The Role of mRNA Quality Control in the Aging of Caenorhabditis elegans

  • The proper maintenance of mRNA quality that is regulated by diverse surveillance pathways is essential for cellular homeostasis and is highly conserved among eukaryotes. Here, we review findings regarding the role of mRNA quality control in the aging and longevity of Caenorhabditis elegans, an outstanding model for aging research. We discuss the recently discovered functions of the proper regulation of nonsense-mediated mRNA decay, ribosome-associated quality control, and mRNA splicing in the aging of C. elegans. We describe how mRNA quality control contributes to longevity conferred by various regimens, including inhibition of insulin/insulin-like growth factor 1 (IGF-1) signaling, dietary restriction, and reduced mechanistic target of rapamycin signaling. This review provides valuable information regarding the relationship between the mRNA quality control and aging in C. elegans, which may lead to insights into healthy longevity in complex organisms, including humans.

간호대학생의 자아탄력성 및 가족탄력성이 성공적 노화 인식에 미치는 영향 (Influence of Ego Resilience and Family Resilience on the Perception of Successful Aging in Nursing Students)

  • Purpose: This study aims to examine the relationship between ego resilience, family resilience, and the perception of successful aging in nursing students; additionally, specific factors relating to the perception of successful aging were identified. Methods: The participants comprised 154 nursing students. Data were collected through the administration of self-reported questionnaires. Data analysis was then performed utilizing a t-test, an ANOVA, Pearson's correlation coefficients, and a multiple regression analysis. Results: Family resilience differed significantly based on the type of residence (p=.020) (i.e., living with grandparents (p=.048)). Perception of successful aging differed significantly based on grade (p<.001). The perception of successful aging showed a positive correlation relative to mode of communication and organizational pattern. The mode of communication accounted for 4.7% of perception of successful aging. Conclusion: The significance of the study on the importance of mode of communication was confirmed by identifying the effects of the perception of successful aging. Nursing students may actively cope with crises through communication; thus, successfully recognizing aging in order to lead a healthy life. Furthermore, having a high perception of successful aging while treating an increasing number of elderly patients at the clinical practice is believed to have a positive effect on patient care.

중국 도시 노인의 노후 상황에 따른 주거 선호 변화 연구 (A Study on Changes of Residential Preferences among Older People Living in Urban China according to Aging Situation)

  • As China has the largest number of elderly people in the world, the issue of the aged people has been a serious matter of concern in Chinese government and society and many studies have been done so far. However, the studies about the dwellings of the aged people that support their basic life are still insufficient. For this reason this study intends to identify the differences in elderly people's preferences according to their situations in aged life. This study surveyed 152 elderly residents aged 50 and over including early aged people in Beijing China about their residential preferences (types of residence, location of residence, size of residence, types of ownership) according to the four types of their situations in aged life, healthy couple, sick couple, healthy single and sick single. The survey conducted from 2013 April 10th to 15th. The results were as follows: Firstly, as the elderly situation became worse, preference for the independent residence decreased very rapidly while the preference for the elderly welfare facilities dramatically increased from the 0 point of the healthy couple and the preference for the elderly welfare facilities marked the highest point in case of the worst situation of sick single. Secondly, among the four different situations preferences for the urban residence were the highest and as the aged people's situation becomes worse, their preferences for urban life increased. Thirdly, healthy couple preferred a large size residence but as their elderly situation becomes worse, their preferences for the large size decreased and their preferences for small size increased dramatically, and in case of elderly singles, preferences for small size residence exceeded their preferences for large size residence. Fourthly, as elderly situation became worse preference for owning residence decreased and preference for rental residence increased and in case of the worst situation of sick single, preference for rental residence was higher than their preferences for their own residence. With this view of point, the residential preference of elderly people changes along with their aging situations. Thus, the government should consider the variable aging market, and develop a variety of aging residence which, can satisfy the different groups of elderly.

전라도 농촌장수지역 거주 70대 노인의 건강상태에 영향을 미치는 환경적 요인에 대한 탐색 연구 (Investigation on Influencing Environmental Factors on Health Status of Korean Septuagenarians Dwelling in Longevity Region in Jeonla Province)

  • Objectives: To evaluate the critical environmental factors on healthy-aging of Korean people, we investigated the significant factors influencing health status of septuagenarians living in rural area of Jeonla province, known to be one of the representative longevity regions in Korea. Methods: We divided subjects into healthy group (36M/25F) or poor-health group (26M/73F) based on self-reported health status, body mass index, a number of prescription, and blood test data. General characteristics, physical measurements, lifestyle, dietary behavior and nutrient intake, physical health and mental health data were statistically compared between the two groups. Results: Average age was not different between healthy group and poor-health group in men and women, respectively. In men, significantly favorable factors to health were observed to be higher education, regular exercise, higher grip strength and walking function, body mass index (${\geq}18.5kg/m^2$), moderate frequency of drinking and eating-out, non-smoking, normal red blood cell (RBC) count, higher serum dehydroepiandrosterone-sulfate (DHEAS) level, good digestive function and appetite, normal hearing function, regular meals, adequate vegetable and fruit intake, diverse food intake, adequate energy and nutrients (protein, vitamin $B_1$, $B_6$, C and E, folate, niacin, P, Zn and K) intake, higher mini-nutrient status assessment (MNA) score and low level of depression. On the other hand, in women, those were literacy, living arrangement, moderate frequency of drinking, healthy teeth, higher grip strength and walking function, bone mineral density, normal RBC and white blood cell (WBC) count, higher DHEAS concentration, higher MNA score, normal cognition and memory function, having snack and adequate fruit intake. Conclusions: These results could be useful to plan effective strategies to increase health-life expectancy of Korean old people living in rural areas.

지역사회 노인의 생활만족도, 건강행위와 성공적 노후간의 관계 (The Relationship of Life Satisfaction, Health Behaviors and Successful Aging in the Elderly)

  • Purpose: This study was conducted in order to the relationship between life satisfaction, health behaviors, and successful aging and factors associated with successful aging. Methods: Participants in this study included 159 elders living in one city. Data on life satisfaction (Choi's Life Satisfaction Scale for Korean elderly, 1984), health behaviors (Choi & Kim's Health Behaviors Scale for Korean elderly, 1997), and successful aging (Kim & Shin's Successful Aging Scale for Korean elderly, 2005) were collected by trained interviewers. For analysis of collected data, the PASW 18.0 program was used, which included descriptive analysis, t-test, ANOVA, Scheffe test, Pearson correlation coefficients, and Multiple regression. Results: Results showed a positive correlation between life satisfaction, health behaviors, and successful aging. Life satisfaction, self-perceived health status, and health behavior explained 60.8% of successful aging. Conclusion: There is a need to help improve health behavior and self-perceived health status of elders. We should also understand life satisfaction over their life span and establish a program that encourages healthy behavior in the community. Accordingly, these efforts will allow for achievement of a more successful aging process for the elderly.

Epigenetics: Linking Nutrition to Molecular Mechanisms in Aging

  • Healthy aging has become a major goal of public health. Many studies have provided evidence and theories to explain molecular mechanisms of the aging process. Recent studies suggest that epigenetic mechanisms are responsible for life span and the progression of aging. Epigenetics is a fascinating field of molecular biology, which studies heritable modifications of DNA and histones that regulate gene expression without altering the DNA sequence. DNA methylation is a major epigenetic mark that shows progressive changes during aging. Recent studies have investigated aging-related DNA methylation as a biomarker that predicts cellular age. Interestingly, growing evidence proposes that nutrients play a crucial role in the regulation of epigenetic modifiers. Because various nutrients and their metabolites function as substrates or cofactors for epigenetic modifiers, nutrition can modulate or reverse epigenetic marks in the genome as well as expression patterns. Here, we will review the results on aging-associated epigenetic modifications and the possible mechanisms by which nutrition, including nutrient availability and bioactive compounds, regulate epigenetic changes and affect aging physiology.

Expanding the aging self: Investigating successful aging among Korean older adults using grounded theory

  • Purpose: This qualitative study aimed to understand older adults' perspectives on successful aging and develop a model of a successful aging process, within the Korean socio-cultural context. Methods: This study used a Grounded Theory approach. Through theoretical sampling, 14 participants were selected from older adults at a public health center and a volunteer institution in Gyeonggi Province, South Korea. Results: The basic social process identified was "expanding the aging self", which was preceded by three phases, namely, adjusting to the changes, developing the valuable self, and embracing the environment. Participants used 2-3 strategies for successful aging in each phase. For adjusting to the changes, participants used strategies such as "adopting a positive attitude", "accepting the changes", and "being at the center of life". To develop the valuable self, "re-identifying capacities", "having something to do", and "advancing to a healthy lifestyle" were used. Participants embraced their environments with "sharing with others" and "embracing younger generations". The causal condition for expanding the aging self was the changes in participants' physical, mental, or psychosocial situations. Conclusion: These findings suggest a theoretical foundation for the development of potential nursing interventions to promote self-care management and the interpersonal relationship for successful aging among Korean elderly individuals.

재난대응조직 연령구성비 변화가 조직효과성 및 서비스질 보장활동에 미치는 영향에 관한 실증분석 - 서울지역 현장활동 소방공무원을 중심으로 - (An empirical study of the impact which the change of age ratio of disaster response organization has on organization effectiveness and quality assurance - Focused on field activities firefighters in Seoul metropolitan city -)

  • Aging of the fire organizations, typical disaster response agencies also will be in serious problems with aging society. It is expected that the ratio of more than fifties will reach 31~40% after 10~15 years in Seoul fire organization. This Aging of firefighters can bring about a serious problem in disaster and safety management, since firefighters require robust fitness and healthy bodies in disaster situations. In this study, after the analysis of aging status and trend of firefighters in Seoul, I have an empirical analysis about the impact this aging has on organization effectiveness and QA. As a result, the increase of the elderly reduced organization effectiveness and QA. In addition, in the group consisting of less than 50, it is expected to increase the organization's effectiveness and QA activities through field activity to improve the professional skills with age, but could not be confirmed. In the group consisting of more than fifties, it was found that aging had a significant negative impact on organizational effectiveness and QA activities as factors, including physical deterioration by aging.

Multi-level remodeling of transcriptional landscapes in aging and longevity

  • In multi-cellular organisms, the control of gene expression is key not only for development, but also for adult cellular homeostasis, and gene expression has been observed to be deregulated with aging. In this review, we discuss the current knowledge on the transcriptional alterations that have been described to occur with age in metazoans. First, we discuss age-related transcriptional changes in protein-coding genes, the expected functional impact of such changes, and how known pro-longevity interventions impact these changes. Second, we discuss the changes and impact of emerging aspects of transcription in aging, including age-related changes in splicing, lncRNAs and circRNAs. Third, we discuss the changes and potential impact of transcription of transposable elements with aging. Fourth, we highlight small ncRNAs and their potential impact on the regulation of aging phenotypes. Understanding the aging transcriptome will be key to identify important regulatory targets, and ultimately slow-down or reverse aging and extend healthy lifespan in humans.
