  • As a part of a study on the development of microbiological methods for petroleum exploration, the distribution of the avarafe survival rate for heterotrophs to various heavy metal ions and hydrocarbons were surveyed and compared by the use of plate count method. In consequence of the avarage survival rate to heavy metal ions(2 hours treatment) and hydrocarbons(1 hour treatment) for heterotrophs isolatinf from soil samples(50cm depth) which located in Doum mountain(A, B and D site) and Aedowon(C site) at Pohang area, the survival rate of heterotrophs for nickel(600ppm), cobalt(500ppm), cadmiun(100ppm), mercury(20ppm), zinc(400 ppm) and lead(500ppm) were 73.7%, 82.6%, 76.8%, 9.5%, 77.8% and 73.6% at A site and 67.9%, 82.5%, 86.0%, 5.8%, 82.5% and 91.7% at B site, 87.8%, 79.8%, 87.5%, 7.0%, 84.2% AND 47.7% AT c SITE, AND 71.8%, 76%, 85.9%, 1,2%, 79.6% AND 88.3% AT D site, respectively. Also the survival rate of heterotrophs from A,B,C and D site to pentane and hexane(each concentration is 20%) were 26.7% and 42.5%, 11.8% and 8.1%, 44.3% and 36.2%, and 12% and 3.5%, respectively. therefore, heterotrophs from B and D site that alternated gravelstone, muddy sandstone and sandstone were higher survival rate to the heavy metal ions than heterotrophs from A site which mainly composed gravelstone. Also, heterotrophs from C site which mainly composed muddy sandstone and once produced natural gas were showed relatively higher survival rate to the heavy metal ions and hydrocarbons than the other sites. Consequently, we confirmed that the distributions of tolerant heterotrophs to heavy metal ions and hydrocarbons were differ from the lithological compositon.

  • Bacterial activities of dam reservoir sediments were assessed using the methods of viable counts, ATP contents, dehydrogenase activity, and oxygen consumption. Viable heterotrophs in collected sediments were observed in the range of $10^6{\sim}10^{10}$ CFU/g dry wt. sediment. All assessed methods showed high activities in sediment samples collected from near dam site. In addition, bacteria increase in sediments amended with cellulose, starch, lipid, and protein compounds. Various ranges of antibiotics and heavy metals resistance bacteria were also detected, especially, 10~100 times less numbers of Cd resistance bacteria were observed compared with those of Pb and Cr. In general, antibiotics resistance groups were in the range of $10^{-1}{\sim}10^{-6}$ times of control.

  • Batch test methods have developed for a long time to measure kinetic and stoichiometric parameters which are required to perform steady state design and mathematical modelling of activated sludge processes. However, at various So/Xo ratios, abnormal behaviors of ordinary heterotrophic organism in batch tests have been reported in many researches. Thus, in this research, abnormal behaviors of heterotrophs in batch tests were investigated at various So/Xo conditions by measuring and interpreting oxygen utilization rate. As So/Xo ratio increased, the calculated values of maximum specific growth rates, ${\mu}_{H,max}$ and $K_{MP,max}$, increased. However, at a certain point of So/Xo (around 10mgCOD/mgMLAVSS), ${\mu}_{H,max}$ and $K_{MP,max}$ values started to decrease. According to this observation, three prominent behaviours of heterotrophs were identified at various So/Xo conditions. (1) At low So/Xo region (below 5 mgCOD/mgMLAVSS), the oxygen utilization rate of heterotrophs in batch tests were almost stable and consequently yielded lower maximum specific growth rate. (2) At high So/Xo region (up to 5~10 mgCOD/mgMLAVSS), oxygen utilization rate incresed sharply with time and indicated more upward curvature than the predicted OUR with conventional activated sludge model, which consists of single hetetrotrophs group. Thus, in this region, competition model of two organisms, fast-grower and slow-grower, seemed to be appropriate. (3) At extremely high So/Xo region (over 10mgCOD/mgMLAVSS), significant oxygen utilization rate was still observed even after depletion of readily biodegradable COD. This might be caused by retarded utilization of intermediates which were generated by self inhibition mechanism in the process of RBCOD uptake.

  • The carbon and nitrogen removal process using PVA-gel (Polyvinyl Alcohol) immobilized microorganisms was studied. The process has been operated under diverse process conditions for 12 months. The process consists of denitrification with internal recycle of 300%, nitrification, aerobic digestion reactors and settling tank. Nitrificatoin and nitrogen removal efficiency according to gel packing ratio and temperature were studied. Yield for Heterotrophs ($Y_H$), decay coefficient for Heterotrophs($b_H$) in aerobic digestion reactor were determined to seize sludge reduction mechanism and compared with typical data of activated sludge process. Then SRT in aerobic digestion reactor was determined on an experimental basis and sludge reduction efficiency was calculated. The process was implemented successfully with sludge reduction efficiency of 92.0~98.5% on a basis of biomass.

  • This study was carried out far isolation and characterization of ammonia oxidizing bacteria (AOB) from aquacultural place and sludges of waste water collected in Pusan. One autotrophic AOB, Nitrosomonas sp. and 8 heterotrophic AOB (2 strains of Bacillus sp., 2 strains of Acinetobacter sp., Xanthomonas sp., Alcaligenes sp., Pseudomonas sp., Sphingobacterium sp.) were isolated. and identified. Variation of total nmmonia nitrogen (TAN) and $NO_2-N$ in mineral salt media containing 10mg/ $\ell$ of NHCl for 15 days in differents 9 strains was measured in order to examine the ablitity of ammonia oxidation. TAN was started to reduce after 4 days incubation and ca. 2 mg/$\ell$ of TAN was decreased after 15 days incubation by Nitrosomonas sp., At that time, $NO_2-N$ was produced to 0.023$\~$0.036 mg/$\ell$. Heterotrophic AOB showed the low ability of ammonia oxidation, 0.02$\~$0,04 mg/$\ell$ of TAN was decreased and $NO_2-N$ was produced to 0.01$\~$0.51 mg/$\ell$ after 15 days. When each strain of 8 heterotrophs was incubated in mimeral salt media containing 10 mg/$\ell$ $NH_4Cl$ and 50 mg/$\ell$ glucose, and 50 mg/$\ell$ $NH_4Cl$ and 5 g/$\ell$ glucose, the diminution of TAN was 1.12$\~$3.38 mg/$\ell$ and 1$\~$20 mg/$\ell$, respectively.

  • The presence of thiosulfate oxidizing bacteria was examined in rhizosphere soils of 19 economically important plant species belonging to 10 different families. The results showed that the thiosulfate oxidizing bacteria were present in all the tested rhizosphere soils, and the total 32 thiosulfate oxidizing bacteria were recovered. Furthermore, the biochemical characterization revealed that 56% and 44% of the isolates belonged to the obligate chemolithoautotrophs and facultative heterotrophs, respectively. The isolates ATSR15P utilized 19.17 mM of thiosulfate and accumulated 11.65 mM of sulfate in the medium. Concurrently, the decrease in pH of the medium was observed. This study comprehensively demonstrates that the active sulfur oxidation is a ubiquitous phenomenon in the rhizosphere of crop plants in Korea.

  • The ecological process-based approach provides a detailed assessment of belowground compartment as one of the major compartment of carbon balance. Carbon net balance (NEP: net ecosystem production) in forest ecosystems by ecological process-based approach is determined by the balance between net primary production (NPP) of vegetation and heterotrophic respiration (HR) of soil (NEP=NPP-HR). Respiration due to soil heterotrophs is the difference between total soil respiration (SR) and root respiration (RR) (HR=SR-RR, NEP=NPP-(SR-RR)). If NEP is positive, it is a sink of carbon. This study assessed the forest carbon balance by ecological process-based approach included belowground compartment intensively. The case study in the Takayama Station, cool-temperate deciduous broad-leaved forest was reported. From the result, NEP was estimated approximately 1.2 t C $ha^{-1} yr^{-1}$ in 1996. Therefore, the study area as a whole was estimated to act as a sink of carbon. According to flux tower result, the net uptake rate of carbon was 1.1 t C $ha^{-1} yr^{-1}$.
