The aim of this study was to investigate the possible antioxidant effect of Spirodela polyrhiza (SP) on rats fed a high fat and high cholesterol diet supplemented with either 5% (SPA group) or 10% (SPB group) SP for 4 weeks. The hepatic SOD activity of the HF group significantly decreased compared to that of the N group, but that of the SPA and SPB groups significantly increased. The GPx activity of the SPA and SPB groups in the liver was significantly greater than that of the HF group, and the hepatic catalase activity of the SPA and SPB groups significantly increased compared to the HF group. The hepatic superoxide radical content of the mitochondria and microsomes of the HF group significantly increased compared to that of the N group, but the contents were reduced in the group that took SP powder. The hepatic hydrogen peroxide content in the cytosol and mitochondria of the SP powder group was lower than in the HF group. The carbonyl content in the mitochondria and microsomes of the SPA and SPB groups was significantly lower than in the HF group. The TBARS values in the liver significantly decreased in the SPA and SPB groups. Spirodela polyrhiza was thus effective in reducing oxidative stress by regulating the hepatic antioxidant enzymes and the free radicals in rats fed high fat and high cholesterol diets.
This study investigated anti-obesity and antioxidant effects of dietary non-fermented soybean crud residue (SCR) and fermented SCR by Monascus pilosus (FSCR) in high-fat induced-obese mice. SCR and FSCR were supplemented with high-fat diet at 2% (wt/wt) dose for 8 weeks. Both SCR and FSCR significantly lowered body weight, epididymal fat weight and weight gain rate compared to high-fat diet control (HC) group and FSCR group showed lowest weight gain rate. In addition, it was observed that serum and hepatic lipid profiles including triglyceride, total cholesterol, LDL-cholesterol and HDL-cholesterol were significantly improved by supplementing SCR or FSCR. Furthermore, SCR and FSCR administration showed increase of glutathione content and decrease of hepatic lipid peroxide content, serum aminotransferase activity, and hepatic xanthine oxidase activity. On the other hand, activities of reactive oxygen species scavenging enzyme such as superoxide dismutase, glutathione S-transferase and glutathione peroxidase in two test groups were higher than those of HC. Lastly, in comparison with SCR, FSCR was more effective in restoring obesity-related biomarkers to normal level in high-diet induced obese mice. In conclusion, the present study indicates that FSCR could have not only anti-obese effects such as inhibition of abdominal fat accumulation, but also protective effects of cardiovascular disease such as atherosclerosis by decreasing serum and hepatic lipid contents. Furthermore, these results suggest that experimental diets in this study could alleviate hepatic damage caused by overproduction of reactive oxygen spices (ROS) due to obesity via inhibition of ROS generating activities and induction of ROS scavenging activities.

To develop functional products based on black garlic, a black garlic extract (BG) of 7 brix, a gaeddongssuk extract (GS) of 0.7 brix and two types of mixtures (MBS-I, black garlic 7 brix : gaeddongssuk 0.7 brix; MBS-II, black garlic 14 brix : gaeddongssuk 1.4 brix, 93:7, v/v) were supplemented to rats training on a treadmill for 4 weeks. Body weight from the training did not decrease during the experimental period. Serum albumin content significantly increased in the groups fed an experimental diet compared to the control. The BUN content significantly decreased in BG and MBS-II groups compared to the control. AST and ALP activities significantly decreased in the groups fed an experimental diet compared to the control. Serum triglyceride and total cholesterol levels in MBS-I and MBS-II groups significantly decreased compared to the control. Lipid levels in the serum and liver tissue were not significantly different between the MBS-I and MBS-II groups. The lipid peroxide content in the serum and liver tissue was significantly reduced in the groups fed all extracts compared to the control; the serum and liver lipid contents was lowest in the MBS-I and MBS-II groups, respectively. Hepatic catalase activity in the GS and MBS groups increased by 1.8~2.3 times compared to the control. SOD and GSH-px activities significantly increased from treatment with the extracts by 1.3~1.5 times and 1.2~1.7 times, respectively. These results indicate that a mixture of BG and GS extracts has higher biological activity than a single supplementation of BG or GS extract. Therefore, the addition of gaeddongssuk to black garlic (MBS-I and MBS-II) is effective as a defense material against oxidative stress. MBS-I may be especially effective for its biological activities.

This study was conducted to examine the effects of dietary xylooligosaccharides on HMG-CoA reductase activity and lipid composition of liver in rat fed high cholesterol diet. Sprague-Dawley male rats weighing 100 $\pm$ 10 g were randomly divided into groups of one normal diet, and four high cholesterol diet containing 1% cholesterol. The high cholesterol (1%) diet groups were classified into xylooligosaccharides free diet (C group), 5% xylooligosaccharides diet (C5XO group), 10% xylooligosaccharides diet (C10XO group), and 15% xylooligosaccharides diet (C15XO grcup) according to the levels of dietary xylooligosaccharides supplementation. The experimental diets were fed ad libitum for 4 weeks. The hepatic lipid contents, cholesterol and triglycerides in xylooligosaccharides supplemented groups were significantly lower than those of C group. An antithrombGsis index, a ratio of n-3 to n-6 fatty acids of liver was significantly increased in 10% xylooligosaccharides supplemented groups compared to that of C group. The activity of hepatic HMG-CoA reductase, a rate limiting enzyme of cholesterol biosynthesis, in xylooligosaccharides supplemented groups was more significantly increased than in C group. These results suggest that dietary xylooligosaccharide may be act as potential substitute for a dietary fiber to improve lipids metabolism in rat fed high cholesterol diet.

To evaluate the antioxidative effect of rice coated with maengjong-juk extract in vivo system, rice coated with maengjong-juk extract diets were fed to NZW rabbit for 16 weeks and lipid peroxidation, protein oxidation, activities of antioxidative enzymes and total glutathione content in tissues were measured. TBARS contents in liver and spleen were significantly decreased in maengjong-juk extract diet group compared to control group, while those in kidney and heart tissue were not significantly different. Maengjong-juk extract diet suppressed the protein oxidation significantly in liver, spleen, kidney and heart tissues. Hepatic total SOD, Cu$.$Zn-SOD and Mn-SOD activities of maengjong-juk extract diets were significantly higher than those of control diet. GSH-Px and catalase activities of maengjong-juk extract diet were higher than those of control, while GR activities show no significant difference between the two groups. Total hepatic glutathione content was significantly increased by maengjong-juk extract diet. According to this study, many antioxidative materials and phytochemicals in maengjong-juk extracts seems to protect tissues from oxidative stress by stimulating antioxidative systems in atherosclerotic rabbit fed high cholesterol diet.
The object of this study is to investigate the toxicity of fenvalerate [(RS)-$\alpha$-cyano-3 -phonoxybenzyl-(RS)-2-(4-ch1orophenyl)-3-methylbutyrate] and the effect of carbaryl on the toxicity of fenvalerate. Rats were treated with fenvalerate (50 mg/kg, 100 mg/kg), carbaryl (50 mg/kg, 100 mg/kg) or mixtures of the two compounds (fenvalerate+carbaryl: 50 mg/kg+50 mg/kg, 50 mg/kg+100 mg/kg) by oral administration for 1~3 weeks. Control groups were treated with corn oil. The experimental results were summarized as follows. 1. LD$_{50}$ values of fenvalerate and carbaryl in male rats were 385 mg/kg and 625 mg/kg respectively. When 50 mg/kg and 100 mg/kg of carbaryl were administratrd, LD$_{50}$values of fenvalerate were 265 mg/kg and 225 mg/kg respectively. 2. Biochemical parameters such as ALT, LDH and glucose in serum were much more increased in the groups treated with mixture than the groups treated with either one of fenvalerate or carbaryl. 3. The groups treated with carbaryl and mixture for 3 weeks, the contents of cytochrome P-450 in the liver were significantly increased. In renal microsomal fractions, however, no significant changes of drug metabolizing enzyme activities were observed. 4. The activities of aniline hydroxylase in hepatic microsomal fractions were increased in the groups treated with fenvalerate and mixture and activity was much more increased in the groups treated with mixture. 5. The activities of ATPase in the groups treated with fenvalerate were decreased than that of groups treated with mixture. TBA values and the activity of glucose-6 -phosphatase in the liver were not significantly changed. 6. In mixture treated groups, the activities of cholinesterase in serum and in the liver were more decreased than those of carbaryl treated groups. The activities of carboxylesterase in serum in the liver were slightly increased in mixture treated groups, but in fenvalerate treated groups, the activities of carboxylesterase were much more increased than those of control groups. 7. As a result of this study, when carbaryl was as the synergist of fenvalerate, carbaryl inhibited the activities of esterases, so the toxicity of fenvalerate was increased.sed.

This study was done to investigate the protective effects of Diospyrus kaki Folium on carbon tetrachloride intoxicated rats. Male Sprague-Dawley rats (200~230g) was used. Experimental groups were divided into normal group and $CCl_{4}$-treated group and then $CCl_{4}$-treated groups were divided into 7 groups: only $CCl_{4}$-treated group, methanol extract pretreated group, butanol fraction pretreated group and $H_{2}O$ fraction pretreated group, $CCl_{4}$ treated groups were injected with $CCl_{4}$ 0.6 mg/kg (ip). The activities of ananine aminotransferasein hexane, chloroform and $H_{2}O$ fraction pretreated groups were significantly decreased (p<0.05) compared to the only $CCl_{4}$-treated group and also activity of aspartate aminotransferase and hepatic malondialdehyde content in $H_{2}O$ fraction pretreated group were significantly decreased (p<0.05) compared to the only $CCl_{4}$-treated group. The activity of alkaline phosphatase in chloroform and $H_{2}O$ fraction pretreated groups were decreased compared to the only $CCl_{4}$-treated group but the difference was not statistically significant. The activity of ${\gamma}$-glutamyltranspeptidase in hexane, chloroform and ethylacetate fraction pretreated groups were significantly decreased (p<0.05) compared to the only $CCl_{4}$-treated group. The contents of cholesterol and triglyceride were not influenced in any fraction pretreated groups, but HDL-cholesterol content in hexane and chloroform fraction pretreated groups were significantly increased (p<0.05) compared to the only $CCl_{4}$-treated group. Therefore, these results demonstrate a possible hepatoprotective role of $H_{2}O$ fraction of Diospyrus kaki Folium methanol extract in carbon tetrachloride intoxicated rats.
In order to investigate radiation effects on the liver, functional changes of liver were analyzed after irradiation. Doses of 10 Gy, 15 Gy and 20 Gy were exposed partially to the liver of male rats(Sprague-Dawley) with X-ray(4MV linear accelerator) at room temperature. On 1, 2, 4 and 8 weeks after irradiation, sera of the animals were compared with those of unirradiated animal by liver function tests. Enzyme activities in sera such as alkaline phosphatase and concentrations of bilirubin in liver function tests. The content of the activities of many enzymes including alkaline phosphatase in sera were increased slightly with increasing exposure dose in all experiments and the activities of these enzymes increased markedly in 20 Gy irradiated groups. The contents of serum bilirubins including direct and indirect bilirubins increased continuously along with the time lapse after irradiation. However, in 20 Gy irradiated group, the content of serum bilirubin decreased slightly during 2 or 4 weeks after irradiation and increased markedly there after. From these above results, functional changes of the liver were induced in all irradiated groups. Damaged liver was recovered along with time collapsed after irradiation to the doses of 10 Gy and 15 Gy while no recovery was deteced within 8 weeks after irradiation to 20 Gy. These results suggest that careful attontion must be paid to liver not to be included in exposure field in radiation therapy.
In the present study, we examined whether four grains including adlay (AD), buckwheat (BW), glutinous barley (GB), and white rice (WR) affect the duration of food residence in the gastrointestinal tract and hepatic enzyme activities in rats fed different combinations of the grains. The rats were raised for 4 weeks on a high fat diet based on the American Institute of Nutrition-93 (AIN-93G) diets containing 1% cholesterol and 20% dietary lipids. Forty male rats were divided into four groups and raised for 4 weeks with a diet containing one of the grains. Corresponding to the dietary fiber contents of the experimental grains, gut transit time was shortest in the rats fed GB and increased in the order of BW, AD, and WR. In addition, the accumulated shortest transit time occurred in the GB group. Gut transit time affected weight gain and major organ weight, as it was closely related to the absorption of nutrients. The level of thiobarbituric acid reactive substance (TBARS) in liver was higher in rats fed WR, AD, BW, and GB, indicating that the other grains decreased oxidative stress in vivo more than WR. Glutathione, glutathione peroxidase, and glutathione S-transferase levels in the AD, BW, and GB groups were significantly higher than those in the WR group. In conclusion, reduced colonic transit time has been implicated in reducing the incidence of colon cancer, as evidenced by populations consuming diets rich in fiber. Whole grains such as AD, BW, and GB may contribute to a significant supply of antioxidants to prevent oxidative stress if they are consumed in large amounts.
An extract of Allomyrina dichotoma larva (ADL), one of the insects used most frequently in traditional Chinese medicine for the treatment of liver diseases such as hepatocirrhosis and hepatofibrosis, was assessed for antioxidant bioactivity in this study. In the current work, we have investigated the protective effects of ADL extracts on tert-butyl hydroperoxide (t-BHP)-induced hepatotoxicity in cultured hepa1c1c7 cells and in the mouse liver. The treatment of the hepa1c1c7 cells with ADL extracts induced a significant reduction of t-BHP-induced oxidative injuries, as determined by cell cytotoxicity, lipid peroxidation (LPO) and reactive oxygen species contents, in a dose-dependent manner. Moreover, ADL extracts evidenced a protective effect against t-BHPinduced oxidative DNA damage, as revealed by the results of the Comet assay in hepa1c1c7 cells. ADL extracts also protected against hydroxyl radical-induced 2-deoxy-d-ribose degradation by ferric ion-nitrilotriacetic acid and $H_2O_2$. In addition, ADL extracts were shown to be able to quench 1,1-diphenyl-2-picrylhydrazyl (DPPH) free radicals. Our in vivo study revealed that ADL extracts pretreatment applied prior to t-BHP administration significantly prevented an increase in the serum levels of hepatic enzyme markers and reduced LPO in the mouse liver in a dose-dependent manner. Taken together, these results suggest that the protective effects of ADL extracts against t-BHP-induced hepatotoxicity may be attributable, at least in part, to its ability to scavenge free oxygen radicals, and to protect against DNA damage due to oxidative stress.
