Directed Mutagenesis of the Bacillus thuringiensis Cry11A Toxin Reveals a Crucial Role in Larvicidal Activity of Arginine-136 in Helix 4

  • Based on the currently proposed toxicity model for the different Bacillus thuringiensis Cry $\delta$-endotoxins, their pore-forming activity involves the insertion of the ${\alpha}4-{\alpha}5$ helical hairpin into the membrane of the target midgut epithelial cell. In this study, a number of polar or charged residues in helix 4 within domain I of the 65-kDa dipteranactive Cry11A toxin, Lys-123, Tyr-125, Asn-128, Ser-130, Gln-135, Arg-136, Gln-139 and Glu-141, were initially substituted with alanine by using PCR-based directed mutagenesis. All mutant toxins were expressed as cytoplasmic inclusions in Escherichia coli upon induction with IPTG. Similar to the wild-type protoxin inclusion, the solubility of each mutant inclusion in the carbonate buffer, pH 9.0, was relatively low When E. coli cells, expressing each of the mutant proteins, were tested for toxicity against Aedes aegypti mosquito-larvae, toxicity was completely abolished for the alanine substitution of arginine at position 136. However, mutations at the other positions still retained a high level of larvicidal activity Interestingly, further analysis of this critical arginine residue by specific mutagenesis showed that conversions of arginine-136 to aspartate, glutamine, or even to the most conserved residue lysine, also abolished the wild-type activity The results of this study revealed an important determinant in toxin function for the positively charged side chain of arginine-136 in helix 4 of the Cry11A toxin.

Correction of microtia with constriction features using a superficial temporal fascial flap combined with a rib cartilage graft

  • Background Microtia with constricted features is characterized by a short helical length of variable severity, upper antihelical or scaphal deficiency, and a downfolded upper ear. No consensus has been reached regarding the most appropriate surgical method for this condition. In this study, we aimed to introduce a simple and safe surgical method for the correction or reconstruction of upper helix ear deformities. Methods Between February 2011 and June 2014, eight patients with microtia with constricted upper helix ear deformity underwent reconstruction of the ear deformity. The upper ear helical framework was constructed by carving and curving the eighth rib cartilage harvested from the ipsilateral chest wall, covering this cartilage with a superficial temporal fascial flap, and adjusting the skin graft to align with the ear contour. To evaluate their satisfaction, patients were asked to complete a questionnaire regarding ear shape, symmetry, position, color, and overall outcome scored on a 5-point scale at 12 months postoperatively. Results None of the patients experienced severe complications in the reconstructed ear. The preoperative and postoperative vertical ear length ratios were 0.88 and 1.02, respectively. And the mean patient satisfaction scores for shape, symmetry, position, color, and overall outcome were 4.2, 4.5, 4.7, 4.4, and 4.6 out of 5 points, respectively. All patients expressed a high level of satisfaction at 12 months postoperatively. Conclusions Our technique provides a good alternative method for the reconstruction of moderate constricted upper helix ear deformities in patients who meet the surgical indications with satisfactory outcomes and few complications.

Structures of antimicrobial peptides HP (2-20) and interactions between HP(2-20) and membrain studied by NMR spectroscopy

  • HP(2-20) (AKKVFKRLEKLEKLFSKIQNDK) derived from the N-terminus of Helicobacter pylori Ribosomal Protein L1 shows potent antimicrobial activity against bacterial, fungi and cancer cells without cytotoxic effect. In order to investigate the relationships between antimicrobial activity and the structures, several analogues have been designed and synthesized. The structures of these peptides in SDS micelles have been investigated using NMR spectroscopy and they revealed that analogue 3 has the longest, well-defined alpha-helix from Val5 to Trp19. NOESY experiments performed on HP and its analogues in nondeuterated SDS micelles show that protons in the indole ring of Trp16 are in close contact with methylene protons of SDS micelles. In order to probe the position of HP and its analogues relative to the SDS micelles, spin-labeled stearate was added. Large effects are observed for the chemical shifts and the intensities of Phe5, Glu9, Phe12, and Trp16 within the helix region by 16-doxylstearate. This result implies that 16-doxylstearate is located in the center of the micelles and the hydrophobic phase of the amphiphilic ${\alpha}$-helix is located in contact with the acyl chains of the micelles. Also, Lys3 and Lys4 at N-terminus and Lys20 at C-terminus may produce an optimal arrangement for electrostatic interactions between the sulfate head groups of the SDS and the positively charged lysyl N$\sub$3/$\^$+/. Interactions between the indole ring of Trp and the membrane, as well as the amphiphilic ${\alpha}$-helical structure of HP induced by Trp at the C-terminus may allow HP to span the lipid bilayer. These structural features are crucial for their potent antibiotic activities.

  • 화학적 살충제는 빠르고 정확하게 해충을 방제하는 효과를 나타내지만, 인간과 다른 유기체에 대한 살충제 잔류의 위험성, 화학적 살충에 대한 저항성을 가진 해충의 출현, 생태계의 교란 등 다양한 부작용이 있다. 이 때문에 생물적 방제를 통한 해충 방제는 효과적인 수단이 될 수 있다. 특히 채소 작물 등에 주로 발생하는 응애는 생산량 저하 및 품질 저하 등과 같은 다양한 문제를 일으킬 수 있다. 특히 진딧물이나 점박이응애는 화학적 살충제에 대해 높은 저항성이 발달되어 문제가 되고 있다. 본 연구에서는 아이비(Hedera helix), 란타나(Lantana camara), 예루살렘체리(Solanum pseudocapsicum)를 통한 천연 식물 추출물의 응애에 대한 생물적 방제 효과를 확인하였다. 각 추출물은 0.5%, 0.05% 주정 및 메탄올로 추출하였으며, 응애 방제 효과(%)는 아이비>란타나>예루살렘체리로 나타났다. 각 추출물은 GC/MS 분석을 통해 생리활성 후보 물질을 확인하였다. 이를 통해 농업 이익 증대, 생산량 유지와 더불어 환경 및 인간과 다른 유기체에 피해를 주지 않으며 효과적으로 응애를 방제 할 수 있는 천연 생물적 방제제로의 가치를 확인하였다.

  • The retraction of anterior teeth is one of the fundamental methods in orthodontic treatment and a proper position and angulation of anterior teeth after the retraction are very important for esthetics, stability, and function of teeth. In this research we analyzed, by Finite Element Method, the stress distribution on the periodontal ligament according to the variation of force and moment applied on the crown and predict the pattern of movement of maxillary central incisor. At the same time, the amount of force and moment caused by activation of the loop which was used for retraction of maxillary central incisor was analyzed by Finite Element Method. We observed the following results: 1) We could control the stress distribution on the periodontal ligament by proper moment/force ratio on maxillary right central incisor and predict the pattern of movement of maxillary right central incisor. 2) The amount of stress on the periodontal ligament as well as the moment/force ratio demanded by each pattern of movement increased as the destruction of alveolar bone was worse. 3) The moment/force ratio demanded by each pattern of movement decreased as the angle between the maxillary central incisor and occlusal plane decreased. 4) The force with the open loop was shown to be large compared to that with the closed loop. Also, the force with the helix decreased by 30% compared to that without the helix. 5) Under the same conditions we observed a larger moment/force ratio when the open loop and/or the helix were used.

Cloning and Sequencing Analysis of the Repressor Gene of Temperate Mycobacteriophage L1

  • The wild-type and temperature-sensitive (ts) repressor genes were cloned from the temperate mycobacteriophage L1 and its mutant L1cIts391, respectively. A sequencing analysis revealed that the $131^{st}$ proline residue of the wild-type repressor was changed to leucine in the ts mutant repressor. The 100% identity that was discovered between the two DNA regions of phages L1 and L5, carrying the same sets of genes including their repressor genes, strengthened the speculation that L1 is a minor variant of phage L5 or vice versa. A comparative analysis of the repressor proteins of different mycobacteriophages suggests that the mycobacteriophage-specific repressor proteins constitute a new family of repressors, which were possibly evolved from a common ancestor. Alignment of the mycobacteriophage-specific repressor proteins showed at least 7 blocks (designated I-VII) that carried 3-8 identical amino acid residues. The amino acid residues of blocks V, VI, and some residues downstream to block VI are crucial for the function of the L1 (or L5) repressor. Blocks I and II possibly form the turn and helix 2 regions of the HTH motif of the repressor. Block IV in the L1 repressor is part of the most charged region encompassing amino acid residues 72-92, which flanks the putative N-terminal basic (residues 1-71) and C-terminal acidic (residues 93-183) domains of L1 repressor.

알라닌 올리고머의 배좌구조에 관한 양자화학적 계산 (Quantum Chemical Calculations on the Conformational Structure of the Alanine Oligomer Model)

  • 본 연구는 올리고펩티드의 연쇄성장 과정에서의 배좌의 변화를 조사하기 위하여 알라닌 올리고머 모델의 2량체 부터 5량체 까지 양자화학 계산(QCC)에 의한 구조최적화 계산을 하였다. 말단기의 영향을 확인하기 위하여 N말단 및 C말단이 공히 amide 구조를 갖는 "amide type" ($CH_3CONH-and-CONHCH_3$)과 N말단만이 methyl 구조를 갖는 "methyl type" ($CH_3NH-and-CONHCH_3$)의 2종류 모델을 준비하였다. 5량체 까지의 구조 최적화 계산 결과 3종류의 형태이성질체로 수렴이 되었다. 안정성이 높은 순서로 각각 ${\alpha}$ 나선형, PPII 유사형, ${\beta}$ 확장형으로 나타났으며, 사슬회전 방향이 서로 반대인 형태이성질체간의 에너지 값은 동일하게 나타났다 (5량체, "amide type" 좌 우회전형: ${\Delta}E=-1.05$, "methyl type" 좌 우회전형: ${\Delta}E=-1.62$). ${\alpha}$ 나선형의 모노머 단위당 에너지변화(${\Delta}E$)는 모노머의 증가와 함께 감소하였다.

  • Experiments were conducted to investigate propagation, shade tolerance, cold resistance and growth rate of evergreen ground cover plants ; Vinca minor K , Ajuga reptans L., Ophiopogon japonicus ker. f nanus hort , and Hedera helix.. The results were as follows : 1.It is convenient to use the growing case for rice seed to grow and transplant Vinca minor seed-ling. The most proper density of transplanting Vinea was 180 plants per 1m$^2$. At the end of growing season, the coverage came up to 90% The rooting ability of Hedera helix was best to cut from April to May (temperature 15~23˚C). The seedling length of Parthenocissus quinguefohlia were irregular and ranged from 5 cm of 200cm. 2. Ajuga reptans L., Ophiopogon j. and Vinca minor L. grew better under 50% light intensity than full sunlight. Particularly, under full sunlight and aestival high temperature, there were a few withering plants in Ajuga reptans L. , and V Vilica minor L. 3. Of Vinca minor K., Ajuga reptans L. , Ophiopogon japonicus ker. f. nanus hort. and Hedera helix. acclimatized in open field, where temperature was from -7~8˚C. There were few plants had damage in low temperature incubator to -16˚C after field acclimatization. 4.Tendrils of Parthenocissus quinguefolia strongly clung to the wire netting stone fence but intruded into the gap and cranny of the precast concrete fence and so clung to, so that it needed to install the bending net on the precast concrete fence.

Crystal Structure of Histidine Triad Nucleotide-Binding Protein from the Pathogenic Fungus Candida albicans

  • Histidine triad nucleotide-binding protein (HINT) is a member of the histidine triad (HIT) superfamily, which has hydrolase activity owing to a histidine triad motif. The HIT superfamily can be divided to five classes with functions in galactose metabolism, DNA repair, and tumor suppression. HINTs are highly conserved from archaea to humans and function as tumor suppressors, translation regulators, and neuropathy inhibitors. Although the structures of HINT proteins from various species have been reported, limited structural information is available for fungal species. Here, to elucidate the structural features and functional diversity of HINTs, we determined the crystal structure of HINT from the pathogenic fungus Candida albicans (CaHINT) in complex with zinc ions at a resolution of $2.5{\AA}$. Based on structural comparisons, the monomer of CaHINT overlaid best with HINT protein from the protozoal species Leishmania major. Additionally, structural comparisons with human HINT revealed an additional helix at the C-terminus of CaHINT. Interestingly, the extended C-terminal helix interacted with the N-terminal loop (${\alpha}1-{\beta}1$) and with the ${\alpha}3$ helix, which appeared to stabilize the dimerization of CaHINT. In the C-terminal region, structural and sequence comparisons showed strong relationships among 19 diverse species from archea to humans, suggesting early separation in the course of evolution. Further studies are required to address the functional significance of variations in the C-terminal region. This structural analysis of CaHINT provided important insights into the molecular aspects of evolution within the HIT superfamily.

소프트웨어 구현에 적합한 스트림 암호의 대수적 공격에 대한 안전성 (Security Analysis of Software-Oriented Stream Ciphers against Algebraic Attacks)

  • 본 논문에서는 최근 제안된 소프트웨어 구현에 적합한 (블록 단위) 스트림 암호는 HELIX, SCREAM, MUGI, PANAMA 알고리즘들의 대수적 공격에 대한 안전성을 분석한다. 대수적 공격은 알려진 입출력 쌍을 가지고 내부 알고리즘의 기본 대수 방정식을 이용하는 방법으로, 과포화된 다변수 연립 방정식을 통하여 변수의 값을 얻고 이를 이용하여 키를 복구해내는 방업이다. 이 분석법은 초기 대수적 특성을 지닌 블록 암호의 분석에 적용되었으나. 이후 블록 암호보다는 스트림 암호의 분석에 보다 용이하게 적용되었다. 그러나 일반적인 알고리즘을 이 분석법을 그대로 적용하기에는 많은 어려움이 존재한다. 따라서 본 논문에서는 최근 제안된 워드 단위의 소프트웨어 구현에 적합한 스트림 암호의 각 알고리즘에 대해 대수적 방정식과 변수의 수를 비교 및 분석함으로써 각 알고리즘의 대수적 공격에 대한 내성을 살펴본다. 또한 이러한 분석을 통해 대수적 공격에 안전한 소프트웨어 구현에 적합한 스트림 암호의 설계 시 고려해야 할 세 가지 설계 고려사항을 제시한다.
