Peptide Profiling and Selection of Specific-Expressed Peptides in Hypoglycemic Sorghum Seed using SELDI-TOF MS (SELDI-TOF MS를 활용한 혈당강하 수수 종자의 펩타이드 프로파일링 및 특이 발현 펩타이드 선발)

  • Sorghum seed is traditionally used as secondary food sources in addition to rice in Korea. While the hypoglycemia regulating phytochemicals have been found in sorghum seed, peptides related with hypoglycemia never been studied before. To obtain the peptide characteristics and the specifically high-expressed peptides in hypoglycemic sorghum seed, peptide profiles of seven hypoglycemic and five non-hypoglycemic sorghum lines bred in RDA were determined using surface-enhanced laser desorption/ionization time-of-flight mass spectrometry (SELDI-TOF MS). The twelve sorghum lines exhibited 104 peptides on CM10 protein chip array (weak cation exchange) and 95 peptides on Q10 (weak cation exchange) in the molecular mass range from 2,000 to 20,000 Da. Heat map via supervised hierarchical clustering of the significantly different peptides (p < 0.01) in peak intensity among the 12 lines effectively revealed the specifically upregulated peptides in each line and distinguished between 7 hypoglycemic and 5 non-hypoglycemic lines. Through the comparison with hypoglycemic and non-hypoglycemic lines, 10 peptides including 2231.6, 2845.4, 2907.9, 3063.5, 3132.6, 3520.8, 4078.8, 5066.2, 5296.5, 5375.5 Da were specifically high-expressed in hypoglycemic lines at p < 0.00001. This study characterized seed peptides of 12 sorghums and found ten peptides highly expressed for hypoglycemic sorghum lines, which could be used as peptide biomarkers for identification of hypoglycemic sorghum.

Phylogenetic characterization of bacterial populations in different layers of oak forest soil (상수리나무림의 토양 층위별 세균군집의 계통학적 특성)

  • We have examined the correlation between the physicochemical and microbiological environment variables for the different layers of oak forest soil in Mt. Gyeryong, Korea. The result shows that there is a high correlation in the environment variables between the soil parameters of the fermented (F) layer and humus (H) layer. In particular, the pH level in the F layer shows a high correlation with C and N, while the various organic acids of the H layer turns out to be closely correlated with soil bacteria density. As we evaluated phylogenetic characteristics of bacterial populations by DGGE analysis with DNA extracted. Total of 175 bands including 43 bands from litter (L) layer, 42 bands from F layer, 43 bands from H layer and 47 bands from rhizosphere (A) layer were selected as the major DGGE band of oak forest soil. Based on the 16S rRNA gene sequences, 175 DGGE bands were classified into 32 orders in 7 phylum. The heat map was analyzed in order to compare the quantity of the base sequences of each order and based on the clustering of the different layers of oak forest soil, the result confirms that the F layer and H layer belong to a different cluster from that of L layer and A layer. Furthermore, it also showed that approximately 50% of the total microbial population in different layers is ${\alpha}$-proteobacteria, which indicates that they belong to the dominant system group. In particular, Rhizobiales, Burkholderiales and Actinobacteriales were observed in all the seasons and layers of oak forest soil, which confirms that they are the indigenous soil bacterial community in oak forest soil.

A Study on Mapping Forest Fire Risk Using Combustion Characteristic of Forest Fuels : Focusing on Samcheok in Gangwon-do (산불연료의 연소특성을 활용한 산불위험지도 작성에 관한 연구 : 강원도 삼척 시를 중심으로)

  • In order to predict about forest fire behavior we constructed a database for combustion characteristic of forest fuels in Samcheok, Gangwon-do and prepared fire risk map and fire risk rating using GIS method in this study. For the mapping autoignition temperature, ignition time, flame duration time, total heat release and total smoke release are selected as the standardized parameters and the overall risk rating was made up of the ignition risk parameters(autoignition temperature, ignition time) and the spread risk parameters(flame duration time, total heat release, total smoke release). Forest fire risk was classified into 5 grades and lower grade of fire risk rating mean to correspond to more dangerous forest fire. As a result, the overall risk rating of Samcheok was classified into three grades from 1 to 3 and Nogok-myeon and Miro-myeon were turned out the most dangerous areas for forest fire. Because of the colony of pine and oak trees and the higher fire loads, the flame propagation will be carried out quickly in these areas.

Fish Distribution Research Using Fishfinder at Fishery Area in the Cheongpyeong Reservoir (어군탐지기를 활용한 청평호 어업 구간의 어류 분포 연구)

  • This study was conducted on October 23, 2020 at the Cheongpyeong Reservoir located in Seorakmyeon, Gapyeong-gun, Gyeonggi-do, and analyzed the horizontal and vertical distribution patterns of fish based on data obtained using fishfinder. The total surface area of fishfinder survey conducted was 782,853 m2, and where the water depth (WD) ranges from 10 m to 12 m is widest which 31.7% of total surface area. As a result of the heat map analysis, fish density was highest at right bank under the Gapyeong-bridge, but there was no specific pattern in horizontal distribution of fish. As a result of vertical distribution of fish analysis, 86.6% of fishes are observed at below 6 m of the fish depth (FD, distance from water surface to fish). As a result of the relative height (RH, water depth-distance from bottom to fish ratio) analysis, there was a tendency that fishes are distributed in near surface area more as the WD increased. This tendency could have various reasons such as water temperature gradient along the water depth, and further studies are required for detailed explanation.

  • This study investigated through eye tracking what gaze path the audience searches through portal sites (Naver, Daum, Zoom, and Nate). As a result of the layout analysis according to the gaze path of the search engine, the four main pages, which can be called to be the gateway to information search, appeared in the form of a Z-shaped layout. The news and search pages of each site use an F-shape, which means that when people's eyes move from top to right in an F-shape, they read while moving their eyes from left to right(LTR), which sequentially moves to the bottom. As a result of analyzing through the heat map, gaze plot, and cluster, which are the visual analysis indicators of eye tracking, the concentration of eyes on the photo and head copy was found the most in the heat map, and it can be said to be of high interest in the information. The flow of gaze flows downward from the top left to the right, and it can be seen that the cluster is most concentrated at the top of the portal site. The website designer should focus on improving the accessibility and readability of the information desired by the user in the layout design, and periodic interface changes are required by investigating and analyzing the tendencies and behavioral patterns of the main users.

A Study for Estimation of High Resolution Temperature Using Satellite Imagery and Machine Learning Models during Heat Waves (위성영상과 머신러닝 모델을 이용한 폭염기간 고해상도 기온 추정 연구)

  • This study investigates the feasibility of three algorithms, K-Nearest Neighbors (K-NN), Random Forest (RF) and Neural Network (NN), for estimating the air temperature of an unobserved area where the weather station is not installed. The satellite image were obtained from Landsat-8 and MODIS Aqua/Terra acquired in 2019, and the meteorological ground weather data were from AWS/ASOS data of Korea Meteorological Administration and Korea Forest Service. In addition, in order to improve the estimation accuracy, a digital surface model, solar radiation, aspect and slope were used. The accuracy assessment of machine learning methods was performed by calculating the statistics of R2 (determination coefficient) and Root Mean Square Error (RMSE) through 10-fold cross-validation and the estimated values were compared for each target area. As a result, the neural network algorithm showed the most stable result among the three algorithms with R2 = 0.805 and RMSE = 0.508. The neural network algorithm was applied to each data set on Landsat imagery scene. It was possible to generate an mean air temperature map from June to September 2019 and confirmed that detailed air temperature information could be estimated. The result is expected to be utilized for national disaster safety management such as heat wave response policies and heat island mitigation research.

Estimation of Potential Evapotranspiration using LAI (LAI를 고려한 잠재증발산량 추정)

  • In the process of a hydrology circulation, evapotranspiration is considered a very important factor to build a plan for the development of water resources and to operate water resources system. This study purposes to estimate daily potential evapotranspiration quantity in consideration of energy factors of the surface by using spatial information such as Landsat TM (ETM+) data, DEM and Landcover. Kyounan-cheon, Han River is selected as a target area, and landcover is divided by vegetation and non-vegetation covered area. Penman-Monteith equation which considers leaf-area index is used to estimate potential evapotranspiration quantity of vegetation covered area. The combination method (energy burget and aerodynamic method) is used in non-vegetation covered area. Among the input data for estimating potential evapotranspiration, NDVI, SR and Albedo is formed by Landsat, TM and ETM+ from 1986 through 2002. ground heat flux is estimated by using NDVI distribution map, LAI distribution map is drawn by using SR distribution map. The result of estimation shows that the average potential evapotranspiration in the whole basin is about 1.8-3.2mm/day per each cell. THe results of estimating potential evapotranspiration quantity by each landcover are as follows; water surface 3.6-4.9mm/day, city 1.4-3.1mm/day, bareland 1.4-3.5mm/day, grassland 1.7-3.7mm/day, forest 1.7-3.0mm/day and farmland 1.8-3.6mm/day. The potential evapotranspiration quantity is underestimated in comparison with observed evaporation data by evaporation pan, but it is considered that it has physical propriety.

A Study on the Domestic Appllication of the Concept of Seed Transfer Zone in the U.S (미국 잠정종자이동구역(Seed transfer zone) 개념의 국내 적용 방안)

  • The seed zone is a map that describes the areas where plant material can be transferred with little risk for properly adapting to a new location. The seed zone study is largely divided into studies based on genetic data and studies based on climatic data. Can be. This study was conducted to establish a temporary domestic seed zone applicable to the entire Korean Peninsula and evaluate its possibility based on the US climate-based seed zone establishment methodology. The temporary seed zone was constructed in the same way as the US case by superimposing the data obtained by dividing the winter minimum temperature into 12 grades and the data obtained by dividing the annual heat: moisture index into 6 grades. As a result of the analysis, 65 temporary seed zones were formed throughout the Korean Peninsula, and the areas of the seed zones representing the smallest and largest areas were 3.0km2 and 29,423.0km2, respectively, and it was confirmed that they had an average size of about 5,064.9km2. Temporary seed zones applied in Korea show a pattern of changes in temperature according to the relatively horizontal forest zone, and it was confirmed that the area where the Baekdu-daegan ecological axis is located has a tendency to show lower dryness than other areas. This study applied the US climate-based seed zone methodology in Korea as a pilot, and confirmed the climatic similarity across the Korean Peninsula. Furthermore, it is expected to provide an optimal seed map that improves the success rate of restoration in the future by revising the seed zone grade suitable for the domestic environment in consideration of the results of this study and the possibility of seed adaptation to the field survey and environmental space.

Distracted Driver Detection and Characteristic Area Localization by Combining CAM-Based Hierarchical and Horizontal Classification Models (CAM 기반의 계층적 및 수평적 분류 모델을 결합한 운전자 부주의 검출 및 특징 영역 지역화)

  • Driver negligence accounts for the largest proportion of the causes of traffic accidents, and research to detect them is continuously being conducted. This paper proposes a method to accurately detect a distracted driver and localize the most characteristic parts of the driver. The proposed method hierarchically constructs a CNN basic model that classifies 10 classes based on CAM in order to detect driver distration and 4 subclass models for detailed classification of classes having a confusing or common feature area in this model. The classification result output from each model can be considered as a new feature indicating the degree of matching with the CNN feature maps, and the accuracy of classification is improved by horizontally combining and learning them. In addition, by combining the heat map results reflecting the classification results of the basic and detailed classification models, the characteristic areas of attention in the image are found. The proposed method obtained an accuracy of 95.14% in an experiment using the State Farm data set, which is 2.94% higher than the 92.2%, which is the highest accuracy among the results using this data set. Also, it was confirmed by the experiment that more meaningful and accurate attention areas were found than the results of the attention area found when only the basic model was used.

A study on the Effectiveness of Urban air temperature Through Citizen Participation (시민참여형 도시온도 모니터링의 실효성에 관한 연구)

  • At the point of implementing policies related to urban heat through the overall environmental assessment of the city using national data, citizen science projects that can collect data in a wide range are emerging for effective policy establishment. Although the utility of citizen data is improving, data quality is a primary concern for researchers employing public participation in scientific research. In this study, validation was conducted based on citizen data acquired in the "Suwon City Heat Map Project", and the applicability to temperature monitoring was confirmed based on the results. As a result of analyzing the validity verification of citizen data using three methods, the data result value is 0.843, RMSE: 0.683℃, and a meaningful value was found within 3km of national data. We found that citizen data utilization is high through the results of this study and These projects are expected to be used as basic data for establishing effective policies or can be reflected in the various planning.
