Improvement of Bronchial Immune Hypersensitivity Reaction by Extracts from Chrysanthemum morifolium and Scutellaria baicalensis

  • This study aimed to discover functional materials derived from resources, which can improve the troublesome symptoms of a bronchus by improving bronchial hyperresponsiveness as air pollution in Korea caused by fine dust and yellow dust is getting worse. Of natural resources grown naturally in Korea, Chrysanthemum morifolium(CM), and Scutellaria baicalensis(SB) have been used as a safe raw material for drinking or medicine for a long time, and it has been found that a combination of CS73 can improve bronchial health function in experimental animal models. Analysis of serum of animal models with asthma induced by ovalbumin (chicken egg albumin) and analysis of cytokine production in BALF (Bronchoalveolar lavage fluid) showed that inflammatory indices IL-1β, IL-4, IL-5, IL-6, IL-10, IL-13, IL-17A, IL-17F, and IL-17E were significantly reduced and that respective production of IL-2 and IFN-γ was significantly increased in the group taking extracts from CS73 when compared with the control group. These results suggested that the combination of CS73 could be used as a natural treatment for asthma. In addition, in the animal models that the combination of CS73 significantly decreased the respective production of IgE, histamine and TSLP when compared with the control group. In experimental models, the ratio of Chrysanthemum morifolium to Scutellaria baicalensis of 7 : 3 had more excellent effect than other combined experimental groups, which suggests that the above combination can be developed as a natural treatment for asthma and is valuable as a pharmaceutical composition with an effect of improving bronchial health, capable of contributing to the public health threatened by fine dust.

Anti-Oxidant and Antiinflammatory Effects of Rosa multiflora Root (찔레나무뿌리(Rosa multiflora root)의 항산화 및 항염증효과)

  • Rosa multiflora thunberg belonging to Rosaceae is widely distributed in East Asia including Korea and Japan, and has been reported to have tormentic acid and rosamultin. To develop a new natural anti-inflammatory agent for cosmetics, we investigated the inhibitory effects of inflammation in Rosa multiflora root (R. multiflora root). The biological activity and anti-inflammatory effects were investigated by water, ethanol, methanol and acetone extracts of R. multiflora root. The measurements of polyphenol content from R. multiflora root were highest in water and acetone extracts, at 57.48 ${\pm}$ 0.88 mg/g and 67.05 ${\pm}$ 0.56 mg/g, respectively. The result of DPPH, ABTS and superoxide anion radical scavenging effects showed over 50% efficacy at 50 ${\mu}g/ml$ in ethanol, methanol and acetone extracts. Hyaluronidase inhibition effect showed over 60% efficacy at 500 ${\mu}g/ml$ in ethanol, methanol, and acetone extracts. Nitric oxide radical inhibition effect of R. multiflora root ethanol extracts showed over 30% efficacy at 500 ${\mu}g/ml$. We investigated the effect of R. multiflora root extracts on nitric oxide (NO) production of inducible nitric oxide synthase (iNOS) in LPS-induced RAW 264.7 macrophage cells. The result showed that R. multiflora root extracts have an inhibitory effect on NO production and iNOS expression and also can be used as an anti-inflammatory agent. These antioxidant and anti-inflammatory effects of R. multiflora root show applicant potential application as a functional cosmetic material.

Composition analysis of raw material constituting the medium for mushroom cultivation (버섯재배용 배지재료의 성분분석)

  • The contents of raw materials which are components of mixed substrate for mushroom cultivation were analyzed to optimize the composition. The pure protein(amino acid) level of soybean meal was the highest, 44.02% followed by those of soybean curd residue(31.5%) and cotton seeds meal(30.6%). The non protein nitrogen(NPN) contents in crude protein of main nitrogen materials were 2.4% for soybean meal and 5.6% for dried soybean curd residue, while those of wheat bran and rice bran used as the carbon source were relatively higher, 17.6% compared to that of nitrogen supplying media. Crude protein content per price was 6.0 for rapeseed meal, indicating that it is high crude protein content per price. Nitrogen-free extract(NFE) considering as an ingredient for mycelial growth were high in alphacorn(72.9%) and wheat bran B(57.2%). Acid detergent fiber(ADF) was high in corncob, 51.88%, its use for cultivation of brown rot fungi including Lentinus lepideus should pay attention because the fungi lack complete lignin degradation activity.

Optimization of extraction conditions of flavonoid compounds from Thyme (Thymus vulgaris Libiatae) (타임으로부터의 플라보노이드 화합물의 추출 조건 최적화)

  • Total polyphenol contents, flavonoid contents, 1,1-diphenyl-2-picrylhydrazyl (DPPH) free radical scavenging activities of ethanol extracts of 20 herbal plants (sage, turmeric, mace, bayleaf, fenugreek, oregano, blackpepper, whitepepper, clove, marjoram, cinnamon, coriander, basil, dillseed, mustard, cadamon, thyme, celery, rosemary, cumin) were analyzed for the screening of high flavonoid-containing plant resource. Thyme extract, showing the highest flavonoid content and a high degree of antioxidant activity, was selected as a bioactive cosmetic material. The total polyphenols and flavonoids contents of thyme extracts were measured as 6.90 mg chlorogenic acid equivalent (CAE)/100 g and 1.71 mg naringin equivalent (NE)/100 g, respectively, and DPPH radical scavenging activities was 90%. Among the tested organic solvents, hexane gave the highest extraction yield. Thus hexane was selected as the most suitable solvent for the extraction of thyme. Response surface method was used to obtain optimized extraction conditions for thyme: reaction temperature of 35.9 ℃, raw material to hexane ratio of 1.63:25 (w/v), and reaction time of 192 min. These predicted extraction conditions was validated by a total flavonoid extraction experiment showing a value equivalent to 96.3% of the predicted total flavonoid content. It is expected that the optimized solvent extraction conditions could be used for the production of flavonoid using thyme.

Improving Effect to Connitive Ability of Cordyceps militaris Extract in PC12 and BV2 cells (PC12와 BV2 세포에서 동충하초 추출물의 인지능력 개선 효과)

  • The aim of this study is to evaluate the efficacy of cordyceps militaris extracts for the improvement of cognitive dysfunction in PC12 and BV2 cells. Cordyceps militaris extracts was prepared by extracting with distilled water. Cell viability was assessed by MTT assay using PC12 cells and BV2 cells. Confirmed effects of L-glutamate induced cytotoxicity test, Acetylcoline (ACh) concentration, and Acetylcolinestase (AChE) activity in PC12 cells. Anti-inflammatory activities of cordyceps militaris extracts was measured through changes in the levels of nitric oxide (NO), and prostaglandin E2 ($PGE_2$) on lipopolysaccharide(LPS)-induced BV2 cell. In addition, we measured the expression of $NF-{\kappa}B$, p38, JNK, and caspase-3 in western blot analysis. Cordyceps militaris extracts showed no cytotoxicity at the concentrations of 1, 10, and $100{\mu}g/m{\ell}$ except for the concentration of $200{\mu}g/m{\ell}$. Cordyceps militaris extracts protected the cell and exhibited significant increases in the ACh concentration and a significant decrease in the AChE activity in L-glutamate induced PC12 cells. Moreover, cordyceps militaris extracts inhibited the productions NO, and PGE2 level and the protein expression of $NF-{\kappa}B$, p38, JNK, caspase-3 in LPS-induced BV2 cells. These results indicate that cordyceps militaris extracts possible prevented and improved cognitive dysfuction symptoms. Thus, cordyceps militaris extracts may be a novel natural material option for the improvement of cognitive dysfunction.

Effect of Seomaeyakssuk (Artemisia argyi H.) Extracts on Anti-pruritic Activities (항가려움증 활성에 섬애약쑥(Artemisia argyi H.) 추출물이 미치는 영향)

  • The purpose of this study was to investigate the anti-pruritic activities such as cell viability and pruritus-related factor using seomaeyakssuk (Artemisia argyi H.; Seomaeyakssuk) extract on MC/9 (mouse mast cell line). Seomaeyakssuk was extracted from hot distilled water. Cell viability was assessed using MTT assay on MC/9 cells. Anti-pruritic activities were measured through changes in the levels of transcription factor (IL4 and IL31) on MC/9 cells. In addition, the expression of linked proteins and histamine was measured. The results confirmed that significant cytotoxicity does not appear in the concentration range of 25, 50, and 100 ㎍/㎖. The levels of IL4 was reduced to 12% (25 ㎍/㎖), 26% (50 ㎍/㎖) and 61% (100 ㎍/㎖). Also, level of IL31 was decreased 33% (50 ㎍/㎖) and 36% (100 ㎍/㎖). In the case of proteins levels decreased significantly IL-4 34%/69% and IL-31 36%/37% at 50 ㎍/㎖ and 100 ㎍/㎖. Histamine decreased by 22, 58% and 61%, respectively, at concentrations of 25, 50, and 100 ㎍/㎖. This results shows possibility of ADD as raw material in anti-pruritic products.

Dietary Fiber and β-Glucan Contents of Sparassis crispa Fruit Fermented with Lactobacillus brevis and Monascus pilosus (유산균 및 홍국균 발효 꽃송이버섯 추출물과 잔사의 식이섬유와 베타 글루칸의 함량)

  • Sparassis (S.) crispa is an edible mushroom abundant in dietary fiber and ${\beta}$-glucan. The aim of this study was to prepare extracts and residues of the fruit bodies of S. crispa fermented with Lactobacillus (L.) brevis and Monascus (M.) pilosus and to measure the remaining dietary fiber and ${\beta}$-glucan. Dried powder of S. crispa containing 64.4 g/100 g total dietary fiber (2.6 g/100 g soluble and 61.8 g/100 g insoluble dietary fibers) and 24.0 g/100 g ${\beta}$-glucan was used as the starting material for the extraction. Raw and fermented S. crispa were extracted with hot water and three kinds of aqueous ethanol (50, 70, and 90%, v/v), respectively. A hot water extract from S. crispa fermented with M. pilosus had greater soluble dietary fiber content (19.3 g/100 g) than that from raw S. crispa with 14.6 g/100 g soluble dietary fiber or that from L. brevis-fermented S. crispa with 8.2 g/100 g soluble dietary fiber. The yield of the extract was 16.6% of intial weight of dried S. crispa. After hot water extraction of S. crispa fermented with M. pilosus, residues containing 90.5 g/100 g total dietary fiber (1.3 g/100 g soluble and 89.2 g/100 g insoluble dietary fibers) were obtained, and the yield was 69.6% of intial weight of dried S. crispa. The residue (31.0 g/100 g) contained more ${\beta}$-glucan than raw S. crispa or M. pilosus-fermented S. crispa (24.4 g/100 g). The resulting hot water extract and residue from S. crispa fermented with M. pilosus would be suitable for use in preparing liquid and powdered health functional foods, respectively.

Data Mining and Construction of Database Concerning Effects of Vitis Genus (산머루 관련 정보수집 및 데이터베이스의 구축)

  • The database for the oriental medicine had been existed in documentation in past times and it has been developed to the database type for random accesses in the information society. However, the aspects of the database are not so diversified and the database for the bio herbal material exists in widened type dictionary style. It is a situation that the database which handles the in-depth raw herbal medicines is not sufficient in its quantity and quality. Korean wild grape is a deciduous plant categorized into the Vitaceae and it was found experimentally that it has various medical effects. It is one of the medical materials with higher potentiality of academic study and commercialization recently because it has a bigger possibility to be applied into diverse industrial fields including the medical product for health, food and beauty. We constituted the cooperative system among the Muju cluster business group for Korean mountain wild grapes, Physiology Laboratory in Kyung Hee University Oriental Medicine and Medical Classics Laboratory in Kyung Hee University Oriental Medicine with a view to focusing on such potentiality and a database for Korean wild grapes was made a touchstone for establishing the in-depth database for the single bio medical materials. First of all, the literatures based on the North East Asia in ancient times had been categorized into the classical literature (Korean literature published by government organization, Korean classical literature, Chinese classical literature and classical literature fro Korean and Chinese oriental medicine) and modern literature (Modern literature for oriental medicine, modern literature for domestic and foreign herbal medicine) to cover the eastern and western research records and writings related to Korean wild grapes and the text-mining work has been performed through the cooperation system with the Medical Classics Laboratory in Kyung Hee University Oriental Medicine. First of all, the data for the experiment and theory for Korean wild grape were collected for the Medline database controlled by the Parliament Library of USA to arrange the domestic and foreign theses with topic for Korean wild grapes and the network hyperlink function and down load function were mounted for self-thesis searching function and active view based on the collected data. The thesis searching function provides various auxiliary functions and the searching is available according to the diverse searching/queries such as the name of sub species of Korean wild grape, the logical intersection index for the active ingredients, efficacy and elements. It was constituted for the researchers who design the Korean wild grape study to design of easier experiment. In addition, the data related to the patents for Korean wild grape which were collected from European Patent Office in response to the commercialization possibility and the system available for searching and view was established in the same viewpoint. Perl was used for the query programming and MS-SQL for database establishment and management in the designing of this database. Antioxidative and Nitrite Scavenging Abilities of Medicinal Plant Extracts (한약재 추출물의 항산화작용 및 아질산염 소거작용)

  • The purpose of this study was to investigate antioxidative and nitrite scavenging abilities of extracts of medicinal plants. Three kinds of medicinal plants, Angelica acutiloba Kitagawa (AA), Akebia quinate Decaisne (AQ), and Caragana chamlagu (CC), were extracted with water and $70\%$ ethanol. The yields of water extracts were increased in the order of Akebia quinate Decaisne ($35.63\%$, Caragana chamlagu ($31.36\%$ and Angelica acutiloba ($8.98\%$). Those of ethanol extracts were Akebia quinate Decaisne ($31.82\%$, Angelica acutiloba Kitagawa ($14.15\%$, and Caragana chamlagu ($5.20\%$). The highest electron donating ability (EDA) at 300-1,000 ppm of water extract was Angelica acutiloba Kitagawa, ranged from $62\%$ to $88\%$ and that of ethanol extract was Akebia quinate Decaisne, ranged from $72\%$ to $87\%$. Nitrite scavenging ability (NSA) of extracts measured at various pH (1.2, 3.0, 4.2, 6.0) showed highest ability in all extracts at pH 1.2 and decreased with increasing pH. The highest NSA of water extracts of 1,000 ppm at pH 1.2 showed $35-40\%$, in Caragana chamlagu and that of ethanol extract was $33-42\%$, in Akebia quinate Decaisne. From these results, Akebia quinate Decaisne was found as a potential raw material baring functionality in the respects of extract yield, EDA and NSA.

Bioactive Compounds, Anti-Oxidant Activities and Anti-Inflammatory Activities of Solvent Extracts from Centella asiatica Cultured in Chungju (충주산 병풀 용매추출물의 활성성분과 항산화 및 항염증 활성)

  • Centella asiatica (CA) has been widely used as herbal plants. It is a valuable resources. The aim of the present study was to evaluate physiological activities of solvent extracts from CA cultivated in Chungju, Korea (Good tiger care). After preparing water (cold-water, CA-WE; hot-water, CA-HWE) and EtOH extracts (50% EtOH, CA-50E; 70% EtOH, CA-70E), their total polyphenol, flavonoid, and triterpenoid contents, and anti-oxidant activities, and anti-inflammatory activities were examined and compared. CA-70E showed the most potent anti-oxidant activity based on ABTS radical scavenging activity and reducing ability assays, while CA-50E and CA-70E showed the highest DPPH radical scavenging activity. Among major triterpenoid glycosides present in CA, madecassoside and asiaticoside contents were found to be the highest in CA-70E, and madecassic acid and asiatic acid were the highest in CA-50E. In LPS-stimulated RAW 264.7 cells, CA-70E showed the highest TNF-α inhibitory activity, although CA-50E and CA-70E similarly inhibited nitric oxide production. Ethanol extracts significantly inhibited IL-6 and IL-8 production more than water extracts using TNF-α/IFN-γ-stimulated HaCaT cells, indicating its better inhibitory against skin inflammation. Therefore, Chungju-CA EtOH extract, especially 70% EtOH extract, has high physiologically active ingredients and potent anti-oxidant and anti-inflammatory activities, suggesting its industrial application as a functional material.
