고 불포화 지방산 식이가 인체 혈청 Cholesterol의 Esterification과 Transfer에 미치는 영향 (Effects of High Polyunsaturated Fat Diet on Human Plasma Cholesterol Esterification and Transfer)

  • Lecithin Cholesterol Acyltransferase(LCAT)와 cholesteryl ester transfer protein(CETP)은 간과 간외조직간의 혈청 cholcsterol 항상성을 유지하는데 중요한 기전인 reverse cholestrol transport system(RCT)에 매우 중요한 인자들이다. 본 연구의 목적은 RCT 기전의 식이 지질 효과를 추정하여 항 고지혈증 치료식이 연구의 중요한 자료를 제공하는데 있다. 4주의 인체대사 실험에서, 새당사 12명의 여성들은 실험 1주 전에 고 포화지질로써 적응시기를 거친 후, 각각 6명씩 high PUFA(corn oil)군과 high SFA(butter)군에 무작위로 배정되었다. Butter군은 total-34(%), esterified-(96%), $HDL_3$-(23%), LDL-(20%) 및 VLDL+LDL-(35%) cholestetol을 감소시켰다. Corn oil군은 esterified(2.5%) 및 LDL-(15%) cholesterol과 triglycerde(27%)를 감소시켰다. Corn oil과 butter fat군간의 식이 효과 차이는 total-(p=0.0001), esterified-(p=0.0001), total HDL-(p=0.005), $HDL_2$-(p=0.01) 및 LDL-(p=0.0001) cholesterol에서 유의적이다. LCAT activity는 두 군에서 변화가 없으나, 4주 후 CETP activity는 butter군에서 2.5배 증가하였다. 이는 VLDL+LDL cholesterol 농도가 butter군에서 증가한 결과와 일치한다. LCAT activity는 corn oil군에서 증가된 HDL의 apo A-I 농도와 free cholesterol과 정의 상관관계가 높은 반면, CETP activity는 total cholesterol과 LDL 및 VLDL+LDL cholesterol과 정의 상관관계가 높았다.

쥐에서 식이 Calcium이 대장 암화관정의 세포증식과 대장점막의 Eicosanoid 및 1,2 -diacylglycerol 수준에 미치는 영향 (Effect of Dietary Calcium on Cell Proliferation and Colonic Mucosal Levels of Eicosanoid and 1,2-diacylglycerol in Colon Carcinogenesis of Rats)

  • The objective of this study was to observe the effect of dietary calcium(Ca) level on colonic mucosal levels of cell proliferation, 1, 2-diacylglycerol(DAG), TXB2, PGE2 and phospholipid fatty acid composition which have been known as biomarkers for colon cancer. One hundred male Sprague Dawley rats, at 7 weeks of age, were divided into two fat type groups. Each group of which was further divided into two Ca level groups. Each rt was intramuscularly injected with 1, 2,-dimenthylhydrazine(DMH) for 6 weeks (total dose of 180mg/kg body weight) and simultaneously fed one of four experimental diets containing 15% dietary fat(corn oil or perilla oil )and 0.3% or 1.0% Ca by weight for 20 weeks. Compared to corn oil, perilla oil significantly reduced cell proliferation by decreasing labeling index, proliferating zone, crypt length in colonic mucosa and colonic mucosa and colonic mucosal levels of DAG, TXB2 . PGE2 and phospolipid (PL) arachidonic acid distribution. The effect of Ca on biomarketrs was different depending on the type of dietary fat comsumed . Ca effect of Ca on biomarkers was different depending on the type of dietary fat comsumed. Ca effect was not significantly shown in the PO group, but it was significant in the CO group in which high Ca(1.0%) decreased the levels of levels of PL-C20 : 4(%), DAG and PGE2 . However , high Ca supplementation had shown only the trends of improving cell proliferation. Overall , high dietary Ca significantly reduced cell proliferation by inhibiting the synthesis of eicosanoid and DAG with reduced distribution of PL-C20 : 4 , which may have resulted in lower activation of PKC through reduced signal transduction. Since a high level of dietary Ca was more effective in reducing the risk factor against colon cancer in corn oil fed rats, it could be suggested that a higher amount of dietary Ca be consumed , especially when more vegetable oil rich in linoleic acid is included in the diet.

식이지방의 종류와 수준에 따라 쥐의 혈장과 조직의 Tocopherol 및 지질과산화상태에 미치는 영향 (Effect of Quality and Quantity of Dietary Fats on the Status of Tocopherol and Lipid Peroxidation of Plasma and Tissue in Rats)

  • The study was to compare the effect of dietary fatty acids on fatty acid profile in tissue and the status of tocopherol and lipid peroxidation, and superoxide dismutase and glutathione peroxidase activities at two fat levels. Male Sprague Dawley rats weighing average 350g(17 weeks) were fed either low fat(LF, 4.3% w/w, 10% kcal) or high fat(HF, 20.8%, w/w, 40% kcal)diet for 6 weeks. The fats used were beef tallow as a source of saturated fatty acid, corn oil for n-6 linoleic acid, perilla oil for n-3 $\alpha$-linolenic acid and fish oil for n-3 eiocosapentatenoic acid(EPA) and n-3 docosahexaenoic acid(DHA). Palsma tocopherol was significantly reduced by fish oil compared to beef tallow at body fat level. However, there was no significant effect on the levels of plasma MDA, RBC MDA and tocopherol, and RBC hempolysis by the type and amount of dietary fat. The peroxidizibility index of fatty acid profile in plasma and liver was increased and liver MDA level was significantly increased by fish oil when dietary fat level was increased. The activities of SOD and GSHPx tended to be increased by perilla oil and fish oil at both fat oil significantly reduced the incorpration of c20:4 and increased the incorporation of c20:5 into liver compared to corn oil. The incorporation of n-3 fatty acids into tissue by perilla oil rich in $\alpha$-linolenic acid was significantly higher tan corn oil and its effect was improved with higher amount of perilla oil in diet by high fat diet. Overall, the lipid peroxidation of tissue could be prevented by tocopherol supplementation when dietary fat level was low in diet. However, at high fat diet, tocopherol supplementation might not be enough to prevent the lipid peroxidation in tissue since the potential for lipid peroxidation was tended to be increased with higher incorporation of higher unsaturated n-3 fatty acids into tissue. Therefore, it could not be recommended to consume large amount of fish oil even with excess amount of tocopherol supplemented to the high fat diet.

Increase in Plasma HDL-Cholesterol Concentration in Goats Fed Sesame Meal Is Related to Ether Extract Fraction Included in the Meal

  • Previously, we reported that a diet including sesame meal (SM) increased plasma total and high-density lipoprotein (HDL)-cholesterol concentrations in goats. In the present study, the components in the sesame meal that can increase plasma total and HDL-cholesterol concentrations have been examined. In experiment 1, we gave goats defatted sesame meal diet (DSM) to investigate the influence of ether extract fraction remained in sesame meal. Corn gluten meal diet (CGM) was also fed to goats as a high-protein diet to examine the influence of high dietary protein level caused by usage of sesame meal. Plasma total and HDL-cholesterol concentrations of goats fed DSM and CGM did not change during experimental periods though they were elevated by feeding SM. In experiment 2, the influence of sesame oil and corn oil added in diets on plasma total and HDL-cholesterol concentrations in goats was investigated. Plasma total and HDL-cholesterol concentrations were increased by feeding both corn oil diet and sesame oil diet. In conclusion, the increase in plasma HDL-cholesterol concentration by feeding sesame meal was resulted by the effect of ether extract fraction including sesame oil or some lipid-soluble components remained in sesame meal.

어유와 비타민 E 보강 수준이 쥐간의 전암성 병변에 미치는 영향 (High Vitamin E Supplement is Needed to Have an Anticarcinogenic Effect of Fish Oil)

  • The influences of fish oil and different levels of vitamin I supplement on hepatocellular chemical carcinogenesis have been studied. Male Sprague-Dawley rats received diethylnitrosamine (DEN)(200mg/kg body weight) and were subjected to two-thirds partial hepatectomy to induce murine chemical hepatocarcinogenic procedure. Placental glutathione S-transferase(GST-P) positive foci area, antioxidant enzymes(Cu/Zn-superoxide dismutase(SOD), catalase, glutathione reductase (GR), total- glutathione peroxidase (TGPx), glutathione S -transferase (GST)), glucose 6-phosphatase (G6Pase) activities, and lipid peroxidation of microsomes(thiobarbituric acid reactive substances (TBARS)) were measured. Experimental animals were fed 15% corn or fish oil with 0, 40, 1,000, 10,000IU vitamin E /kg diet for 8 weeks. Vitamin E supplements decreased the area of GST-P positive foci in both groups. The higher the vitamin E levels, the smaller the area of GST-P positive foci were noticed. Compared to 0 IU vitamin E, 40 IU in corn oil and 1,000 IU in fish oil groups were effective in decreasing G57-P positive foci area. Fish oil groups tended to have smaller area of GST-P positive foci. fish oil groups showed lower body weight, lower activities of Cu/Zn-SOD and TGPx, higher TBARS contents, higher activities of GST, catalase, G6Pase, GR and higher liver/body ratio than corn oil groups. As the level of vitamin I increased, GST-P positive foci count, catalase activities, and TBARS tended to decrease. G6Pase activities tended to increase in both groups. At higher vitamin E levels, GST activities tended to decrease in fish oil groups. These results suggest that vitamin I has suppressive offects on hepatocellular chemical carcinogenesis probably through antioxidant eH:cts decreasing TBARS contents, $H_2O$$_2$, and organic peroxides. fish oil tended to have greated suppressive offects than corn oil on hepatocellular carcinogenesis. Influence of Xanthan, Emulsification Temperature, and Environmental Stresses on the Preparation of Water-in-Corn Oil Emulsions Droplets Coated by Polyglycerol Polyricinoleate

  • The purpose of this study was to prepare stable water-in-corn oil (W/O) emulsion droplets coated by polyglycerol polyricinoleate (PGPR). W/O emulsions (20 wt% aqueous phase, 80 wt% oil phase containing 8 wt% PGPR) were produced by high pressure homogenization (Emulsions 1), however, appreciable amount of relatively large water droplets (d>$10{\mu}m$) were found. To facilitate droplet disruption, viscosity of each phase was adjusted: (i) increased the viscosity of aqueous phase by adding 0.1 wt% xanthan (Emulsions 2); (ii) decreased the viscosity of oil phase and aqueous phase by heating them separately at $50^{\circ}C$ for 1 hr immediately before emulsification (Emulsions 3). Homogenizing at the elevated temperature clearly led to a smaller water droplet size, whereas xanthan neither improved nor adversely affected on the microstructures of the emulsions. In addition, the Emulsions 3 had good stability to droplet aggregation under shearing stress, thermal processing, and long term storage.

옥수수 기름의 제조공정별 벤조피렌 함량 변화 (Changes in Benzo(a)pyrene Content During Processing of Corn Oil)

  • 옥수수기름의 제조공정 단계별 공정유 및 이 과정에서 발생되는 부산물에 함유되어 있는 B(a)P 함량을 측정하고 이의 제거방안에 대하여 연구하였다. 옥수수기름에서 높은 수준의 B(a)P가 검출되는 원인은 옥수수에 함유되어 있는 B(a)P의 80% 이상이 옥수수 배아에 집중적으로 분포하는데 따른 현상이었다. 이로부터 얻어진 원유를 일반적인 정제공정을 거칠 경우 최종 탈취유에서는 $2.15{\mu}g$/kg가 검출 되었다. 따라서, 옥수수기름에서 일반적인 정제공정에 의해서는 현행 우리나라의 식용유 중 B(a)P 함유량 $2.0{\mu}g$/kg 이하의 법적 규격을 충족시키기 어려움을 쉽게 알 수 있었다. 이에 탈색공정에서 사용하는 2%(w/w) 내외의 산성백토 대신 산성백토:활성탄소=90:10(w/w)의 혼합분을 처리하여 $0.09{\mu}g$/kg 수준의 탈취유를 얻을 수 있었다. 이 때, 활성탄소의 처리량은 산성백토 대비 10% (w/w) 수준이 적정하였으며, 이의 증가에 따른 B(a)P 제거율 증가는 기대하기 어려웠다. 처리하는 활성탄소의 입도가 B(a)P 제거에 미치는 영향이 컸는데, $50{\sim}100$ mesh 수준의 입도를 갖는 것이 최적의 조건임을 확인하였다.

  • The study was designed to observe the effect of dietary calcium and fats on plasma cholesterol level, hepatic microsomal fluidity and HMG-CoA reductase activity as well as the excretion of fecal bile acids and neutral sterols in 1, 2-dimethylhydrazine(DMH)-treated rats. Male Sprague Dawley rats, at 7 weeks of age, were divided into 2 groups, 0.3% and 1.0% Ca levels and each group again subdivided into 2 groups of corn oil and perilla oil. Each rat was intramuscularly infused with DMH for 6 weeks to give total dose of 180mg/kg body weight and also fed experimental diet containing 15%(w/w) different fit and Ca(0.3% or 1.0%) for 20 weeks. High dietary calcium(1.0%) did not significantly influence on plasma cholesterol as well as hepatic microsomal fluidity and HMG CoA reductase activity, but significantly reduced the excretion of total bile acid per gram of faces and increased the excretion of total neutral sterol. However, high dietary Ca reduced the excretion of secondary bile acid(deoxycholic and lithocholic acids) which was known as promoter for colon cancer. Perilla oil rich in n-3 ${\alpha}$-linolenic acid significantly decreased plasma cholesterol by increasing hepatic microsomal fluidity compared with corn oil, but did not influence on HMG CoA reductase activity. Perilla oil did not influence on fecal excretion of total and primary bile acids, but reduced the excretion of secondary bile acids. Therefore, it could be recommended to consume more fish product and food rich in calcium and use more perilla oil in meal preparation to prevent from coronary hear disease and colon cancer especially when high fit diet has been practiced. 양파 메탄올 추출물의 생리활성 및 항산화효과 (Physiological Activity and Antioxidative Effect of Methanol Extract from Onion (Allium cepa L.))

  • Onion was extracted by methyl alcohol and some physiological activities and antioxidative effect of the extracts on soybean oil, corn oil, lard and shortening were compared with commercial antioxidants such as $\alpha$-tocopherol and BHT. The electron donating ability was about 70% at concentration above of 30mg/mL. Nitrite scavenging effects of all concentrations diminished at higher pH, while in the case of pH 1.2, it showed a nitrite scavenging effect of more than 80% af concentration above of 30mg/mL. And also, the angiotensin I-converting enzyme inhibitory effect was good above 80%. The soybean oil, corn oil, lard and shortening containing different levels of the onion extract were stored at 6$0^{\circ}C$ to evaluate the antioxidative activity of onion extracts. Peroxide value, acid value and TBA value of each oil were detected. Onion extract was very effective to retard oxidation of soybean oil, corn oil and lard, and especially, the very high antioxidative effect was observed for lard.

  • Triglyceride compositions of cottonseed, soybean, sesame, perilla corn and rapeseed oils have been determined by a high performance liquid chromatographic analysis. An optimum condition was obtained by using a ALC/GPC 244 type, from Waters Association, Japan with $\mu$ Bondapak $C_{18}(1/4^{'}\times1^{'})$ column. A similar distribution pattern of triglycerides was found in cottonseed, soybean, sesame, rapeseed and corn oils. It was noted that $C_{40}$, $C_{42}$ and $C_{44}$ were the major components in these seed oils, except perilla oil. The results showed that contents $O_{40}-C_{48}$ triglyceride types in cottonseed, sesame and corn oil were within $2.23{\sim}41.24%$ and $C_{38}-C_{48}$ triglyceride types in soybean oil were within $3.01{\sim}10.02%$ and $C_{34}-C_{46}$ triglyceride types in rapeseed oil were within $2.38{\sim}28.68%$.
