Fermentative Water Purification based on Bio-hydrogen (생물학적 수소 발효를 통한 수처리 시스템)

  • Among various techniques for hydrogen production from organic wastewater, a dark fermentation is considered to be the most feasible process due to the rapid hydrogen production rate. However, the main drawback of it is the low hydrogen production yield due to intermediate products such as organic acids. To improve the hydrogen production yield, a co-culture system of dark and photo fermentation bacteria was applied to this research. The maximum specific growth rate of R. sphaeroides was determined to be $2.93h^{-1}$ when acetic acid was used as a carbon source. It was quite high compared to that of using a mixture of volatile fatty acids (VFAs). Acetic acid was the most attractive to the cell growth of R. sphaeroides, however, not less efficient in the hydrogen production. In the co-culture system with glucose, hydrogen could be steadily produced without any lag-phase. There were distinguishable inflection points in the accumulation of hydrogen production graph that resulted from the dynamic production of VFAs or consumption of it by the interaction between the dark and photo fermentation bacteria. Lastly, the hydrogen production rate of a repeated fed-batch run was $15.9mL-H_2/L/h$, which was achievable in the sustainable hydrogen production.

A Study on the Filtration of Swine Anaerobic Digestate Using Multi-Layered Compost Beds (다층구조의 퇴비단을 이용한 돈분뇨 슬러리 혐기소화액의 여과효과에 대한 연구)

  • The objective of this study was to verify whether SCB(Slurry Composting & Bio-filtration) system can be applied for the treatment of anaerobic digestion(AD) wastewater and also, to identify the most effective set among three filtration compost beds tested. Results can be summarized as these; (a) When AD wastewater was sprayed on the top of beds which were mainly composed of sawdust and/or other media and, subsequently, filtrates collected and analyzed, there were large drop in the values of Electric Conductivity(EC), Total Suspended Solid(TSS), Biochemical Oxygen Demand(BOD), and Chemical Oxygen Demand(COD). In contrast, Total Nitrgen(T-N) and Total Phosphorus(T-P) were progressively elevated. We consider these changes as positive if the filtrate are to be utilized as liquid fertilizer. (b) When three sets of filtration beds (T1, T2, T3) were compared for their effectiveness, no significant difference was found among them. These indicate that expensive sawdust can be replaced in part with cheaper media such as woodchip, rice husks, or others. (c) At early stage of operation (within 20 days), BOD in filtrates were maintained at high level probably due to the lack of microbial activity. During the same stage, T-N, T-P was at low level but, were elevated to higher levels thereafter. These data, when combined, indicate that the filtration system needs at least a couple of weeks for the optimized microbial functioning. (d) The temperatures of the experimental beds were progressively dropped as the experiment continued through the fall season, although filtration effectiveness was not noticeably influenced.

Method of Reducing Separation Membrane Fouling Using Microbubbles (마이크로버블을 이용한 분리막 파울링 저감방법)

  • Due to water shortages caused by water pollution and climate change, total organic carbon (TOC) standards have been implemented for wastewater discharged from public sewage treatment facilities. Furthermore, there is a growing interest and body of research pertaining to the reuse of sewage treatment water as a secure alternative water resource. The membrane bio-reactor (MBR) method is commonly used for advanced wastewater treatment because it can remove organic and inorganic ions and it does not require or emit any chemicals. However, the MBR process uses a separation membrane (MF), which requires frequent film cleaning due to fouling caused by a high concentration of mixed liquor suspended solid (MLSS). In this study, process improvement and microbubble cleaning efficiency were evaluated to improve the differential pressure, water flow, and MF fouling, which are the biggest disadvantages of operating the MF. The existing MBR method was improved by installing a precipitation tank between the air tank and the MBR tank in which raw water was introduced. Microbubbles were injected into a separation membrane tank into which the supernatant water from the precipitation tank was introduced. The microbubble generator was operated with a 15 day on, 15 day off cycle for 5 months to collect discharged water samples (4L) and measure TOC. As the supernatant water from the precipitation tank flowed into the separation membrane tank, about 95% of the supernatant water MLSS was removed so the MF fouling from biological contamination was prevented. Due to the application of microbubbles to supernatant water from the precipitation tank, the differential pressure of the separation membrane tank decreased by 1.6 to 2.3 times and the water flow increased by 1.4 times. Applying microbubbles increased the TOC removal rate by more than 58%. This study showed that separately operating the air tank and the separation membrane tank can reduce fouling, and suggested that applying additional microbubbles could improve the differential pressure, water flow, and fouling to provide a more efficient advanced treatment method.

Developments of Water Treatment System by Biological Fluidized Bed for Water Reuse Aquaculture (생물학적 유동층을 이용한 어류양식 순환수의 처리씨스템 개발)

  • The experimental study was made to propose the treatment method of wastewater in the high-density fish culture system. The BOD to COD ratios of effluents were almost same to 0.65 in the eel-farm, but were various in the farm rearing together with tilapia etc. A BOD rate curve of the eel-farm effluent could be described mathematically by the equation, $BODu=14.1(1-10^{-0.222t})+30.9(1-10^{-0.035(t-8)})$. Nitrification in Biological Fluidized Bed(BFB) system to treat the fish-farm wastewater could be reduce ammonium level up to $65{\sim}79\%$ when ammonium loading rates were between 0.014 and 0.075g $NH_4/g$ BVS-day. Nitrification efficiency was decreased by organic matters in the wastewater when ammonium loading was low(0.014 g $NH_4/g$ BVS-day). T-N removal ratios were decreased to increase loading in denitrification process, because of low C/N ratio. Based on much higher biological mass concentrations, BFB system takes many advantages of a practical viewpoint, such as stability of treatment efficiency and reduction of necessary site area for the facility, as compared with conventional treatment systems.

Characteristics of Ammonia in Alkaline Stabilization Facility of Sludge from Sewage Treatment Plant (하수처리오니 알칼리 안정화 처리시설에서의 암모니아 발생특성)

  • The characteristics of ammonia generated from alkaline stabilization facilities was investigated which are for organic sewage sludge from wastewater treatment plants. The highest concentration of ammonia was found in mixing and curing process in alkaline stabilization facility and ammonia mainly showed a range of 87.78 ppm($66.62mg/m^3$) to 1,933 ppm($1,467.01mg/m^3$) by detection tube. This is presumed to occur because nitrogen oxides are converted into ammonia as the sewage sludge is mixed with lime. In some facilities, hydrogen sulfide and methyl mercaptan were detected in relatively high concentrations, but odor materials except ammonia were not detected in most of the facilities. The concentration of ammonia caused by process was generally high in the order of "mixing > curing > output > storage > drying > input." It was found that odor compounds are removed by wet absorption using sulfuric acid and sodium hypochlorite in the 5 alkaline stabilization facilities currently in operation. Each facility was designed to meet the concentration of after-treatment emission in 1 ppm($0.76mg/m^3$), 50 ppm($37.95mg/m^3$) or 100 ppm($75.89mg/m^3$), but no facility satisfied the design standard for their emssion limit. In case of ammonia, some workplaces in alkaline stabilization facilities exceeded the exposure limits established by the Ministry of Labor. It appears that proper ventilation should be provided for the safety of workers in future. No odor compound including ammonia was found by detection tubes in the border of the facilities, but trace amounts of odor compounds are expected to exist, given the current operational status of facilities.

Isolation of High Yielding Alkaline Protease Mutants of Vibrio metschnikovii Strain RH530 and Detergency Properties of Enzyme

  • Abstract A facultative alkalophilic gram-negative Vibrio metschnikovii strain RH530, isolated from the wastewater, produced several alkaline proteases (VAP) including six alkaline serine proteases and a metalloprotease. From this strain, high yielding YAP mutants were isolated by NTG treatment. The isolated mutant KS1 showed nine times more activity than the wild-type after optimization of the culture media. The production was regulated by catabolite repression when glucose was added to the medium. The effects of several organic nitrogen sources on the production of the YAP were investigated to avoid catabolite repression. The combination of 4% wheat gluten meal (WGM), 1.5% cotton seed flour (eSF), and 5% soybean meal (SBM) resulted in the best production when supplemented with 1% NaCl. The YAP showed a resistance to surfactants such as $sodium-{\alpha}-olefin$ sulfonate (AOS), polyoxy ethylene oxide (POE), and sodium dodecyl sulfate (SDS), yet not to linear alkylbenzene sulfonate (LAS). However, the activity of the YAP was restored completely when incubated with LAS in the presence of POE or $Na_2SO_4$. The YAP was stable in a liquid laundry detergent containing 6.6% SLES (sodium lauryl ether sulfate), 6.6% LAS, 19.8% POE, and stabilizing agents for more than two weeks at $40^{\circ}C$, but the stability was sharply decreased even after 1 day when incubated at $60^{\circ}C$. A washing performance test with the YAP exhibited it to be a good washing power by showing 51 % and 60% activity at $25^{\circ}C{\;}and{\;}40^{\circ}C$, respectively, thereby indicating that the YAP also has a good detergency at a low temperature. All the results suggest that the YAP produced from the mutant strain KSI has suitable properties for use in laundry detergents.rgents.

The Measurement of Biochemical Methane Potential in the Several Organic Waste Resources (유기성 폐자원별 메탄 생산 퍼텐셜 측정 연구)

  • This research studied the bio-methane potential of several waste biomass materials as alternative sources for biogas production, and the laboratory procedure for measuring the biochemical methane potential was described. The wastes from four agro-industries (sewage, livestock, food wastewater treatment sludge and cattle rumen substance generating in slaughter house) were evaluated as substrates for the assay of biochemical methane potential. In order to estimate the ultimate methane yield, two empirical equations (modified Gompertz equation and exponential equation) was investigated. The ultimate methane yield of sewage, livestock, food sludge and lumen substance estimated by the modified Gompertz equation were 0.086, 0.147, 0.146, and 0.121 L $CH_{4}\;g^{-1}\;VS_{added}$, respectively. The ultimate methane yield estimated by the exponential equation were 0.109, 0.246 and 0.174 L $CH_{4}\;g^{-1}\;VS_{added}$ in sewage, livestock sludge and lumen substance. And the ultimate methane yield estimated by the exponential equation showed more high values in the range of 26.7 ~67.3% than the ultimate methane yield estimated by the modified Gompertz equation.

Behavior of Soluble Microbial Products in a Submerged Membrane Separation Activated Sludge Process (침지형 막분리 활성오니법에 있어서 생물대사성분의 거동)

  • A laboratory-scale experiment was conducted to investigate the effect of soluble microbial products(SMP) on permeate flux in the submerged membrane separation activated sludge process. Continuous and batch filtration test were operated to understand mechanism of relationship between membrane fouling and SMP. Synthetic wastewater(phenol) was used as a carbon source. Hydraulic retention time(HRT) and mixed-liquor volatile suspended solids(MLVSS) of the reactor were kept at 12 hours and 9.000mg VSS/L, respectively. Batch filtration tests ($J_{60}/J_o$) using the mixed liquor from reactor showed that the increase of accumulated SMP concentration in the reactor caused to the decreasing permeate flux and the increasing of the adhesion matters which form cake and gel layer. The resistance value of cake layer was measured $2.9{\times}10^{10}{\sim}4.0{\times}10^{10}(1/m)$, this value showed more significant effect on flux drop than that of among other resistance layers. Batch phenol-degradation experiment was conducted to observe SMP type expected $SMP_{nd}$ and $SMP_{e}$ (SMP resulted from endogenous cell decomposition), these are non-biodegradable high molecular weight organic matter and playa significant role in permeate flux drop. Also, SMP concentration was accumulated as increased of HRT against flux drop.

1,3-Propanediol Fermentation using the by-Products from Fat Industry (글리세롤을 함유한 유지산업 부산물의 1,3-propanediol 발효)

  • 1,3-Propanediol as a bifunctional organic compound could be used in polymerization reactions producing polyesters and polyurethanes. Byproduct containing high concentration of glycerol from fat industry was used to produce 1,3-propanediol in lower production cost as well as waste treatment. In this study, various attempts were made to increase 1,3-propanediol production under different conditions using Klebsiella pneumoniae ATCC 15380. The conversion yield and byproduct formation were influenced significantly by the fermentation pH and temperature. The optimal glycerol and nitrogen concentration for 1,3-propanediol production were found to be 25 a/L and 1%(w/v), respectively. The formation of 1,3-propanediol was optimal at pH 6.0 and temperature $35^{\circ}C$. 1,3-Propanediol production from byproduct from 2.5% glycerol was lower than that of 2.5% commercial glycerol and amounted only to 9.84 a/L from byproduct, while to 12.13 a/L from commercial glycerol.

A Study on the Odor Characteristics and Identification of Microbial in Biological Swine Manure Treatment Process by Bioreactor (생물반응기를 도입한 돈분뇨의 생물학적 처리공정에서 악취발생 특성 및 미생물동정에 관한 연구)

  • A bioreactor filling with pellets and stones was equipped to the swine manure treatment system, which is expected to emit high concentration of odor in the process of the organic wastewater treatment system, and in comparison with the activated sludge process as the control process, the reactor operation state, treatment water quality and odor emission concentration were measured. The reactor using the bioreactor proved to be much more stable in the bubble condition, treatment water transparency, etc, and BOD removal efficiency was also much better. The removal efficiency of T-N and T-P, however, showed little difference in the two reactors. Odor, as a result of examining $NH_3-N$, $NH_3$ concentratio, and complex odor, was 4 times to 24 times less emitted in the system using bioreactor than in the activated sludge system. $H_2S$, methyl mercaptan, dimethyl sulfide, and dimethyl disulfide were not found or were found in only 5 ppbs in each reactor and showed little difference between the two reactors. In the bioreactor process, Bacillus sp./ Pseudomonas sp. species were mainly found and in the activated sludge process, acterium sp. Chryseobacterium sp. species were mainly found.
