Monoclonal antibody to the Escherichia coli heat-stable enterotoxin(ST) was produced to develop a rapid and convenient diagnostic method to the ST. The toxin was purified from culture supernatant of enterotoxigenic E. coli O148H28($ST^+/LT^+$) and conjugated to bovine serum albumin(BSA). The ST-BSA conjugate was used to immunize BALB/c mice and the immune spleen cells from these mice were fused with $P3{\times}63$ Ag8.V653 plasmacytoma cells. Hybridomas were screened by ELISA and positive hybridomas were cloned by limiting dilution. Finally, one stable clone (AS36) specific to ST was selected for further growth and characterization. Antibody titers of culture supernatant and ascitic fluid from BALB/c mice were 1:1,024 and 1:20,480 respectively in ELISA. The isotype and subclass of monoclonal antibody was IgG1 in sandwich ELISA. To test the neutralizing effect of monoclonal antibody on toxin activity of ST, mixture of ascitic fluid and ST was assayed by infant mouse assay and this monoclonel antibody was proved to be a neutralizing antibody. The titer of ascitic fluid which completely neutralized biological activity of 4 units of ST was 1:4. Purified ST was quantitatively measured by competitive ELISA and minimum amount of ST detectable by this assay was 250pg, which was an amount six-fold smaller than that detectable by infant mouse assay. Four reference strains of enterotoxigenic E. coli from WHO were detected by competitive ELISA and highly specific, sensitive and reproducible result was obtained.
The outbreak of haemorrhagic specticemia and lesions in dermis, frequently associated with keratitis have occured on rock sea bream (Oplegnathus fasciatus) reared in Jeju island. A bacterium isolated from the diseased fish identified as Pseudomonas sp. by morphologic, cultural, and biochemical tests. The bacterium was identified Pseudomonas anguilliseptica according to 16S rDNA sequence analysis. The highest growth rate of this strain was obtained at 20 to $\25^circ{C}$ although the disease occurred mostly below $16^\circ{C}$. The challenge test by ip injection revealed that this bacterium was pathogenic to rock sea bream. All experimental fish in challenged by ip injection died within 21 days but there was no death in control group. The bacterium was sensitive to Doxycycline, Chloramphenicol, Ciprofloxacin, Erythromycin, Flumequine, Nalidixic acid, Norfloxacin, and Ofloxacin.
The present study was carried out to reveal the characteristics of organism responsible for so called vibriosis prevailing in warm water season among cultured kuruma prawn Penaeus japonicus. A bacterium was isolated from the heart, lymphoid organ and muscle of the diseased kuruma prawn. Six strains obtained from diseased kuruma prawn in some culture farms in Namhae and Anhung from august to october in 1989 were submitted to the morphological, biochemical and physiological characterization. All the strains were gram-negative, nonsporning short rods with one polar flagellum. Glucose was fermented with no gas production by these strains. Some distinguishing features of the organisms were negative to lysine, arginine, ornithine decarboxilization test. In general, the temperature $20{\sim}27^{\circ}C$, the NaCl concentration of 2~3% and pH of 6~8 were optimal for the growth in peptone water. The organisms were sensitive to the vibrio static agent 0/129. The isolated should be identified as Vibrio sp.. No essential differences in histopathological finding were noted between the naturally and the experimentally infected prawns. Most characteristic pathological changes were extensive necrosis caused by severe bacteria invasion and multiple formation of meranized nodules in the lymphoid organ.
The intestinal microflora of humans is an extraordinarily complex mixture of microorganisms, the majority of which are anaerobic microorganisms. The distribution of amylolytic microorganisms in the human large intestinal tract was investigated in various individuals of differing ages using anaerobic culture techniques. A large percentage of the amylolytic microorganisms present belonged to the Genus Bifidobacteria. The number of Bifidobacteria increased significantly at two years of age. Adults and children above 2 years old carried about $0.8{\times}10^9-2.0{\times}10^{10}$ colony forming units (CFU/gram) of amylolytic Bifidobacteria. Among these amylolytic Bifidobacteria, Int-57 was chosen for further studies. Between 65% and 85% of the amylase produced was secreted and the remaining amylase was bound to the cell wall facing the outside. Amylase production could be induced by starch in a stable form. When cells were grown on maltose or glucose, amylase production was much lower than on starch and amylase activity disappeared after 24 hours growth on these media. Partially purified enzymes showed optimum activity at a temperature of $50^{\circ}C$ and at an optimum pH of 5.5, respectively. Heat treatment at $70^{\circ}C$ for 30 minutes almost completely inactivated amylase. The hydrolysis products of starch were mainly maltose and maltotriose. Soluble starch, amylose, amylopectin, and $\gamma$-cyclodextrin($\gamma$-CD) were easily hydrolyzed. The rate of hydrolysis of $\alpha$-CD and $\beta$-CD was slower than that of $\gamma$-CD. Carboxymethyl cellulose, $\beta$-1, 3-glucan and inulin were not hydrolyzed.

Strain BH5 was isolated from naturally fermented Kimchi and identified as a bacteriocin producer, which has bactericidal activity against Micrococcus flavus ATCC 10240. Strain BH5 was identified tentatively as Lactococcus lactis by the API test and some characteristics. Lactococcus lactis BH5 showed a broad spectrum of activity against most of the non-pathogenic and pathogenic microorganisms tested by the modified deferred method. The activity of lacticin BH5, named tentatively as the bacteriocin produced by Lactococcus lactis BH5, was detected at the mid-log growth phase, reached its maximum during the early stationary phase, and decreased after the late stationary phase. Lacticin BH5 also showed a relatively broad spectrum of activity against non-pathogenic and pathogenic microorganisms as tested by the spot-on-lawn method. Its antimicrobial activity on sensitive indicator cells was completely disappeared by protease XIV or ${\alpha}$-chymotrypsin. The inhibitory activities of lacticin BH5 were detected during treatments up to 100$^{\circ}C$ for 30 min. Lacticin BH5 was very stable over a pH range of 2.0 to 9.0 and was stable with all the organic solvents examined. The cell concentration and bacteriocin production in strain BH5 were maximum when grown at 30$^{\circ}C$ in a modified MRS medium supplemented with 0.5% tryptone, 1.0% yeast extract, and 0.5% beef extract as nitrogen sources. It demonstrated a typical bactericidal mode of inhibition against Micrococcus flavus ATCC 10240. Lacticin BH5 was purified through ammonium sulfate precipitation, ethanol precipitation, and CM-Sepharose column chromatography. The apparent molecular mass of lacticin BH5 was estimated to be in the region of 3.7 kDa, by the direct detection of bactericidal activity after SDS-PAGE. Mutant strain NO141 which was isolated by nitrosoguanidine mutagenesis produced about 4 fold more bacteriocin than the wild type.
Al2O3/t-ZrO2 particulate composites were prepared by sintering at 150$0^{\circ}C$ and 1$600^{\circ}C$ for 2h in air and microstructure and mechanical properties of the composites were investigated. Although most ZrO2 particles existed at Al2O3 grain boundaries a few ZrO2 particles within Al2O3 grains. Al2O3 grain growth was depressed due to the pinning effect by ZrO2 particles. During sintering coarsening of intergranular ZrO2 particles occurred as a results of the elimination of ZrO2 intraagglomerate grain boundaries and the coalescence of dragged ZrO2 particles by migrating Al2O3 grain boundries. Changes in mechanical properties of Al2O3 composites were dependant on microstructure of Al2O3 matrix and on size and structure of dispersed ZrO2.
In this study, the effects of Mulberry leaves extract (ME) addition on the quality of Kimchi was investigated during 30 days of fermentation at $5^{\circ}C$. ME was added to salted cabbage at concentrations of 0% (C), 0.4% (ME1), 0.8% (ME2) and 1.2% (ME3) (w/w). The pH of Kimchi added above 0.8% ME was higher than that of Kimchi without ME after 12 days of fermentation. During overall fermentation, the titratable acidity of Kimchi with 1.2% ME remained at the lowest level. The degree of salinity decreased with increasing amount of added ME, whereas control groups showed the highest salinity. In L, a and b values of samples, the highest Figures of L and b were observed in control groups, and ME3 showed the highest value of a. In addition, the cutting force during the fermentation period was higher in all treated groups compared with control groups, and ME3 showed the highest hardness value. Moreover, growth of lactic acid bacteria and total bacteria were inhibited by addition of ME. In the sensory assessment, 0.4% ME improved the quality of Kimchi, showing the highest taste score and overall preference. Therefore, addition of 0.4% ME appears to be an acceptable approach to enhance the quality of Kimchi without reduction of acceptability.
Background: The incidence of halitosis has a prevalence of 22-50% throughout the world and is generally caused by anaerobic oral microorganisms, such as Fusobacterium nucleatum, Clostridium perfringens, and Porphyromonas gingivalis. Previous investigations on the structure-activity relationships of ginsenosides have led to contrasting results. Particularly, the antibacterial activity of less polar ginsenosides against halitosis-related bacteria has not been reported. Methods: Crude saponins extracted from the Panax quinquefolius leaf-stem (AGS) were treated at $130^{\circ}C$ for 3 h to obtain heat-transformed saponins (HTS). Five ginsenoside-enriched fractions (HTS-1, HTS-2, HTS-3, HTS-4, and HTS-5) and less polar ginsenosides were separated by HP-20 resin absorption and HPLC, and the antimicrobial activity and mechanism were investigated. Results: HPLC with diode-array detection analysis revealed that heat treatment induced an extensive conversion of polar ginsenosides (-Rg1/Re, -Rc, -Rb2, and -Rd) to less polar compounds (-Rg2, -Rg3, -Rg6, -F4, -Rg5, and -Rk1). The antimicrobial assays showed that HTS, HTS-3, and HTS-4 were effective at inhibiting the growth of F. nucleatum, C. perfringens, and P. gingivalis. Ginsenosides-Rg5 showed the best antimicrobial activity against the three bacteria, with the lowest values of minimum inhibitory concentration and minimum bactericidal concentration. One major reason for this result is that less polar ginsenosides can more easily damage membrane integrity. Conclusion: The results indicated that the less polar ginsenoside-enriched fraction from heat transformation can be used as an antibacterial agent to control halitosis.
Benomyl has been used to control Mycogone perniciosa for several years, the reduced effect of Benomyl against Mycogone perniciosa has been reporteod here. These experiments were carried out to substitute Benomyl with other chemicals. The isolates of Mycogone perniciosa those Benomyl tolerant are proved to be sensitive to Vitathiram, Difolatan, Ambam and RH-2161. However, these chemicals brought undisirable effects on the cultivation. Basamid and Vapam are found not to be harmful to Agaricus bisporus' mycelial growth in casing soil, these chemicals were treated up to 100 ppm of concentration for 15days in advance of use. Although Mycogone perniciosa was tolerant to Benomyl, Basamid and Vapam were very effective and enabled it to produce higher levels of mushroom sporophores.
Six unrecorded yeasts, Cryptococcus festucosus 41-3, Cryptococcus heveanensis 56-4, Debaryomyces nepalensis 95-4, Issatchenkia occidentalis 142-1, Dioszegia zsoltii 39-1, and Kwoniella europala 47-2 were screened from 108 yeasts isolated from flowers and fruits in orchards of Yesan-gun, Chungcheongnam-do, Korea. The morphological and cultural characteristics of these unrecorded yeasts were investigated. They had various shapes, including ellipsoidal, globose, and oval, and also had budding mode in vegetable reproduction, except I. occidentalis 142-1 (fission mode). K. europaea 47-2 only formed pseudomycelium. D. zsoliti 39-1 did not grow in yeast extract-malt extract medium, potato dextrose medium, and vitamin-free medium. C. festucosus 41-3 grew well in 5% NaCl-containing yeast extract-peptone-dextrose medium and had a growth pH range of 7.0~10.0. Three unrecorded yeasts Ogataea polymorpha HB45-1, Rhodotonula hinnulla HB62-2, and Cryptococcus rajasthanensis HB80-4 were screened from 51 yeasts isolated from flowers in Hanbat arboretum in Daejeon city, Korea. They were globose in shape and did not form pseudomycelium. In addition, O. polymorpha HB45-1 and C. rajasthanensis HB80-4 had budding mode in vegetable reproduction. All of them grew well in vitamin-free medium and C. rajasthanesis HB80-4 also grew in 50% glucose and 5% NaCl-containing YPD medium.
