국내 유통중인 약용작물의 생물학적 위해요소 모니터링 (Monitoring of Biological Hazards in Herbal Crops from Korean Market)

  • Background: The public has increasing concerns about herbal crops owing to insufficient information on biological hazards such as foodborne pathogens. Therefore, the objective of this study is the development of a herbal crop quality control system through monitoring with biological hazard analysis. Today, it is estimated that millions of people become ill every year from food contamination. The public demands agricultural products of stable and consistent quality. Governments have the responsibility of establishing the standards, legislation and enforcement programs necessary to control food quality and safety. However, research on the biosafety of herbal crop products is still insufficient. Therefore, the implementation of monitoring systems with high standards is critical for public safety. Methods and Results: In this study, we collected 52 samples of herbal crop products, and conducted both quantitative and qualitative biological hazard analysis. With biological hazard analysis, aerobic bacteria, Staphylococcus aureus, Salmonella spp., Escherichia coli, Coliforms, and Listeria spp. could be detected. Conclusions: Herbal crops were found to be contaminated with aerobic bacteria at $3.69{\pm}0.32log\;CFU/g$. Staphylococcus aureus, Salmonella spp., Escherichia coli, Coliforms, and Listeria spp. were not detected in any of the samples. This research suggests that continuous monitoring of biological hazards is required to improve the quality of herbal crops.

탕전실의 조제 관리 및 운영 기준에 대한 연구 (A Study on the Management and Dispensing Standards of Herbal Medicine in Herbal Dispensaries)

  • Objective : Herbal dispensaries, where medications are prepared for patients, require specific preparation and operations management for safe herbal medicines. This study aims to propose improvements in the herbal medicine preparation and operation of the herbal dispensaries as series study of the "A Survey on the Management Status of Extramural Herbal Dispensaries" Methods : We first checked current management regulations. In addition, we compared and analyzed related laws which are "State management norm of Traditional Chinese Medicine dispensaries", "Enforcement Rule of Medicinal Product Safety" and " Hazard Analysis and Critical Control Point(HACCP)", to introduce the assessment items of herbal dispenses evaluation and certification system. Results : In this study, we found that the current regulations, "Guideline on Installation and Utilization of Extremal Herbal Pharmaceutical Facility and Shared-use of Herbal Dispensary", are insufficient to ensure the quality and safety of herbal medicines. We suggested articles for the proper management of herbal dispensaries and these were introduced to the Herbal dispensary evaluation and certification system. Conclusion : We recommend evaluation articles in Herbal dispensary evaluation and certification systems need to be popular among herbal dispensaries, then Korean herbal medicine could restore credibility from the people.

목단피의 감별을 위한 ITS-PCR 분석 (ITS-PCR Analysis for the Discrimination of Moutan Cortex)

  • The application of PCR analysis on the herbal medicine Moutan Cortex (Paeonia suffruticosa Andrews) was evaluated by the comparison of the genetic relationship based on the DNA sequence with Paeoniae Radix (Paeonia lactiflora Pallas) following development of specific primers. Moutan Cortex and Paeoniae Radix were distinguished through the PCR analysis based on the internal transcribed spacer (ITS-PCR) from nuclear ribosomal DNA region. The 294 bp PCR products both of Moutan Cortex and Paeoniae Radix was amplified by MIF1 and MIR1. And a Moutan Cortex specific 225 bp PCR amplification product was amplified by MIF2 and MIR1 primers. The 225 bp sequence could be successfully amplified from Mortan Cortex of dried herbal preparations. PCR analysis based on ITS (ITS-PCR) may be an efficient tool for the discrimination of Moutan Cortex.

생약복합제 GCSB-5의 품질 표준화를 위한 오가피 및 우슬의 함량 분석 (Quantitative Analysis of Acanthopanacis Cortex and Achyranthis Radix for Standardization of GCSB-5 Preparation)

  • In this study, we have investigated the HPLC analysis methods and quantitative analysis of standard compounds for quality standardization of a medicinal crude drug GCSB-5, a herbal formulation consisting of 6 medicinal plants (Acanthopanacis Cortex, Achyranthis Radix, Ledebouriellae Radix, Cibotii Rhizoma, Glycine Semen, Eucommiae Cortex) which are used in traditional medicine to treat various bone disorders. HPLC analysis methods of acanthoside D(Acanthopanacis Cortex), 20-hydroxyecdysone(Achyranthis Radix) which were known standard compounds among 6 medicinal plants were developed on crude material and product. And validation of HPLC analysis methods were conformed for verification of HPLC methods by check to specificity, linearity, intra-day precision, inter-day precision and accuracy following ICH guideline. Content of acanthoside D and 20-hydroxyecdysone on raw material of GCSB-5 were decided at 0.577-0.578 mg/g and 0.311-0.312 mg/g. And we confirmed that content of acanthoside D and 20-hydroxyecdysone on GCSB-5 preparation were 0.302-0.303 mg/capsule and 0.113-0.115 mg/capsule.

HF-IFF: TF-IDF를 응용한 병증-본초 연관성(relevancy) 측정과 본초 특성의 시각화 -청강의감 방제를 대상으로- (HF-IFF: Applying TF-IDF to Measure Symptom-Medicinal Herb Relevancy and Visualize Medicinal Herb Characteristics - Studying Formulations in Cheongkangeuigam -)

  • Objectives : We applied the term weighting method used in the field of data search to quantify relevancy between symptoms and medicinal herbs, and, based on this, we aim to introduce a method of visualizing the characteristics of medicinal herbs. Methods : We proposed HF-IFF, an adaptation of TF-IDF, which is a term weighting measurement method adapted in the field of data search. Using this method, we deduced relevancy between symptoms and medicinal herbs In Cheongkangeuigam that was published in 1984 by organizing the medical theory of Cheongkang, Kim Younghoon, and visualized this as a graph in order to compare the characteristics of medicinal herbs used for different symptoms. Results : HF-IFF is the product of HF and IFF, where HF is the frequency of the relevant medicinal herb for a set of symptoms, and IFF is the inverse of the number of formulations (FF) containing that herb. A total of 251 types of medicinal herb are used in Cheongkangeuigam, and 1538 formulations are classified according to 67 types of symptom. The overall mean for HF-IFF was 0.491, with a maximum of 4.566 and a minimum of 0.013. Conclusions : In spite of several limitations, we were able to use HF-IFF to measure relevancy between symptoms and medicinal herbs, with formulations as an intermediate. We were able to use the quantified results to visually express the characteristics of the herbs used for symptoms by bubble chart and word-cloud from HF-IFF.

찔레뿌리 추출물을 주성분으로 하는 천연물 조성물의 발모촉진 효과에 관한 전임상 및 임상시험 연구 (The Preclinical and Clinical Effects of Herbal Product Containing Rosa mutiflora Roots Extracts as a Main Component on the Hair Growth Promotion)

  • In this study, to develop hair growth agents using natural products which has excellent ability to promote hair growth effect and fewer side effect, animal experiment and clinical trials was performed to evaluate hair growth promotion effect of herbal product containing $Rosa$ $mutiflora$ roots extracts as a main component (RMHP). 4-week-old male C57BL/6 mice were removed the dorsal hair using thioglycolate, and applied 15% EtOH solution as a negative control, 5% minoxidil as a positive control and RMHP to dorsal skin. In the results of macroscopy and photo-interpretation, RMHP group recorded 100% (+++++) of hair growth was proved to significantly stimulate hair growth against 80% (++++) negative control group. 37 patients were treated with RMHP and evaluated the therapeutic effect at 16 weeks. Hair density was significantly increased at 16 weeks after applying RMHP ($125.0{\pm}4.9\;FU/cm^2$) compared to before treatment ($104.3{\pm}4.7\;FU/cm^2$, p < 0.05), and hair thickness were also significantly increased ($0.066{\pm}0.003$ mm) than before treatment ($0.055{\pm}0.002$ mm, p < 0.05). The result of clinical photo-interpretation using 7-point rating scale assessment, after 16 weeks clinical symptoms were evaluated to significantly improve with $1.23{\pm}0.05$ (p < 0.05). Therefore, the results of this study were observed that RMHP have hair loss prevention effect and hair growth promotion effect to hair loss patients.

  • Good Laboratory Practice(GLP) is becoming more and more important in the research and development of Oriental Pharmacy(OP) and its globalization. If a OP product is to be registered as Over-the-Counter(OTC) drug and enter international markets, the safety and efficacy studies conducted according to GLP requirements is necessary. The article introduces the content of GLP requirements and the recent development of GLP. The safety and efficacy assessment for OP or herbal medicines under GLP are also covered. This paper also briefly describes the areas that should be covered by GLP regulation and the areas that do not need to follow GLP requirements.

KHP 기준에 의한 보험용 단미엑스산제의 지표성분함량 확인연구 (A quantitative analysis of marker compounds in single herb extracts by the standard of KHP)

  • Objectives : Since single extract powders was released at 1987, the insurance fee has not been changed, but the price of raw material has been increasing. According to this, Pharmaceutical company couldn't invest on quality of the product, so, the quality of single extracts went down and lost the consumer's trust. We checked the contents of marker ingredients in single extract thereby to recover the reliability of insurance-covered herbal preparations. Methods : we bought total twelve products of eight different single extracts of two main pharmaceutical companies among total 65 kinds of single extract powder stipulated in KHP (The Korean Herbal Pharmacopoeia) monograph III at Jan. 2013. Assays of selected single extract powders are performed by KHP regulation. And we surveyed price fluctuation of relevant herb raw materials from 2005 to 2012. Results : Among twelve products, eight single extract powders were suitable by the KHP regulation. But four products didn't reach the content amount of KHP. Marker contents in the single extracts product of Pueraria Root, Licorice, Peony root and Scutellaria Root of A company were 70%, 1%, 23.7% and 75.1%, respectively. Conclusions : We can acertain whether there's a quality problem in the insurance-covered single extract powders. But, A company is no longer producing these improper single extract powders. As a medicine, single extract powders needs to be strictly quality controlled by the company, and regularly monitored by the KFDA.

Multiplex Polymerase Chain Reaction을 이용한 당귀 종 판별 (Development of Multiplex Polymerase Chain Reaction Assay for Identification of Angelica Species)

  • Background: Angelica gigas, A. sinensis, and A. acutiloba are commercially important in the herbal medicine market, and among them, A. gigas has the highest economic value and price. However, their similar morphological traits are often used for fraud. Despite their importance in herbal medicine, recognition of the differences between Angelica species is currently inadequate. Methods and Results: A multiplex polymerase chain reaction (PCR) method was developed for direct detection and identification of A. gigas, A. sinensis, and A. acutiloba. The gene for the distinction of species was targeted at ITS in the nucleus and trnC-petN gene in chloroplasts. The optimized multiplex PCR in the present study utilized each Angelica species-specific primer pairs. Each primer pair yielded products of 229 base pairs (bp) for A. gigas, 53 bp for A. sinensis, 170 bp for A. acutiloba. Additionally non-specific PCR products were not detected in similar species by species-specific primers. Conclusions: In the present study, a multiplex-PCR assay, successfully assessed the authenticity of Angelica species (A. gigas, A. sinensis, and A. acutiloba). and whole genome amplification (WGA) was performed after DNA extraction to identify, the species in the product. The detection method of raw materials developed in the present study could be applied to herbal medicine and health functional food management.
