Variation in Tree Growth Characteristics, Pilodyn Penetration, and Stress-wave Velocity in 65 Families of Acacia mangium Trees Planted in Indonesia

  • Growth characteristics [stem diameter (D), tree height (H)] and wood properties [Pilodyn penetration (P) and stress-wave velocity (SWV)] were measured for 65 families of 6-year-old Acacia mangium trees planted in Indonesia, in order to characterize their variation in D, H, P, and SWV. Therefore, the correlations between the measured characteristics were also determined, and their significant differences observed. Furthermore, their low to moderate values of narrow-sense heritability was obtained, and the results indicated the characteristics to be genetically controlled in A. mangium. In addition, highly significant positive correlations were observed among the growth characteristics, suggesting a close relationship, while there was no significant association between the growth characteristics and P, as well as SWV, indicating their independent. Therefore, these results demonstrate a potential for the improvement of both growth and wood properties of A. mangium trees, using the appropriate breeding programs. In addition, 18 families showed good performance in D and SWV, signifying their positive prospect of being considered as plus trees for the next generation breeding cycles.

  • Two experiments were conducted to determine the effects of $MnSO_4$ on controlling harmful microorganisms in vitro and in vivo. The in vitro experiment was conducted to examine the effects of manganese sulfate $(MnSO_4)$ on the reduction of Escherichia coli (E. coli) and Staphylococcus aureus (S. aureus) by growth stimulation of Pediococcus acidilactici (P. acidilactici; lactic acid bacteria). Manganese ion (0.003 %) was found to stimulate the growth of P. acidilactici in the In Vitro system. When E. coli and S. aureus were grown in a mixture with P. acidilactici, their numbers were reduced. This may be the result of a reduction of pH in the medium as a result of better growth of P. acidilactici due to stimulation by the Mn ion. The in vivo experiment was conducted to determine the effects of $MnSO_4$ in diets on controlling harmful microorganisms in fecal samples of pigs. There were no significant differences for the microbial numbers (i.e., total microorganisms, E. coli, lactic acid bacteria and S. aureus) in feces of pigs fed $MnSO_4$ compared to feces of pigs fed the control diet through 7 days. However, on day 7 of experiment, the pH of feces in pigs fed $MnSO_4$ (0.1%) decreased faster than pigs fed the control diet.

  • As a basic study to elucidate nutritional physiology and composition of synthetic medium of red rotting bacteria, Erwinia carotovora, of ginseng, the effects of hydrogen ion concentration, various kinds of carbon sources, nitrogen source, micrometallic salts and it's concentration on the gorwth of the bacteria were investigated and the results were as follows. Optimal pH in the basal medium for the growth of the bacteria was 6.5. After incubation the pH in culture media was neutralized. Among the various kinds of carbon sources, sucrose, glucose mannitol, but organic acids were not utilized effectively as nutrients. After incubation the pH turned acidic. Alanine as organic nitrogen sources nad ammonium sulfate as inorganic nitrogen promoted the growth, but L-valine and sodium nitrite were the least effective. Ferric chloride 1.0mg/dl and ferrous sulfate 100mg/dl were the most effective as micrometallic sources. Control and boric acid were the least effective. New synthetic medium based on the above results was follows ; Alanine 1.0g, $KH_2PO_4\;1.0g, \;sucrose\;30.0g, \;MgSo_4{\cdot}7H_2O\;0.5g, \;FeCl_3{\cdot}6H_2O\;1.0mg\;thiamine\;200{\gamma}g$, and distilled water 1000ml, pH6.5.

  • The toxic compounds of sorghum stem extracts were isolated by different organic solvents and pH, and characterized and quantified in terms of their inhibition of seed germination and seedling growth in Echinochloa colona(L.) Link and radish(Raphanus sativus L.). Sequential partitioning of stem extract with various organic solvents with increasing polarity showed that all fractions of hexane, ethyl ether, methylene chloride, ethyl acetate, and the aqueous remainder inhibited germination and seedling growth in E. colona. Of the five fractions, the ethyl ether fraction had the greatest inhibitory effect on E. colona. Further separation of the ethyl ether fraction at different pH(pH 2-11) showed that phytotoxic compounds were acidic. The result indicates that the phytotoxin present in the stem extract may be nonpolar and acidic.

  • This study was investigated to determine the antimicrobial activity of ethanol extract of green tea, and of solvent fractionated ethanol extract on Escherichia coli O157:H7. MIC value of ethanol extracts and solvent fractionated ethanol extract (hexane, ethyl-acetate, chloroform and water) were $2000{\mu}g/ml,\;0,\;500{\mu}g/ml,\;2000{\mu}g/ml,\;1000{\mu}g/ml$, respectively. The antimicrobial activity of ethyl acetate fraction against E. coli O157 :H7 ranged from $250-2000{\mu}g/disk$. The antimicrobial activity was significantly increased as concentration dependent. When E. coli O157:H7 was pH-adjusted TSB containing $1000{\mu}g/ml$ ethyl acetate, E. coli O157:H7 was significantly inhibited at initial pH of 10, whereas the pathogen grew well in the presence of pH 4.5-pH 9. The inhibitory effect of the ethyl acetate fraction on the growth of E. coli O157 :H7 was investigated. Growth of both strain in the tested conditions were rapidly occurred up to 12 h, but no growth was occurred in the presence of $250-1000{\mu}g/mL$ of ethyl acetate fraction for 72 h.

The Value of Ultrasonographic Endometrial Measurement in the Prediction of Pregnancy Outcome in In Vitro Fertilization (체외수정시술 주기에서 자궁내막발달과 착상에 관한 연구)

  • The condition of the endometrium is an important factor which may influence the success or failure in IVF-ET. This study was undertaken for evaluation of the value of endometrial growth as an early predictor for the success of IVF. Ultrasonographic endometrial measurement were performed in 43 IVF cycles that conceived, 101 cycles that did not with an IVF-ET There was no significant difference in the endometrial thickness and the serum concentration of estradiol in the pregnant versus nonpregnant group(10.4 vs. 9.9 mm: 2348 vs. 2017 pg/ml no hCG administration day). No correlation was found between the ultrasound image and serum estradiol levels around the time of hCG administration(r=0.54, p=0.13 no Day 2; r=0.45, p=0.14 no Day 1). The duration of gonadotropin treatment, number of follicles, number of oocytes retrieved, and fertilization rate were not statistically different in the two groups, however, there was a significant difference in the number of embryos in the pregnant versus nonpregnant group)p< 0.05). A higher pregnancy rate and ongoing pregnancy rate occured with an endometrial thickness over 11 mm compared with below 7mm(p< 0.05, p< 0.005). however, no significant differences were noted in the implantation rate and abortion rate among the groups that classified according to their endmetrial thickness. The endometrial growth(${\Delta}$) from hCG administration day(DO) to D6 was greater in the women who achieved pregnancy than in the nonpregnant group(p< 0.01). There were no significant differences in serum estradiol levels, implantation rate, pregnancy rate, and abortion rate among the groups that classified according to the pattern of echogenesity of endometrium, however, significantly higher ongoing pregnancy rate was noted in group A, B compared with group C.(p< 0.0001, p< 0.001) These results suggest that there were no ultrasonographically detectable differences in the patterns of endometrial growth and development around the time of hCG administration in patients who conceive versus those that do not in IVF-ET.

Influence of Dietary Oligosaccharides on Growth Performance and Intestinal Microbial Populations of Piglets

  • An experiment was conducted to determine the effects of dietary oligosaccharides on performance and intestinal microbial populations of piglets. Ten litters of piglets were assigned to five groups randomly, with two litters per group. The control group was fed with corn-soybean basal diet. Oligosaccharides was added to the basal diet at the level of 0.05%, 0.1%, 0.2% and 0.35% respectively to form four experimental diets. The experiment was conducted with two periods. The first period (suckling period) was from 7 to 28 days of age and the second period (weanling period) was from 28 to 56 days of age. Fresh fecal samples were collected at 21 days of age and assayed for Escherichia coli concentration, pH and moisture content. Three pigs per group were slaughtered at 42 days of age and cecum, colon, and rectum content samples were collected and assayed immediately for Escherichia coli and Bifidobacterium concentration, pH and moisture content. The results showed that dietary oligosacchrides decreased fecal Escherichia coli population and pH significantly (p 0.05), but did not affect performance and fecal moisture content during suckling period. 0.1% oligosaccharides for weanling pigs increased growth and improved feed conversion ratio together with a reduction of diarrhea (p 0.05), but 0.35% oligosaccharides did not affect growth performance. 0.1% and 0.2% oligosaccharides for weanling pigs had a suppression to Escherichia coli colonization in rectum and an enrichment to Bifidobacterium in colon (p 0.05). Oligosaccharides decreased significantly (p<0.05) rectum moisture content, but did not affect cecum, colon and rectum pH.

Tree Ring Ca/Al as an Indicator of Historical Soil Acidification of Pinus Densiflora Forest in Southern Korea

  • BACKGROUND: Soil acidification, which is known to be one of the reasons of forest decline, is associated with decreases in exchangeable Ca and increases in Al concentration, leading to low Ca/Al ratio in soil solution. As tree rings are datable archives of environmental changes, Ca/Al ratios of annual growth ring may show decreasing pattern in accordance with the progress of soil acidification. This study was conducted to investigate Ca/Al pattern of Pinus densiflora tree ring in an attempt to test its usefulness as an indicator of historical soil acidification. METHODS AND RESULTS: Three P. densiflora tree disks were collected from P. densiflora forests in Jeonnam province, and soil samples (0-10, 10-20, and 20-30 cm in depth) were also collected from the tree locations. Soils were analyzed for pH and exchangeable Ca and Al concentrations, and Ca/Al was calculated. Annual growth rings formed between 1969 and 2007 were separated and analyzed for Ca/Al. Soil Ca/Al was positively (P<0.01) correlated with soil pH, suggesting that soil acidification decreased Ca while increasing Al availability, lowering Ca/Al in soil solution. The Ca/Al of tree rings also showed a decreasing pattern from 18.2 to 5.5 during the period, and this seemed to reflect historical acidification of the soils. CONCLUSION(s): The relationship between soil pH and Ca/Al and the decreasing pattern of Ca/Al of tree ring suggest that Ca/Al of tree ring needs to be considered as a proxy of the progress of soil acidification in P. densiflora forest in southern Korea.

Study on the rumen fermentation, growth performance and carcass characteristics according to the supplementation of lupin flake in Hanwoo steers

  • This study was conducted to determine the rumen fermentation dynamics of lupin flakes and elucidate the effects of lupin flake supplementation on the growth performance, blood metabolites, and carcass characteristics of Hanwoo steers. In vitro and in situ trials of lupin grains and lupin flakes were conducted using three Hanwoo cows with rumen fistulas. The feeding trial included 40 early-fattening Hanwoo steers randomly divided into four groups: control, T1, T2, and T3. Their formula feed contained 0%, 3%, 6%, and 9% lupin flakes, respectively. In vitro rumen pH and ammonia concentrations were lower in the lupin flake group than in the lupin grain group after 6 and 24 h of incubation, respectively (p < 0.05). Concentrations of propionate, butyrate, and total volatile fatty acids were higher in the lupin flake group than in the lupin grain group after 12 h of incubation (p < 0.05), as was the crude protein disappearance rate at 9 and 12 h of rumen fermentation (p < 0.05). Supplementation with lupin flakes did not affect the average daily gain. Compared to that in the control group, dry matter intake was lower in the lupin flake-supplemented groups (p < 0.05); the feed conversion ratio was lower in T2 and T3 (p < 0.05); and plasma total protein concentration in 29-month-old steers was lower in T1 and T3 (p < 0.05). Plasma triglyceride concentration was lower in the lupin flake-supplemented groups than in the control group (p < 0.05). The incidence rate of yield grade A was higher in T1 and T2 than in the control group; the incidence rate of meat quality 1+ grade or higher was highest in T2. The carcass auction price was higher in T2 than in the other groups. Overall, compared to whole lupin grains, lupin flakes seem to more substantially affect rumen ammonia concentrations and crude protein disappearance rate. Additionally, we suggest that supplementation with 6% lupin flake formula feed exerts positive effects on the feed conversion ratio, yield grade, and quality grade of Hanwoo steers.
