두송실에 의한 충치균의 유기산 생성 억제효과 (Inhibition Effect by Juniperus rigida S. et Z. on Organic Acids Production from Streptococcus mutans)

  • 생약에서 충치균에 대한 항균성 물질을 개발하기 위하여 수종의 야생생약을 수집하여 항균활성을 측정해 보았다. 그 중에서 충치균에 강한 항균활성을 나타낸 두송실(Juniperus rigida S. et Z.)을 methanol로서 추출하고 극성이 다른 수종의 용매로서 분획하여, 충치균에 대한 항균활성과 유기산의 생성억제효과를 측정해 본 결과, n-hexane, chloroform 분획에서 활성이 나타났다. 충치균에 의하여 생성된 유기산은 주로 lactic acid와 citric acid로 전체의 $80{\sim}90%$를 차지하였는데 두송실의 methanol추출물과 n-hexane분획물을 1 mg/ml 처리한 시험군에서 각각 89.3, 90.8%의 유기산 생성 억제효과를 나타내었다.

천년초 줄기의 항균, 항산화 및 항암효과에 관한 연구 (The Effects of Antimicrobial, Antioxidant, and Anticancer Properties of Opuntia humifusa Stems)

  • 본 연구는 천년초 줄기를 이용하여 항균, 항산화, 항암 등 천년초의 생리활성 효과를 연구하였으며 그 결과는 다음과 같다. 천년초 줄기의 추출물중 gram 양성균인 4가지 균주(Rhodococcus equi, Staphylococcus aureus, Clostridium perfringens, Bacillus cereus)에서 항균 효과가 나타난 것은 메탄올과 핵산층이었으며, 4가지 균주 중 Bacillus cereus를 제외한 3종에 대해서는 우수한 항균효과를 나타냈다. 천년 초 줄기의 peroxynitrite($ONOO^-$) 제거능 측정결과 핵산층을 제외한 메탄올층, 부탄올층, 수층의 항산화 활성은 천년초 줄기 추출물의 농도가 증가할수록 항산화 효과가 유의적으로(p<0.05) 높게 나타났다. 천년초 줄기 추출물의 각 용매별 DPPH 유리기 소거능은 농도가 증가함에 따라 소거능도 유의적으로(p<0.05) 증가하였으며, 부탄올>메탄올>수층>핵산층 순으로 높게 나타났다. 천년초 줄기의 암세포 증식 억제 효과에서 피부암 세포의 경우 $50\sim100{\mu}g/mL$의 농도에서 메탄올층과 핵산층이 수층과 부탄올층에 비하여 유의적으로(p<0.05) 높게 나타났다. 간암세포인 HepG2의 경우 추출물 $100{\mu}g/mL$의 농도를 기준으로 메탄올층이 유의적으로(p<0.05) 높았고, 핵산층>부탄올층>수층의 순으로 나타났다. 대장암세포인 HT-29 역시 메탄올층이 유의적으로 가장 높았고, 핵산층>수층>부탄올층 순으로 나타났다. 유방암세포인 MCF-7은 메탄올층>핵산층>수층>부탄올층 순으로 효과가 높게 나타났다. 이와 같은 결과로 볼 때 천년초줄기의 항균, 항산화, 항암 효과가 높게 나타나 줄기를 활용한 항균제품이나 여러 가지 가공제품을 개발하는데 기초 자료로 활용할 수 있다.

연잎 추출물의 항균 효과 (Antimicrobial Effect of the Fractions Extracted from a Lotus (Nelumbo nucifera) Leaf)

  • 본 연구에서는 연잎 추출분획물의 항균 활성을 benzoic acid와 비교, 검토하고자 하였다. $70\%$ ethanol 추출물을 hexane, chloroform, ethyl acetate, butanol, water로 순차 분획하여 총 페놀함량을 비교한 결과 ethyl acetate분획물이 가장 높은 함량을 보여주었다. 항균활성 또한 ethyl acetate 분획물에서 가장 우수한 활성을 보여주었으며 benzoic acid 보다 높은 활성을 나타냈다. Gram 양성균과 음성균간 항균력의 차이는 보이지 않았다. Liquid culture에서 생육저해 효과를 측정한 결과 추출물의 농도가 800 ppm이상일 경우 생장이 완전히 억제되었다.

해양 미세조류 Nannochloropsis oculata 추출.분획물의 ACE, α-glucosidase 및 암세포 저해 활성 (ACE, α-Glucosidase and Cancer Cell Growth Inhibitory Activities of Extracts and Fractions from Marine Microalgae, Nannochloropsis oculata)

  • Extracts of the marine microalgae Nannochloropsis oculata were obtained using 80% methanol (MeOH) and water. The 80% MeOH extract was further fractionated with n-hexane, chloroform, ethyl acetate (EtOAc), n-butanol (n-BuOH), and water to isolate the active fraction. Seven samples were prepared and their angiotensin converting enzyme (ACE), $\alpha$-glucosidase, and cancer cell growth inhibitory activities in vitro were determined. The most profound ACE inhibitory activity was observed in the chloroform fraction, while the others had moderate effects. By contrast, greater $\alpha$-glucosidase inhibitory activity was found in the EtOAc fraction, n-hexane fraction, and water extract of N. oculata. The antiproliferative effects of the extracts and fractions against HL-60, U937, CT-26, and SK-Hep1 cancer cells were also determined. The n-BuOH fraction had the strongest antiproliferative effects on CT-26 cells in a time-dependant manner (P<0.05). These results suggest that the extracts and fractions from N. oculata could be used as a potential functional food or as pharmaceutical ingredients.

Bioactivity-Guided Fraction from Viscera of Abalone, Haliotis discus hannai Suppresses Cellular Basophils Activation and Anaphylaxis in Mice

  • Basophils and mast cells are specialized effector cells in allergic reactions. Haliotis discus hannai (abalone), is valuable seafood. Abalone male viscera, which has a brownish color and has not been previously reported to show anti-allergic activities, was extracted with acetone. Six different acetone/hexane fractions (0, 10, 20, 30, 40, and 100%) were obtained using a silica column via β-hexosaminidase release inhibitory activity-guided selection in phorbol myristate acetate and a calcium ionophore, A23187 (PMACI)-induced human basophils, KU812F cells. The 40% acetone/hexane fraction (A40) exhibited the strongest inhibition of PMACI-induced-β-hexosaminidase release. This fraction dose-dependently inhibited reactive oxygen species (ROS) production and calcium mobilization without cytotoxicity. Western blot analysis revealed that A40 down-regulated PMACI-induced MAPK (ERK 1/2, p-38, and JNK) phosphorylation, and the NF-κB translocation from the cytosol to membrane. Moreover, A40 inhibited PMACI-induced interleukin (IL)-1β, IL-6, and IL-8 production. Anti-allergic activities of A40 were confirmed based on inhibitory effects on IL-4 and tumor necrosis factor alpha (TNF-α) production in compound (com) 48/80-induced rat basophilic leukemia (RBL)-2H3 cells. A40 inhibited β-hexosaminidase release and cytokine production such as IL-4 and TNF-α produced by com 48/80-stimulated RBL-2H3 cells. Furthermore, it's fraction attenuated the IgE/DNP-induced passive cutaneous anaphylaxis (PCA) reaction in the ears of BALB/c mice. Our results suggest that abalone contains the active fraction, A40 is a potent therapeutic and functional material to treat allergic diseases.

  • The fluctuation integrals which give useful information in the structure of solution are associated with the mixed direct correlation integral ($C_{12}$) known. Using its weighted arithmetic mean of $C_{11}$ and $C_{22}$ and the activity coefficient model, the fluctuation integrals on solute-solute, solvent-solute, and solvent-solvent can be calculated in the function of mole fraction. In this work, several binary mixtures containing c-hexane were tested and the results on the fluctuation integrals were rather good.

  • 80% MeOH extract of black rices fractionated with n-hexane, EtOAc, and n-BuOH. Copper-induced LDL oxidation inhibition assay and ACAT inhibition assay examined with fractions. n-Hexane and EtOAc fractions showed high inhibition activity. We divided n-hexane fraction into three sub-layers(H1 - H3) and EtOAc into eight sub-layers(E1 - E8). H1, H2, E6, and E7 are showed higher inhibition activity than standard in Lp-PLA2 inhibition assay and H1 and E6 are showed higher ACAT inhibition activity than standard.

Antimutagenic Compounds Indentified from Chloroform Fraction of Persimmon Leaves

  • Methanol extract of dried persimmon leaves was refractionated using hexane, chloroform, ethylacetate, n-butanol aqueous fractions. Among these chloroform fraction showed the highest inhibition rate on the mutagenicities of aflatoxin B₁(AFB₁) and 3,2' -dimethyl-4-amino-bipheny1(DMAB) in Salmonella typhimurium TA 100. Chloroform fraction was further fractionated into eight fractions by silica gel column chromatography and thin layer chromatography(TLC). The fraction 5 on TLC exhibited the highest antimutagenic activities in AFB₁and DMAB. 2,4-Decadienal, dihydro-4-methyl-2(3H)-furanone, hexanoic acid 1,4-bis(1-methy1 ethy1)benzene, heptanoic acid, phenol, octanoic acid, nonanoic acid and benzoic acid were tentatively identified from this antimutagenic fraction by GC-MS.

울금색소 추출물의 항균성과 물질분리 동정 (Antibacterial Activity and Identification of the Active Compound from Tumeric Extract)

  • To identify the antimicrobial activity, of tumeric and its active compound tumeric was fractionated into four groups dichloromethane extract, hexane fraction, methanol soluble fraction and residue's extract. They were tested for antibacterial activity against E. coil and S. aureus and the methanol soluble fraction was found lo be the most active fraction. Compound I, II and III were isolated from TLC and silica gel column chromatography in the methanol soluble fraction. These compounds were analyzed by $^1H-NMR\;and\;^{13}C-NMR$ spectra and identified as curcumin I, II and III. They were also tested for antimicrobial activity against E. coli and S. aureus. Curcumin I was the must active curcuminoids due to the phenolic and methoxyl$(OCH_3)$ moiety in the same molecular structure.

자근에 함유된 복강 마크로파지의 탐식능 활성분획 (An Active Fraction on Phagocytosis of Murine Peritoneal Macrophage in Lithospermi Radix)

  • The phagocytic activity of murine peritoneal macrophages was determined by lucigenin chemiluminescence with luminometer and engulfment of fluorescein-conjugated E. coli particles. 70% MeOH extract of Lithospermi Radix was fractionated successively with hexane, methylene chloride, n-BuOH and water. The water fraction (m.w. 500 to 1,000) enhanced the lucigenin chemiluminescence and the engulfment of fluorescein-conjugated E. coli particles in murine peritoneal macrophages. The water fraction suppressed the production of nitric oxide in the macrophages. These results suggest that an active fraction of phagocytosis in Lithos-permi Radix is the water fraction and the molecular weight is 500 to 1,000.
