Antioxidant Activity of Ripe Fruit Components of Rubus coreanus: Extraction Using Porous Polymer Resins (다공성 합성흡착제를 이용한 복분자 딸기(Rubus coreanus) 농축액의 항산화 효과)

  • This study was carried out to develop a method of removing unnecessary sugars and high-molecular weight soluble components from water extract by using various highly porous polymer resins: these resins are widely used as adsorbents for polyphenolic compounds. Each anthocyanin-rich fraction (ARF) that was eluted from column packed with 4 different resins (SEPABEADS SP207, 700, 850 and Diaion HP 20) was obtained from an aqueous extract of the ripe fruits of Rubus coreanus, which is well-known in Korea as "Bok-bun-ja". Among the above mentioned resins, Diaion HP20 had the highest efficacy and provided maximum yield of the ARF. The ARF eluted from the column packed with HP 20 exhibited the strongest antioxidant activity in vitro. Bioactive phytochemicals extracted from Rubus coreanus were investigated using the on-line HPLC-$ABTS^{{\cdot}+}$ system, and the resulting 6 peaks were identified as radical-scavenging components. By using liquid chromatography-mass spectrometry, 5 peaks were obtained, and these were identified as cyanidin-3-sambubioside, cyanidin-3-glucoside, cyanidin-3-xylosylrutinoside, cyanidin-3-rutinoside and pelargonidin-3-rutinoside. These results indicate that the use of appropriate porous resin (Diaion HP 20) leads to an increase in the yield of bioactive components and enhancement of their biological properties.

  • Biogas yields point of view, the possibility of reusing excess sludge treated by thermal pretreatment for the purpose of improving the efficiency of the anaerobic digestion process has been investigated in recent year. Thermal pretreatment technology was considered as a pretreatment technique to improve excess sludge properties because of the solubilization of particulate organics. As a view point of sludge reduction and recycle, however, many researchers focused on the ability of particulate hydrolysis and COD solubilization under a high temperature, and few reports have addressed on the physical/chemical characteristics changing. This research was performed to evaluate the effects of a various temperature and chemical additives on carbon formation and fractionation in treated slurry from thermal pretreatment. Based on the results, it was revealed that oxidants injection and temperature changing have significantly caused the change of carbon fractions in slurry from thermal pretreament. Especially, it was considered that the efficiencies of particle hydrolysis increased with the increase of the reaction temperature. Low molecular weight(Mw < 350 g/mol) organic carbon formation increased with the increase of oxidants injection. It was expected that results of this research will provide an overview of the characteristics of thermal pretreatement for excess sludge reduction and recycle.

Isolation of Anticarcinogenic Isoflavone-conjugated Glycoproteins from a Submerged Liquid Culture of Agaricus blazei Mycelia by the Autolysis Process (신령버섯균사체 액체배양물의 자가분해에 의한 항암성 isoflavone-conjugated glycoprotein 분리)

  • Most beta-glucans obtained from various fruit bodies of mushrooms and mushroom mycelial cultures have high-molecular weight glycoproteins, conjugated with beta-glucans. We report that isoflavone-conjugated glycolproteins (designated as gluvone) were isolated and exhibited stronger anticarcinogenic activities. Agaricus blazei mycelia (ABM) was cultured in a liquid medium containing soybean flakes for 14 days. The liquid culture was autolyzed by incubating at $53^{\circ}C$ (pH 5.5) for 3 h. A crude glycoprotein (CGP) fraction with a cytotoxic effect on a mouse ascite cancer cell line (S-180) and a human breast cancer cell line (MCF-7) was isolated from the autolyzed ABM cultures by 80% ethanol treatment. Gluvone was isolated from the CGP with Sephadex G-75 column chromatography. It exhibited a stronger anticancer effect than CGP against the S-180 cell-induced female ICR mouse ascites carcinogenesis. Gluvone with 9,400 daltons was identified as a glycoprotein conjugated with isoflavone. According to HPLC and GC analysis, in conjunction with $^1H$-NMR spectral analysis, it contained 60% carbohydrates (glucose, fructose, and ribose), 31% protein, and 2% isoflavone (daidzein and genistein), which is a novel material. These results indicate that a strong anticarcinogenic gluvone was isolated from the autolyzed product of a submerged liquid culture of ABM, suggesting that autolysis could be a useful tool to produce antitumor agents.

  • An one percent sodium carbonate extract prepared from sclerotia of Poria cocos activated the proliferation of the T lymphocytes as measured by mixed lymphocyte responses(MLR). The active fraction, PCSC22, was isolated from an one percent sodium carbonate extract by a combination of fractionation procedures, including ethanol precipitation and chromatographies on column of DEAE-cellulose and Sephadex G50. Carbohydrate and peptide contained in PCSC22 were 78 : 22% in ratio. On employing gel filtration high performance liquid chromatography, PCSC22 exhitited a homogeneous peak with an average molecular weight of 8 kDa. The sugar moiety of PCSC22 was composed with mannose (92%), galactose (6.2%) and arabinose (1.3%), which might be indicated as heteromannan. Fifteen amino acids were found in peptide moiety of the polysaccharide and aspartic acid, serine, and valine were major components. PCSC22 activated the primary proliferation of T lymphocytes measured by mixed lymphocyte responses, the antibody production of the B lymphocytes and the secretion of nitric oxide from macrophage cell line, RAW264.7.

Development and Full-scale Application of the Alternative Carbon Source Based on the Substrate Compatibility (미생물 순응 호환성에 기반한 대체탄소원 개발 및 실용화 사례)

  • J sewage treatment plant (WWTP) in Busan has used methanol as an external carbon source for the biological denitrification process. Methanol is widely used. but rather expensive and very dangerous in handling. Therefore, it has been required that the economic alternative carbon source must be developed. By-product from a fine chemical industry can be Purified by removing high molecular weight substances using the ultrafilter membrane separation process and RBDCOD fraction becomes $98{\sim}99%$ of COD substances in the purified by-product. The purified by-product containing three types of alcohols, methanol, prophylenglycol and methoxypropanol; showed similar chemical characteristics to the methanol, a main external carbon source, in biodegradation pathway. Shown above, the compatibility between main and alternative carbon sources has been achieved. Also very short or no adaptation period is necessary in the case of exchanging these carbon sources. The compatibility between external carbon sources is an essential element for stabilizing WWTP operations. During the full-scale application test of the by-product, the alternative carbon source line got on par with the treatment efficiency of the methanol line. With the test result, J-WWTP changed methanol to a fine chemical by-product, in two out of three J-WWTP lines. Moreover, it is expected that 55.4% of the external carbon source cost reduction can be achieved in the alternative carbon source applied lines.

Antibacterial and Antibiofilm Activities of Diospyros malabarica Stem Extract against Streptococcus mutans (Streptococcus mutans에 대한 인도감나무 줄기 추출물의 항균활성 및 생물막 형성 억제 효과)

  • The objective of this study was to evaluate the potential of Diospyros malabarica stem extract, a natural materials, in oral health material. With this aim in mind, thin layer chromatography (TLC), TLC-bioautography, high-performance liquid chromatography (HPLC), electrospray ionization-mass spectrometry (ESI-MS), scanning electron microscopy (SEM), and real-time qPCR were performed. The antibacterial activity of D. malabarica stem extract against Streptococcus mutans KCTC3065 was confirmed in an n-hexane fraction with low polarity. The molecular weight of the antibacterial compound was estimated to be 188 by ESI-MS analysis. The inhibitory effects of the extract on biofilm formation and gene expression related to biofilm formation of S. mutans were determined by SEM and real-time PCR analysis. The extract inhibited the formation of S. mutans biofilms at D. malabarica stem extract concentrations of 1 mg/ml, as shown by SEM. The real-time PCR analysis showed that the expression of the gtfC gene, which is associated with biofilm formation, was significantly decreased in a dose-dependent manner. Based on the above results, it can be concluded that D. malabarica stem extracts, a natural materials, can be used in oral health products to suppress the formation of biofilms by inhibiting tooth adhesion of S. mutans, a causative agent of dental caries.

Discrimination and Hordein Polypeptide Patterns of Malting Barley Varieties Using UPLC (UPLC 분석을 이용한 맥주보리 품종의 호데인 단백질 분석 및 품종 판별)

  • Ultra-performance liquid chromatography (UPLC) was used to assess the hordein protein fraction of malting barley. C-hordeins (barley prolamins) were extracted with 70% ethanol (EtOH) and 55% isopropyl alcohol (IPA, 2-propanol), and B-hordeins were extracted with the same alcohols in 1.0% dithiothreitol (DTT). High molecular weight (HMW) prolamins (D-hordeins) were extracted with 50% IPA with 1M Tris-HCl (pH 8.0). The same protein patterns were observed in both the experimental extraction solutions (EtOH and IPA). However, the patterns of hordein, extracted with EtOH and IPA containing 1.0% DTT, differed slightly. C- and B-hordeins extracted from those solutions were analyzed. Twenty-six malting barley varieties developed in Korea were analyzed using UPLC. The varieties were divided into seven groups according to hordein patterns of retention time 16 min to 18 min, and 20 varieties showed unique patterns.

Studies on the Physical and Chemical Denatures of Cocoon Bave Sericin throughout Silk Filature Processes (제사과정 전후에서의 견사세리신의 물리화학적 성질변화에 관한 연구)

  • The studies were carried out to disclose the physical and chemical properties of sericin fraction obtained from silk cocoon shells and its characteristics of swelling and solubility. The following results were obtained. 1. The physical and chemical properties of sericin fraction. 1) In contrast to the easy water soluble sericin, the hard soluble sericin contains fewer amino acids include of polar side radical while the hard soluble amino acid sach as alanine and leucine were detected. 2) The easy soluble amino acids were found mainly on the outer part of the fibroin, but the hard soluble amino acids were located in the near parts to the fibroin. 3) The swelling and solubility of the sericin could be hardly assayed by the analysis of the amino acid composition, and could be considered to tee closely related to the compound of the sericin crystal and secondary structure. 4) The X-ray patterns of the cocoon filament were ring shape, but they disappeared by the degumming treatment. 5) The sericin of tussah silkworm (A. pernyi), showed stronger circular patterns in the meridian than the regular silkworm (Bombyx mori). 6) There was no pattern difference between Fraction A and B. 7) X-ray diffraction patterns of the Sericin 1, ll and 111 were similar except interference of 8.85A (side chain spacing). 8) The amino acids above 150 in molecular weight such as Cys. Tyr. Phe. His. and Arg. were not found quantitatively by the 60 minutes-hydrolysis (6N-HCI). 9) The X-ray Pattern of 4.6A had a tendency to disappear with hot-water, ether, and alcohol treatment. 10) The partial hydrolysis of sericin showed a cirucular interference (2A) on the meridian. 11) The sericin pellet after hydrolysis was considered to be peptides composed with specific amino acids. 12) The decomposing temperature of Sericin 111 was higher than that of Sericin I and II. 13) Thermogram of the inner portioned sericin of the cocoon shell had double endothermic peaks at 165$^{\circ}C$, and 245$^{\circ}C$, and its decomposing temperature was higher than that of other portioned sericin. 14) The infrared spectroscopic properties among sericin I, II, III and sericin extracted from each layer portion of the cocoon shell were similar. II. The characteristics of seriein swelling and solubility related with silk processing. 1) Fifteen minutes was required to dehydrate the free moisture of cocoon shells with centrifugal force controlled at 13${\times}$10$^4$ dyne/g at 3,000 R.P.M. B) It took 30 minutes for the sericin to show positive reaction with the Folin-Ciocaltue reagent at room temperature. 3) The measurable wave length of the visible radiation was 500-750m${\mu}$, and the highest absorbance was observed at the wave length of 650m${\mu}$. 4) The colorimetric analysis should be conducted at 650mu for low concentration (10$\mu\textrm{g}$/$m\ell$), and at 500m${\mu}$ for the higher concentration to obtain an exact analysis. 5) The absorbing curves of sericin and egg albumin at different wave lengths were similar, but the absorbance of the former was slightly higher than that of the latter. 6) The quantity of the sericin measured by the colorimetric analysis, turned out to be less than by the Kjeldahl method. 7) Both temperature and duration in the cocoon cooking process has much effect on the swelling and solubility of the cocoon shells, but the temperature was more influential than the duration of the treatment. 8) The factorial relation between the temperature and the duration of treatment of the cocoon cooking to check for siricin swelling and solubility showed that the treatment duration should be gradually increased to reach optimum swelling and solubility of sericin with low temperature(70$^{\circ}C$) . High temperature, however, showed more sharp increase. 9) The more increased temperature in the drying of fresh cocoons, the less the sericin swelling and solubility were obtained. 10) In a specific cooking duration, the heavier the cocoon shell is, the less the swelling and solubility were obtained. 11) It was considered that there are differences in swelling or solubility between the filaments of each cocoon layer. 12) Sericin swelling or solubility in the cocoon filament was decreased by the wax extraction.. 13) The ionic surface active agent accelerated the swelling and solubility of the sericin at the range of pH 6-7. 14) In the same conditions as above, the cation agent was absorbed into the sericin. 15) In case of the increase of Ca ang Mg in the reeling water, its pH value drifted toward the acidity. 16) A buffering action was observed between the sericin and the water hardness constituents in the reeling water. 17) The effect of calcium on the swelling and solubility of the sericin was more moderate than that of magnecium. 18) The solute of the water hardness constituents increased the electric conductivity in the reeling water.

Cloning and Characterization of Phosphomannomutase/Phosphoglucomutase (pmm/pgm) Gene of Vibrio anguillarum Related to Synthesis of LPS (Lipopolysaccharide 생합성에 관여하는 Vibrio anguillarum의 phosphomannomutase/phosphoglucomutase 유전자 cloning과 특성)

  • The phosphomannomutase/phosphoglucomutase gene (pmm/pgm) of Vibrio anguillarum (the causative agent of fish vibriosis) was cloned, and the open reading frame corresponded to a protein with 446 amino acids. The pmm/pgm gene showed a significant degree of sequence homology with the previously reported genes from V. mimicus, V. vulnificus, V. splendidus, and V. harveyi, with 92.3%, 91.4%, 89.9%, and 89.9% amino acid identity, respectively. By reverse transcriptase-polymerase chain reaction, we found that the pmm/pgm gene was upregulated under cold stress condition. The PMM/PGM protein is known to catalyze the interconversion between mannose-1-phosphate and mannose-6-phosphate or glucose-1-phosphate and glucose-6-phosphate, which are important intermediates for lipopolysaccharide (LPS) biosynthesis. To confirm the role of PMM/PGM in the LPS biosynthetic pathway, we constructed a knock out mutant by homologous recombination. The respective LPSs were isolated from the V. anguillarum wild-type and mutant strains, and changes were compared by subjecting them to sodium dodecyl sulfate polyacrylamide gel electrophoresis. Based on the different patterns of the LPSs, we expect the pmm/pgm gene to have an important role in LPS biosynthesis. The pmm/pgm-deficient mutant of V. anguillarum will contribute to further studies about the role of LPS in V. anguillarum pathogenesis.
