Characterization of Biomass-Based Foam Structures for Home-Meal-Replacement Containers (가정간편식 용기용 바이오매스 기반 발포구조체의 특성에 관한 연구)

  • A series of foamed plastic sheets containing biomass (as HMR container) were developed via different foaming process temperatures, and their density, porosity, WVTR, and pore morphology were evaluated. Thermal stability of samples during re-heating the food in oven, change in morphology, density, porosity, and WVTR were investigated using a simulated thermal shock process according to MIL-STD-883E assay. As such, the pore size of samples was generally increased with increasing temperature of the foaming process. It can be explained that as foaming temperature increased, the viscosity of molten resins and the repulsive force against pore expansion decreased. In addition, an increase in the thermal shock cycle reduced the pore size and WVTR, while density increased because high temperature treatment that softened the sheet matrix was followed by a low temperature incubation, which contracted the matrix, thereby changing the physical and morphological properties of samples. However, an insignificant change in density was observed and WVTR tended to be decreased, indicating that as-prepared foamed plastic sheets could be used as a high thermal stable container for HMR application. Therefore, it found that the properties of newly developed HMR containers containing biomass were dependent on the foaming process temperature. Moreover, to better understanding of these newly developed containers, further investigations dealing with foaming process temperature based on various food items and cooking conditions are needed.

Production of Phenyl Lactic Acid (PLA) by Lactic Acid Bacteria and its Antifungal Effect

  • Phenyllactic acid (PLA) which is known as antimicrobial compound can be synthesized through the reduction of phenylpyruvic acid (PPA) by lactate dehydrogenase (LDH) of lactic acid bacteria (LAB). LAB producing PLA was isolated from Korea Kimchi and identified to Lactobacillus plantarum SJ21 by 16 rRNA gene sequence analysis. Cell-free supernatant (CFS) from L. plantarum SJ21 was assessed for both the capability to produce the antimicrobial compound PLA and the antifungal activity against four fungal pathogens (Rhizoctonia solani, Aspergillus oryzae, Botrytis cinerea, and Collectotricum aculatum). PLA concentration was investigated to be 3.23mM in CFS when L. plantarum SJ21 was grown in MRS broth containing 5mM PPA for 16 h. PLA production also could be promoted by the supplement of PPA and phenylalanine in MRS broth, but inhibited by the supplement of 4-hydroxyphenylpyruvic acid and tyrosine as precursors. Antifungal activity demonstrated that all fungal pathogens were sensitive to 5% CFS (v/v) of L. plantarum SJ21 with average growth inhibitions ranging from 27.32% to 69.05% (p<0.005), in which R. solani was the most sensitive to 69.05% and followed by B. cinerea, C. aculatum, and A. oryzae. The minimum inhibitory concentration (MIC) for commercial PLA was also investigated to show the same trend in the range from $0.35mg\;mL^{-1}$ (2.11 mM) to $0.7mg\;mL^{-1}$ (4.21 mM) at pH 4.0. The inhibition ability of CFS against the pathogens was not affected by heating or protease treatment. However, pH modification in CFS to 6.5 caused an extreme reduction in their antifungal activity. These results may indicate that antifungal activities in CFS were caused by acidic compounds like PLA or organic acids rather than proteins or peptides molecules.

Effect of Bambusae caulis in Liquamen on Streptozocin-Induced Diabetic C57BL/6 Mice

  • Bambusae caulis in Liquamen is one of the important herbal medicine produced by heating bamboo indirectly and is used for treatment of stroke, hypertension, and diabetes etc. Recently the mechanism of clinical effects on Bambusae caulis in Liquamen has been studied. This experiment was conducted to confirm the clinical effects of Bambusae caulis in Liquamen on type 1 diabetes and its related mechanism. We divided C57BL/6 mice into 3 groups and induced them to be type 1 diabetes by injection of streptozocin into peritoneum. The dosage of each group was 150 mg/kg once only, 140 mg/kg once only and 40 mg/kg for 5 days respectively. The two groups injected streptozocin for once took orally Bambusae caulis in Liquamen after the induction of diabetes, and the other one group was given Bambusae caulis in Liquamen during the diabetes inducing period. As the result, the two diabetes-induced groups showed blood glucose decreasing effect by Bambusae caulis in Liquamen on an average, but they didn't show the signiftcant differences statistically. But Bambusae caulis in Liquamen showed the anti-diabetic effect suppressing blood sugar rising trend during the diabetes inducing peried (P<0.05). The anti-oxidative effect of Bambusae caulis in Liquamen was measured with the hypoxanthine/xanthine oxidase (HX/XOD) system. The quantity of ROS was measured using DCFDA reagent indirectly. As the result, $10\%$ solution of Bambusae caulis in Liquamen showed anti-oxidative effect by scavenging $93.4\%$ superoxide as compared with control group. It is suspected that the anti-oxidative effect of Bambusae caulis in Liquamen suppressed the increase of blood glucose in the diabetes-inducing group. These results could be useful data to understand the effect of Bambusae caulis in Liquamen on type 1 diabetes and type 1 diabetes developing because ROS were closely connected with the induction and complications of diabetes.

  • To develope a rapid processing method for fish sauce, processing conditions of fish sauce from sardine waste was investigated. The chopped waste was homogenized and hydrolyzed by commercial proteolytic enzymes such as Complex enzyme-2000($2.18\cdot10^4$ U/g solid) and Alcalase($1.94\cdot10^4$ U/g solid) in a cylindrical vessel with 4 baffles and 6-bladed turbine impeller. Optimal temperature for the case of hydrolysis with Complex enzyme-2000 was 50 and that with Alcalase was $55^{\circ}C$. In both cases, the reasonable pH, amount of water for homo-genization, enzyme concentration and hydrolyzing time were 8.0, $40\%$ (W/W), $3\%$ and 100 min, respectively. Heating of the filtrated hydrolysate for 2 hours at $90^{\circ}C$ with $6\%$ of invert sugar was suitable for pasteurization of the hydrolysate and inactivation of enzymes. Flavor, taste and color of the hydrolysate was improved during the thermal treatment in which the browning reaction products might participate and result in antioxidative and bactericidal effects. Combined use of $0.005\%$ of Caryophylli flos with invert sugar was also effective for the improvement of taste. Yield of the fish sauce based on the total nitrogen in the raw sardine waste was $91.2\~92.3\%$ and $87.2\~87.8\%$ of the total nitrogen in the fish sauce was in the form of amino nitrogen. The pH, salinity and histamine content of the fish sauce prepared with $15\%$ of table salt were $6.1\~6.2$, $14.2\~14.4\%$ and less than $10mg\%$, respectively. The fish sauce was stable during the storage of 60 days at $26\pm3^{\circ}C$ on bacterial growth and its quality was also maintained.

  • The purpose of this study is to establish effective conditions for controlling $CH_4$, $N_2O$ emission from organic Waste / wastewater treatment processes. Continuous and batch experiments were conducted to treat the micro algae from polluted and eutrophicated lakes through the thermophilic oxic process. The microalgae used were mainly Microcystis sp.(collected from eutrophic lake) and Chlorella sp. (cultured in laboratory) Wasted cooking oil was added by aid-heating source. Physico-chemical components of sludges and microalgae were analyzed. In batch experiments, air supply was changed from 50ml/min to 150ml/min. The temperature. water content and drained water were affected by the air flow rate at initial stage. However, there was almost no influence of air flow rate on them in middle and last stages. At air flow rate of 100ml/min, the degradation rate of organic material was higher than that at other air flow rates. $CO_2$ concentration in exhaust was proportional to the strength of aeration, especially at initial stage when degradation was active. $CH_4$ with low concentration was detected only at starting stage when air diffusion was not enough. $N_2O$ production was not affected by variation of air supply. In continuous experiments no matter what the dewatering methods (with PAC and without PAC) and media (wood chip and reed chip) were changed, $N_2O$ was almost not affected by variation of injected air. Result showed that the reed chips using for lake purification could be used as media for thermophilic oxic process in lake and marshes area. $CO_2$ concentration was not so much affected by the change of dewatering methods and media types. $CH_4$ was not detected in the experimental period. So it can be shown that the thermophilic oxic process had been well operated in wide handling conditions regardless of media and dewatering methods.

Changes in Days to Drying and Some Chemical Components by Different Drying Methods in Paeoniae radix (건조방법에 따른 작약근 건조 소요일수 및 성분 변화)

  • Seven different drying methods were tested in peony roots of Euisung cultivar, harvested in February, in three year's old plant. The roots were selected in length and diameter and half of the samples were removed cork-layers to compare the effects of cork-layer in processing of drying. The 3$0^{\circ}C$ heat drying without cork-layer reduced in days to drying by five days compared to those of the with cork-layers at the same temperature. The 5$0^{\circ}C$ heat drying after cork-layers removed was the most effective in days to drying. In quality of skin color of the 5$0^{\circ}C$ heating was worse to compare with the lower drying temperature. In the drying at room temperature and the heat drying at lower temperature, the paeoniflorin content in drying after cork-layers removed were higher than that of the drying with cork-layers. However, in the boiling water treatment, the paeoniflorin contents in drying after cork-layers removed were lower than those of with cork-layers. In heat drying, paeoniflorin content showed a decreasing tendency to increase of drying temperature. Total sugars in the peony roots showed a decreasing tendency according to the drying temperature increasing, but starch concentration showed a increasing tendency at the same condition. Concentrations of crude protein, crude fiber and crude ash were showed no differences in various drying methods and the materials with or without cork-layers. Relationships between the paeoniflorin and total sugars, and the paeoniflorin and starch were different significantly by the materials of cork-layers removed or not. The 30~4$0^{\circ}C$ heat drying without cork-layers was the most advisable condition for drying in paeoniflorin concentration, days to drying and skin color after drying.

Investigation on Economical Feasibility for Energy Business of Waste Water Sludge Discharged in 'A' Industrial Complex (A-산업단지 발생 슬러지의 에너지화를 위한 경제성 검토)

  • Industrial complexes in Korea have been vigorously established by economic development plan and development policy of industry in 1960s. Recently, Korean government has promoted Eco Industrial Park (EIP) project to recycle by-products and wastes in industrial park In this study, we analyzed the physical and chemical properties for the sludges discharged from A industrial complex. And we investigated the economic feasibility and environmental impact of sludge to energy facilities. The analysis results indicated that the petrochemical industry were 92% in sludge production, the highest treatment amount was landfill, followed by incineration and recycling and then ocean disposal. Wastewater sludge and process sludge samples are collected and analyzed to use as basic data on economic feasibility and environmental impact. Weighted average heating value of sludge samples was 3,891kcal/kg. Based on this data, installation and operation costs, operation returns of operating the drying facility are estimated, compared with cogeneration facility. And this study examines how the payback period of each simulation(total 8 case) with the important parameter changes. As a result, it was found that what needs the shortest payback period is 3years with connection of drying facility and cogeneration facility based on the government's financial subsidy system.

Thermal Inactivation of Sodium-Habituated Staphylococcus aureus in Ready-to-Heat Sauces

  • The objective of this study was to evaluate the effect of sodium habituation on thermal resistance of Staphylococcus aureus in various ready-to-heat (RTH) sauces. The strain mixture of S. aureus strains KACC10768, KACC10778, KACC11596, KACC13236 and NCCP10862 was habituated up to 9% of NaCl. The inocula of NaCl-habituated and non-habituated S. aureus were inoculated in 5 g portions of pork cutlet, meat and Carbonara sauces at 7 Log CFU/g, and the samples were vortexed vigorously. The inoculated samples were then exposed to 60 and $70^{\circ}C$ in a water-bath, and survivals of total bacteria and S. aureus were enumerated on tryptic soy agar and mannitol salt agar, respectively, every 30 min for 120 min. At 60oC, the cell counts of total bacteria and the significant difference in survivals between sodium-habituated and non-habituated S. aureus were observed only in the Carbonara sauce; the tailing effect, which is the period of no reduction of bacterial cell counts, was observed in pork cutlet, meat and Carbonara sauces subjected to $60^{\circ}C$. At $70^{\circ}C$, total bacterial populations and sodium-habituated and non-habituated S. aureus cell counts in meat and Carbonara sauce also significantly decreased (p<0.05) after 30 min of heat treatment, followed by the obvious tailing effect. Sodium-habituated S. aureus cell counts in meat and Carbonara sauces were higher (p<0.05) than those of non-habituated S. aureus at $70^{\circ}C$. The results indicate that sodium habituation of S. aureus cells may increase the thermal resistance of the pathogen in RTH sauces; moreover, heating RTH sauces for a short time before serving may not sufficiently decrease the cell counts of S. aureus, particularly for sodium-habituated strain.

  • Vitrification technology has been widely applied as one of effective processing methods for wastes generated in nuclear power plants. The advantage of vitrifying for low- and intermediate-level radioactive wastes has a large volume reduction and good durability for the final products. Recently, a filter using on HVAC(Heating Ventilating & Air Conditioning System) is composed with media (glass fiber) and separator (aluminum film) has been studied the proper treatment technology for meeting the waste disposal requirement. Present paper is a feasibility study for the filter vitrification that developing of the glass compositions for filter melting and melting test for physicochemical characteristic evaluation. The aluminum metal of film type is preparing with 0.5 cm size for proper mixing with glass frit, glass fiber is also preparing with 1 cm size within crucible. The glass compositions should be developed considering molten glass are related with wastes reduction. Glass compositions obtained from developing on glass formulation are mainly composed of $SiO_2$ and $B_2O_3$ for aluminum metal. A variety of factors obtained from the glass formulation and melting test are reviewed, which is feeding rate and glass characteristics of final products such as durability for implementing the wastes disposal requirement.

  • In order to prepare digested Protein source for the Weanling Food from soybean, an attempt was made to decompose steamed soybean protein to amino acids and peptides by protease and cellulase produced from Aspergillus niger and Aspergillus sojae. In this paper, the optimum condition for digestion of soybean protein were studied and also investigated the effects of decolorization of it. As the results, followings were obtained; 1. As steaming conditions, a treatment under 15 lb of pressure and 10 minutes of heating shows most effective. 2. The optimum pH of Asp, sojae enzyme for the digestion of soybean protein is 6.0, while that of Asp. niger enzyme is 4.4. In successievly-decomposing with Asp. sojae and Asp. niger, it shows the most effective on ratio of water-soluble-nitrogen to total nitrogen and amino-nitrogen to total nitrogen than any other separate treatments. 3. The suitable amount of the enzyme solution to that of the soybean substrate paste, in volume, is 1 : 2. 4. Digestion ratio of soybean protein indicates the gradual and steady effects of increasing time of digestion, but 8 hour-digestion regarding to putrefaction was suitable. 5. The most effective decolorization was successively passed on culumns of active carbon and anion exchanger (Dowex 2-x-8) at room temperature. In separate treatments, the effective order of decolorization was as follows; (Dowex 2-x-8)>Active carborn>Amberite IR-120 6. The powder type of the soy protein source obtained by concentration below $60^{\circ}C$ contains 12.51% of moisture, 66.31% of protein, 4.25% of fat, 12.75% of carbohydrate, 4.18% of ash.
