송어 간세포와 생쥐 간세포에서 TCDD가 CYP1A1 유전자 발현에 미치는 영향 비교 연구 (Comparative Study of TCDD Effect on CYP1A1 Gene Expression in Trout and Mouse Liver Cells)

  • In mammalian, cytochrome P4501A1 (CYP1A1) is very important for metabolism of xenobiotics such as PAHs(Polycyclic aromatic hydrocarbon) and heterocyclic amine, and it is induced by environmental contaminants such as PAHs, TCDD(2,3,7,8-tetrchlorodibenzo-p-dioxin) and 3-MC (3-methylcholanthrene). In fish, like mammalian, when it is exposed to environmental contaminants, they cause specific and sensitive induction of CYP1A. Therefore, induction of CYP1A in fish and mammalian is widely used as a biomarker for exposure of environmental contaminants. In this study, to compare the function of Cyp1a1 in fish with it in mammalian, we have used rainbow trout(Oncorhynchys mykiss) hepatoma cells (RTH-149) and mouse hepatocyte (Hepa-I). in order to examine induction of Cyp1a1 by TCDD, we have used the bioassay system. We examined effects of TCDD on the Cyp1a1-luciferase reporter gene activity, 7-ethoxyresorufin O-deethylase(EROD) activity and Cypa mRNA level.

In vitro Antioxidant and Cytoprotective Activities of the Extract of Dangyuja (Citrus grandis Osbeck) Leaves

  • The antioxidant activities of the extracts of dangyuja (Citrus grandis Osbeck) leaves were evaluated. The highest phenolic content was obtained from the ethyl acetate fraction (EF) (202.1$\pm$0.8 mg GAE/g dried extract) and it exhibited the highest 1,1-diphenyl-2-picrylhydrazyl (DPPH) free radical scavenging activity. The cytoprotective effects of EF on oxidative damage induced by tert-butyl hydroperoxide (t-BHP) in a human hepatoma cell line, HepG2 cells, were investigated to understand the intracellular antioxidant mechanisms. Treatment of HepG2 cells with EF prior to oxidative stress was found to inhibit reactive oxygen species (ROS) generation, lipid peroxidation, and DNA damage in a dose-dependent manner. Gas chromatography-mass spectrometry (GC-MS) studies on EF resulted in tentative identification of 19 compounds representing 94.3% of the total content. Taken together, these results demonstrated that EF has excellent antioxidant activities and thus dangyuja leaves have great potential as a source for natural antioxidant which can be applied in food products.

  • MCH-201 was isolated from the mycelium of Streptomyces sp. Ba16 as a potent effector on proliferation and morphology of human breast tumor cell line, MCF-7. Morphological change could be observed at concentration between 2.5${\mu}$g/ml and 250pg/ml and showed cytotoxic effect at the concentration of more than 5${\mu}$g/ml. This compound also showed inhibitory effect on DNA synthesis of hepatoma cells, Hepa 1c1c7, and strong cytotoxic effect on proliferation of human tumor cell lines, A549 and XF498.

Application of Nanotechnology to Korean Black-Red Ginseng: Solubility Enhancement by Particle Size Reduction

  • In order to investigate whether the particles reduced to almost nano grade might affect the chemical and physical properties of organic materials, whole Korean Black-Red Ginseng was pulverized into almost nano size and then ginsenosides, minerals, carbohydrates, lipids and proteins in the ultrafine particles were compared with those in the regular particles as control. The mean size of the ultrafine particles was in the 350 nm range, while that of the regular particles was $127{\mu}m$. More ginsenosides, minerals, carbohydrates, lipids and proteins were detected in the ultrafine particles than in the regular particles. Interestingly, more lipids from the ultrafine particles dissolved in the water than those from the regular particles in the ethanol. Absorption and transport of carbohydrate, lipid or antioxidant activity across the intestinal wall using everted intestine sacks of mice was also enhanced by particle size reduction at the almost nano scale. More cytotoxic effect against hepatoma cell growth by ultrafine particles was also found. These results could be used as the basic data for the understanding and evaluation of the effects of organic nanomaterials on the human health.

암성 통증 치료를 위한 경막외 카테터 터널 거치법 (Cancer Pain Management with Epidural Tunnelling)

  • Continuous epidural analgesia with intermittent morphine injection is a good technique for the management of intractabie cancer pain. Since epidural tunnelling was introduced for the first time to our institution in 1988, it has been widely used for the cancer pain management. Recently epidural tunnelling was introduced for the keeping an inserted catheter for a long period of time. Twenty-five patients with terminal cancer had been used an epidural tunnelling at the pain clinic of Dongsan Medical Center during the period of 3 months in 1991. Among the 25 patients, 14 were male and 11 were female, and most of them were at the ages of 6th decade. The causes of pain were from stomach cancer in 12, CBD cancer in 3, hepatoma in 2, pancreatic cancer in 2, uterine cervical cancer in 2, and miscellaneous cancer in 4 cases respectively. The outcome of treatment were excellent results in 14(56%), good in 9(36%), and moderate in 2.patients(8%). Accordingly satisfactory pain relief was achieved in 92% of the patients. It is suggested that this long-term management of intractable cancer pain by epidural bupivacaine and morphine with epidural tunnelling is satisfactory and reliable.

생간탕(生肝湯)의 효능(效能)에 관(關)한 임상적(臨床的) 연구(硏究) (A Clinical Study on the Effects of Saenggan-tang)

  • In order to study the effect on liver diseases, Saenggan-tang has been applied to 26 patients of chronic hepatitis, 9 patients of liver cirrhosis, 5 patients of acute hepatitis, 8 patients of alcoholic liver diseases, 1 patient of hepatoma total 49 patients visiting the first internal medicine department of the hospital of Oriental Medicine, Kyung Hee University. The Saenggan-tang was taken every 4 weeks (ecept acute hepatitis every 2 weeks). 1. Saenggan-tang had decrease on activity of SGOTF SGPT, with the passage of time, 4, 8, and 12 weeks, showing statistically significant effect. 2. Saenggan-tang revealed sharp decrease on levels of total bilirubin and alkaline phosphatase no showing significance in relation to a few cases 3. Serum protein total and albumin levels were in normal limit before or after Saenggan-tang treatment 4. Triglyceride level was lowered remarkably after Saenggan-tang treatment, showing significance in alcoholic liver disease group whereas no significance in chornic hepatitis Judging from above results, it is proved that Saenggan-tang has curative effect of liver diseases such as acute, chronic hepatitis, liver cirrhosis, alcobolic liver diseases and so on.

  • Hepatitis B virus (HBV) infection is one of the serious problems in Southeast Asia including Korea because it causes chronic hepatitis, which can easily be transformed In fatal conditions such as cirrhosis and hepatoma. Even though lots of informations on structural characteristics and gene expression mechanisms have been accumulated, the mechanism for HBV-induced hepatocellular injury which is believed to be the consequences of the immunological response is not well understood. In order tn perform immunopathological studies for prevention and treatment of HBV infection, we designed transgenic mice as a disease model which can mimic HBV infection, In this study, a promoter-HBV DNA fragment for the preparation of HBV transgenic mice has been constructed. To add a proper enzyme site on 5' end of HBV gene, total HBV (subtype adr) gene was inserted into BamHI site of pBluescript SK vector and reextracted by PstI-SacI treatment A liver-specific promoter, rat ${\alpha}$ 2u globulin gene promoter, was insrted to pBluescript SK vector and reextracted by BamHI-PstI treatment, Promoter-HBV DNA was constructed by ligation of two fragments using identical PstI sites. For large scale production of promoter-HBV DNA, it was inserted to BamHI-SacI site of pBluescript SK vector.

  • Ethanol extracts from Xanthii Fructus (XFE) and Prunellae Spica (PSE) were investigated for the effects on the induction of cancer chemoprevention-associated enzymes. The following effects were measured: (a) induction of quinone reductase (QR) (b) induction of glutathione S-transferase (GST) (c) reduced glutathione (GSH) level. XFE and PSE were potent inducers of quinone reductase activity in Hepa1c1c7 murine hepatoma cells. Glutathione levels were increased with XFE and PSE. In addition, glutathione S-transferase activity was increased with XFE. However, GST activity was not increased with PSE. These results suggest that XFE and PSE have chemopreventive potentials by inducing quinone reductase and increasing GSH levels.

하고초 에탄올추출물이 유방암 예방효소계에 미치는 영향 (Effect of Ethanol Extract from Thesium chinense Tunczaninov on Chemopreventive Enzymes of Breast Cancer)

  • Ethanol extract from Thesium chinense Tunczaninov (TCTE) was tested for breast cancer chemopreventive activity by measuring 7,12-dimethylbenz[a]anthracene (DMBA) - induced cytochrome P450 1A1 activity, induction of quinone reductase and glutathione S-transferase, and glutathione level. TCTE significantly inhibited cytochrome P45O 1A1 activity at the concentration of 90 and 150 mg/ml. TCTE induced quinone reductase activity in a dose-dependent manner in a concentration range of 3-150 mg/ml. In addition glutathione S-transferase activity and glutathione level were increased with TCTE in cultured murine hepatoma Hepa1c1c7 cells. These results suggest that TCTE has breast cancer chemopreventive potential by inhibiting cytochrome P45O 1A1 activity, inducing quinone reductase and glutathione S-transferase activities, and increasing GSH level.

결명자의 타크린으로 유발한 간 세포독성 보호 성분 (Hepatoprotective compounds of Cassiae Semen on tacrine-induced cytotoxicity in Hep G2 cells)

  • Tacrine is an acetylcholinesterase inhibitor that is approved for the treatment of Alzheimer's disease. However, tacrine treatment for Alzheimer's disease results in reversible hepatotoxicity in 30-50% of patients, which seriously limits its clinical use. Accordingly, the identification of constituents in natural products that have protective effects on tacrine-induced hepatotoxicity would be valuable. In the present study, an immortalized human hepatoma cell line, HepG2 was employed to screen for agents that protect against tacrine-induced hepatotoxicity. The bioassay-guided fractionation of water extract of Cassiae Semen furnished two anthraquinones, aurantio-obtusin (1) and obtusifolin (2). Compounds 1 and 2 showed hepatoprotective effects with the protection ratio values of 55.3 +/- 0.5% and 41.2 +/- 0.8% at $160{\mu}M$, respectively.
