Kinetic analysis of Drosophila Vnd protein containing homeodomain with its target sequence

  • Homeodomain (HD) is a highly conserved DNA-binding domain composed of helix-turn-helix motif. Drosophila Vnd (Ventral nervous system defective) containing HD acts as a regulator to either enhance or suppress gene expression upon binding to its target sequence. In this study, kinetic analysis of Vnd binding to DNA was performed. The result demonstrates that DNA-binding affinity of the recombinant protein containing HD and NK2-specific domain (NK2-SD) was higher than that of the full-length Vnd. To access whether phosphorylation sites within HD and NK2-SD affect the interaction of the protein with the target sequence, alanine substitutions were introduced. The result shows that S631A mutation within NK2-SD does not contribute significantly to the DNA-binding affinity. However, S571A and T600A mutations within HD showed lower affinity for DNA binding. In addition, DNA-binding analysis using embryonic nuclear protein also demonstrates that Vnd interacts with other nuclear proteins, suggesting the existence of Vnd as a complex.

  • The use of error compensation techniques has been recognized as an effective way in the improvement of the accuracy of a machine tool. The laser measurement method for identifying position errors of machine tool has the disadvantages such as high cost, long calibration time and usage of volumetric error synthesis model. Accordingly, this paper deals with analysis of the position errors of a machine tool using ball bar test without using complicated error synthesis model. Statistical analysis method was adopted in this paper for deriving position errors using hemispherical helix ball bar test.

De Novo Design and Their Antimicrobial Activity of Stapled Amphipathic Helices of Heptapeptides

  • In this study we designed and synthesized several heptapeptides that are enforced to form an amphipathic helix using all-hydrocarbon stapling system and evaluated their antimicrobial and hemolytic activities. The antimicrobial activity showed clear structure-activity relationships, confirming the importance of helicity and amphipathicity. Some stapled heptapeptides displayed a moderate antimicrobial activity along with a low hemolytic activity. To our best knowledge, although not highly potent, these stapled peptides represent the shortest helical amphipathic antimicrobial peptides reported to date. The preliminary data obtained in this work would serve as a good starting point for further developing short analogs of amphipathic helical antimicrobial peptides.

와이어 코일이 삽입된 나선형 내면가공관의 열전달 및 압력강하 특성 (Characteristics of Heat Transfer and Pressure Drop for Spirally Indented Tubes with Wire Coil Inserts)

  • The characteristics of heat transfer and pressure drop through tubes has been investigated experimentally for a compound heat transfer enhancement. The test tubes were spirally indented tubes with wire coil inserts which had a various combinations of pitch and helix angles. Pure water was used as working fluids for the experiments, Heat transfer coefficients and friction factors of the test tubes were evaluated from the values of measured temperatures, flow rates and pressure drops. An performance evaluation was performed to find an optimal combination of spirally indented tubes with wire coil inserts. When the helix angle of wire coil insert are $71^{\circ}-72^{\circ}$, the best heat transfer enhancement was shown. The friction factor was 9 - 13 times higher than those in smooth tubes, and the heat transfer was enhanced a maximum of 500%.

  • MatR in Rhizobium trifolii is a malonate-responsive transcription factor that regulates the expression of genes, matABC, enabling decarboxylation of malonyl-CoA into acetyl-CoA, synthesis of malonyl-CoA from malonate and CoA, and malonate transport. According to an analysis of the amino acid sequence homology, MatR belongs to the GntR family The proteins of this family have two-domain folds, the N-terminal helix-turn-helix DNA-binding domain and the C-terminal ligand-binding domain. In order to End the malonate binding site and amino acid residues that interact with RNA polymerase, a site-directed mutagenesis was performed. Analysis of the mutant MatR suggests that Arg-160 might be involved in malonate binding, whereas Arg-102 and Arg-174 are critical for the repression activity by interacting with RNA polymerase.

  • STD11 is one of difficult-to-cut materials and its cutting characteristic data is not built enough. A bad cutting condition of it leads to low productivity of die and mould, so it is necessary to evaluate the machining characteristics of STD11. In this paper, the relations of the geometry of ball-end mill and mechanics of machining with it are studied. The helix angle of ball-end mill varies according to a location of elemental cutting edge in the cutting process are difficult to calculate accurately. To calculate instantaneous cutting forces, it is supposed that the tangential, radial and axial cutting force coefficients are functions of elemental cutting edge location. Elemental cutting forces in the x,y and z direction are calculated by coordinate transformation. The total cutting forces are calculated by integrating the elemental cutting forces of engaged cutting edge elements. This model is verified by slot and side cutting experiments of STD11 workpiece which was heat-treated to HRC45.

  • 본 논문에서는 인터체인지나 램프시설 등에서 볼 수 있는 공간 곡선부재들을 나선형 곡선부재로 이상화시켰다. 나선의 기하학적 특성치와 초기 곡률을 고려하여 평형방정식을 유도하였으며 모우드 형상함수를 가정하여 단순지지된 나선형 곡선부재의 규준화된 고유진동수를 계산할 수 있는 모우드 방정식을 유도하였다. 이러한 모우드 방정식을 이용하여 나선형 곡선부재의 고유진동수, 모우드 형상 그리고 고유진동수의 분포를 규명하였다.

Identification of Three Positive Regulators in the Geldanamycin PKS Gene Cluster of Streptomyces hygroscopicus JCM4427

  • In the Streptomyces hygroscopicus JCM4427 geldanamycin biosynthetic gene cluster, five putative regulatory genes were identified by protein homology searching. Among those genes, gel14, gel17, and gel19 are located downstream of polyketide synthase genes. Gel14 and Gel17 are members of the LAL family of transcriptional regulators, including an ATP/GTP-binding domain at the N-terminus and a DNA-binding helix-turn-helix domain at the C-terminus. Gel19 is a member of the TetR family of transcriptional regulators, which generally act to repress transcription. To verify the biological significance of the putative regulators in geldanamycin production, they were individually characterized by gene disruption, genetic complementation, and transcriptional analyses. All three genes were confirmed as positive regulators of geldanamycin production. Specifically, Gel17 and Gel19 are required for gel14 as well as gelA gene expression.

헬리컬 스프링의 자유진동 해석 (Free Vibration Analysis of Helical Springs)

  • Free vibration analysis of helical springs was performed by the use of the commercial finite element analysis program, ANSYS. The investigation of national frequency was focused on the effect of various parameters such as boundary conditions, spring indices, number of coil turns and helix angles which are considered to affect the free vibration of a spring. The finite element method was validated by comparison with the result of a previouosly published literature. The similarity of frequency trend was shown among three boundary conditions: clamped-clamped, free-free, simpliy supported-simply supported but there was no similarity in light of mode shapes among them. Several modes showed similar frequencies on and near the frequencies identified by the natural frequency formula of Wahl. Natural frequencies increased with spring indices and number of turns decreasing and with helix angles increasing. The results investigated by finiete element method were compared with the experemental result and theoretical result and showed a good agreement among them.
