흰쥐의 합곡부위(合谷部位)에 황련약침(黃連藥鍼)이 TNBS로 유발된 대장염(大腸炎)에 미치는 영향(影響) (Effects of Rhizoma Coptidis Herbal Acupuncture applied to HapGok $(LI_4)$ on TNBS-induced Colitis in rats)

  • Objectives : Ulcerative colitis and Crohn's diseases are chronic inflammatory disease of the gastrointestinal tract identified mainly for diarrhea, stomachache, hemafecia. The source and pathologic mechanism about ulcerative colitis and Crohn's disease are still unknown but stress, infection, a genetic factor, environmental factors etc. may be the cause of chronic inflammatory disease of the gastrointestinal tract recently. Methods : Because ulcerative colitis and Crohn's disease is recognized as Ha-ri(下痢) or Jang-Byok(腸?) in Oriental Medicine, Rhizoma Coptidis Herbal acupunctures were injected in hapgok $(LI_4)$ which is used to treat intestinal disease in clinic. All animals were subjected to the injection of saline $(300\;{\mu}l,\;500\;{\mu}l)$ for a study control and TNBS $(300\;{\mu}l,\;500\;{\mu}l)$ into the lumen of the colon, 8cm proximal to the anus through the intestine. Rhizoma Coptidis Herbal (20 mg/ml, 0.4 ml) acupuncture solution was injected to the $LI_4$ (both hands) at the secondary injection time of TNBS in rats. And body weight, RBC count, WBC count, total protein, Paw edema rate, rate of protein leakage into CMC-pouch fluid, IgG levels and IgM levels were observed to study the effects of Rhizoma Coptidis Herbal acupuncture in hapgok $(LI_4)$. Results : In results, Rhizoma Coptidis Herbal acupuncture in hapgok $(LI_4)$ on TNBS-induced colitis reduced the rate of weight loss but had no effect on RBC and WBC count. And it reduced total protein concentration, paw edema, rate of protein leakage into CMC-pouch fluid, IgG levels and IgM levels. Conclusion : Data suggest that injection of Rhizoma Coptidis herbal acupuncture solution into hapgok has significant therapeutic effect on the colitis.

족삼리(足三里)에 시술(施術)한 한연초(旱蓮草) 약침(藥鍼)의 면역증진(免疫增進)과 항암작용(抗癌作用)에 관한 연구(硏究) (The effect of anti-cancer and Immune response improvement of ELP(Eclipta prostrata) Herbal-Acupuncture into Chok-samni(St36))

  • To study the effects of anti-cancer, anti-metastasis and immune response improvement of herbal-acupuncture with Eclipta prostrata diffuse herba infusion solution(ELP-HAS), we injected ELP-HAS into Chok-samni(St36) of C57BL/6 mice implanted intravenously with B16-F10 melanoma. We have reached the following conclusion through the effect on the number of $CD25^+/CD4^+$, $CD8^+/CD3e^+$, $CD69^+/B220^+$, $NK1.1+/CD3e^+$ cells in mouse PBMCs, the effect on the pulmonary colony number, and the effect on MST(Median Survival Time) and ILS(Increase of Life Span) of C57BL/6 mice implanted intravenously with B16-F10 melanoma The results were obtained as follows: 1. In the experiment groups treated with ELP(Eclipta prostrata) Herbal-Acupuncture, the spleen cell proliferation in Balb/c mouse was significantly increased compared with control group. 2. In the experiment groups treated with ELP(Eclipta prostrata) Herbal-Acupuncture, the percentage of $CD25^+/CD4^+$, $CD8^+/CD3e^+$, $CD69^+/B220^+$, $NK1.1^+/CD3e^+$ cells in C57Bl/6 mouse PBMCs was increased compared with control group. 3. In the experiment groups treated with ELP(Eclipta prostrata) Herbal-Acupuncture, the pulmonary colony number of C57BL/6 mice implanted intravenously with B16-F10 melanoma was decreased significantly compared with control group. 4. In the experiment groups treated with ELP(Eclipta prostrata)Herbal-Acupuncture, MST(Median Survival Time) and ILS(Increase of Life Span) of C57BL/6 mice implanted intravenously with B16-F10 melanoma were increased significantly compared with control group.

족삼리(足三里) 의이인약침(薏苡仁藥鍼)이 Collagen으로 유발(誘發)된 생쥐의 관절염(關節炎)에 미치는 영향(影響) (Effects of Herbal-acupuncture with Coicis Semen Solution at Joksamni (ST36) on Collagen-induced Arthritis in DBA/1J Mouse)

  • Objectives & Methods : The purpose of this study is to observe the effects of Coicis Semen Herbal-acupuncture solution (CS-HAS) at the Joksamni (ST36) on the collagen-induced arthritis in the DBA/1J mouse. The author performed several experiments to analyze the effects of CS-HAS on arthritis; change of the weight; the spleen size and adhesion rate; serum cytokine levels; serum antibody levels; changes of immunocyte counts; the histological changes of joint. Results : In the Coicis Semen Herbal-acupuncture (CS-HA), arthritis index, the incidence of arthritis, and the degree of joint edema were decreased. In CS-HA, there was no weight loss. The size of the spleen, adhesion rate, and the edema and transformation of joint were lowered. In CS-HA, the levels of IL-1${\beta}$, IL-6, TNF-${\alpha}$, IFN-${\gamma}$, IgG, IgM, and anti-collagen II in serum and the levels of IFN-${\gamma}$, IL-4, IL-10 in spleen were significantly decreased. In CS-HA, the expression ratios of $CD45^+$ to $CD3e^+$ and $CD8^+$ to $CD4^+$ were decreased. Also, the overall $CD4^+/CD8^+$ cell ratio was lowered in spleen. Ratios of the $CD4^+/CD25^+$, $CD45^+/CD69^+$ cells were decreased in lymph nodes. In addition, ratios of the $CD3^+/CD69^+$, $CD11b^+/Gr-1^+$ cells were also decreased in synovium. In the histological study, the cartilage destruction and synovial cell proliferation, and collagen fiber destruction were decreased with CS-HA treated group. Conclusions : From the results mentioned above, it is suggested that CS-HA at the ST36 has several significant effects on the collagen-induced arthritis.

A Study on Single Dose Toxicity of Intravenous Injection of Mecasin Herbal Acupuncture

  • Objectives : This study was conducted to analyze the single dose toxicity of Mecasin (Gami-Jakyak Gamcho buja Decoction) herbal acupuncture administered in the vein of Sprague-Dawley rats. Methods : All experiments were performed at the Medvill, an institution licensed to conduct nonclinical studies, under the Good Laboratory Practice (GLP) regulations. Sprague-Dawley rats were chosen in this pilot study. In the experiment, Sprague-Dawley rats were divided into four groups of five male and five female animals per group. Doses of Mecasin herbal acupuncture, at 0, 500, 1,000, and 2,000 mg/kg, were given to the experimental groups, and a dose of normal saline solution, at 2 ml/kg, was administered to the control group. Mecasin herb acupuncture and normal saline were injected into the vein at once, and we observed mortality, clinical signs, weights, and gross findings for 14 days after injection. This study was conducted under the approval of the Institutional Animal Ethics Committee. Results : There is no death or abnormality in any of the four groups. All groups put on weights favorably. There are no significant gross findings in necropsy examinations. Conclusions : The above results showed that intravenous injection of 500-2,000 mg/kg of Mecasin herb acupuncture did not cause any changes in weight or, in the results of necropsy examinations, in mortalities. Therefore, the toxicity of Mecasin herb acupuncture was not confirmed, and the presumptive lethal dose of Mecasin herb acupuncture was higher than 2,000 mg/kg. The outcomes suggest that treatment with Mecasin herbal acupuncture is relatively safe. Further evaluations on this subject are needed to yield more concrete evidence.

녹용약침액(鹿茸藥鍼液)의 위암세포주(胃癌細胞柱)에 대한 Oligonucleotide Chip 분석 (Oligonucleotide Chip Analysis of Cervi parvum cornu Herbal-acupuncture Solution (CPC-HAS) on SNU484 carcinomar cells)

  • Objectives: It has long been known about the osteogenic effect of CPC-HAS(cervi parvum cornu herbal-acupuncture solution) on bone tissues. However, it has not been determined the effect of CPC-HAS on cancer cells. The purpose of this study is to screen the CPC-HAS mediated differentially expressed genes..in cancer cells such as SNU484 gastric cancer cell lines. Oligonucleotide microarray approache was employed to screen the differential expression genes. Methods: CPC-HAS was prepared by boiling and stored at $-70^{\circ}C$ until use. Cells were treated with various concentrations of CPC-HAS (0.1, 0.5, 1.5, 10, 20 mg/ml) for 24 h. Cell toxicity was tested by MTT assay. To screen the differentially expressed genes in cancer cells, cells were treated with 1.5 mg/ml of CPC-HAS. For oligonucleotide microarray assay, total RNA was used for gene expression analysis using oligonucleotide Genechip(Human genome U133 Plus 2.0., Affimatrix Co.). Results: It has no cytotoxic effects on SNU484 cell in all concentrations(0.l, 0.5, 1.5, 10, 20 mg/ml). In oligonucleotide microarray assay, in SNU484 cells, the number of more than twofold up-regulated genes was 5 while, the number of more than twofold down-regulated genes was 10. Conclusions: This study showed the screening of CPC-HAS mediated differentially regulated genes using combined approaches of oligonucleotide microarray. The screened genes will be used for the better understanding of the therapeutic effects of CPC-HAS on cancer fields.

음곡(KI10) 편축 약침의 LPS 유도 급성신장염 억제 효과 (Anti-Inflammatory Effect of Polygoni Avicularis Herba Herbal-Acupuncture at KI10 on LPS-Induced Nephritis in Rats)

  • Objectives : This study aimed to evaluate the effects of Polygoni avicularis Herba Herbal-acupuncture( PaH-HA) at KI10(Umgok) on nephritis induced by lipopolysaccharide(LPS) in rats. Methods : Rats were allocated into normal, control, and 3 experimental groups. The rats in the control group were intra-peritoneally injected with LPS for nephritis induction. The rats in the groups of experiment 1, experiment 2, and experiment 3 were treated with single needle prick, saline injection, and PaH-HA, respectively at KI10 three times for a week and then injected with LPS. To evaluate the effects of PaH-HA at HI10, WBC count in blood, serum CINC-1, renal TNF-${\alpha}$ and renal MPO were measured. Results : Needle prick at KI10 suppressed the increase of WBC in blood and CINC-1 in serum of LPS-stimulated rats. Saline injection at KI10 suppressed the increase of WBC in blood. PaH-HA at KI10 suppressed the increase of WBC in blood, CINC-1 in serum, and MPO in kidney of LPS-stimulated rats. Conclusions : PaH-HA at KI10 has an anti-inflammatory effect on LPS-induced nephritis in rats and there may be a synergism between KI10(Umgok) stimulation and PaH-HA Solution.

족삼리(足三里) 강활약침(羌活藥鍼)이 생쥐의 Collagen-induced arthritis에 미치는 영향 (A Study on the Effects of Herbal-acupuncture with Notopterygii Radix solution at ST36 on CIA in Mice)

  • Objective & Methods : This study is performed to observe the effect of Herbal-acupuncture with Notopterygii Radix Herbal-Acupuncture Solution(NR-HAS) at Joksamni(ST36) on Collagen II-induced arthritis (CIA) in DBA/1J mice. Result : 1. The highest survival rate of mice lung fibroblasts were measured in the 1% NR-HAS, and the expression of $TNF-{\alpha}$ in synovial cells were significantly decreased in the 1% and 10% NR-HAS. 2. The incidence of arthritis and the spleen weight were significantly decreased by Notopterygii Radix Herbal-acupuncture(NR-HA) at ST36. 3. The levels of IL-6, $INF-{\gamma},\;TNF-{\alpha}$, IgG, IgM, anti-collagen II in serum of CIA mice were significantly decreased by NR-HA at ST36. 4. In histology, the cartilage destruction and synovial cell proliferation were decreased by NR-HA at ST36, and the collagen fiber expressions in the NR-HA I II groups were similar with that of the normal group. 5. In lymph node, the expression ratios of $CD3e^+\;to\;CD19^+$ cell and $CD4^+\;to\;CD8^+$ cell in the NR-HA I II groups were similarly maintained as those in the normal group. 6. In lymph node, $CD69^+/CD3e^+$ cells and $CD11a^+/CD19^+$ cells were decreased by NR-HA at ST36. 7. In the articular joint, $CD11b^+/Gr-1^+$ cells were decreased by NR-HA at ST36. 8. NR-HA at ST36 did not make a considerable difference in DBA/1J mice without CIA 9. Throughout the overall experimental result, NR-HA I group showed more predominant effect than the NR-HA II group. Conclusion : These results suggest that NR-HA at ST36 has an effect to control synovial cell proliferation and cartilage destruction in rheumatoid arthritis, as well as prophylaxis is important to treat rheumatoid arthritis in clinic.

자하거(紫河車) 약침액(藥鍼液)이 Lipopolysaccharide로 처리된 RAW 264.7 대식세포주(大食細胞柱)의 유전자(遺傳子) 발현(發顯)에 미치는 영향(影響) (Microarray Analysis of Gene Expression in Raw 264.7 Cells Treated with Hominis Placenta Herbal-Acupuncture Solution)

  • Hominis Placenta has a broad array of clinical applications in Korean medicine, including treatment of inflammatory conditions such as rheumatoid arthritis. The purpose of this study is to explore the global gene expression profiles in human RAW 264.7 cell lines treated with Hominis Placenta herbal-acupuncture solution (HPHAS) using microarray analysis. The RAW 264.7 cells were treated with lipopolysaccharide (LPS), HPHAS, or both. Of the 8,170 genes profiled in this study, with a cut-off level of two-fold change in the expression, 72 genes (CTD1, regulating synaptic membrane exocytosis 2, etc.) were upregulated and 135 genes(splicing factor, arginine/serine-rich 1, actinin, alpha 1, etc.) downregulated following LPS treatment. One gene (acrosin) was upregulated and 12 genes (phospholipase A2, group IB, neurofilament, heavy polypeptide 200kDa, etc.) were downregulated following HPHAS treatment. Eleven genes (RAB27A, member RAS oncogene family, eosinophil peroxidase, etc.) were upregulated and 16 genes (V-maf musculoaponeurotic fibrosarcoma oncogene homolog G (avian), RW1 protein, etc.) were downregulated following co-stimulation of HPHAS and LPS. It is thought that microarrays will play an ever-growing role in the advance of our understanding of the pharmacological actions of HPHAS in the treatment of arthritis. Further studies, however, are required to concretely prove the effectiveness of HPHAS.

  • Objectives & Methods : The purpose of this study is to observe the effects of Gentianae macrophyllase Radix herbal-acupuncture solution(GR-HAS) at Joksamni(ST36) on Collagen-induced arthritis(CIA) in DBA/1J mice. The author performed several experimental items to analyze arthritis evaluation, change of weight, spleen size and adhesion rate, change of cytokine level, IgG, IgM and anti-collagen II , change of immunocyte count, histological change of CIA mouse joint related with RA. Results : 1. In the GR-HA group, the arthritis index, the incidence of arthritis and joint edema were significantly decreased. 2. In GR-HA group, there were no weight loss and similarly maintained as normal group. Spleen size, adhesion rate and the edema and transformation of knee joint were low and similarly maintained as normal group. 3. In the GR-HA, cytokine level, IgG, IgM and anti-collagen II were significantly decreased. 4. In the GR-HA, the change of immunocyte count were similarly maintained as normal group. 5. In the histological changes of the CIA mouse joint, the cartilage destruction and synovial cell proliferation were decreased in the GR-HA(Hematoxylin and eosin stain). The collagen fiber expressions in the GR-HA were similar with that of the normal group(Masson's Trichrome stain) Conclusions : These results suggest that GR-HA at ST36 has an effect to control immune reaction and suppress inflammation, synovial cell proliferation and cartilage destruction in rheumatoid arthritis.

반하(半夏) 약침액(藥鍼液)이 사람 기관지 상피세포의 TARC 분비에 미치는 효과 (Effect of Pinelliae Rhizoma Herbal Acupuncture on the Release of Thymus and Activation-Regulated Chemokine(TARC) in Human Bronchial Epithelial Cell)

  • 사람 기관지 상피세포에 알러지 환경을 유발 하고자 사이토카인을 처리하여 TARC의 분비를 유도하고, 이 케모카인 분비에 반하(半夏) 약쇄액(藥鎖液)이 미치는 효과를 실험한 결과 다음과 같은 결론을 얻었다. 1. 사람의 기관지 상피 세포주에 각각 IL-4, TNF-${\alpha}$, IFN-${\gamma}$ 및 IL-$1{\beta}$를 처리하는 경우와 IL-4와 TNF-${\alpha}$, INF-${\alpha}$와 IFN-${\gamma}$, IFN-${\gamma}$와 IL-$1{\beta}$를 병용 처리할 경우 TARC의 분비량를 측정한 결과 IL-4와 TNF-${\alpha}$와 TNF-${\alpha}$와 IFN-${\gamma}$를 병용 처리하였을 경우 TARC의 분비량이 유의하게 증가하였다. 2. 반하(半夏) 약쇄액(藥鎖液) 처리군의 24시간 및 48시간에서 통계적으로 유의한 감소를 관찰할 수 있었다. 3. 반하(半夏) 약쇄액(藥鎖液)에 의한 TARC 분비억제를 관찰 한 결과 농도의존적인 분비 감소 효과를 관찰 할 수 있었다. 4. MTT assay법을 이용한 세포 독성 측정에선 대조군과 반하(半夏) 약침액(藥鍼液) 처리군간에 유의성이 없었으므로 50, 100 및 200${\mu}g/m{\ell}$의 농도에선 세포독성이 없었음을 관찰할 수 있었다. 이에 반하(半夏) 약광액(藥鑛液)은 TARC 케모카인 억제를 통해 천식에 대한 치료효과를 나타낼 수 있을 것으로 사려된다.
