body weights in any diet groups. Percent weight and cell size of the abdominal white adipose tissue in mice on the high fat diet with capcaicin were significantly lower compared with those in mice on the high fat diet with water. However, percent brown adipose tissue weight per body weight in mice on the high fat diet was not affected by capsaicin. Capsaicin reduced the levels of s-triglyceride and s-total cholesterol in the pellet diet or high fat diet groups. There was no difference in the s-protein levels between the capsaicin group and the control water group. These data indicate that 1) orally administered capsaicin has a reducing effect on the blood triglyceride and total cholesterol levels, and 2) capsaicin has lowering effects on the body weight, percent weight and cell size of the abdominal white adipose tissue.
Effects of powdered siho(bupleuri radix) on serum and liver lipid composition and antioxidative capacity were investigated in rat fed high oxidized fat. Sixty male Sprague-Dawley rats weighing 161.25$\pm$2.51g were blocked into four groups according to body weight and raised seven weeks with basal diet(normal group), basal diet and 10% oxidized fat(control group), basal diet, 10% oxidized fat and 2% powdered siho(2% powdered siho group) and basal diet, 10% oxidized fat and 3% powdered siho(3% powdered siho group). Feed intake, body weight gain and feed efficiency were not significantly different among oxidized fat diet groups. The values of total cholesterol and triglyceride in plasma and liver showed no significantly different(p>0.05) in the oxidized fat diet groups. Plasma HDL-cholesterol showed atendency to increase in powdered siho diet groups. The values of thiobarbituric acid in plasma and liver were lower in the powdered siho diet groups than control group. Plasma GOT and GPT activity showed a tendence to decrease in powdered siho groups. Liver glutathione peroxidase activity showed a tendence to increase in powdered siho diet groups. Thus, it seems that powdered siho had some antioxidative elements.

This experimental study was designed to investigate the effects of Yukgunja-tang(YGJT) on the change of weight and serum total cholesterol, HDL-cholesterol, LDL-cholesterol, triglyceride, free fatty acid, total lipid, phospholipid level in obese mice induced by high fat diet. I fed normal group fed normal diet and administered DDW 0.2㎖ during 7 weeks, control group fed high fat diet and administered DDW 0.2㎖ during 7 weeks, sample A group fed high diet and administered YGJT 300mg/kg 0.2㎖ during 7 weeks, sample B group fed high diet and administered YGJT 500mg/kg 0.2㎖ during 7 weeks. The results were as follows ; 1. Sample A and Sample B were significantly decreased body weight(4weeks) and serum free fatty acid level in comparison with control group. 2. Sample A was significantly decreased body weight(7weeks), serum total cholesterol level and serum total lipid level, but significantly increased serum HDL-cholesterol level in comparison with control group. 3. Sample A was decreased serum LDL-cholesterol level, serum triglyceride level and serum phospholipid level in comparison with control group. 4. Sample B was increased serum HDL-cholesterol level in comparison with control group. 5. Sample B was decreased body weight(7weeks), serum total cholesterol level, serum LDL-cholesterol level, serum triglyceride level, serum total lipid level and serum phospholipid level in comparison with control group. According to above results, I suggest YGJT is able to be used for the herbal medication of obesity.
Objectives: This study was performed to evaluate the effects of fermented Samjung-hwan (SJH) extracts on weight, serum lipids and blood glucose. Methods: SJH was fermented using three different probiotic bacterial strains (Lactobacillus plantarum [LP], Leuconostoc mesenteroides [LM], Bifidobacterium longum [BL]) separately. Thirty-six rats were divided into normal, control (high fat diet), SJH-UF (high fat diet+unfermented SJH 200 mg/kg), SJH-LP (high fat diet+LP fermented SJH 200 mg/kg), SJH-LM (high fat diet+LM fermented SJH 200 mg/kg) and SJH-BL (high fat diet+BL fermented SJH 200 mg/kg). For 8 weeks later, we examined body weight, total cholesterol, high-density lipoprotein (HDL)-cholesterol and blood glucose. Results: The control group showed significantly increased weight gain compared with normal group and SJH-LP and BL groups had less weight gain than control group, significantly. In the lipid serum tests, control group showed significantly increased total cholesterol levels compared with normal group and only SJH-LP represented decreased total cholesterol levels compared with control group. However there was no significant change in the HDL-cholesteol levels. In the blood glucose tests, that of control group significantly incereased more than that of normal group, SJH-BL showed significantly decreased blood glucose levels compared with control group. Conclusions: SJH-LP, SJH-BL showed weight control effect, SJH-LP decreased TC and SJH-BL reduced blood glucose.

High-fat diet up-regulates either insulin resistance or triglycerides, which is assumed to be related to the expression of peroxisome proliferator-activated receptor (PPAR)-${\alpha}$ and PPAR-${\gamma}$. The beneficial effects of vitamin E on insulin resistance are well known; however, it is not clear if vitamin E with a high-fat diet alters the expression of PPAR-${\alpha}$ and PPAR-${\gamma}$. We investigated the effects of d-${\alpha}$-tocopherol supplementation on insulin sensitivity, blood lipid profiles, lipid peroxidation, and the expression of PPAR-${\alpha}$ and PPAR-${\gamma}$ in a high-fat (HF) diet-fed male C57BL/6J model of insulin resistance. The animals were given a regular diet (CON; 10% fat), a HF diet containing 45% fat, or a HF diet plus d-${\alpha}$-tocopherol (HF-E) for a period of 20 weeks. The results showed that the HF diet induced insulin resistance and altered the lipid profile, specifically the triglyceride (TG) and total cholesterol (TC) levels (P < 0.05). In this animal model, supplementation with d-${\alpha}$-tocopherol improved insulin resistance as well as the serum levels of TG and very-low-density lipoprotein-cholesterol (VLDL-C) (P < 0.05). Moreover, the treatment decreased the levels of malondialdehyde (MDA) in the serum and liver while increasing hepatic PPAR-${\alpha}$ expression and decreasing PPAR-${\gamma}$ expression. In conclusion, the oral administration of d-${\alpha}$-tocopherol with a high-fat diet had positive effects on insulin resistance, lipid profiles, and oxidative stress through the expression of PPAR-${\alpha}$ and PPAR-${\gamma}$ in a high-fat diet-fed male mice.
