Influence of gene flow from GM to non-GM soybeans by the size of the pollen donor

  • The use of genetically modified (GM) crops has increased continuously over the world, and concerns about the potential risks of GM crops have also risen. Although, until now, GM crops have not been cultivated commercially in Korea, it is necessary to develop technology for the safe evaluation of GM crops. In this study, we investigated the influence of gene flow from GM to non-GM soybeans by the size of the pollen donor. In the experimental design, GM soybeans were placed in the center as a pollen donor and non-GM soybeans were placed in four directions as the pollen receivers. Three sizes of pollen donor were designed as $90cm{\times}90cm$, $180cm{\times}180cm$, and $360cm{\times}360cm$. A total 22,719 seeds were collected from non-GM soybeans, and 14 hybrids were finally obtained through herbicide resistance screening and PCR analysis. The highest hybridization rate was 0.78% at a distance of 15 cm from a $360cm{\times}360cm$ GM pollen donor, and the farthest distance of hybridization was 180 cm from a GM pollen donor which was $360cm{\times}360cm$ in size. Ten hybrids were found among the 14 hybrids at the $360cm{\times}360cm$ pollen donor size, 3 hybrids at $180cm{\times}180cm$, 1 hybrid at $90cm{\times}90cm$. From these results, it could be concluded that with the larger pollen donor size, more hybridization occurred in soybeans.

Establishment of Foliar Application Assays for Developing Natural Herbicides (천연물 제초제 개발을 위한 전식물체 수준의 경엽처리 검정법 확립)

  • This study was carried out to establish an improved bioassay system, whole-plant bioassay which is more effective in developing natural herbicides for foliar treatment such as herbicidal essential oils. Two bioassay systems using four weed species (Echinochloa crus-galli, Digitaria sanguinalis, Aeschynomene indica, and Abutilon theophrasti), spraying method and spotting method, were established. Spraying method is applicable if the amount of test compounds is enough, while spotting method is useful for the small amount of test compounds. The initial application rate was desirable at $2,500{\sim}5,000\;{\mu}g\;mL^{-1}$. Herbicidal activities were higher in the NOP treatment when compared to the Tween 20 treatment. To efficiently evaluate volatile compounds such as essential oils, if the compound-treated pots were incubated in dew chamber for about 10hrs, better results were obtained in the degree and stability of herbicidal responses. When the efficiency of bioassay systems established in this study was compared, the spraying method was minimized four times to the conventional method that has beed used for screening of synthetic compounds in KRICT. On the other hand, in the spotting method, screening for development of a natural herbicides was possible even in level of 1/100 test volume and 1/200 amounts of test compound compared to the spraying method.

Screening of Herbicidal Activity from Aqueous Extracts of Coronopus didymus (냄새냉이 수용성추출물의 제초활성 탐색)

  • This study was carried out to allelopathic effects of aqueous extracts on Coronopus didymus in order to investigate the competitive dominant in plant ecosystem and possibility application in natural herbicide. Number of species and species diversity for close to patch of C. didymus was decreased gradually site #1 (7, 1.76), site #2 (5, 1.34) and site #3 (5, 1.25). It was generally decreased the relative germination ratio (r=-0.731, p<0.01), the mean germination time, the relative elongation ratio (r=-571, p<0.01, r=-0.730, p<0.01), the relative fresh weight (r=-0.743, p<0.01), development of root hairs of receptor plants by concentration of the aqueous extracts from C. didymu. But they were different from the growing regions, the kind of receptor plants and the treatment of the aqueous extracts. Especially, it was differently effected among growing regions that inhibited more radicle than shoot by the aqueous extracts concentration of C. didymus. Total phenolic compound in the aqueous extracts of C. didymus analyzed about $23.0{\pm}1.1mg/g$. Total phenolic compounds of soil in survey area was increased gradually site #1 ($0.072{\pm}0.002mg/g$), site #2 ($0.082{\pm}0.003mg/g$) and site #3 ($0.092{\pm}0.004 mg/g$). We think that the aqueous extracts of C. didymu showed allelopathic effects on other plants. Therefore, C. didymu hold the competitive dominant of plant ecosystem in Jeju Island and possibility application of natural herbicide.

Germination Characteristics of Diplachne fusca seeds (갯드렁새(Diplachne fusca) 종자의 발아특성에 관한 연구)

  • The germination characteristics of Diplachne fusca (L.) P. Beauv. seeds were investigated under different seed storage conditions to find out reliable system for maintaining the seeds with high and uniform germination rate and thus for possible use of the seeds in herbicide screening with a continuous seed supply. When the seeds were stored under wet-low temperature($4^{\circ}C$) condition, the germination rate was 88% after 4-week-storage. The germination rate slightly declined after the storage for longer than 3 months. Dry seeds stored at room temperature exhibited very low germination rate. The wet-low temperature treatment was effective for inducing the germination of the seeds which had been stored under dry-room temperature condition for 4 months. The germination rate was 70% after 2-week- storage under the wet-low temperature condition. The germination rate was much higher under an alternate temperature condition than under a continuous temperature condition. The optimum temperature was 35/$25^{\circ}C$(14/10hrs). The seeds had a capability to germinate under NaCl-treated condition even at a concentration of 1.0%, whereas the germination of Echinochloa crus-galli seeds was completely inhibited by 0.5% NaCl. This result indicates that D. fusca has an advantage over E. crus-galli to survive in reclaimed 1and from the sea.

The Initial Growth of Scirpus planiculmis and its Herbicidal Response for the Screening (Screening을 위한 새섬매자기(Scirpus planiculmis) 초기생육(初期生育) 및 제초제(除草劑) 반응성(反應性))

  • The initial growth and herbicidal responses of Scirpus planiculmis were examined under both laboratory and greenhouse condition. Initial growth of S. planiculmis at two different soils(soil A and B), was compared with the growth at normal paddy soil, and growth rates at two soils were 32% and 86% as compared to the growth rates at normal paddy soil. Two different soils were soil A(rice-uncultivable) and soil B(rice-cultivable). Transplanted(2.5 leaf stage) and direct seeded rice(Dongjinbyeo) could not grow at soil A. The growth of transplanted rice at soil B was almost same as the growth at normal paddy soil, but the growth of direct seeded rice was 50% of the growth at normal paddy soil. S. planiculmis among the perennial weeds was most tolerant to NaCl, and Cyperus serotinus, Eleocharis kuroguwai, Sagittaria pygmaea, Sagittaria trifolia, Potamogeton distinctus were followed. The intial growth rate of S. planiculmis was more rapid than C. serotinus. Cutting types and times of corm reduced 20-40% of the initial growth of S. planiculmis. During the initial growth of S. planiculmis, the growth of S. planiculmis was severly inhibited by the earlier removal of corm from seedlings, and it represents that the corm plays an important role in the initial growth of this species. Bentazone among 14 commercialized herbicides showed the best safety on direct seeded rice and the highest control effect agaist S. planiculmis.

Studies on Germination Characteristics and Differential Responses to Herbicides in Setaria Spp. (강아지풀 종류별 종자발아특성과 제초반응 차이)

  • To supply seeds with a good quality as plant materials for herbicide screening and to know whether the germination characteristics could be associated with a differential response to herbicides, germination characteristics and differential responses to herbicides were investigated with 3 species of a genus Setaria ; Yellow foxtail(Setaria glauca P. Beauv, SETGL), Giant foxtail(Setaria faberi Hetrm, SETFA), and Green foxtail(Setaria viridis P. Beauv, SETVI). Degree of dormancy was high in the order of SETGL, SETVI and SETFA. The dormancy of SETGL seed was relatively well removed by room temperature and drying storage, but SETFA and SETVI by low temperature and wetting storage(stratification). For breaking dormancy of SETGL, SETVI and SETFA, it was necessary for being kept under the above storage conditions for at least 2, 4 and 4-5 months, respectively. When the dormancy-breaked seeds were transfered to low temperature($4^{\circ}C$) and drying condition, SETGL showed germination rate of 96% even after 2 month storage. However, SETVI and SETFA showed a decreased germination of 54% and 69%, respectively, with a decreased velocity of germination, indicating that secondary dormancy might be induced. On the other hand, a significant change in germination rate was not observed as the seeds were transfered to room temperature($25^{\circ}C$) and drying condition. The germinability of SETGL seed began to decline from 6th year after storage in room temperature and drying condition. All of 3 species showed relatively high germination rate at alternating temperature of $30^{\circ}C$/$20^{\circ}C$(14hr/10hr) and their germination were not increased by light. All of 3 species exhibited similar responses to cycloxydim, sethoxydim and primisulfuron in greenhouse experiment. In contrast, SETVI and SETFA were relatively susceptible to fenoxapropethyl, SETFA to fluazifop-butyl, SETGL and SETFA to clorimuron-ethyl, and SETGL to EK-2612. The difference in herbicidal response among 3 species was the highest in the treatment of EK-2612. These results suggest that there is no a consistent tendency in responses of 3 species to herbicides which have the same target site. And the relationship between germination characteristics and differential responses to herbicides was not found.
