Soybean transgenic plants with ectopically expressed AtABF3 were produced by Agrobacterium-mediated transformation and investigated the effects of AtABF3 expression on drought and salt tolerance. Stable Agrobacterium-mediated soybean transformation was carried based on the half-seed method (Paz et al. 2006). The integration of the transgene was confirmed from the genomic DNA of transformed soybean plants using PCR and the copy number of transgene was determined by Southern blotting using leaf samples from $T_2$ seedlings. In addition to genomic integration, the expression of the transgenes was analyzed by RT-PCR and most of the transgenic lines expressed the transgenes introduced. The chosen two transgenic lines (line #2 and #9) for further experiment showed the substantial drought stress tolerance by surviving even at the end of the 20-day of drought treatment. And the positive relationship between the levels of AtABF3 gene expression and drought-tolerance was confirmed by qRT-PCR and drought tolerance test. The stronger drought tolerance of transgenic lines seemed to be resulted from physiological changes. Transgenic lines #2 and #9 showed ion leakage at a significantly lower level (P < 0.01) than ${\underline{n}}on-{\underline{t}}ransgenic$ (NT) control. In addition, the chlorophyll contents of the leaves of transgenic lines were significantly higher (P < 0.01). The results indicated that their enhanced drought tolerance was due to the prevention of cell membrane damage and maintenance of chlorophyll content. Water loss by transpiration also slowly proceeded in transgenic plants. In microscopic observation, higher stomata closure was confirmed in transgenic lines. Especially, line #9 had 56% of completely closed stomata whereas only 16% were completely open. In subsequent salt tolerance test, the apparently enhanced salt tolerance of transgenic lines was measured in ion leakage rate and chlorophyll contents. Finally, the agronomic characteristics of ectopically expressed AtABF3 transgenic plants ($T_2$) compared to NT plants under regular watering (every 4 days) or low rate of watering condition (every 10 days) was investigated. When watered regularly, the plant height of drought-tolerant line (#9) was shorter than NT plants. However, under the drought condition, total seed weight of line #9 was significantly higher than in NT plants (P < 0.01). Moreover, the pods of NT plants showed severe withering, and most of the pods failed to set normal seeds. All the evidences in the study clearly suggested that overexpression of the AtABF3 gene conferred drought and salt tolerance in major crop soybean, especially under the growth condition of low watering.
To investigate the research state and characteristics of dinoflagellate cyst along the Korean coastal waters, this work analyzed 45 studies conducted in the Korean coasts and basin, the south of Jeju, East China Sea including some part of the Yellow Sea. It was found that the research on dinoflagellate cyst began in the later half of the 1980s in Korea, and that the research areas focused on eutrophied regions where a red tide occurred or on the seas where coastal development occurred. In other words, no research on the East Sea was found. In terms of research contents, there were various studies on analysis of cyst assemblage, tracking of the changes in marine environment like process of eutrophication, roles as a seed population in occurrence of a red tide, creation of cyst by use of sediment trip and tracking of its changes, morphological changes by the change of pH concentration in the hypoxia zone of eutrophied region, germination of a specific species, and other international-level studies. Species composition and cell density also varied in the Korean coastal waters. However, much research on dinoflagellate cyst in Korea is not systematic, and is fragmentary and individual and hencesuffers from great limitations. The research results so far have focused only on specific regions, and sporadic research had been conducted by specific research groups. Dinoflagellate cyst of surface sediments represents the accumulation index of pelagic environment in the sea and is a useful index to understand the marine environment efficiently. In conclusion, Korea requires organized human resources and collaborative research on dinoflagellate cyst which in turn should be considered as a component of marine ecosystem and as an essential aspect of marine biology.

There are various method for evaluating the durability life of concrete structures due to salt damage. The best way is to perform a corrosion test for a rebar embedded in concrete specimen was exposure to marine environment. However, this method has the disadvantage that it takes a long period of time. Also, accelerated corrosion test which was complemented complements the time-consuming weakness is limited to apply because it could not reveal a correlation between long-term exposure test. Accordingly, the purpose of this study is to derive a correlation coefficient between cycle drying-wetting accelerated corrosion test and long-term exposure test. Corrosion initiation time was measured in four types of concrete samples, i.e., two samples mixed with fly ash(FA) and blast furnace slag(BS), and the other two samples having two water/cement ratio(W/C = 0.6, 0.35) without admixture(OPC 60 and OPC 35). The accelerated corrosion test was carried out by two case, i.e., one is a cyclic drying-wetting method(case 1), and the other is a artificial seawater ponding test method(case 2). Whether corrosion occurs, it was measures using half-cell potential method. The results indicated that case 1 is to accelerated the corrosion of rebar about 24~36% as compared with case 2, then the corrosion of rebar embedded in concrete occurred according to the order of OPC60, FA, BS, OPC35. Correlation coefficient between accelerated corrosion test and long-term exposure test, case 1 is 4.23 to 5.42, and case 2 is 6.54 to 7.82.
Purpose: Venous stasis ulcer is the most severe form of chronic venous insufficiency and this commonly appears in the lower limb. Pharmacological therapy, reconstruction of the venous system, surgical management, cellular therapy and compression therapy are known as the treatments of venous stasis ulcer, but relapses are common, which make it a typical chronic wound. We report here on a case of recurrent venous stasis ulcer that healed with compression therapy without any other treatment. Methods: A 35-year-old man with a 13 years history of venous stasis had developed an ulcer on the distal third portion of the lower left limb which was developed 12-year before enrollment in this study. He had been treated with vacuum assist closure, 2 times of cell therapy and 3 times of skin graft for 8 years, but the lesion recurred. From November, 2008 compression therapy was done with the 3M $Coban^{TM}$ 2 Layer Compression System (3M, St. Paul, USA). The ulcer at that time was oval shaped and $3{\times}4$ cm in size. A comfort layer bandage was applied from the proximal phalanx of the great toe to the knee. A compression layer bandage was applied on the previous layer with it being overlapped one half the width of the comfort layer bandage. The dressing was changed every 4 days and the change was recorded with photography. Results: A total of 12 $Coban^{TM}$ 2 Layer Compression Systems were used. The size of the ulcer decreased to $2.5{\times}2.5$ cm in one month, to $2{\times}2$ cm in 2 months, it was $1{\times}1.8$ cm in size at 3 months and it completely healed in 4 months. Conclusion: The venous stasis ulcer was completely healed using the 3M $Coban^{TM}$ 2 Layer Compression System. This method was easy to apply, made the patient comfortable and it provided an excellent compression effect. As in the previous studies, this compression therapy has been proven to play an important role for the treatment and prevention of venous stasis ulcer.
The experiments were carried out to investigate the biocontrol potential of Lysobacter capsici YS1215 on root-knot nematode and to characterize its lytic enzyme activities. L. capsici YS1215 showed chitinase and gelatinase activities on the medium containing 0.5% chitin or 0.5% gelatin as substrates. Cell growth of L. capsici YS1215 was highest at 6 days, and the highest activities of chitinase (4.0 unit $ml^{-1}$) and gelatinase (7.43 unit $ml^{-1}$) were observed on 3 and 5 days after incubation, respectively. To investigate the effect of L. capsici YS1215 on tomato growth and nematode infection, the plants in pot trial were treated with bacterial culture (BC), half of bacterial culture (HBC), only bacterial medium (BM), tap water (TW) and commercial nematicide (CN). HBC treatd plants showed the higher shoot fresh weight and dry weight on $5^{th}$week after incubation while BM, HBC and BC had consistently higher values than TW at $9^{th}$ week. HBC appeared to be the highest shoot fresh length at $9^{th}$ week. Both CN and BC showed lower number of egg mass, root gall, and population of juveniles in soil compared to BC, HBC, BM and TW. These results suggest that L. capsici YS1215 with its strong ability of lytic enzyme production can be one of the most significant candidates for biocontrol agents against root-knot nematodes.
Purpose: Use of radiotherapy combined with chemotherapy is increasing in hypopharyngeal cancer. However, many show residual tumor after radiotherapy. Timing for treatment evaluation and salvage therapy is essential. However, optimal timing for salvage surgery has not been suggested. In this study, we tried to evaluate optimal timing for salvage surgery. Methods and Materials: Patients who were diagnosed with hypopharyngeal squamous cell carcinoma between 2006 and 2015 were retrospectively analyzed. All patients received definitive radiotherapy with or without chemotherapy. Response of all treated patients were analyzed at 1, 3, and 6 months after radiotherapy. Any patients with progression before 6 months were excluded. Results: A total of 54 patients were analyzed. Complete remission (CR) rates at 1 month (CR1), 3 months (CR3) and 6 months (CR6) were 66.7%, 81.5%, and 90.7%, respectively. Non-CR at 1 month (NCR1), 3 months (NCR3), and 6 months (NCR6) showed poor locoregional recurrence-free survival rates (1-year rates of 63.7%, 66.7%, and 0.0%, respectively) compared to CR1, CR3, and CR6 (1-year rates 94.3%, 88.0%, and 91.5%, respectively). Particularly significant differences were seen between CR6 and NCR6 (p < 0.001). Of 10 patients with NCR3, 5 showed CR at 6 months (NCR3/CR6). There was no statistical difference in locoregional recurrence-free survival between CR3 and NCR3/CR6 group (p = 0.990). Conclusion: Our data suggest half of patients who did not show CR at 3 months eventually achieved CR at 6 months. Waiting until 6 months after radiotherapy may be appropriate for avoiding additional salvage therapy.

Objective : The purpose of the present study was to compare inter-fragmentary compression pressures after fixation of a simulated type II odontoid fracture with the headless compression Herbert screw and a half threaded cannulated lag screw. Methods : We compared inter-fragmentary compression pressures between 40- and 45-mm long 4.5-mm Herbert screws (n=8 and n=9, respectively) and 40- and 45-mm long 4.0-mm cannulated lag screws (n=7 and n=10, respectively) after insertion into rigid polyurethane foam test blocks (Sawbones, Vashon, WA, USA). A washer load cell was placed between the two segments of test blocks to measure the compression force. Because the total length of each foam block was 42 mm, the 40-mm screws were embedded in the cancellous foam, while the 45-mm screws penetrated the denser cortical foam at the bottom. This enabled us to compare inter-fragmentary compression pressures as they are affected by the penetration of the apical dens tip by the screws. Results : The mean compression pressures of the 40- and 45-mm long cannulated lag screws were $50.48{\pm}1.20N$ and $53.88{\pm}1.02N$, respectively, which was not statistically significant (p=0.0551). The mean compression pressures of the 40-mm long Herbert screw was $52.82{\pm}2.17N$, and was not statistically significant compared with the 40-mm long cannulated lag screw (p=0.3679). However, 45-mm Herbert screw had significantly higher mean compression pressure ($60.68{\pm}2.03N$) than both the 45-mm cannulated lag screw and the 40-mm Herbert screw (p=0.0049 and p=0.0246, respectively). Conclusion : Our results showed that inter-fragmentary compression pressures of the Herbert screw were significantly increased when the screw tip penetrated the opposite dens cortical foam. This can support the generally recommended surgical technique that, in order to facilitate maximal reduction of the fracture gap using anterior odontoid screws, it is essential to penetrate the apical dens tip with the screw.

Rice is the most important cereal crop not only in supplying the basic staple food for more than half of the world's population but also as a model plant for functional genomic studies of monocotyledons. Although rice transformation method using A. tumefaciens has already been widely used to generate transgenic plants, the transformation rate is still low in most Korean elite cultivars. We made several modifications of the standard protocol especially in the co-cultivation step to improve the efficiency of the rice transformation. The co-culture medium was modified by the addition of three antioxidant compounds (10.5 mg/L L-cysteine, 1 mM sodium thiosulfate, 1 mM dithiothreitol) and of Agrobacterium growth-inhibiting agent (5 mg/L silver nitrate). Co-cultivation temperature ($23.5^{\circ}C$ for 1 day, $26.5^{\circ}C$ for 6 days) and duration (7 days) were also changed. The plasmid of pMJC-GB-GUS carrying the GUS reporter gene and the bar gene as the selectable marker was used to evaluate the efficiency of the transformation. After co-cultivation, a high level of GUS gene expression was observed in calli treated with the modified method. It is likely that those newly added compounds helped to minimize the damage due to oxidative bursts during plant cell-Agrobacterium interaction and to prevent necrosis of rice cells. And the transformation rate under the modified method was also remarkably increased approximately 8-fold in Heungnambyeo and 2-fold in Ilmibyeo as compared to the corresponding standard method. Furthermore, we could produce the transgenic plants stably from Ilpumbyeo which is a high-quality rice but its transformation rate is extremely low. Transformation and the copy number of transgenes were confirmed by PCR, bar strip and Southern blot analysis. The improved method would attribute reducing the effort and the time required to produce a large number of transgenic rice plants.
Dengue virus is a typical mosquito-borne virus, and the half of the world's population is exposed to infection. Dengue virus causes relatively mild symptoms such as dengue fever. However, when not treated properly, it is known to cause severe symptoms such as dengue hemorrhagic fever and dengue shock syndrome with a mortality rate of over 20%. Development of dengue virus detection technology is very important because it is reported that early diagnosis of dengue fever can lower the mortality rate to less than 1%. In this study, patent search related to dengue virus detection technology was conducted in Korea, USA, Europe, Japan, and China. The quantitative analysis of 69 validated patents from the searched patents was conducted by country, year, and patent holder. In addition, in-depth analysis was carried out by classifying into three categories: molecular diagnostics, immuno-diagnostics, and cell culture-based diagnostics from all validated patents. From these results, we analyzed the patent trend related to dengue virus detection and dengue fever diagnosis technology and discussed the features and limitations of molecular diagnostics and immuno-diagnostics at present level. Furthermore, we discussed the direction of technology development and future prospects to overcome limitations.
This study investigated the effect of the addition of pomegranate concentrate to yogurt on the growth of pathogenic and lactic acid bacteria. The concentration of the MRS broth was adjusted to one-half and used for an experiment. Pomegranate concentrate was added at concentrations of 4%, 2%, 1%, and 0.5%, which significantly promoted the growth of Lacto-coccus cremoris, Weissella cibaria, Weissella paramesenteroides, Lactobacillus plantarum, Lactobacillus acidophilus, Streptococcus thermophillus, Lactobacillus bulgaricus, and Lactobacillus lactis. The growth of lactic acid bacteria increased with higher concentrations of pomegranate. However, the addition of pomegranate concentrate inhibited the growth of Escherichia coli KCCM11587, E. coli KCCM11591, E. coli KCCM11596, and E. coliKCCM11600. Yogurt with added pomegranate concentrate demonstrated optimal conditions compared to that of the control without the addition. Particularly, the viable cell count of lactic acid bacteria was significantly higher in the yogurt with pomegranate concentrate. Furthermore, the viability of the lactic acid bacteria in the yogurt with pomegranate concentrate was higher than that of the control without the addition of concentrate during storage.
