흑삼의 제조 및 항암효과 (Preparation of Black Ginseng and its Antitumor Activity)

  • The purpose of this study is to prepare black ginseng and evaluate its antitumor activity. In order to achieve such aim, 5 year fresh ginsenges were steamed at 95'E for 3 hr in pottery apparatus and dried at $60^{\circ}C$ for 12-36 hr. This process was repeated again nine times in same condition. Among the ginseng saponins in black ginseng, the amount of Ginsenoside $Rg_3$ was examined by HPLC. 10.05 mE of Ginsenoside $Rg_3$ was obtained from 1 g of dried black ginseng prepared. The extract of black ginseng exhibited stronger cytotoxic activity against MCF-1, HT-1080 and Hepa 1C1C7 tumor cell lines in vitro than the extract of red ginseng. Also, the extract of black ginseng exhibited stronger antitumor activity(33%) in BDFl mice bearing Lewis lung carcinoma cells(LLC) than the extract of red ginseng(23%). From these results, it was concluded that Black ginseng had antitumor activity suggesting its application for the prevention and treatment of cancer.

Significance of $p27^{kip1}$ as potential biomarker for intracellular oxidative status

  • Our previous proteomic study demonstrated that oxidative stress and antioxidant delphinidin regulated the cellular level of $p27^{kip1}$ (referred to as p27) as well as some heat shock proteins in human colon cancer HT 29 cells. Current study was conducted to validate and confirm the regulation of these proteins using both in vitro and in vivo systems. The level of p27 was decreased by hydrogen peroxide in a dose-dependent manner in human colon carcinoma HCT 116 (p53-positive) cells while it was increased upon exposure to hydrogen peroxide in HT 29 (p53-negative) cells. However, high concentration of hydrogen peroxide (100 ${\mu}M)$ downregulated p27 in both cell lines, but delphindin, one of antioxidative anthocyanins, enhanced the level of p27 suppressed by 100 ${\mu}M$ hydrogen peroxide. ICR mice were injected with varying concentrations of hydrogen peroxide, delphinidin and both. Western blot analysis for the mouse large intestinal tissue showed that the expression of p27 was upregulated by 25 mg/kg BW hydrogen peroxide. To investigate the association of p27 regulation with hypoxia-inducible factor 1-beta (HIF-$1{\beta}$), the level of p27 was analyzed in wild-type mouse hepatoma hepa1c1c7 and Aryl Hydrocarbon Nuclear Translocator (arnt, HIF-$1{\beta}$)-defective mutant BPRc1 cells in the absence and presence of hydrogen peroxide and delphinidin. While the level of p27 was responsive to hydrogen peroxide and delphinidin, it remained unchanged in BPRc1, suggesting that the regulation of p27 requires functional HIF-$1{\beta}$. We also found that hydrogen peroxide and delphinidin affected PI3K/Akt/mTOR signaling pathway which is one of upstream regulators of HIFs. In conclusion, hydrogen peroxide and antioxidant delphinidin seem to regulate intracellular level of p27 through regulating HIF-1 level which is, in turn, governed by its upstream regulators comprising of PI3K/Akt/mTOR signaling pathway. The results should also encourage further study for the potential of p27 as a biomarker for intracellular oxidative or antioxidant status.

들깨의 볶음처리와 산가수분해에 의한 세포모델계 Quinone Reductase 활성유도능의 변화 (Modulation of Cellulalr Quinone Reductase Inducibility by Roasting Treatment and Acid Hydrolysis of Perilla)

  • 발암물질의 해독에 관여하는 2상 효소계의 지표효소인 QR을 활성화시키는 암예방성분의 존재여부를 탐색한 이전의 연구에서 들깨박의 메탄올 추출물이 hepa1c1c7 cell에서 높은 QR 유도활성을 나타내었다. 본 연구에서는 탈지공정에 앞서 실시되는 볶음과정에서 비활성상태로 존재하던 QR inducer가 활성상태로 유리되는 것으로 가정하고 산가수분해와 볶음처리를 한 후, 각각의 메탄올 추출물에 대한 QR 유도활성을 측정하였다. $100^{\circ}C$에서 30, 60, 120분간 산가수분해시킨 날들깨박의 메탄올 추출액은 가수분해시간이 증가할수록 세포의 QR 활성유도능이 증가하는것으로 나타났다. $180^{\circ}C$$200^{\circ}C$에서 5, 10, 20분간 볶은 들깨박의 경우, QR유도 활성은 날들깨박에 비해 높았으며, 묶음시간이 걸어질수록 증가하는 경향을 보였다. 볶은 들깨박의 메탄올 추출액은 농도가 증가함에 따라 비례적으로 QR 효소 활성을 증가시켰다. 탈지들깨박이 동물조직의 QR 효소활성에 미치는 영향을 생쥐를 이용하여 확인한 결과, 볶은 들깨박의 메탄올 추출물은 QR 효소활성을 간과 위에서 유의적으로 증가시켰고, 날들깨박은 간과 폐에서 유의적으로 효소활성을 증가시켰다. 이렇듯 들깨박은 동물의 여러기관에서 암예방의 지표효소인 QR을 유도하는 것으로 나타나, 발암 물질로부터 생체를 보호하는 물질을 함유하고 있을 가능성이 높은 것으로 추정된다.

파파야씨 추출물 및 분획물의 항산화, QR 활성 (Antioxidant Capacity and Quinone Reductase Activity of Methanol Extracts and Fractions from Papaya Seed)

  • 본 연구에서는 파파야씨의 메탄올 추출몰과 분획물을 이용하여 항산화 활성 및 quinone reductase (QR) 활성을 측정하였다. 파파야씨의 추출물 및 분획물의 총 폴리페놀 함량은 17.74~125.99 ${\mu}g/mg$이며, 총 플라보노이드 함량은 1.60~32.69 ${\mu}g/mg$으로 나타났으며, ethyl acetate (EtOAc) 분획층의 총 폴리페놀 및 플라보노이드 함량이 다른 분획층과 비교했을 때 가장 높은 것으로 나타났다. ${\alpha}$,${\alpha}$-Diphenyl-${\beta}$-picrylhydrazyl (DPPH) radicals, 2,2'-azino-bis (3-ethylbenzthiazoline-6-sulfonic acid) (ABTS) and hydrogen peroxide를 이용하여 파파야씨 메탄올 추출물 및 분획물의 항산화 활성을 조사한 결과, EtOAc 분획층에서 가장 높은 free radical 억제능을 보였다. 또한 EtOAc 분획층 12.5~50 ${\mu}g/ml$의 농도에서 QR의 유도활성을 조사한 결과, 50 ${\mu}g/ml$의 농도에서 3.3배 정도의 QR 유도능을 보였다. 따라서 파파야씨의 EtOAc 분획층에 존재하는 물질들은 QR inducer 로써의 역할이 기대된다.
