표고버섯 열수 추출물이 발암원을 급여한 흰쥐의 간 기능 관련 효소활성에 미치는 영향 (Effects of Hot Water Extracts from Lentinus edodes on Hepatic Functional Enzyme Activities in the Rat Fed Butter Yellow(p-Dimethylaminoazobenzene))

  • 표고버섯의 열수 가용성 다당류(PS)와 간 장해물질이며 발암성을 갖는 butter yellow를 흰쥐의 식이에 6주간 투여하여 혈장과 간의 효소활성 및 과산화지질의 함량을 측정한 결과는 다음과 같다. 1. 혈장 중 GOT 활성은 better yellow 첨가군(BO군)보다 PS의 투여군(BP군 : BO+PS)이 비교적 낮은 수준을 나타내었으며 ${\gamma}$-GTP와 GPT 활성에서도 BO군에 비하여 BP군에서 유의하게 낮은 활성을 나타냈다. 2. 간장 중 glutathion peroxidase, catalase 및 lactate dephdregenase 활성 또한 기초식이군(NO군)에 PS를 토여한 군(NP군)은 NO군과 유의한 차이가 없었으나 BP군은 BO군에 비하여 유의하게 낮은 수준이었다. SOD활성은 각 군간에 유사한 수치로 유의적인 차리를 보이지 않았다. 3. 간장의 microsomal 분획분 및 cytosol 분획분 중의 glutathione S-transferase 활성도는 BP군이 BO군보다 유의하게 낮은 수준을 보였으나 better yellow를 첨가하지 않은 NO군에 비하여 상당히 높은 수준을 나타내었다. 4. 간장 내 glutathione과 malondialdehyde의 함량 또한 PS의 투여로 생성이 저하되는 것으로 나타났다. 이상의 결과를 보아 표고버섯의 열수 추출물은 생체조직의 지질의 과산화를 방지하므로써 better yellow에 의한 간장장애의 치료 또는 독성을 완화하는 효과가 있는 것으로 사료된다.

Nutritional and Hormonal Induction of Fatty Liver Syndrome and Effects of Dietary Lipotropic Factors in Egg-type Male Chicks

  • This experiment was conducted with male chicks to investigate the influence of hormones and nutrients on the development of fatty liver syndrome (FLS) as well as the effects of dietary lipotropic factors on hepatic fat accumulation and lipogenic enzyme gene expression. A total of two-hundred sixteen 4-wk-old Hy-Line male chicks were divided into six groups and fed an experimental diet (T1, low-energy diet with low levels of lipotropic factors; T2, high-energy diet with low levels of lipotropic factors; T3 and T5, low-energy diet with high levels of lipotropic factors; T4 and T6, high-energy diet with high levels of lipotropic factors) for six weeks. The chicks in T5 and T6 groups were treated with intramuscular injections of estradiol benzoate for three days prior to biopsy and clinical analysis of FLS. Chicks treated with estrogen had significantly greater liver weights than untreated chicks. The abdominal fat contents were increased in chicks consuming high-energy diets as compared to those consuming low-energy diets. Treatment with estrogen significantly increased the concentrations of serum cholesterol, triacylglycerol and phospholipid (p<0.05). The hepatic triacylglycerol levels were tenfold higher in the estrogen treated chicks than in the untreated chicks. There were no significant differences in malondialdehyde levels between the treatment groups. Estrogen treatment dramatically increased the levels of fatty acid synthetase, acetyl-CoA carboxylase and ApoB mRNA. The results indicated that treatment with exogenous estrogen in growing male chicks induced hepatic fat accumulation, which might be partially due to increased lipogenic enzyme gene expression.

반복적인 Betaine 투여가 간독성 및 Cytochrome P-450 의존성 약물대사효소계 활성에 주는 영향 (The Effect of Repeated Betaine Treatment on Hepatotoxicity and Cytochrome P-450 Dependent Drug Metabolizing Enzyme System)

  • Betaine is one of the major water-soluble components in Lycii Fructus. In the present study the effect of repeated betaine treatment on the hepatotoxicity and the cytochrome P-4 50-dependent enzyme system was examined in adult female rats. Administrations of betaine (100 or 1,000mg/kg/day, ip) to rats repeatedly for 4 or 9 days did not evoke hepatotoxic response as determined by increases in glutamic pyruvic transaminase(GPT) and glutamic oxaloacetic transaminase(GOT) activities measured 24 hours following the final dose of betaine. The activities of aminopyrine N-demethylase, p-nitroanisole O-demethylase and p-nitrophenol hydroxylase as well as the contents of cytochrome P-450 were determined in hepatic microsomes of rats treated with betaine(1,000mg/kg/day, ip) for 4 or 9 days. Repeated treatment of rats with betaine for a period of 4 days induced a marginal decrease in the contents of cytochrome P-450, but did not influence the activities of p-nitrophenol hydroxylase, p-nitroanisole O-demethylase, or aminopyrine N-demethylase. Extension of the betaine treatment to 9 consecutive days failed to alter the parameters for hepatic drug metabolizing activity determined in the present study. Since repeated large doses of betaine were demonstrated to be tolerated by rats without showing any toxicity or changes in drug metabolizing enzyme activities in the liver, this compound appears to be relatively safe to animals upon long-term ingestion.

성게 부위별 및 그 추출물의 간 해독과 항산화 활성 효과 (Hepatic Detoxification and Antioxidant Activity in Sea-urchin Roe and Ethanol Extract of Roe)

  • Sea-urchins (Anthocidaris crassispina) are widely distributed in the East Sea of Korea. The aim of this study was to evaluate the hepatoprotective effects of sea-urchin roe on bromobenzene (BB)-induced liver damage in rats. The antioxidative and detoxifying properties of sea-urchin roe in BB-poisoned rat liver was examined by chemical analysis of serum aminotransferase (AST, ALT), glutathione S-transferase (GST), $\gamma$-glutamylcystein synthetase, glutathione reductase, epoxide hydrolase, amino-N-demethylase (AD), aniline hydrolase (AH) enzyme activity, as well as lipid peroxide and glutathione contents. Sea-urchin roe inhibited the increase of serum AST, ALT enzyme activity. Increasing lipid peroxide contents and AD and AH activities were significantly decreased in ethanol extract of sea-urchin roe. GST, $\gamma$-glutamylcystein synthetase, glutathione reductase and epoxide hydrolase enzyme activities increased in sea-urchin roe-fed group, compared with the BB-treated group. These results suggest that sea-urchin roe facilitates recovery from liver damage by enhancing antioxidative defense mechanisms and hepatic detoxication metabolism.

맹종죽(Phyllostachys Pubescens)추출물 코팅쌀이 Atherogenic 식이를 섭취한 C57BL/6 마우스의 항산화 시스템에 미치는 영향 (Effect of Maengjong-Juk (Phyllostachys Pubescens) Extract Coated Rice Diet on Antioxidative System of C57BL/6 Mice Fed Atherogenic Diet)

  • To evaluate the antioxidative effect of maengjong-juk (Phyllostachys pubescens) extract coated rice in vivo system, maengjong-juk extract coated rice diets were fed to C57BL/6 mice for 16 weeks. Plasma total antioxidative capacity, hepatic lipid peroxidation, protein oxidation, activities of antioxidative enzymes and total glutathione content were measured. Plasma total antioxidative capacity was elevated significantly in maengjong-juk extract diets supplemented group in a dose dependant manner. Hepatic TBARS contents were significantly decreased in maengjong-juk extract diets supplemented group compared to high cholesterol group. Maengjong-juk extract coated rice diets suppressed the protein oxidation significantly in liver. Activities of hepatic antioxidative enzymes such as total SOD, CuㆍZn-SOD, Mn-SOD, GSH-Px and catalase activities of maengjong-juk extract coated rice diets were significantly higher than those of high cholesterol diet. Total hepatic glutathione content was significantly increased by maengjong-juk extract coated rice diets administration. According to this study, numerous antioxidative materials and phytochemicals containing in maengjong-juk extracts appear to protect antioxidative systems in C57BL/6 mice fed bamboo extract coated rice diet. 식이성 Tungstate가 사염화탄소 투여에 의한 흰쥐 간 손상에 미치는 영향 (Effect of Dietary Tungstate on the Liver Damage in $CCl_4$-treated Rats)

  • To evaluate the role of xanthine oxidase in liver damage by CCl4, a group of rats were fed tungstate for a month, which suppressed the activities of xanthine oxidase in serum and liver. Control group of rats were fed standard diet without tungstate. Liver damage was induced both in tungstate fed and control groups by two intraperitoneal injections of CCl4 at the level of 0.1ml/100g body weight at intervals of 24 hours. Increases in the levels of serum alanine aminotransferase by CCl4 were significantly smaller in tungstate fed rats than in control rats. Concomitantly, histopathologic changes were less in tungstate fed rats than in control ones. In rats either treated with CCl4 or not, hepatic type O xanthine oxidase activities were remarkably reduced by tungstate feeding. Hepatic aniline hydroxylase activities were higher in rats fed tungstate than control rats when animals were not treated with CCl4, but the enzyme activities were lower in tungstate fed rats than control when they were treated with CCl4. Neither tungstate feeding nor CCl4 treatment caused any significant changes in hepatic glutathione contents, and activities of hepatic glutathione S-transferase, glutathione peroxidase and superoxide dismutase. It is concluded xanthine oxidase reaction augment CCl4 induced liver damage via oxygen free radical system.

들깨유 옥수수유의 섭취가 2-Acetylaminofluorene을 투여한 쥐 간에서 소포체막의 지방산 조성과 Cytochrome P-450 함량, Glutathione S-transferase 활성도에 미치는 영향 (Effects of Perilla and Corn Oil Diets on the Hepatic Microsomal Fatty Acid Composition, Cytochrome P-450 Contents, and Glutathione S-transferase Activities in 2-Acctylaminofluorcne Treated Rats)

  • This paper examines the effects of dietary fats on the fatty acid composition and market enzyme activites during liver damage in 2-acetylaminofluorene treated rats. Weaning Sprague-Dawley male rats were fed the diet of beef tallow(BT source of sturated fatty acid) corn oil(CO source of n-6 fatty acid) and perilla oil(PO source of n-3 fatty acid) at the level of 15% fat. Ten days after feeding 2-acetylaminofluorene(2-AAF) was injected intraperitoneally twice every week at the level of 50mg/kg body weight for 7 weeks. Liver microsomal and cytosolic fractions were collected to determine the microsomal fatty acid composition lipid peroxide(malondialdehyde MDA) contents glucose 6-phosphatase(G6 Pase) activity cytochrome(Cyt) P-450 contents and cytosolic glutathione S-transferase(G6 Pase) activity cytochrome(Cyt) P-450 contents and cytosolic glutathione S-transferase(GST) activity. The fatty acid composition in microsomal fraction was reflected by different dietary fats. By 2-AAF treatment linoleic acids were increased regardless of the diet MDA contents were higher in CO group than it was in BT group. However 2-AAF treatment decreased MDA contents in all dietary groups. G6Pase activity of BT group was higher than those of the other gropus. CO group had the highest Cyt P-450 contents and 2-AAF treatment lowered Cyt P-450 contents only in CO gropu GST activites were higher in CO than in BT group whereas the enzyme activites were increased by 20AAF treatment in all dietary groups. These results suggest that dietary fats and 2-AAF treatment in all dietary groups,. These results suggest that dietary fats and 2-AAF treatment affect microsomal fatty acid composition The enzyme activities concerned with liver damage were influenced differently by dietary fats and 2-AFF treatment Although PO diet contains much more polyunsaturated fatty acids than CO diet PO diet doesn't cause more oxidant stress compared with CO diet in these data.

오디, 뽕잎 및 누에의 혼합비율에 따른 Streptozotocin 유발 당뇨쥐에서의 항산화 효과 및 지질대사개선 효과 (Antioxidant Effects and Improvement of Lipid Metabolism of Mulberry fruit, Mulberry Leaves and Silkworm Powder with Different Mixing Ratios in Streptozotocin-Induced Diabetic Rats)

  • This study was conducted to investigate the effects of mulberry fruit, mulberry leaves and silkworm powder with different mixing ratios on hepatic antioxidative system and lipid metabolism in streptozotocin-induced diabetic rats. Sprague-Dawley male rats weighing $100{\pm}10g$ were induced diabetic by 50 mg/kg bw streptozotocin and randomly assigned to following experimental groups; normal diet group (DM), 0.3% and 0.6% mulberry fruit diet groups (F and 2F), 0.3% mulberry leaves diet group (M), 0.3% silkworm powder diet group (S), 0.15% mulberry fruit+0.15% mulberry leaves diet group (FM), 0.15% mulberry fruit+0.15% silkworm powder diet group (FS), 0.1 % mulberry fruit+0.1 % mulberry leaves+0.1% silkworm powder diet group (FMS). The experimental diets were fed for 4 weeks. Hepatic SOD activity was not changed significantly by any of single or combined supplementations of mulberry fruit, leaves and silkworm powder but GSH-px and catalase activities were increased by the groups supplemented with two or three of the test ingredients (FM, FS, FMS) as compared with the DM group. Hepatic TBARS value was not reduced significantly by any of the supplementations but lipofuscin contents were significantly reduced in the FM, FS and FMS groups as compared with the DM group. Hepatic mitochondria and microsomal carbonyl values were reduced by the single and combined supplementations of the test ingredients. Hepatic HMG-CoA reductase activities were increased in the all supplementation groups as compared with the DM group. Hepatic total lipid and triglyceride contents were increased but cholesterol contents reduced in the supplemented groups. The effects on the enzyme activities, peroxide or its products and lipid contents were most remarkable in the FMS group. In conclusion, mulberry fruit, mulberry leaves and silkworm powder have the favorable effects on antioxidative system and lipid metabolism in the diabetic liver and the mulberry fruit, leaves and silkworm powder with equal ratio exert the synergistic effect expectedly to prevent diabetic complications.

The Effect of Supplementation of Persimmon Leaf Extract on Lipid and Antioxidant Metabolism in Rats Fed a High-cholesterol Diet

  • Fruits and vegetables reportedly have a protective effect against hyperlipidemia and oxidative disease. Accordingly, this study aimed to investigate the lipid-lowering effect and antioxidative capacity of persimmon leaf extract (PLE) in rats fed a high-cholesterol diet. Male rats were fed a high-cholesterol (1% wt/wt) or high-cholesterol diet supplemented with Lovastatin (0.02% wt/wt) or PLE (0.2% wt/wt) for 5 weeks. The concentration of plasma total cholesterol was significantly lower in the PLE group than in the lovastatin group. However, the concentration of plasma HDL-cholesterol and the ratio of HDL-cholesterol/total-cholesterol (%) were significantly higher in the PLE group than in the control group. The PLE supplement also significantly lowered the contents of hepatic cholesterol and triglyceride. In comparing fecal sterol contents, the PLE group saw a significant increase of both neutral and acidic sterol compared to the other groups. The PLE supplement significantly lowered plasma GOT and GPT activity, which ave indices of hepatic toxicity. Plasma TBARS concentration was significantly lower in the PLE group than in the control group, while hepatic TBARS level was not significantly different between the groups. In a comparison of hepatic antioxidant parameters, SOD, catalase and GSH-Px activity were significantly higher in the PLE group than in the control group. However, the PLE supplement significantly towered antioxidant enzyme activity in the erythrocyte. Furthermore, these results suggest that supplementation of PLE promoted the excretion of fecal sterols, thereby leading to decreased absorption of dietary cholesterol. In addition, PLE may play an important role in regulating antioxidative capacities by altering SOD and ChT activity.

흰쥐에 있어서 사염화탄소에 의한 간손상에 allopurinol의 영향 (Effect of Allopurinol Pretreatment on the Liver Damage in $CCl_4$-treated Rat)

  • To evaluate the effect of xanthine oxidase on liver injury by $CCl_4$, liver damage was induced both in allopurinol pretreated rats (500 mg/kg. ip) and control group by twice intraperitoneal injection of $CCl_4$ (0.1 ml/100 g body wt. 50% in olive oil) at interval of one day. Increases in the levels of serum alanine aminotransferase and liver weight/body weight (%) by $CCl_4$ were significantly smaller inallopurinol pretreated rats than in control whereas the hepatic microsomal glucose-6-pholphatase activities were significantly higher in allopurinol pretreated rats than control group by $CCl_4$ treatment. These results indicates that allopurinol pretreatment may reduce the liver damage in $CCl_4$ intoxicated rats. In rats either with $CCl_4$or not, hepatic type O xanthine oxidase activities were significantly reduced by allopurinol pretreatment and the increasing rate of these enzymes to each control was remarkably lower in allopurinol pretreated rats than control. Liver cytosolic protein contents and aniline hydroxylase, aminopyrine demethylase activities were higher in allopurinol pretreated rats than coirol rats when animals were treated with $CCl_4$. On the other hand, neither allopurinol pretreated nor $CCl_4$ treatment caused any significant changes in hepatic superoxide dismutase and catalase activities. Hepatic glutathione contents were higher in $CCl_4$-treated rats than control, but no significant changes were found in both between the allopurinol treated rats and $CCl_4$-treated rats pretreated with allopurinol, and glutathione and glutathione S-transferase activities were significantly reduced in $CCl_4$-treated rats than control whereas these enzyme activities showed on significant change in both between allopurinel treated and $CCl_4$-treated rats pretreated with allopurinol. It is concluded that xanthine oxidase reaction system augment $CCl_4$ induced liver injury via even oxygen free radical system.
