한약 복합추출물 HCE-2가 알코올을 투여한 쥐의 알코올 대사 및 간기능에 미치는 영향 (Protective Effect of the Herbal Combination HCE-2 on Alcohol Metabolism and Alcohol-induced Liver Injury in Ethanol-loaded Rats)

  • This study was performed to investigate the medicinal effects of the herbal combination extracts-2 (HCE-2), consisting of Artemisia capillaris Thunb., Lonicera japonica Thunb., Prunella vulgaris var. lilacina, and Hovenia dulcis Thunb. on the alcohol-induced liver injury in rats. The rats were randomly divided into four groups: normal group (n =6), non-treated control group (n =6), saline-treated group (n =6) and the herbal combination extract (HCE-2)-treated group (n =6). The rats in the alcohol-loaded groups were orally administered with ethanol at a daily dose of 4 g/kg-body weight for 5 weeks. Thirty minutes before the ethanol injection, saline or herbal combination extracts was administered by using a gastrogavage. Blood and liver tissue samples were taken out from the hearts and livers of the rats, respectively, on 15th and 38th days. The activities of serum aspartate aminotransferase (AST) and alanine aminotransferase (ALT) were measured using an enzyme-linked immunosorbent assay (ELISA). We also investigated the protective effect of the herbal combination extracts by Hematoxylin-Eosin staining on histological sections of rat liver. In this study, the oral administration of the herbal combination extracts significantly reduced the serum levels of AST and ALT, which had been raised by alcohol-induced liver injury. Histological analysis and apparent observation of liver also showed the preventive effect of the herbal combination extracts in a chronic alcohol-induced rat model. Theses results revealed that the herbal combination extracts effectively prevented hepatic damage consequent to the chronic exposure to repetitive administration of ethanol and could be used as a primary resource of a health beverage or herbal medicine, alleviating the alcohol-induced hepatic injury and hangover symptoms.

조직공학적 골을 위한 애프리플라본을 함유한 다공성 지지체의 제조 및 그 특성 (Preparation and Characterization of Ipriflavone-Loaded Poly(L-lactide-co-glycolide) Scaffold for Tissue Engineered Bone)

  • 이프리플라본은 이소플라본의 파생물로서 골의 재흡수를 방지하여 골의 재형성을 방해함으로써 골 형성에 도움을 준다. 이프리플라본은 칼슘의 양을 안정적으로 증가시킴과 함께 골수 줄기 세포의 작용으로 세포층에 칼슘을 침착시킨다. 조직공학적 골을 형성시키기 위해 락타이드-글리를라이드 공중합체 (PLGA)에 이프리플라본을 함유시킨 지치체를 용매 캐스팅/염 추출법으로 제조하였다. 수은 다공도계, 주사 전자 현미경, 시차 주사 열량계, X선 회절기를 이용하여 특성결정을 수행하였고, 이프리플라본이 함유된 지지체와 이프리플라본이 함유되지 않은 지지체를 면역이 결핍된 쥐의 피하에 삽입하여 이들의 골 형성을 비교하였다. 조직을 hematoxylin & eosin, 본쿠사 염색과 면역화학적 염색법인 콜라겐 I 형과 오스테오칼신 염색을 하였다. 이프리플라본이 함유된 담체의 다공도는 91.7% 이상이었고, 평균 다공 크기도 101 $\mu\textrm{m}$였다. PLGA로만 제조된 지지체와 이프리플라본을 50% 함유시킨 지지체를 동물 실험을 수행한 결과 이프리플라본은 피하 층과 다른 연조직에서 미분화 줄기 세포가 칼슘 침착, 골아 세포, 골상으로의 유도에 더 많은 영향을 주는 것을 관찰하였다. 결론적으로 이프리플라본을 함유한 지지체에서 이프리플라본이 골형성에 중요한 요인으로 작용한다고 사료된다.

허혈-재관류에 의해 유도된 백혈구-혈관내피세포 유착에 대한 Videomicroscopy 영상소견 (Leukocyte-Endothelial Cell Adhesion Induced by Ischemia and Reperfusion Observed with in vivo Videomicroscopy)

  • Purpose : Recent evidence suggests a possible role for leukocytes in brain injury following ischemia and reperfusion. This study examined the temporal profile of ischemic tissue damage and leukocyte response after transient middle cerebral artery occlusion(MCAO) with reperfusion in the mouse. Methods : Focal cerebral ischemia was made by temporary occluding of the stem of the proximal MCA. Two groups of the mouse were investigated : (1) sham operation(n=10), and (2)those having the arterial occlusion released after 90 minute(n=20). By 4 hours(n=10) and 24 hours(n=10) after the onset of ischemia-reperfusion, fluorescein videoimages were under-taken in the pial venules of the mouse using a closed cranial window technique. Rhodamine 6G was administered as a $80-100{\mu}l/min$ i.v. loading dose and a $30-40{\mu}l/min$ i.v. maintenance dose in saline to selectively label circulating leukocytes. Neuropathologic evaluation for brain injury was accomplished using the histochemical stain 2,3,5-triphen-yltetrazolium chloride(TTC) and hematoxylin and eosin(H & E) stain. Results : The mean number of adherent leukocytes to cerebral venules in the 90 minutes MCAO and 24 hours reperfusion group were $306{\pm}24$ compared with $72{\pm}8$ in the sham operation group. In the TTC staining method, the cortical infarct affecting 34.8% of hemispheric volume were created in all of animals (n=10) undergoing 90 minute MCAO with 24 hours reperfusion, but the infarcted area were not found in the other(sham operation and 90 minute MCAO with 4 hours reperfusion)groups. In the H & E stain, the brain tissue following 90 minute MCAO with 4 hours reperfusion revealed only a pyknosis of the nuclei with shrunken cytoplasm, but infiltrated leukocytes were not observed. After 24 hours of reperfusion, a many leukocytes were infiltrated within parenchyma and blood vessles. Conclusions : These findings demonstrate the feasiblity of continous in vivo monitoring of leukocyte adherence in cerebral venules and suggest that reperfusion induced leukocyte adherence to venular endothelium may contribute to tissue injury following focal cerebral ischemia.

아연 전처리가 시간 경과에 따른 카드뮴 유도 고환 독성에 미치는 보호 효과 (Preventive Effects of Zinc Pretreatment in the Time-course of Cadmium-induced Testicular Toxicity in the Rat)

  • Cadmium (Cd) is known to exert gonadotoxic and spermiotoxic effects. The present study was performed to investigate the morphological effects and metallothionein (MT) expression by zinc pretreatment in the course of time of cadmium-induced testicular injury in rat. Fifty male Spraque-Dawley rats weighing 160~180 g were divided into two groups : saline-pretreated cadmium group and zinc-pretreated cadmium group. Rats of two groups received subcutaneous injection of saline and 100 mg/kg $ZnSO_4$ at 0, 2, 5 and 8 hrs intervals respectively. Cadmium chloride (4.5 mg/kg $CdCl_2$) was administrated intraperitoneally at 2 hrs after zinc injection and rats were killed 0, 12, 24, 48 and 72 hrs later. Testicular tissue damages, interstitial (Leydig) cells status and MT expression were determined using hematoxylin-eosin stained sections and a computerized image analysis system on sections immunostained with a mouse anti-metallothionein respectively. Zinc pretreatment was significantly reduced testicular damages in five pathological categories after cadmium administation. The number of surviving interstitial cells was significantly higher in the zinc-pretreated group than in the saline-preatreated group at 48 and 72 hrs after cadmium administration. Non-damaged testis showed the positivity of MT staining in spermatogenic cells, Sertoli cells and endothelium of blood vessel, but not in the Leydig cells. The positivity of MT staining in saline-pretreated group was significantly reduced at 24 hrs after cadmium administration, whereas zinc-pretreated group showed strong MT positive staining similar to the 0 hr by 42 hrs after cadmium administration. In damaged testis, MT positive staining was also observed in the Leydig cells of both groups. These results suggest that a major preventive effect of zinc against cadmium-induced testicular toxicity may be due to its ability to reduce the cytotoxicity of cadmium in spermatogenic cells and Leydig cells by inhibiting the susceptibility of the testis to cadmium but not MT production by cadmium.

C57BL/6 쥐 외상성 뇌손상 모델에서 뇌 손상 정도에 따른 조직병리학적 변화 및 신경행동학적 특징 (Histopathological and Neurobehavioral Characterization in Adult Mice Exposed to Traumatic Brain Injury)

  • Purpose: Traumatic brain injury (TBI) is a significant cause of morbidity and mortality worldwide. Severity of the initial insult is one of the most significant factors affecting outcome following TBI. In order to investigate the mechanisms of cellular injury and develop novel therapeutic strategies for TBI, we designed a standardized animal TBI model and evaluated histological and functional outcomes according to the degree of impact severity. Methods: Male adult C57Bl/6 mice underwent controlled cortical impact (CCI) at varying depths of deflection (1.0-2.0 mm). We performed hematoxylin and eosin staining at 7 days after recovery from TBI. Neurobehavioral characterization after TBI was analyzed by the Barnes maze test, passive avoidance test, open field test, rotarod test, tail suspension test, and light/dark test. Results: We observed a graded injury response according to the degree of deflection depths tested (diameter, 3 mm; velocity, 3 m/s; and duration, 500 ms) compared to sham controls. In the Barnes maze test, the severe TBI (2 mm depth) group showed reduced spatial memory as compared with the sham and mild TBI (1 mm depth) groups at 7 days after TBI. There was a significant difference in the results of the open field test and light/dark test among the three groups. Conclusion: Our findings demonstrate that the graded injury responses following TBI resulted in differential histopathological and behavioral outcomes in a mouse experimental CCI model. Thus, a model of CCI with histologic/behavioral outcome analysis may offer a reliable and convenient design for preclinical TBI research involving mice.

The Protective Effects of Statins towards Vessel Wall Injury Caused by a Stent Retrieving Mechanical Thrombectomy Device : A Histological Analysis of the Rabbit Carotid Artery Model

  • Objective : Endovascular mechanical thrombectomy (MT) has been regarded as one of the standard treatments for acute ischemic stroke caused by large vessel occlusion. Despite the wide use of stent retrievers for MT, arterial intimal damage caused when deployed stent is pulled has been a certain disadvantage. We hypothesized that statin could protect and stabilize vessel damage after endovascular MT using a stent retriever. In this animal study, we observed the protective effects of the statins towards MT-induced vessel wall injury. Methods : Twenty-eight carotid arteries of fourteen rabbits were used in the experiments with MT using stent retriever. We divided the rabbits into four groups as follows : group 1, negative control; group 2, positive control; group 3, statin before MT; and group 4, statin after MT. After MT procedures, we harvested the carotid arteries and performed histomorphological and immunohistochemical analyses. Results : In histomorphological analysis with hematoxylin and eosin and Masson's trichrome stain, significant intimal thickening (p<0.05) was observed in the positive control (group 2), compared to in the negative control (group 1). Intimal thickening was improved in the statin-administered groups (groups 3 and 4 vs. group 2, p<0.05). We also observed that statin administration after MT (group 4) resulted in a more effective decrease in intimal thickness than statin administration before MT (group 3) (p<0.05). We performed immunohistochemical analysis with the antibodies for tumor necrosis factor-alpha (TNF-α), cluster of differentiation (CD)11b, and CD163. In contrast to the negative control (group 1), the stained percentage areas of all immunological markers were markedly increased in the positive control (group 2) (p<0.05). Based on statin administration, the percentage area of TNF-α staining was significantly reduced (p<0.05) in group 3, compared to the positive control group (group 2). However, significant differences were not observed for CD11b and CD163 staining. In group 4, no significant differences were observed for TNF-α, CD11b, and CD163 staining (p≥0.05). The differences in the percentage areas of the different markers between the statin-administered groups (groups 3 and 4) were also not revealed. Conclusion : We presented that statin administration before and after MT exerted protective effects towards vessel wall injury. The efficacy of statins was greater post-administration than pre-administration. Thus, statin administration in routine prescriptions in the peri-procedural period is strongly advised.

흰쥐에서 SAL5의 알코올성 지방간 형성에 미치는 영향 (Effect of SAL5 on chronic ethanol-induced fatty liver model)

  • Objective : In this study, we investigated the effect of SAL5(mixing extracts of Schisandra chinensis Baillon, Artemisia capillaris Thunb., and Aloe vera Linne) on chronic ethanol-induced fatty liver model. Methods : Sprague-Dawley male rats were fed Liber-DeCarli (normal), ethanol liquid diet (control), SAL5 (200 mg/kg). We administrated the SAL5 on chronic ethanol-induced fatty liver model for 5 weeks. We measured alkaline phosphtase (ALP), alanine transminase (ALT), aspartate transminase (AST) and ${\gamma}-glutamyl$ transpeptase (${\gamma}-GTP$) in serum and triglyceride (TG), superoxide dismutase (SOD), catalase, glutathione (GSH) and malondialdehyde (MDA) level in liver. Liver histopathology was examined by Hematoxylin-eosin and Oil red O staining of the fixed liver tissues. Real-time PCR was performed to measure the mRNA expression of inflammatory cytokines and MMP-2, MMP-9. Results : SAL5 administration resulted in significantly decreased liver marker enzymes activities of alanine transminase (ALT), ${\gamma}-glutamyl$ transpeptase (${\gamma}-GTP$) in serum and triglyceride (TG) activities in liver. The control group decreased the activities of superoxide dismutase (SOD), catalase (CAT) with the reduced level of glutathione (GSH) in liver. On the other hand, SAL5 group increased the activities of SOD, CAT and the level of GSH. SAL5 delayed the development of an alcoholic fatty liver by reversing fat accumulation in the liver, as evidenced in histological observations. The gene expression of mRNA were significantly decreased at the $IL-1{\beta}$, $TNF-{\alpha}$, NOS-II and MMP-2 by SAL5. Conclusions : These results indicate that SAL5 might have protective effect chronic ethanol-induced fatty liver models.

근관충전재(根管充塡材)의 매식(埋植)이 골조직(骨組織)에 미치는 영향(影響)에 관(關)한 실험적(實驗的) 연구(硏究) (AN EXPERIMENTAL STUDY OF THE EFFECTS ON THE BONE TISSUE BY IMPLANT OF ROOT CANAL FILLING MATERIALS)

  • The purpose of this study was the observe the toxic effects of root canal sealers in 108 white rats. Experimental animals were divided into control and experimental groups. Theree representative types of materials, such as AH26, Z.O.E. and F.R. were used in this study. Cavities were prepared on the left mandibular area of 108 white rats. Three different sealers were placed in as experiment and bone cavities were left without filling as control. The experimental animals were sacrificed by cervical dislocation at the intervals of 1, 3, 7, 14, 28 and 49 days after filling. Each specimen was fixed with 10% neutral formalin solution, decalcified with 5% nitric acid, embedded in paraffin and sectioned 5-7${\mu}$. in thickness. The paraffin sections stained with Hematoxylin - Eosin were observed through the ordinary light microscope. The results were as follows; 1. Slight toxic effect to surrounding tissue were found in every experimental specimen. 2. AH26 showed the highest inflammatory response, and F.R. showed the lowest inflammatory response which subsided and replaced by fibrosis at 4 weeks after filling. 3. The cavity filled materials, such as implanted root canal sealers, blood clots and necrotic tissue, showed a tendency to be absorbed gradually proportioned to the experimental periods. A small amount of cavity filled materials were observed in the bone cavities after 4 weeks. 4. Fibroblastic proliferation began to produce fibrous capsule around the bone cavity in 2 weeks after filling. Fibrosis was prominent at 4 weeks after filling. 5. Osteoblastic activity of surrounding bone was observed at first in 2 weeks after filling and prominent in 4 weeks after filling. Osteoblastic activity showed an increasing effect as the time prolonged. 6. Surrounding tissue of the bone cavities showed the features of tissue destruction and had very severe inflammatory response at an initial stage. Above-mentioned appeared to be recovered gradually proportioned to the experimental periods.

골유착 이전에 악정형력이 임프란트 주위조직에 미치는 영향 (THE EFFECT OF DENIAL ORTHOPEDIC FORCE TO IMPLANTS ON BONE TISSUE BEFORE COMPLETE OSSEOINTEGRATION)

  • 저자들은 임프란트 매식 후 골유착을 위한 초기 치유기간 이전에 악정형력이 임프란트 주위 조직에 미치는 영향을 관찰하기 위하여 가토 12마리의 양측 대퇴골에 임프란트를 식립하고 2주, 4주 그리고 6주후에 각각 300g의 악정형력을 Ni-Ti close coil spring을 이용하여 매식된 임프란트에 4주동안 가하고 관찰한 후 다음과 같은 결론을 얻었다. 1. 모든 실험군의 임프란트는 4주간의 악정형력 적용후에도 안정된 견고성을 유지하였다. 2. 2주 실험군에서 대조군에 비해 임프란트와 골조직 사이의 섬유조직 증식이 많이 관찰 되었으나 특이할만한 염증소견은 관찰되지 않았다. 3. 4주 실험군, 대조군에서는 2주 실험군, 대조군과 각각 비교하였을 때 보다 많은 양의 골재생이 관찰 되었으며 실험군과 대조군 모두에서 임프란트와 골조직 사이의 섬유조직은 관찰하기 힘들었다. 4. 6주에서는 실험군과 대조군 사이에 뚜렷한 차이가 발견되지 않았다. 이상의 결과로 보아 임프란트 주위의 골조직 재생이 충분하지 않아도 골의 양과 질이 우수해 임프란트 식립시 견고한 초기고정을 얻을 수 있다면 골유착이 완성되는 초기 치유기간 이전에도 치과교정적 고정원으로 사용가능할 것으로 사료된다.
