Inhibition of Hepatic Triglyceride Accumulation and Stimulation of Alcohol Metabolism by the Herbal Extract Containing Phaseoli radiati semen in Rats Fed Ethanol (급성 알콜 투여 흰쥐에서 녹두 함유 복합생약제제의 간 중성지방 축적억제 및 알콜대사 촉진 효과)

  • An ethanol administration causes hepatic triglyceride accumulation in rats. To assess whether the herbal extract containing Phaseoli radiati semen(herbal extract) inhibit s the triglyceride accumulation in the liver, we determined the hepatic triglyceiide levels in rats fed ethanol and the herbal extract. In addition, the blood ethanol concentrations and the activities of hepatic alcohol dehydrogenase(ADH) and aldehyde dehydrogenase(ALDH) were measured to determine the effects of the herbal extract on alcohol metabolism in rats. The administration of the herbal extract markedly reduced the triglyceride levels elevated by ethanol in the liver as well as in the serum. The herbal extract remarkably lowered blood ethanol concentrations in a dose-dependent manner. The ADH activities decreased by ethanol were recovered to the normal level by the herbal extract treatment. Moreover, the ALDH activities slightly decreased by ethanol increased beyond the normal level by the herbal extract treatment. We conclude that the herbal extract inhibits the hepatic triglyceride accumulation and stimulates alcohol metabolism by preventing ADH and ALDH from inhbition by the ethanol administration in the rat liver.

  • Objective : This experimental study was performed to investigate the effect of Hwangryunhaedoktang Herbal-acupuncture & extract on Staphylococcus aureus and Staphylococcus epidermidis that induce keratitis. Methods : MIC was measured by dropping to $50{\beta}^{\circ}$ according to density Hwangryunhaedoktang Herbal-acupuncture & extract(100%, 10%, 1%, 0.1%). Anti-bacterial potency was measured by the size of inhibition zone With change of volume. Results : 1. MIC on Staphylococcus aureus in Hwangryunhaedoktang Herbal-acupuncture & extract were not disappear all sample. 2. MIC on Staphylococcus epidermidis in Hwangryunhaedoktang Herbal-acupuncture & extract were not disappear all sample. Conclusions : The present author think that Hwangryunhaedoktang Herbal-acupuncture must be extract a new method for anti-bacterial potency.

Antimicrobial activity and inhibition effect on the artificial dental plaque formation of herbal medicine extract (생약 추출물의 항균력 및 인공치태형성 억제효과)

  • This study was executed to evaluate the effects of natural herbal medicine extract on the antimicrobial activity and the artificial plaque formation. Schisandra chinensis, Sophora flavescens, Dryopteris crassirhizoma and Aristolochia contorta were extracted by 80% MeOH respectively and each concentration(5mg, 10mg, 20mg) of herbal medicine extract was tested to show the antimicrobial activity against S. mutans. To show the inhibition effect of herbal medicine extract on the artificial dental plaque formation each extract of 0.4% concentration was added into M 17 broth and inoculated with $5.0{\times}10^6$ of S. mutans. After 24 hour incubation each weight of artificial dental plaque was measured and compared with each other. Results obtained are as follows: 1. Dryopteris crassirhizoma showed very intense antimicrobial effect but Schisandra chinensis, Sophora flavescens and Aristolochia contorta showed a little difference against control. 2. Regarding artificial plaque formation inhibiting activity by each herbal extract at 0.4% concentration, Dryopteris crassirhizoma showed the perfect inhibition effect. Schisandra chinensis and Aristolochia contorta followed in order but Sophora flavescens showed the worst value. In conclusion, Dryopteris crassirhizoma showed strong antimicrobial effect against S. mutans and significant inhibiting effect on the artificial dental plaque formation.

Single Oral Dose Toxicity Test of Fermented Samchulgeonbi-tang Extract in ICR mice (ICR 마우스를 이용한 발효삼출건비탕의 단회투여 독성에 대한 연구)

  • Objectives : Samchulgeonbi-tang (shenzhujianpi-tang) has been prescribed as one of traditional herbal medicine for treatment of stomach diseases since ancient time in Korea. Samchulgeonbi-tang extract was fermented by Lactobacillus spp. for improving the effect. However, the toxicity and safety of fermented Samchulgeonbi-tang (FS) extract were not confirmed. Therefore, this study was performed to evaluate the acute toxicity and safety of FS extract. Methods : To evaluate the acute toxicity and safety of FS extract, several doses of FS extract, 0, 500, 1000 and 2000 mg/kg, were orally administered to 20 male and 20 female ICR mice, respectively. After treatment with FS extract, we observed mortality, general toxicity, behavior and change of body weight for the 14 days. After 14 days of oral administration, all mice were sacrificed and hematological parameters were analyzed from blood serum. Results : In present study, the toxic signs such as mortality or abnormal behaviors by FS extract were not observed. There are no significant differences between FS-treated group and control group in body weight, organ weights, and hematological parameters. Conclusions : The remarkable adverse effects by FS extract were not observed in ICR mice. Also, any death was not occurred at all treated FS doses, 500, 1000 and 2000 mg/kg. Therefore, the approximate lethal dose (ALD) of FS extract may be more than 2000 mg/kg.

Effects of Herbal treatment for In-patients with Mild Fever: Retrospective Clinical Study

  • Objectives: This study was designed to investigate the effect of herbal extract medicines for inpatients with fever symptoms in an oriental medicine hospital. Methods: Medical records of inpatients who experienced over $38^{\circ}C$ of fever from July 2010 to August 2011 has been requested with deleted personal identifiable information. The requested data were analyzed by patients' general characteristics, administrated western/herbal medicines, and changes in the body temperature of four administration groups (group 1, herbal extract medicine only; group 2, western medication(antibiotics/antipyretics) only; group 3, combination of herbal extract medicine and western medication; group 4, no medication control). The SPSS 19.0 was used for statistical analysis and p-value of less than 0.05 was regarded significant. Results: The body temperature has significantly dropped over time in the herbal extract medicine only group (p<0.001) along with the antibiotics/antipyretics only group. When both treatments were combined, stronger antipyretic effect was shown compared to the sole treatment of herbal extract medicine or antibiotics/antipyretics. Conclusion: The use of herbal extract medicines may be effective in alleviation of fever.

A quantitative analysis of marker compounds in single herb extracts by the standard of KHP (KHP 기준에 의한 보험용 단미엑스산제의 지표성분함량 확인연구)

  • Objectives : Since single extract powders was released at 1987, the insurance fee has not been changed, but the price of raw material has been increasing. According to this, Pharmaceutical company couldn't invest on quality of the product, so, the quality of single extracts went down and lost the consumer's trust. We checked the contents of marker ingredients in single extract thereby to recover the reliability of insurance-covered herbal preparations. Methods : we bought total twelve products of eight different single extracts of two main pharmaceutical companies among total 65 kinds of single extract powder stipulated in KHP (The Korean Herbal Pharmacopoeia) monograph III at Jan. 2013. Assays of selected single extract powders are performed by KHP regulation. And we surveyed price fluctuation of relevant herb raw materials from 2005 to 2012. Results : Among twelve products, eight single extract powders were suitable by the KHP regulation. But four products didn't reach the content amount of KHP. Marker contents in the single extracts product of Pueraria Root, Licorice, Peony root and Scutellaria Root of A company were 70%, 1%, 23.7% and 75.1%, respectively. Conclusions : We can acertain whether there's a quality problem in the insurance-covered single extract powders. But, A company is no longer producing these improper single extract powders. As a medicine, single extract powders needs to be strictly quality controlled by the company, and regularly monitored by the KFDA.

A study on the effects of herbal acupuncture with Liriopis Tuber extract on airway inflammation in the mouse induced with bronchial asthma

  • Objectives: Herbal acupuncture has been administered with Liriopis Tuber extract on the point of BL 13 (Pyesu) to treat bronchial asthma and a certain degree of clinical benefits have been observed but lacking scientific substantiation. Methods: The present report describes on Th1 cytokine (Interleukin-2, Interferon-gamma), Th2 cytokine, (Interleukin-4, Interleukin-5), and IL-12 in bronchoalveolar lavage fluid (ELISA). Five groups were devised to study the effects of herbal acupuncture with Liriopis Tuber extract at BL 13 (Pyesu) for airway inflammation in the mouse model with bronchial asthma. Results shows that herbal acupuncture with Liriopis Tuber extract at BL 13 increased Th1 cytokine (Interleukin-2) in allergic sensitization and allergic challenge, and decreased Th2 cytokine (Interleukin-2, Interleukin-5) in allergic sensitization.

Antioxidant Activities of Ulmi cortex Extracts According to Ethanol Contents (에탄올 함량변화에 따른 유백피 추출물의 항산화 활성)

  • Objectives : This study was performed to find best extraction solvent for application of Ulmi cortex to food or herbal medicine as an antioxidant only using water, ethanol and their mixtures. Methods : The Ulmi cortex extracts were prepared using water and 10%, 20%, 30%, 40%, 50%, 60%, 70%, 80%, 90% and 100% (v/v) ethanol, and were evaluated yields, total polyphenol contents, DPPH and ABTS radical scavenging activities, lipid peroxidation activities, and catechin and epicatechin contents. Results : Among the Ulmi cortex extracts, the yield was highest in water extract (8.9%) and lowest in ethanol extract (3.8%). The yield of 30% ethanol extract (8.5%) also was very high to similar with water extract. The total polyphenol content was highest in the 30% ethanol extract ($253.6{\mu}g/mg$ extract) and lowest in water extract ($109.0{\mu}g/mg$ extract). The DPPH radical scavenging activity was highest in ethanol extract (IC50, $8.53{\mu}g/ml$), ABTS radical scavenging activity was highest in 60% ethanol extract (IC50, $3.08{\mu}g/ml$), and the inhibition of lipid peroxidation was highest in 70% ethanol extract (IC50, $7.96{\mu}g/ml$). As ethanol content of extraction solvent increased from 0% to 30%, the antioxidant activities were remarkably increased whereas from 30% to 100%, the antioxidant activities were increased or decreased a little. Conclusions : The findings of the present study suggest that 30% ethanol is best solvent for extraction of Ulmi cortex, considering yield, polyphenol content, and antioxidant activities with extraction cost.

Effect of Cinnamomi Ramulus Herbal Acupuncture on the Type 1 Hypersensitivity (계지약침(桂枝藥鍼)이 Type 1 Hypersensitivity에 미치는 영향)

  • Objectives : We studied anti-allergic effects of Cinnamomi Ramulus(CR) herbal acupuncture and Cinnamomi Ramulus extract. Methods : In vivo, animals were herbal-acupunctured with CR at both ST36s three times for 5 days. Then, we induced active systemic anaphylatic shock using compound 48/80 in ICR mice, and passive cutaneous anaphylaxis using anti-DNP IgE in Sprague Dawley rat. In vitro, we measured cell viability, ${\beta}$ -hexosaminidase release and the expressions of IL-4, TNF-${\alpha}$ and COX-2 mRNA in RBL-2H3 cells after treatment of various concentrations of CR extract. Results : In vivo, CR herbal acupuncture pretreatments at both ST36s inhibited compound 48/80-induced active systemic anaphylatic shock. Passive cutaneous anaphylaxis was inhibited by CR herbal acupuncture pretreatments at both ST36s and optional points. In vitro, CR extract treatments did not affect on cell viability and inhibited ${\beta}$-hexosaminidase release. CR extract treatments also decreased the expressions of IL-4, TNF-${\alpha}$ and COX-2 mRNA in RBL-2H3 cells. Conclusions : These results suggest that CR herbal acupuncture and CR extract should be beneficial in the inhibition of allergic inflammatory response.

Effect of Root Extract of Lythrum salicaria L. on Liver Function of Rat Acutely Administrated with Alcohol (털부처꽃 뿌리 추출물의 단회투여가 급성 알코올 투여 흰쥐의 간기능에 미치는 영향)

  • This experiment was conducted to verify whether one time feeding of Lythrum salicaria root crude extract (LSR extract) exhibits liver protecting activities in acutely ethanol administrated rat. Experiment groups were composed of normal, negative control (alcohol control), 2 positive control (Hovenia dulcis extract 900 mg/kg and milk thisle 100 mg/kg), betulinic acid (20 mg/kg), a compound separated from LSR, and 3 LSR (100, 300, 900 mg/kg) groups. LSR treated groups showed decrease (p < 0.05) in serum triglyceride by dose-dependent manner. The content of serum albumin and the activity of ADH and ALDH in LSR extract fed rats were increased (p < 0.05) dependently on the administration amounts. Our study indicated that one time supplement of LSR downregulates oxidative stress and shows liver protective activity in the acute alcohol-fed rats.
