Effect of Corticosterone Pretreatment on the Production of LPS-Induced Inflammatory Mediators in Hepa1c1c-7 Cells (Hepa1c1c-7 Cell에서 리포폴리사카라이드로 유도된 염증성 매개인자 생산에 있어서 코르티코스테론 전처리 효과)

  • Endotoxemia induces production of inflammatory mediators and acute phase proteins, leading to multiorgan injury and systemic inflammation. Hypothalamic-pituitary-adrenal (HPA) axis activation and glucocorticoids (GCs) release modify endotoxemia-induced inflammatory responses. In the present study, we investigated whether pre-exposure of GCs influences endotoxin-induced production of inflammatory mediators in hepatocytes. Hepa1c1c-7 cells were pretreated with low concentrations of corticosterone for 24 h and then cultured without corticosterone in the presence or absence of LPS. Our results demonstrated that LPS alone significantly enhanced production of IL-6 and CRP but reduced vascular endothelial growth factor (VEGF) compared to controls. Combination of corticosterone pretreatment and LPS significantly upregulated production of IL-6, IL-$1{\beta}$, and VEGF but downregulated CRP compared to those in LPS alone. These findings suggest that in low concentration of corticosterone-preexposed hepatocytes, endotoxemia may induce upregulation of IL-6, IL-$1{\beta}$, VEGF and but downregulation of CRP.

Cytotoxic Effect of Artemisia capillaris Extracts on the Cancer Cells on in vitro (인진쑥 추출물의 세포독성 효과)

  • Artemisia capillaris THUNB is a perennial herb that belongs to the family Compositae spp and the most common plant among the various herbal folk remedies used in treatment of abdominal pain, hepatitis, chronic liver disease, jaundice and coughing in Korea. This experiment was conducted to investigate cytotoxic effects of Artemisia capillaris extracts on the Hepa-1c1c7 and Sarcoma 180 cancer cells on in vitro experimental tests. On in vitro tests using MTT assay and SRB assay, the extracts showed prominent cytotoxic effects on the two kinds of cancer cell lines, respectively. Antihumor effects appeared in the concentration of over $250{\mu}g/mL$ of both ethanol and ethyl acetate extract, over $500{\mu}g/mL$ of methanol extract, over $5000{\mu}g/mL$ in water extract and over 50% cytotoxicity on the Hepa-1c1c7 and Sarcoma 180. The results suggest that Artemisia capillaris extracts have prominent cyotoxic effects on the cancer cell lines Hepa-1c1c7 and Sarcoma 180.

Chemopreventive Effect of Gamdutang Aqua-Acupuncture Solution (감두약침액의 암예방 효과)

  • Gamdutang aqua-acupuncture solution (GAS), Gamdutang water-extracted solution (GWS) and Dae-Gamdutang aqua-acupuncture solution (DGAS) were prepared and tested for chemopreventive potentials. GAS was potent inducer of quinone reductase (QR) activity in Heapa1c1c7 murine hepatoma cells in culture, whereas GWS is less potent. GAS, GWS and DGAS were significantly induced QR activity in cultured rat normal liver cell, Ac2F. Glutathione (GSH) levels were increased about 1.8, 1.0 and 1.1 fold with GAS, GWS and DGAS in Hepa1c1c7 cells, respectively. In addition glutathione S-transferase (GST) activity was increased with GAS, GWS and DGAS. The effects of GAS, GWS and DGAS on the growth of Acanthamoeba castellanii were tested. Proliferation of A. castellanii was inhibited by GAS, GWS and DGAS at concentrations of 1 $\times$ and 5$\times$. These results suggest that GAS has chemopreventive potential by inducing QR and GST activities, increasing GSH levels and inhibition of polyamine metabolism.

Suppression of CYP1A1 Expression by Naringenin in Murine Hepa-1c1c7 Cells

  • Naringenin, dietary flavonoid, is antioxidant constituents of many citrus fruits. In the present study, we investigated the effect of naringenin on 2,3,7,8-tetrachlorodibenzo-p-dioxin (TCDD)-inducible CYP1 A 1 gene expression in mouse hepatoma Hepa-1c1c7 cells. Naringenin alone did not affect CYP1A1-specific 7-ethoxyresorufin O-deethylase (EROD) activity. In contrast, the TCDD-inducible EROD activities were markedly reduced upon concomitant treatment with TCDD and naringenin in a dose dependent manner. TCDD-induced CYP1A1 mRNA level was also markedly suppressed by naringenin. A transient transfection assay using dioxin-response element (DRE)-linked luciferase and electrophoretic mobility shift assay revealed that naringe-nin reduced transformation of the aryl hydrocarbons receptor(AhR) to a form capable of specif-ically binding to the DRE sequence in the promoter of the CYP1A1 gene. These results suggest the down regulation of the CYP1A1 gene expression by either naringenin in Hepa-1c1c7 cells might be antagonism of the DRE binding potential of nuclear AhR.

Effect of Sophorae Flos Aqua-acupuncture Solution on the Quinone Reductase and Glutathione S-transferase Activities of Hepa 1c1c7 Cells (괴화약침액이 간세포의 Quinone redutase 와 Glutathione S-transferase 활성에 미치는 영향)

  • Sophorae Flos aqua-acupuncture solution(SFAS) was prepared and tested for the induction of quinone reductase and glutathione S-transferase activities and glutathione. SFAS significantly induced QR activity at the concentrations of $0.5{\times},\;1{\times}\;and\;3{\times}$ in cell culture. However, GST activity in murine Hepa 1c1c7 cells was slightly increased with SFAS. SFAS increased GSH levels.

Induction of Quinone Reductase by Extracts of Traditional Medicinal Plants (Quinone reductase 유도활성 측정에 의한 약용식물의 항암활성 검색)

  • Induction of phase II enzymes is a major mechanism of chemoprevention. The induction levels of quinone reductase (QR) activity in cultured murine hepatoma (Hepa 1c1c7) cells by 80%-methanol extracts of traditional medicinal plants were measured. Among the tested 81 plants, the extracts of Aralia continentalis, Magnolia obovata, and Viscum album were found to induce QR activities over 250%. The maximum induction levels obtained were 401.9%, 270.5%, and 301.8% by treatments of the extracts of A. continentalis $(318\;{\mu}g/ml)$, M. obovata $(53.8\;{\mu}g/ml)$ and V. album $(80.6\;{\mu}g/ml)$, respectively. These QR induction activities were more potent than those of the known QR inducers, t-butylhydroquinone (170.1%) and ${\beta}-naphthoflavone$ (320.0%).

  • Phase II enzymes are transcriptionally induced by synthetic chemical agents and natural products, and such induction plays critical roles in protection against chemical carcinogens and other toxic xenobiotics. To discover natural products for use as cancer chemopreventive agents, the ability of Citrus aurantium Linn (Jikak) to induce activities of quinone reductase (QR) and glutathione S-transferase (GST) in wild-type murine hepatoma cell line (Hepa 1c1c7) and Ah-receptor-defective mutant of the same cell line (Bprcl) was investigated. Hexane and chloroform fractions of C. aurantium Linn (Jikak) at doses not exhibiting cytotoxicity were effective inducers of QR (${\sim}1.8-fold$) and GST (${\sim}1.5-fold$) in Hepa 1c1c7 cells, whereas showed low QR induction potency in Bprcl cells, which indicates they have weak monofunctional action. Results suggest C. aurantium Linn (Jikak) as potentially useful cancer chemopteventive agent.

Effect of Aqua-acupuncture Solution of Medicinal Plants on Induction of Anticarcinogenic Phase II Enzymes (생약(生樂) 약침액(藥鍼液)이 암예방(癌豫防) 효소계(酵素系)에 미치는 영향(影響))

  • Induction of phase II enzymes such as quinone reductase (QR) or glutathione S-transferase (GST) is considered a major mechanism of protection against initiation of carcingenesis. The induction of detoxification enzymes and glutathione were studied with Lonicerae Flos aqua-acupuncture solution (LFAS), Angelicae gigantis Radix aqua-acupuncture solution (AGRAS), and Gamdutang aqua-acupunture solution (GAS) in murine hepatoma cells grown in microtiter plate wells. LFAS, AGRAS and GAS were potent inducers of QR activity. LFAS was induced about 2.6-fold at concentration of $3{\times}$. AGRAS and GAS were also induced about 2.6-, 1.8-fold at concentration of $5{\times}$, respectively. In addition, GST activity was increased with LFAS, AGRAS, and GAS. GSH levels were increased about 2-fold with LFAS at concentration of $5{\times}$, 1.3-fold with AGRAS at concentration of $3{\times}$, and 1.2-fold with GAS at concentration of $5{\times}$. These results suggested that LFAS, AGRAS, and GAS may act as blocking agents against carcinogenesis by induction of phase II marker enzymes.
