Theoretical Studies for the Supercritical CO2 Solubility of Organophosphorous Molecules: Lewis Acid-Base Interactions and C-H···O Weak Hydrogen Bonding

  • Exploring the basic concepts for the design of CO2-philic molecules is important due to the possibility for “green” chemistry in supercritical CO2 as substitute solvent systems. The Lewis acid-base interactions and C?H…O weak hydrogen bonding were suggested as two key factors for the solubility of CO2-philic molecules. We have performed high level quantum mechanical calculations for the van der Waals complexes of CO2 with trimethylphosphate and trimethylphosphine oxide, which have long been used for metal extractants in supercritical CO2 fluid. Structures and energies were calculated using the MP2/6-31+G(d) and recently developed multilevel methods. These studies indicate that the Lewis acid-base interactions have larger impact on the stability of structure than the C?H…O weak hydrogen bonding. The weak hydrogen bonds in trimethylphosphine oxide have an important role to the large supercritical CO2 solubility when a metal is bound to the oxygen atom of the P=O group. Trimethylphosphate has many Lewis acid-base interaction sites so that it can be dissolved into supercritical CO2 easily even when it has metal ion on the oxygen atom of the P=O group, which is indispensable for a good extractant.

Measurement of solubility and miscibility of R-410A/POE oil mixture (R-410A/POE 오일 혼합물의 용해도 및 상용성 측정)

  • The solubility and miscibility measurement apparatus was developed and used to obtain data for refrigerant/oil mixture. The solubility and miscibility data for R-410A/68 ISO VG polyol ester (POE) oil mixture are obtained over the temperature range from -20 to $60^{circ}C\;with\;10^{\circ}C$ intervals and the oil concentration range from 0 to 90 wt%. Using the experimental data, an empirical model was developed to predict the solubility relations for R-410A/POE oil mixture at equilibrium. In the R-410A/Solest 68 oil mixture, the average root-mean-square deviation between measured data and calculated results from the empirical model is 3.4% and in the R-4104/EMKARATE RL 68H oil mixture, that is 2.86%. For R-410A/68 ISO VG POE oil mixture immiscibility was usually observed at the low oil concentrations(5~30 wt%) over the all experimental temperature range($-20~60^{circ}C$) and at the high oil concentrations(50~90 wt%) over the low experimental temperature range($-2O~0^{circ}C$).

Effect of Proteolytic Enzymes and Ginger Extract on Tenderization of M. pectoralis profundus from Holstein Steer

  • The effects of proteolytic enzymes (bromelain and bromelain+papain) and a ginger extract were assessed on collagen content and solubility, thermal shrinkage temperature of connective tissue, pH, cooking loss, drip loss, and Warner-Bratzler shear force (WBSF) of M. pectoralis profundus isolated from the beef brisket cut. Both proteolytic enzymes and ginger extract led to a significant increase in cooking loss and collagen solubility compared with untreated controls. On the other hand, the peak ($T_p$) thermal shrinkage temperature markedly decreased in all treatments compared with those in controls. Samples treated with bromelain, bromelain + papain, and ginger extract showed a significant decrease in WBSF by 36%, 40%, and 37%, respectively, compared with untreated controls. Our findings suggest that ginger extract are useful for post-mortem tenderization of meat containing high levels of collagen, compared to control even though, bromelain and bromelain + papain treatments have higher collagen solubility than ginger extract.

  • The synthesis and physicochemical properties of a novel cephalosporin, DWC-751 are described. DWC -751, (6R , 7R)-7-[ (Z)-2-(2-aminothiazol-4-yl)-2- methoxyiminoacetamido]-3-[(1-methylbenzotriazol-3-ium) methyl]-ceph-3-em-4-carboxylate monosulfate($IV_{\alpha}$) was conveniently obtained by the conversion of compound (IV) into the crystalline monosulfate. Adjusting pH 4.8-5.2 in aqeous solution of the crude crystalline, compound(IV) in the form of a crystalline pentahydrate was prepared with a high degree of purity. The influence of the various organic and inorganic acids on the solubility of compoud(IV) and its salts, was examined. Particularly, the solubility of DWC-751 was 92 mg/mι at pH 1.7 and 233 mg/mι at pH 3.0. DWC-751 showed a broad antimicrobial spectrum against gram-positive and negative bacteria.

Effect of Polyoxyethylene Alkyl Esters on Permeation Enhancement and Impedance of Skin

  • In this work, we have investigated the effect of polyoxyethylene alkyl ester nonionic surfactants on percutaneous permeation enhancement of a model drug, ketoprofen. We also investigated the mechanism involved in the enhancement using impedance and solubility measurement. Three groups of nonionic surfactants with different ethylene oxide content were studied. The permeation results showed that all surfactants enhanced the percutaneous absorption, irrespective of the molecular weight. The permeation results from PEG-45 monostearate (PEGMS45) were rather unexpected. Impedance and solubility results indicate that the mechanism involved in the enhancement of permeation by PEG-10 monooleate (PEGMO10) and PEGMS45 is rather different. The results from PEGMS45 suggest that it could be a potential candidate as a skin penetration enhancer with high molecular weight, which may poses less skin irritation and systemic side effect than the smaller surfactant molecules. Overall, this work provided some useful information on percutaneous transport enhancement and the mechanistic insights involved in skin permeation for these nonionic surfactants.

  • The stability and solubility of piroxicam in propylene glycol (PG), polyethylene glycol (PEC), and PG-water cosolvents have been studied by using high performance liquid chromatography. The degradation rate followed an apparent first-order kinetic and the reaction rate constants at 70, 80, and $90^{circ}C$ were determined. From these rate constants, the activation energy and the rate constant of piroxicam at $25^{circ}C$ in pure PG calculated by Arrhenius equation were 23.34 kcal/mole and $7.0\;{\times}\;10^{-4}\;day^{-1}$, respectively. Both of PG and PEG increased the solubility of the drug, but PEG was more effective.

  • The tensile properties, acetic acid solubility and degree of swelling in distilled water of cellulose/chitosan and sericin/chitosan film blended by mixing chitosan acetic acid solution with cellulose solution or sericin solution were investigated and the effect of crosslinking agent on solubility and degree of swelling were also considered. From the experimental results, the model of intermolecular bond is proposed. Tensile modulus of 100% cellulose film is high but the tensile strength and elongation are low. The elongation of 100% chitosan film is high but tensile modulus and strength is low. But it is possible to make film having same or higher tensile strength and modulus compared to that of 100% cellulose film by mixing cellulose and chitosan or by mixing sericin and chitosan. Chitosan is solved in 5vol % acetic acid solution but cellulose and sericin are not solved. Degree of swelling of chitosan in distilled water is higher than that of cellulose and sericin. Lower than 40wt% chitosan content, the solubility of cellulose/chitosan film in 5vol % acetic acid solution shows lower expected value but higher in case of sericin/chitosan film.

  • Alginate-chitosan blend containing coconut-based activated carbon was prepared as a drug delivery carrier in order to improve the loading and releasing capacity of the drug. The activated carbon was incorporated as effective adsorbent for drug due to the extremely high surface area and pore volume, high adsorption capacity, micro porous structure and specific surface activity. Alginate-chitosan blend containing coconut-based activated carbon showed the sustained release for a longer period. Alginate-chitosan blend showed higher release of drug as the pH increased and higher release of drug as the content of chitosan decreased due to the pH-dependent solubility of blend components.

Chitosan as a Flocculant: An Approach to Improve its Solubility for Efficient Harvesting of Microalgae

  • Chitosan is a promising flocculant for microalgae harvesting, but its scale-up application is not economically supported yet. Low solubility of chitosan in microalgae suspension demands high dosage (as a flocculant) to destabilize the cells, and thus, increases the cost of microalgae harvesting. This study identifies efficient solvents for the chitosan, and optimizes the concentration of solvents and chitosan dose to improve the harvesting efficiency. Chitosan was dissolved in different acids, and subsequently used as a flocculant. The flocculant efficacy was measured in terms of harvesting efficiency and reduction in chemical oxygen demand (COD) of the microalgae suspension. It was found that chitosan dissolved in 0.05 M HCl showed the highest harvesting efficiency ($89{\pm}0.87%$) at only 30 mg/L of dosage. In comparison, 270 mg/L of $FeCl_3{\cdot}6H_2O$ was required to attain $86{\pm}0.083%$ of the harvesting efficiency. $H_2SO_4$ dissolved chitosan required high flocculant dose (150 mg/L) and resulted in relatively low harvesting efficiency ($77{\pm}0.11%$). It was concluded that the efficacy of chitosan is solvent dependent, and the selection of proper solvent can decrease the dosage requirement for microalgae harvesting.

  • Some soluble species may not be solubility-limited or congruent-released with the matrix species. For example, during the operation of the nuclear reactor, the fission products can be accumulated in the fuel-cladding gap, voids, and grain boundaries of the fuel rods. In the waste package for spent-fuel placed in a geologic repository, the high solubility species of these fission products accumulated in the“gap”, e.g. cesium or iodine are expected to dissolve rapidly when ground water penetrates fuel rods. The time and space dependent mass transport for high solubility nuclides in the gap is analyzed, and its numerical illustrations are demonstrated. The approximate solution that is valid for all times is developed, and validated by comparison with an asymptotic solution and the solution obtained by the numerical inversion of Laplace transform covering the entire time span.
