  • The effect of prolonged treatment with Mori Folium column fraction(MFCF) on intestinal ${\alpha}$-glycosidase activity has been studied in mice made hyperglycemic and hyperinsulinemic by feeding a high carbohydrate-containing diet. Mice were treated for 10 week with or without MFCF, added to the high carbohydrate-containing diet at 50mg/100g food. While MFCF had no effect on body weight, it prevented the rise in glycemia and insulinemia. Maltase, sucrase and lactase activities were measured in intestinal homogenates of proximal, middle and distal segments of jejunoileum. Following 10 week of MFCF administration, MFCF significantly induced all three enzyme activities especially at middle and distal segments. The mechanisms responsible for these changes and their potential biochemical implications remain to be determined.

Effects of Garcinia cambogia Extract Feeding on Body Weight and Lipid Profiles in Rats Fed a High-carbohydrate or High-fat Diet

  • This study was designed to evaluate the effects of high-carbohydrate (HC) or high-fat (HF) diet with hydroxycitric acid (HCA)-containing Garcinia cambogia in rats. Seventy male Sprague-Dawley rats were randomly divided into 7 groups and raised with experimental diets containing different HCA levels (0, 1.6, and 3.2%) and calorie sources (carbohydrate, fat) for 8 weeks. Energy intake was significantly reduced in rats fed a HC diet with HCA compared to the HC diet without HCA. Body weight gain was significantly reduced in HCA groups. In the diet groups, plasma total lipid and triglyceride (TG) levels of the HCA groups were significantly lower than those of the group without HCA. There were no significant differences in energy intake and plasma lipid profile in HF groups. These results suggest that HC diet with HCA was more effective in the reduction of energy intake, body weight gain, and plasma lipid contents than those of HF diet with HCA.

  • The effects of silkworm extract on body weight, blood glucose and insulin levels, and an intestinal glycosidase activity were studied in high carbohydrate diet administered mice. Hyperglycemia and hyperinsulinemia represented in high carbohydrate diet administered mice disappeared when silkworm extract(50 mg) was added to 100 g diet for 10 week period. These results suggest that silkworm extract may resolve the insulin resistance by lowering serum insulin level resulting from inhibition of intestinal glycosidase activity. Silkworm extract coadministered with high carbohydrate diet to mice for 10 weeks significantly induced maltase, sucrase and lactase acivities, especially middle and distal portion. Further studies about regulatory mechanisms of glycosidase by silkworm extract are needed.

Carbohydrate Intake Associated with Risk Factors of Coronary Heart Disease in the Adults: NHANES III (성인의 만성질환관련 탄수화물 식사지침 연구)

  • Recent research reported health risks associate with high carbohydrates diets. Objectives of this study were to evaluate in a cross-sectional study if high carbohydrate diet is associated with coronary heart disease(CHD) risk factors: examined blood concnetration of triglyceride(TG), total cholesterol, high density lipoprotein cholesterol(HDL-C), plasma glucose, systolic blood pressure(BP), body mas index(BMI), wasit-hip ratio(WHR) and waist-stature ratio(WSR). Using the most recent US National Health and Nutrition Examination Survey(IIINHANES III) data, the nationally representative US population (3772 men, 4095 women of 25-64 years of age) was divided into low vs. high carbohydrate diet groups(below 40% vs. above 60% energy intake from cab carbohydrates) and compared by the CHD risk factors. Triglyceride was higher(p<0.001) in the high carbohydrate group, whereas high density-lipoprotein cholesterol(HDL-C)was lower(p<0.01) in the high carbohydrate diet group. In plasma glucose, there was no significant differences between high carbohydrate diet and low carbohydrate diet. In adiposity(BMI, WHR and WSR), it also showed no significant differences, After adjustment for age, ethnicity, alcohol and smoking in upper 60%-carbohydrate diet, Odds Ratio of TG and HDL-C were 1.42 and 1.23 in men and 1.22 and 1.17 in women. 50-60% carbohydrate diet was associated with decreased risk of CHD. Dietary guidelines for Koreans recommend 60-70% of total energy from carbohydrate, as Koreans traditionally consumed high carbohydrate diets. In a cross-sectional population of adults, diets containing 55-60% energy from carbohydrate were suggested as a dietary guideline of carbohydrate intake for Koreans.   • The current study was undertaken to determine the effects of the ratios of dietary fat to carbohydrate and energy restriction on insulin sensitivity in the growing rats. Male rats weighting 80-90g were fed experimental diets for two weeks. Rats were killed and epiddymal adipose tissue were removed and sliced. Explants of adipose tissues were incubated for 2 hours in KRB(Krebs-Ringer bicarbonate) buffer containing various concentrations of human insulin and [U-14C]glucose. Insulin sensitivity was determined as glucose conversion to total lipids (lipogenesis) during 2 hr incubation. Exp't I : Effects of Ratios of Fat to Carbohydrate on Insulin Sensitivity. Eighteen male rats were fed 3 diets for 2 weeks. Diet 1 was low fat-high carbohydrate (4% soybean oil and 66.5% cornstarch) ; diet 2, medium fat-medium sarbohydrate(12% soybean oil and 58.5% cornstarch) ; diet 3, high fat-low carbohydrate (20% soybean oil and 50.5% cornstarch). Insulin sensitivity was higher in the order of LF-HC, MF-HC and HF-LC diet groups (p<0.05), i.e, lipogenesis was higher at all insuline concentration in the explants from rats fed LF-HC diet. However, thers was no significant difference in body weight gain and epididymal adipose tissue weight among treatments. Exp't II ; Effects of Energy Restriction on Insulin Sensitivity. Twelve rats were grouped into ad libitum feeding and restricted feeding(70% of ad libitum). The experimental diet was medium fat-medium carbohydrat diet as used in the Exp't I. Restricted feeding group tended to show higher insulin sensitivity compared to ad libitum group. However, there was no statistical difference between two groups. As expected, body weight gain and epididymal adipose tissue were higher in the ad libitum group. In summary, the resutls of the current study showed that the epididymal adipose tissue taken from the rats fed low fat-high carbohydrate diet showed higher insulin sensitivity compared to those fed high fat-low carbohydrate, and that resticted feeding tended to elevate insulin sensitivity in these tissues.

Effects of ᴅ-allulose on body fat accumulation in rats fed severely carbohydrate-restricted diets containing beef tallow or soybean oil

  • Purpose: The carbohydrate-restricted diet has been recognized to be effective into preventing and alleviating lifestyle-related diseases, such as obesity and type 2 diabetes. The rare sugar ᴅ-allulose is a functional monosaccharide with anti-obesity effects. In the present study, we examined the effects of dietary ᴅ-allulose on body fat accumulation in rats fed severely carbohydrate-restricted diets containing high concentrations of different fats, beef tallow, or soybean oil. Methods: Male Wistar rats (n = 35, 3-week-old) were divided into 5 groups: One chow-fed control (C) group, and four carbohydrate-restricted groups, namely, beef tallow (B), beef tallow + ᴅ-allulose (BA), soybean oil (S), and soybean oil + ᴅ-allulose (SA), with free access to the diet and water for 8 weeks. The B and BA diets contained 23% beef tallow and 2% soybean oil, whereas the S and SA diets contained 25% soybean oil. Furthermore, the BA and SA diets contained 5% ᴅ-allulose. Results: The final body weight, weight gain, and food intake were significantly higher, and food efficiency was significantly lower in the control group compared to the other carbohydrate-restricted groups. Intra-abdominal adipose tissue, carcass fat, and total body fat weights were not influenced by dietary fat type or ᴅ-allulose supplementation, except for the epididymal adipose tissue weight. In contrast, carbohydrate restriction suppressed body weight gain in rats, but remarkably increased body fat accumulation. Conclusion: Under carbohydrate-restricted conditions, no anti-obesity effects of dietary ᴅ-allulose were observed, regardless of the dietary fat type. The causes of these effects are unknown. However, they may be influenced by a very low carbohydrate and high protein diet. Further research is required to elucidate the effects of ᴅ-allulose under various nutrient compositions with different fat, carbohydrate, and protein energy ratios.

Effect of Dietary Proteins without Vitamin Premix Supplementation on the Growth and Body Composition of Juvenile Black Rockfish Sebastes schlegeli (비타민프리믹스 무첨가시 사료 중 단백질원이 조피볼락(Sebastes schlegeli) 치어의 성장과 체성분에 미치는 영향)

  • A feeding experiment was conducted to determine the nutritional value of various dietary protein sources without a vitamin premix supplementation in juvenile black rockfish, Sebastes schlegeli. Diets containing similar quantities of lipid and carbohydrate with 50% crude protein from fish meal (FM)/soybean meal (SBM)/corn gluten meal (CGM), FM only, casein/gelatin, high FM/low casein and low FM/high casein were fed to rockfish along with a commercial rockfish diet. The diet containing only 65.6% FM and the commercial rockfish diet produced significantly greater weight gain (WG) and feed efficiency (FE) than all other semipurified diets (SPDs); the economical diet with 52.8% FM, 10% SBM and 5% CGM produced lower WG than the diet containing FM and commercial rockfish diet, but FE and survival were similar both diets. Intermediate responses were observed for rockfish fed the SPD containing 32.8% FM and 27.4% casein, while SPDs containing 10% FM and 41.8% casein and 38.5% casein and 10% gelatin yielded the lowest WG and FE. Some differences in tissue indices and body composition of rockfish were induced by the diets containing various protein sources and levels without the vitamin premix supplementation.

Ca and P Balance in Korean Female Adolescents (청소년기 여자의 칼슘과 인 평형 연구)

  • Intakes and excretions of calcium and phosphorus were determined for 8 female adolescents(aged 16.3$\pm$0.5y; body mass index 20.4$\pm$1.3kg/$m^2$; body fat 33.3$\pm$2.5%; bone mineral density of lumbar spine in L2-L4; 0.96$\pm$0.08g/$\textrm{cm}^2$) when they consumed diets basal and high in calcium for 6 days each. All subjects consumed a basal Ca diet containing 800mg, Korean RDA level of the subjects, and a high Ca diet containing 1200mg, RDA plus 2 SDs of calcium intake. The diets provided 58% of energy intake as carbohydrate, 25% as fat, and 17% as protein. Food, urine, and fecal composites were collected during the last 3 days of each feeding period. Fecal excretion of calcium was significantly greater on the high Ca than on the basal Ca diet. Hence, apparent absorption rate was significantly lowered from 40.9% on the basal Ca diet to 33.1% on the high Ca diet. There was no significant difference in calcium retention between the two diet periods but it tended to be greater on the high Ca diet. However, excretions and retention of phosphorus did not show any significant difference even though intake was significantly increased from 1,253mg on the basal Ca diet to 1,583mg on the high Ca diet. The results indicate that higher calcium intake than the Korean RDA level is recommended for adolescents to meet peak bone mineral accretion and attain a desirable level of calcium retention. Effects of a Very Low Carbohydrate (Pork Rind-Based) Diet on Weight Gain, Serum Levels of Cholesterol, Triacylglycerol, Glucose, Ketone Bodies and Insulin and Body Composition in Adult Rats

  • A study was carried out to determine the effect of a very low-carbohydrate diet on weight gain, body composition, and serum levels of cholesterol, triacylglycerol, glucose, ketone bodies and insulin. Twenty rats (mean initial weight, 212 g) were divided into two groups and each group was assigned a conventional high-carbohydrate diet (control) or a very low-carbohydrate diet containing 59.8% ground pork rind snack and fed the diet for four weeks. Average daily body weight gain was not different between the two groups during the first two weeks, but was significantly lower in rats fed the very low-carbohydrate diet than in those in the control group during third (p<0.05) and fourth weeks (p<0.01). Feedintake as well as energy intake was lower in rats fed the very low-carbohydrate diet than in those in the control group. The very low-carbohydrate diet reduced (p<0.01) serum triacylglycerol (34$\pm$83 vs 82$\pm$8 mg/l00 mL) and insulin (3.90$\pm$0.53 vs 7.60$\pm$0.61 $\mu$IU/mL) levels, while increasing (p<0.01) ketone body level (368$\pm$25 vs 236$\pm$24 $\mu$mol/L), compared with the control. Serum glucose and total cholesterol levels were not different (p>0.05) between the two dietary treatments. Proximate analysis of carcasses showed that the very low-carbohydrate diet decreased (p<0.01) body fat (26.1$\pm$1.04 vs 30.5$\pm$0.86%), while increasing (p<0.01) body protein (63.1$\pm$0.94 vs 59.4$\pm$0.70%) contents. Results indicate that short-term feeding of a very low-carbohydrate diet is beneficial for alleviating risk factors known to involve cardiovascular diseases or artherosclerosis. However, more studies with model animals as well as humans are recommended to examine the long-term health benefits of low-carbohydrate diets.

Effects of Different Dietary Carbohydrate Sources on Growth and Body Composition of Juvenile Snail (Semisulcospira gottschei)

  • To test the effect of different carbohydrate sources, wheat flour, used as the carbohydrate source in the control diet, was replaced with 30% glucose, 30% maltose, 30% cellulose or 30% $\alpha$-starch. After feeding trial for 8 weeks, no significant differences were found in growth and proximate composition of edible fraction of the snail fed diets containing different carbohydrate sources. All the tested carbohydrates are shown as good dietary carbohydrate source.
