Studies on the Calcium Phosphopeptide in Milk Casein (우유 Casein 중의 Calcium Phosphopeptide에 관한 연구)

  • The economical producing method of casein phosphopeptide (CPP) and the physicochemical properties related to the solubilization of calcium were studied. Firstly, The compositions of the purified CPP-III were 13.1% of nitrogen, 2.3∼2.4% of phosphate and the ratio of N/P was 5.4∼5.6. In consideration of economic aspects, the preparation method of the CPP- I and II which were lower purity than the CPP-III was established. The physico-chemical property of the CPP was compared with the enzymically dephosphorylated CPP. CPP and polyglutamate effectively inhibited the formation of insoluble calcium phosphates at physiological pH.

Formation and Characterization of Casein Phosphopeptide/Chitosan Oligosaccharide NanoComplex (케이신 포스포펩티드/키토올리고당 나노 복합체 형성과 특성 연구)

  • The objectives of this study were to manufacture casein phosphopeptide (CPP)/chitosan oligosaccharide (CSO) nanocomplexes and to investigate the impacts of manufacturing variables, such as CPP concentration and pH, on their morphological and physicochemical characteristics. Transmission electron microscopy (TEM) and particle size analysis were used to assess the morphological and physicochemical properties of the CPP/CSO nano-complexes, respectively. Based on the images obtained by TEM, the spherical shapes of the CPP/CSO nanocomplexes ranged from 50 to 150 nm. As the concentration of CPP was increased and the pH was decreased, the average particle size of the nanocomplexes significantly (p<0.05) increased. The CPP/CSO nanocomplexes had a highly uniform distribution with a polydispersity index value of less than 0.3. In addition, they had a negative surface charge with a zeta-potential value between -17 and -26 mV. The CPP/CSO nanocomplexes showed good stability during the freeze-drying process. In conclusion, CPP/CSO nanocomplexes were successfully manufactured, and the CPP concentration and pH were found to be key factors that affected their morphological and physicochemical properties.

Application of Casein Phosphopeptide/Chitosan Oligosaccharide Nanocomplex to Dairy Foods (케이신 포스포펩티드/키토올리고당 나노 복합체의 유식품 적용 연구)

  • The aim of this research was to investigate the potential application of casein phospho-peptide (CPP)/chitosan oligosaccharide (CSO) nanocomplexes to dairy foods. The physical stability of CPP/CSO nanocomplexes during storage in model dairy foods including milk and yogurt was assessed by measuring the size and polydispersity index of the nanocomplexes. Encapsulation efficiency and in vitro vitamin D release from CPP/CSO nanocomplexes during gastrointestinal digestion were determined using HPLC. CPP/CSO nanocomplexes with increased CPP concentrations and decreased pH displayed significantly increased average particle size. During storage in model dairy foods, CPP/CSO nanocomplexes prepared with lower CPP concentrations and raised pH exhibited excellent physical stability. Vitamin D encapsulation efficiency increased significantly (p<0.05) as CPP concentration and/or pH decreased. Less than 3% vitamin D were released under gastric digestion conditions in vitro, while 91% of encapsulated vitamin D was released by 2 h of incubation under intestinal conditions, indicating that CPP/CSO nanocomplexes could effectively protect vitamin D from gastric conditions for delivery to the intestines. In conclusion, CPP/CSO nanocomplexes can be applied to dairy foods as an effective vitamin D delivery system.

  • This study compared tooth's remineralization using enamel surface artificially demineralized with 0.1M lactate and HCL solution using Vicker's Hardness Number(VHN) to compare CPP-ACP and remineralization of nano-sized Carbonate Apatite's initial caries. Using pH circulation models divided into 0% nano-CA, 5% nano-CA, 10% nano-CA, 10% CPP-ACP and D.W. they were treated for 5 minutes, three times a day for 14 days to get the following results. 1. There were no significant differences among the initial surface hardness of samples demineralized surface of front tooth in 5 groups. and all 5 groups' surface hardness reduced significantly after demineralization of enamel. 2. When inquiring into hardness changes through pH circulation model, the highest hardness change was in 5% nano-CA group. Also. 10% nano-CA and 10% CPP-ACP groups increased significantly. but there was no significant difference statistically. In generalizing the above experiment results, nano-sized Carbonate Apatite showed remineralization, and compared to 10% CPP-ACP group, 5% nano-CA had remineralization to artificial caries. thus implies that when we develop method to contact with tooth of nano-CA in the future, it is expected to gain synergy effect on function of saliva, a natural remineralization material.

  • Sasang constitution experts utilize voice characteristics as an auxiliary measure for deciding a person's constitutional group. This study aims at establishing a relationship between speech features and the constitutional groups by subjective listening evaluation of voice characteristics. A speech database of 841 speakers whose constitutional groups have been already diagnosed by Sasang constitution experts was constructed. Speech features related to speech source and vocal tract filter were extracted from five vowels and one sentence. Statistically significant speech features for classifying the groups were analyzed using SPSS. The features contributed to constitution classification were speaking rate, Energy, A1, A2, A3, H1, H2, H4, CPP for males in their 20s, F0_mean, CPP, SPI, HNR, Shimmer, Energy, A1, A2, A3, H1, H2, H4 for females in their 20s, Energy, A1, A2, A3, H1, H2, H4, CPP for male in the 60s, and Jitter, HNR, CPP, SPI for females in their 60s. Experimental results show that speech technology is useful in classifying constitutional groups.

  • For the production of Casein Phosphopeptide(CPP) inhibiting the insolubility of calcium, 10% sodium caseinate was treated with 1.5% of protease from Streptococcus sp.. Optimal conditions and productivity for the CPP production, and properties of the CPP were compared with tryptic hydrolysates of sodium caseinate. Optimum conditions of pH, temperature and reaction time were 8.0, 50C, 4 hrs, respectively. Under these conditions the productivity of CPP was 23% and Molecular weight of CPP was ranged from 3, 000 to 17, 000. The results also showed that the insolubility of calcium was completely inhibited by using 1.5 times of CPP for the amount of calcium.

  • Optimum conditions for production of casein phosphopeptides (CPP) from sodium casenate by immobilized cell culture of Streptococcus faecalis var. liquefaciens were investigated. Immobilized cells were made by mixing 60% sodium alginate solution with an equal volume of culture broth at the end of exponential phase and subsequently dropping the mixture into $CaCl_{2}$ solution. Optimum conditions for CPP production by the immobilized cells were the same as those ($50^{\circ}C$, pH 7.0, and 10% substrate concentration) by the crude enzyme solution from the supernatant of culture broth. Optimum loading volume of the immobilized cells into a batch reactor was 30% (w/v). Using a continuous reactor loaded by the immobilized cells under the identified optimal conditions, we were able to produce CPP continuously up to 30 days with a maximum CPP conversion efficiency of 20%.

  • The purpose of this study was to analyze spectral versus cepstral measurements in esophageal speakers. The comparison between the measurements in thirteen male esophageal speakers was compared with the control group of thirteen normal speakers using the sustained vowel /a/. The main results can be summarized as below: (a) the CPP and L/H ratio of the esophageal group were significantly lower than those of the control group (b) the CPP was significantly correlated with the spectral parameters such as jitter, shimmer, NHR and VTI, and (c) the ROC analysis showed that the threshold of 10.25dB for the CPP achieved a good classification for esophageal speakers, with 100% perfect sensitivity and specificity. Thus, it was known that cepstral-based acoustic measures such as CPP, may be more reliable predictors than other spectral-based acoustic measures such as jitter and shimmer. And it was found that cepstral-based acoustic measures were effective in distinguishing esophageal voice quality from normal voice quality. This research will contribute to establishing a baseline related to speech characteristics in voice rehabilitation with laryngectomees.

Immunohistochemical study of CPP32 (Caspase-3) in the spinal cords of rats with experimental autoimmune encephalomyelitis (자기면역성 뇌척수염 조직에서 CPP32의 면역조직화학적 관찰)

  • The aim of this study was to evaluate the involvement of CPP32 (caspase-3), one of the death-related enzymes, in the course of experimental autoimmune encephalomyelitis (EAE). EAE was induced in Lewis rats immunized with an emulsion of rat spinal cord homogenate with complete Freunds adjuvant supplemented with Mycobacterium tuberculosis (H37Ra, 5mg/ml). The expression of CPP32 in the spinal cords of rats with EAE was studied. In normal rat spinal cords, CPP32 is constitutively, but weakly, expressed in neurons and some neuroglial cells. In the EAE spinal cords, many inflammatory cells were positive for CPP 32, and the majority of CPP32(+) cells were identified as ED1(+) macrophages. During this stage of EAE, the number of CPP32(+) cells in brain cells, including neurons and astrocytes, increased, and these cells also had increased CPP32 immunoreactivity. CPP32 immunor eactivity was not always matched with apoptosis of inflammatory cells in EAE lesions. We speculate that CPP32, which is constitutlvely expressed in brain cells, increases in response to neuroimmunological stimulation in both brain neuronal cells and inflammatory cells. The functional role of CPP32 in neuroimmunological disorders is discussed.
