Anti-oxidative and Anti-inflammatory Effect of 7 Herbal Extracts and Methods of Herbal Formula Compositioning (7종 한약재 추출물의 항산화 및 항염증 효과와 이를 이용한 방제 구성에 대한 고찰)

  • Objectives : Herbal formulas are complicated to analyze and difficult to compose because of its mixed, complex features. So we discussed about how to compose herbal formulas effective and efficient by analyzing various effectiveness of each herbal extracts. Methods : To evaluate the effectiveness of herbal formula, anti-oxidative and anti-inflammatory effectiveness were mathematically analyzed. DPPH, superoxide anions, Nitric Oxide scavenging activity was measured to evaluate the effectiveness of 7 herbal extracts. And next, cytotoxic activity of extracts on RAW 264.7 cells were measured using MTS assay. To asses anti-inflammatory effect, nitric oxide and $PGE_2$ production were measured. Based on these anti-oxidative and anti-inflammatory experiment result, mathematical analyzation were carried out with constOptm function, and determined efficacy-maximizing ratio. Results : This mathematical analysis based formula showed significantly outstanding effectiveness than other formulae. And estimated tendency of anti-inflammatory effectiveness was matched with real effectiveness. Conclusions : So, mathematical analysis can be available to evaluate and estimate the effectiveness of herbal formulae.

Stimulation of the Immune Response by Yin-Tonifying Formula (대표적인 보음지제(補陰之劑)의 면역 활성화 비교 연구 - 육미지황탕, 자음강화탕, 쌍화탕 -)

  • Objectives: Three yin-tonifying formulae (Ssanghwa-tang, Yukmijihwang-tang and Jaeumganghwa-tang) were applied to investigate their immunological activities on antigen (Ag)-specific or Ag-non-specific immune responses in the murine macrophage cell line (RAW 264.7) and in ovalbumin (OVA)-immunized mice. Methods: This study was carried out in nitricoxide (NO) synthesis in RAW 264.7 cells and cellular proliferation in mouse splenocytes in association with three herbal formulas. C57BL/6 mice were immunized intraperitoneally with OVA/aluminum ($100\;{\mu}g/200\;{\mu}g$/mouse) on days 1, 8, and 15. Three herbal formulas were administrated to mice orally for 3 weeks from day 1. On day 22, OVA-, lipopolysaccharide (LPS)-, and concanavalin A (Con A)-stimulated splenocyte proliferation and antibodies (OVA-specific antibodies of the IgG, IgG1, and total IgM classes) in plasma were measured. Results: All three yin-tonifying formulas significantly enhanced cellular proliferation by LPS and Con A in splenocytes from OVA-immunized mice (p<0.001). Also, these herbal formulas all significantly enhanced plasma OVA-specific IgG, IgG1, and total IgM levels compared with the OVA/Alum group. Conclusion: These results suggested that the three yin-tonifying formulae could be used as stimulators of immune response.

Literature Review of Herbal Medicines on Treatment of Obesity Since 2000;Mainly about Ephedra Herba (2000년 이후 비만치료에 사용되는 처방(處方) 및 본초(本草)에 대한 문헌연구;마황(麻黃)을 중심으로)

  • Objectives : This study has been done to know which herbs and herbal formulae are frequently used for the treatment of obesity in Korea and overseas since 2000. Methods : We conducted a computerized literature search in KISS (Korean studies Information Service System), Pubmed, Journal of Oriental Association for Study of Obesity ,and Journal of Oriental Rehabilitation Medicine(2000 -Apr. 2006) using search key words obesity, obese, weight loss, weight control combined with herb, herbal formula. Result and conclusion : Seventy-two studies have been identified(50 Korean papers and 22 pubmed papers) and 81 herbs have been found. In USA it was mainly used as single herb for food, but in Korea it was mainly used as herbal formula. For single herb, ephedra(麻黃) was the most studied single herb both in Korea and the USA. For herbal formulae, Sasang Constitutional Medicine(SCM, 19times) was the most frequently used herbal formula, in which the herbs for Tae-eum type person like Taeyeumjowui-tang(太陰調胃湯, 7times) and Choweseungcheng-tang(調胃升淸湯, 5times) were mainly prescribed. The next was Chegameuiin-tang(體減薏苡仁湯, 11times) and Bangpungtongsungsan( 防風通聖散, 3times). Calculating each herb consisted in each herbal formula, Coicis Semen(薏苡仁), Angelicae gigantis Radix(當歸), and Glycyrrhizae Radix(甘草), Raspberry Ketone, Cnidii Rhizoma(川芎), and Ephedra Herba(麻黃) turned out to be the most frequently used herb.

Analysis of the low-chapter prescription in 『Bangyakhappyun』 (『방약합편(方藥合編)』 하통(下統) 처방(處方)의 방제분석에 대한 고찰(考察))

  • Objectivs : This report describes the studies as follows through the research of 163prescribtions in low-chapter from Bangyakhappyun. Methods : Analysis the 163prescribtions of basic prescribtions, therapeutic area, symptoms and pathology. Result : Many prescriptions are contained in the diseases areas such as defecate diseases, women diseases, internal diseases, exogenous febrile diseases. And prescriptions in the low-chapter are used in the areas such as eye, aggregation, mouth and tongue, teeth, hand, back, neck, emergency. Eighty-five basic prescriptions are analyzed from the low-chapter and the ratio of nineteen prescriptions containing Ijintang, Pyeongwisan, Oryungsan, Samultang, Sunggitang, Jichultang are over 60%. The efficacy classifications containing basic prescriptions frequently used are researched in the following order: heat-clearing and fire-purging formulae, digestant formulae, phlegm-dispelling formulae, qi-regulating formulae and blood-activating formulae. And tonifying and replenishing formulae are rarely used. Most of the prescriptions in the low-chapter are used for the excess syndrome and few of them are used for the deficiency syndrome. And six excesses, food damage, phlegm are the major causes of excess syndrome. Conclusions : As s result of study on analysis all prescriptions in low-chapter from Bangyakhappyun. We can understand more about basic prescriptions, symptoms of prescriptions and pathology.

The Application of High Performance Thin Layer Chromatography for Herbal Formula Standardization (한약처방의 표준화를 위한 HPTLC의 활용)

  • Objective: The purpose of this study is to expatiate on high performance thin layer chromatography (HPTLC) as a simple, easy and scientific method for evaluation and standardization of herbal formulae. Methods: Through retrieving HPTLC application for herbal formulae in the literatures, the current situation of HPTLC application, and potential as well as limitation of HPTLC as a standardization method for multi-herbal drug was studied. Results: HPTLC is a speedy, inexpensive and well-operable tool for possessing multi-capability on component identification, separation, quantification and purification compared to other methods, such as high performance liquid chromatography (HPLC), gas chromatography (GC). Conclusions: HPTLC is considered as an available and convenient method for quality control and standardization of multi-herbal drugs. Thereby, this method could be recommended to widely applicate in the traditional Korean medicine.

Effect of Bojungikgitang on Starvation Stress in Mice (보중익기탕(補中益氣湯)이 생쥐의 기아(飢餓)stress에 미치는 영향)

  • Objectives: We investigated the effect of Bojungikgitang on the change of the corticosterone and the rectal temperature of the mice induced by starvation stress. Methods: After administration of Bojungikgitang three times, mice were starved. The corticosterone and rectal temperature were measured after 36.5 hours starvation stress. Results : The plasma corticosterone level in Bojungikgitang(0.25g/Kg) was decreased comparing with the Control group and the rectal temperature in the Bojungikgitang(1.0g/Kg) was increased comparing with the Control group. Conclusion : it might be recognized that Bojungikgitang has anti starvation stress effect, and also it might be needed further study in various viewpoints.

Effects of Lycii Radicis Cortex in HPA-Axis System on the Forced Swimming Test (지골피(地骨皮)가 강제수영부하실험에서 HPA-Axis System에 미치는 영향)

  • In order to investigate the antidepressant effects of Lycii Radicis Cortex on the change of HPA-Axis system, the forced swimming test was performed. The expressions of CRF and c-Fos in the PVN were measured with immunohistochemical method and the concentration of ACTH in Serum was evaluated with ELISA method. And the results obtained were as follows. Results : 1. The duration of immobility in the forced swimming test was significantly decreased in the LRC100 group and the LRC400 group(P<0.001). 2. The expressions of CRF and c-Fos were significantly reduced in the LRC100 group and the LRC400 group(P<0.001). And the concentration of ACTH in Serum were significantly reduced in the LRC 100 group(P<0.05). According to the results above mentioned, it can be considered that Lycii Radicis Cortex has antidepressant effects.

Effects of three different formulae of Gamisoyosan on lipid accumulation induced by oleic acid in HepG2 cells

  • Background: Gamisoyosan (GSS) is an herbal formula which has been used to treat women's diseases for several hundred years in Korea. GSS is one of the three most common prescriptions among women and is used to treat menopausal symptoms. Fatty liver disease is also common in postmenopausal women and can precede more severe diseases, such as steatohepatitis. The present study compared the effects of GSS on fatty liver using three different formulae, Dongui-Bogam (KIOM A), Korean Pharmacopeia (KIOM B) and Korean National Health Insurance (KIOM C). Methods: In oleic acid-induced HepG2 fatty liver cells, cellular lipid accumulation, triglycerides and total cholesterol were measured after treatment with three GSS formulae and simvastatin as a positive control. To investigate the phytoestrogen activity of GSS, MCF-7 cells were treated with GSS, and hormone levels were quantified. Also, qualitative analysis was performed with UPLC. Results: All types of GSS decreased cellular lipid accumulation. KIOM A was slightly less effective than the other two GSS formulae. KIOM B and KIOM C decreased cellular triglycerides more effective than simvastatin, but KIOM A did not affect cellular triglycerides. Cellular total cholesterol was decreased by all GSS and simvastatin. GSS showed phytoestrogen activity in MCF-7 cells. From the UPLC analysis data, geniposide, paeoniflorin and glycyrrhizin were detected form three GSS formulae. Conclusion: These results suggest that all GSS formulae have a beneficial effect on fatty liver disease during menopause and that differences of formula have no effect on the efficacy of the prescription.

A Case Report of Tinnitus Treated by Socheongryong-tang based on Shanghanlun Provisions (『상한론(傷寒論)』 변병진단체계(辨病診斷體系)에 근거하여 소청룡탕(小靑龍湯) 투여 후 호전된 이명(耳鳴) 증례 1례)

  • Objective : The purpose of this study was to report the improvement of a patient with tinnitus treated by herb-formulae medication based on Shanghanlun provisions. Methods : According to a disease pattern identification diagnostic system based on Shanghanlun provisions, a patient was diagnosed with Taeyang-byung, 40th provision and was administered Socheongryong-tang herbal medication for 45 days. The numeric rating scale (NRS) was used to estimate the response. Results : The NRS score changed from 10 to 0 and the tinnitus disappeared by 40th provision of Socheongryong-tang selected according to Shanghanlun provisions. Conclusions : This case report suggests that the two words in the 40th provision of Shanghanlun, '表不解, 水氣'(exterior do not release, water qi) indicate a close relationship between economic difficulty and stuffiness of being in water, which affect tinnitus in this case.

Textual Research : Indication and Dosage of Panax Ginseng during Lactation (모유수유 중 인삼의 적응증과 용량에 관한 기존한약서 고찰)

  • Purpose: To provide basic informations for guidelines of Ginseng usage during lactation. Method: Based on a regulation of scope and preparation of herbal prescriptions by Ministry of health and welfare of Korea, we selected the formulae for postpartum care and disease in 7 Korean Medical Classics. And we searched the number of formulae including Ginseng, dosage and indications of Ginseng in formulae in those books. Results: The range of Ginseng dosage during lactation from medical classics is 1.5-18.75g/day for medical purposes. Indications of Ginseng are lethargy, excessive bleeding, asthma, fever, cold, pain, dizziness, mental disorder, spasm, digestive problem, constipation, diarrhea, urinary incontinence, edema, breast engorgement, lack of breastmilk, breast ulcer, etc. But Ginseng had not been used in the case of stroke, epistaxis, beginning of mastitis, tumor of lower abdomen. Conclusions: Ginseng should be recommended less than 1.5g/d as a nutritional supplement. If breastfeeding woman wants to take Ginseng as a medication or as a tonic, she should consult with a official specialist about proper prescription and dosage instead of risky random medication.
