To investigate protective activity in pepper plants, which were pre-inoculated with arbuscular mycorrhizal (AM) fungi Glomus intra radices (Gi), against pathogenic strain Phytophthora capsici (Pc), pathogenesis-related (PR) proteins and antioxidant enzymes were examined. The growth of root and shoot was the highest in peppers inoculated with G. intraradices, compared with non-inoculated control plants and those challenged by the pathogen with and without mycorrhizae after nine days of infection. Mycorrhizal colonization rate was reduced by about 10% in pathogen-challenged plants, but disease pressure was reduced. The activities of PR proteins, $\beta$-1- 3-glucanase and chitinase, were increased in Pc-treated plants compared to Gi+Pc-treated plants in leaves, but those in roots were suppressed. Superoxide dismutase activity and $H_2O_2${/TEX> content in Gi+Pc and Pc-treated plants were gradually increased in leaves. However, those in roots continuously increased up to 5 days, and then decreased dramatically. Peroxidase activity in leaves and roots increased after P. capsici infection both in plants inoculated with or without G. intraradices. These results suggest that AM fungi, G. intra radices, potentially act as one of the protective agents against plant pathogens. Changes of PR proteins and antioxidative enzymes in mycorrhizae-inoculated pepper appear to be regulated differently in leaves and roots by pathogen infection.
This study aimed to determine the optimum dietary crude protein level in a typical diet for fattening pigs fed ad libitum under normal climate conditions in Central Vietnam. One hundred and ninety two gilts of Mong Cai local breed (MC), $F_1$ Large White${\times}$Mong Cai and $F_2$ crossbreds of (Landrace${\times}$Mong Cai)${\times}$Large White were used. At the start of the experiment, Mong Cai pigs weighed 12 kg at 11 weeks of age, $F_1$ pigs 12.1 kg at 8 weeks of age and $F_2$ pigs 12.2 kg at 8 weeks of age. Four diets differing in crude protein (CP) content (10.1, 13.1, 16.1 and 18.9% in DM) were formulated from rice bran, corn meal, cassava meal and fish meal. Calculated digestible energy content of the diets ranged from 13.5 to 13.8 MJ per kg DM. Pigs were housed individually in pens of $2.5\;m^2$ each and had ad libitum access to feed in a trough as well as water in bowls. The final weights after a growing period of 150 days were 66, 86 and 96 kg for MC, $F_1$ and $F_2$, respectively. Feed intake of MC pigs was highest at 13.1% CP while $F_1$ and $F_2$ had the highest feed intake at 16.1% CP. The results showed that for MC the maximum gain was obtained at levels between 13 to 16% CP. For the $F_1$ the maximum gain was at dietary protein levels of 16-17%. For $F_2$ the max gain was obtained at CP levels of 16 to 18%. Feed conversion was highest in MC pigs (~4.0) followed by $F_1$ (~3.3) and $F_2$ (~3.1), and within genotypes was lowest at the optimum CP level (p<0.05). Back fat thickness in MC (33.1 mm), $F_1$ (23.0 mm) and $F_2$ (20.5 mm) pigs was different and within genotypes was the lowest at intermediate CP levels. In conclusion, increasing the dietary crude protein contents in practical diets for pigs in Vietnam can increase production on small holder farms. Optimal performance for MC, $F_1$ and $F_2$ pigs is achieved at different dietary crude protein contents.
A bacterial strain, designated B301T and isolated from raw chicken meat obtained from a local market in Korea, was characterized and identified using a polyphasic taxonomic approach. Cells were gram-negative, non-motile, obligate-aerobic coccobacilli that were catalase-positive and oxidase-negative. The optimum growth conditions were 30℃, pH 7.0, and 0% NaCl in tryptic soy broth. Colonies were round, convex, smooth, and cream-colored on tryptic soy agar. Strain B301T has a genome size of 3,102,684 bp, with 2,840 protein-coding genes and 102 RNA genes. The 16S rRNA gene analysis revealed that strain B301T belongs to the genus Acinetobacter and shares highest sequence similarity (97.12%) with A. celticus ANC 4603T and A. sichuanensis WCHAc060041T. The average nucleotide identity and digital DNA-DNA hybridization values for closely related species were below the cutoff values for species delineation (95-96% and 70%, respectively). The DNA G+C content of strain B301T was 37.0%. The major respiratory quinone was Q-9, and the cellular fatty acids were primarily summed feature 3 (C16:1 ω6c/C16:1 ω7c), C16:0, and C18:1 ω9c. The major polar lipids were phosphatidylethanolamine, diphosphatidyl-glycerol, phosphatidylglycerol, and phosphatidyl-serine. The antimicrobial resistance profile of strain B301T revealed the absence of antibiotic-resistance genes. Susceptibility to a wide range of antimicrobials, including imipenem, minocycline, ampicillin, and tetracycline, was also observed. The results of the phenotypic, chemotaxonomic, and phylogenetic analyses indicate that strain B301T represents a novel species of the genus Acinetobacter, for which the name Acinetobacter pullorum sp. nov. is proposed. The type strain is B301T (=KACC 21653T = JCM 33942T).
The study area, Kwangneung Experiment Forest (KEF) is located on the west-central portion of Korean peninsula and belongs to a cool-temperate broadleaved forest zone. At the old-growth deciduous forest near Soribong-peak (533.1m) in KEF, we have established a permanent plot and a flux tower, and the site was registered as a KLTER site and also a KoFlux site. In this study, we aimed to present basic ecological characteristics and synthetic data of carbon budgets and flows, and some monitoring data which are essential for providing important parameters and validation data for the forest dynamics models or biogeochemical dynamics models to predict or interpolate spatially the changes in forest ecosystem structure and function. We made a stemmap of trees in 1 ha plot and analyzed forest stand structure and physical and chemical soil characteristics, and estimated carbon budgets by forest components (tree biomass, soils, litter and so on). Dominant tree species were Quercus serrata and Carpinus laxiflora, and accompanied by Q. aliena, Carpinus cordata, and so on. As a result of a field survey of the plot, density of the trees larger than 2cm in DBH was 1,473 trees per ha, total biomass 261.2 tons/ha, and basal area 28.0 m2/ha. Parent rock type is granite gneiss. Soil type is brown forest soil (alfisols in USDA system), and the depth is from 38 to 66cm. Soil texture is loam or sandy loam, and its pH was from 4.2 to 5.0 in the surface layer, and from 4.8 to 5.2 in the subsurface layer. Seasonal changes in LAI were measured by hemispherical photography at the l.2m height, and the maximum was 3.65. And the spatial distributions of volumetric soil moisture contents and LAIs of the plot were measured. Litterfall was collected in circular littertraps (collecting area: 0.25m2) and mass loss rates and nutrient release patterns in decomposing litter were estimated using the litterbag technique employing 30cm30cm nylon bags with l.5mm mesh size. Total annual litterfall was 5,627 kg/ha/year and leaf litter accounted for 61% of the litterfall. The leaf litter quantity was highest in Quercus serrata, followed by Carpinus laxiflora and C. cordata, etc. Mass loss from decomposing leaf litter was more rapid in C. laxiflora and C. cordata than in Q. serrata litter. About 77% of C. laxiflora and 84% of C. cordata litter disappeared, while about 48% in Q. serrata litter lost over two years. The carbon pool in living tree biomass including below ground biomass was 136 tons C/ha, and 5.6 tons C/ha is stored in the litter layer, and about 92.0 tons C/ha in the soil to the 30cm in depth. Totally more than about 233.6 tons C/ha was stored in DK site. And then we have drawn a schematic diagram of carbon budgets and flows in each compartment of the KEF site.

Prolific females require better nutrition and feeding practice because of larger litter size and the substantial decline in body fat. Life-time pig output will be compromised if body protein and fat are not properly managed. First litter females are especially vulnerable because they can loose ${\geq}15%$ of whole-body protein. Conservation of body protein mass during first lactation minimizes wean to estrus interval and increases second litter size (up to 1.2 pigs). The ability to influence litter-size by amino acid nutrition is a new dimension in our understanding. A P2 fat depth below 12 mm at farrow and below 10 mm at wean compromised wean to estrus interval (>2 d) and next litter size (0.5 to 1.5 pigs) in sows. It is now clear that a 'modest' excess of feed during the first 72 h of pregnancy decreases embryo viability so that the potential for an increased litter size at birth is not realized. The capacity for milk production by prolific young sows is 25% higher than the standard used previously (NRC, 1988). First litter females averaged 9.82 kg milk/d for a 21 d lactation. Second and third litter counterparts averaged 10.35 kg/d. Milk production was 95% of peak by 10 d of lactation and sows were in greatest negative energy and lysine balance during the first 6 d. Nearly 45% of the total loss in body protein occurred within the first 6 d, but this could reduced to 30-35% by using a more aggressive feeding strategy after parturition. There appear to be 2 phases in lactation for lysine need (d 2-12 vs 12-21). Feeding to the higher level alleviates the second litter size decline. The lysine requirement for lactation can be predicted with accuracy, but we are not able to predict the second limiting amino acid. Mammary uptake of valine relative to lysine and recent work with practical diets suggest that the recent NRC (1998) pattern is realistic and that threonine and valine could be co-limiting for corn-soy diets for prolific sows nursing 10-11 pigs. Empirical studies are needed to refine the ideal pattern so that synthetic lysine can be used with more confidence. Milk fat output for the elite sow is extraordinary and poses an unnecessarily high energetic cost. Methods that reduce mammary fat synthesis will benefit the sow and may enhance piglet growth.
The extracts from approximately 40 different Korean traditional medicines were prepared to investigate the antimicrobial activities against poultry disease-related bacteria. Among tested, the extracts of Schizandra chinensis (SC), Melia azedarach (MA), Caesalpinia sappan (CS) and Rhus javanica (RJ) exhibited significant antimicrobial activities against Salmonella gallinarum, whereas the extracts of Elsholtzia ciliata (EC), Myristica fragrans (MF), Alpinia katsumadai (AK), Poncirus trifoliata (PT), Prunella vulgaris (PV), CS and RJ exhibited antimicrobial activities against Staphylococcus aureus. Minimum inhibitory concentrations (MIC) of MA, CS and RJ extracts against S. gallinarum were 1.2 mg/ml, whereas MIC of RJ extract for S. aureus was 0.6 ㎎/ml, which was the lowest among tested. The antimicrobial activities of SC and RJ extracts against S. gallinarum were reduced, but those of AK and CS extracts against S. aureus were not affected by heating treatment. The antimicrobial activities of SC extract against S. gallinarum and those of EC, PT and RJ extracts against S. aureus were stable by acid treatment but unstable by alkaline treatment. those of CS extract was not effected by either acid or alkaline treatment. The growth of all bacteria was significantly inhibited within 24 hours by the addition of at least 100 ppm and 300 ppm of RJ and CS extracts, respectively, compared with the control group. In conclusion, these findings suggest that RJ and CS extracts may play important roles for antimicrobial activities against poultry disease-related bacteria.
We developed laminated cylindrical scaffolds composed of poly(lactide-co-glycolide)(PLGA) and keratin, and investigated their potential for tissue engineering and disk regeneration. The scaffold was designed to have two parts, i.e. inner cylinder and outer disk, to mimic a natural disk. The outer disk was composed of PLGA and the inner cylinder was prepared using PLGA film or PLGA/keratin hybrid film. In this study, we investigated the effects of keratin on the growth and proliferation of annulus fibrous(AF) cells in the cylindrical scaffolds. Scaffolds containing PLGA/keratin films showed a significantly higher cell proliferation and expression of collagen I and II than the counterpart with PLGA films. Keratin containing scaffolds also exhibited an excellent mechanical strength, demonstrating that keratin influences the proliferation of annulus fibrous cells. The results provide valuable information on PLGA/keratin films for tissue engineered disk regeneration.
This study was carried out to provide information for establishing counter measures of soil pollution through analysis of Cd. Cu, Pb, and Zn in paddy soils and brown rice. Cadmium, Cu, Pb, and Zn contents in soils were analyzed and distribution maps for these heavy metals were prepared. Heavy metal contents in brown rice were also measured. Average contents of Cd, Cu, Pb, and Zn in surface paddy soils extracted with 0.1 N HCl were 7.4, 35.8, 98.9, and $118.8mg\;kg^{-1}$, respectively. These were 9 times (Cu) to 50 times (Cd) higher than the background level of heavy metals in unpolluted paddy soils in Korea. The contents of Pb and Zn were lower than those measured in 1980, whereas Cd content did not decrease. The levels of heavy metal contamination in paddy soils may not affect growth or yield of rice plant, however, Cd contents indicated a level of serious concern to humans. The average contents of Cd, Cu, Pb, and Zn in brown rice were 0.38, 2.38, 1.31 and $22.31mg\;kg^{-1}$, respectively.
The objective of the study was to investigate the effects of water treatment sludge (WTS) on the growth of soybean and carrot, and uptake of some inorganic components in the plants. WTS was incorporated to field at the rates of 0, 1, 3, and 5 tons per 10a on the basis of dry weight. With the application of the sludge, OM and CEC in the soil increased slightly while the concentration of available phosphorous decreased. And heavy metals, including Pb, Cd, Cr, As and Hg, were not detected at the harvest stage in crops. With an increase in the application of the sludge, the concentration of phosphorous in the seed of soybean and the root of carrot tended to be decreased. There seems no correlation between rate of application of the sludge and uptake of Al in the plants. The yield of soybean was the highest when applied to one ton sludge and the yield on carrot increased with an increase in rate of applied sludge. Also, the concentration of carotene increased with an increased application of the sludge. But, application of WTS showed to deteriorate the visual quality with an irregularity of carrot's surface. This results indicate that application of WTS will be give a positive and negative effects on soil and crops.

The aim of this study is to compare cariogenic characteristics of fluoride-sensitive Streptococcus mutans [fluoride-sensitive (FS) S. mutans ] and fluoride-resistant Streptococcus mutans [fluoride-resistant (FR) S. mutans] in the presence of sucrose, and to evaluate its effect on cariogenic biofilm formation. S. mutans ATCC 25175 was continuously cultured in trypticase soy broth (TSB) containing NaF (70 ppm) for 40 days to generate FR S. mutans. FS and FR S. mutans were inoculated in TSB with or without 2% sucrose, and optical density and pH were measured every hour. An oral biofilm was formed using saliva bacteria and analyzed through confocal laser scanning microscopy and CFU count. Finally, the expression of glucosyltransferases genes of both S. mutans was investigated through RT-PCR. FR S. mutans exhibited slower growth and lower acidogenicity in the presence of sucrose compared to FS S. mutans . Both cariogenic and single species biofilm formation was lower in the presence of FR S. mutans, along with reduced number of bacteria. FR S. mutans showed significantly low levels of gtfB, gtfC, and gtfD expression compared to FS S. mutans . On the basis of results, FR S. mutans may be less virulent in the induction of dental caries.
