The Effects of Lactic Acid Bacteria on the Growth and Aflatoxin $B_1$ Production of Aspergillus parasiticus (유산균이 Aspergillus parasiticus의 성장과 Aflatoxin $B_1$ 생성에 미치는 영향)

  • This study was performed to investigate the competitive effect of Leuconostoc mesenteroides and Lactobacillus plantarum on the growth and aflatoxin $B_1$ production of Aspergillus parasiticus. Leu. mesenteroides and Lac. plantarum were grown with A. parasiticus in a modified APT broth at $28^{\circ}C$ for 9 days. It was observed that the growth of A. parasiticus got lower when the two bacteria were inoculated simultaneously than when the mold grew alone. The pH of mixed culture of Leu. mesenteroides and Lac. plantarum showed significantly lower value than the pH of the pure culture of A. parasiticus (p<0.05). The acidity of the mixed culture group significantly increased compared with the control group (A. parasiticus alone group) (p<0.05). The aflatoxin $B_1$ production was significantly decreased in the mixed culture group than in the A. parasiticus alone group (p<0.05). Leu. mesenteroides showed the more efficient effect than Lac. plantarum. These results indicate that the two lactic acid bacteria have inhibitory effect on the growth and aflatoxin $B_1$ production of Aspergillus parasiticus.

Effects of pH of soil medium on the growth and nutrient absorption of cultivated and native Chinese chives plants (토양배지의 pH가 재배 및 자생 부추류의 생육과 양분흡수에 미치는 영향)

  • This experiment was conducted to investigate the effects of pH on the mineral nutrient uptake and growth of the four Chinese chives species. The Chinese chives species used in the experiment were the cultivated species grown in the farm (cultivated Allium tuberosum) and three wild species of wild Allium tuberosum, A. thunbergii and A. senescens. The pH levels of soil medium were set to be 4.5, 6.5, and 7.5. Fresh weight(FW) of cultivated A. tuberosum was highest at all pH levels. The increase of soil pH increased the FW of the wild A. tuberosum and A. thunbergii, but no difference was noted for the A. tuberosum and A. senescens. Plant height was higher in the order of wild A. tuberosum, A. thunbergii, and cultivated A. tuberosum and A. thunbergii. Notably plant height of the wild A. tuberosum increased significantly by the pH increase. The Zn content of the wild A. tuberosum was shown to be significantly higher than that of the other species and increased with the increase of soil pH. This indicates that there is a close relationship between the plant height and Zn content in Chinese chives plant. Principal component analysis for characterizing closely related A. species using the factors of plant growth and amounts of nutrients uptake showed that the cultivated A. and wild A. tuberosum were in the $4^{th}$ quadrant of the graph which are classified as the same species, while A. senescens and thunbergii was in $1^{st}$ and $3^{rd}$ quadrant indicating different species, respectively.

Various levels of copra meal supplementation with β-Mannanase on growth performance, blood profile, nutrient digestibility, pork quality and economical analysis in growing-finishing pigs

  • Background: To reduce use of main feed ingredient like corn, soy bean meal (SBM) and wheat, alternative ingredients has been studied like copra meal (CM). Production amount of CM which has been high makes CM to be an alternative feed stuff. However, low digestibility on AA and low energy content by high fiber content can be an obstacle for using CM. This experiment was conducted to evaluate the effects of CM supplementation with ${\beta}$-mannanase on growth performance, blood profile, nutrient digestibility, pork quality and economic analysis in growing-finishing pigs. Methods: A total of 100 growing pigs ([Yorkshire ${\times}$ Landrace] ${\times}$ Duroc) averaging $31.22{\pm}2.04kg$ body weight were allotted to 5 different treatments by weight and sex in a randomized complete block (RCB) design in 5 replicate with 4 pigs per pen. Treatments were 1) Control (corn-SBM based diet + 0.1% of ${\beta}$-mannanase (800 IU)), 2) CM10 (10% copra meal + 0.1% ${\beta}$-mannanase (800 IU)), 3) CM15 (15% copra meal + 0.1% ${\beta}$-mannanase (800 IU)), 4) CM20 (20% copra meal + 0.1% ${\beta}$-mannanase (800 IU)) and 5) CM25 (25% copra meal + 0.1% ${\beta}$-mannanase (800 IU)). Four phase feeding program was used: growing I (week 1-3), growing II (week 4-6), finishing I (week 7-9) and finishing II (week 10-12). Results: In growth performance, there was no significant difference among treatments during whole experimental period. In growingI phase, G:F ratio tended to increase when CM was increased (P = 0.05), but ADG and ADFI tended to decrease in finishingII phase (linear, P = 0.08). Also, increasing CM reduced ADG (linear, P = 0.02) and feed efficiency (linear, P = 0.08) during the whole finishing period. In blood profiles, BUN was linearly increased as CM increased (linear, P = 0.02) at growingII period. In digestibility trial, there was no significant difference in dry matter, crude fat, crude ash and nitrogen digestibility. However, crude protein digestibility was decreased linearly (linear, P = 0.02). In economic analysis, feed cost per weight gain and total feed cost per pig were reduced in overall period when CM was provided by 25% (linear, P = 0.02). Conclusion: CM with 0.1% of ${\beta}$-mannanase (800 IU) could be supplemented instead of corn and SBM up to 25% without detrimental effects on growth performance and pork quality of growing-finishing pigs.

Physiological Characteristics and Optimized Culture Condition of the Mycelia of Inonotus mikadoi. (황갈색시루뻔버섯(Inonotus mikadoi)의 균사체 배양 최적 조건 및 생리학적 특성)

  • The culture condition and medium composition for the enhanced mycelial growth of Inonotus mikadoi IMSNU 32058 were investigated. The optimal temperature and pH for the mycelial growth were $27^{\circ}C$ and pH 4.5, respectively. Among the complex media tested, the malt extract medium and Phellinus igniarius medium were very good for mycelial growth of I. mikadoi. When Czapek-Dox medium was used as a minimal medium for cultivation of mycelia, xylose, raffinose and carboxymethyl cellulose were very excellent as a carbon and energy source. With respect to carbohydrate, sucrose and glucose were very good carbon sources. In general, organic complex nitrogen sources were better than other inorganic ones. As the organic complex nitrogen sources tested, yeast extract, soytone, proteose peptone and bacto peptone were the best as a source of organic nitrogen. When ammonium sulfate as an inorganic source of nitrogen was used, the enhanced mycelial growth was shown. p-Aminobenzoic acid was proved to be most appropriate source of vitamin. After the mycelia of I. mikadoi was cultivated at $27^{\circ}C$ for 5 days in MEM broth (pH 4.5), the activities of both exomycelial and endo-mycelial enzymes were determined. Among endomycelial enzymes assayed, the specific activity of laccase was much higher than those of other enzymes. When the fungus was grown in MEM broth, exomycelial specific enzyme activity of laccase was comparatively high. However, little or no enzyme activities of protease, chitinase and lipase were found.

  • The seed formation and growth of $\alpha$-ferric oxyhydroxide with aerial oxidative precipitation from aqueous solution of ferrous sulfate with KOH, NaOH, $Na_2CO_3$ and $K_2CO_3$ as precipitants have been studied by free pH drift experiment. It has been shown that all precipitants give same particle formation and growth path, and average particle length from KOH and NaOH as precipitants was about 1.5 times shorter than that of $K_2CO_3$ and $Na_2CO_3$. When initial mole ratio, $R_o=[Fe^{2+}]_o/[OH^-]_o$ of KOH was decreased the particle was grown oxyhydroxide seed growth from aqueous solution of ferrous sulfate with KOH has been studied. The influence of the air flow rate, reaction temperature and initial mole ratio, $R_o=[Fe^{2+}]_o/[OH^-]_o$, on the kinetics of seed growth are investigated by static pH experiment. The oxidation rate of seed growth increased with increase in the air low rate, reaction temperature and initial mole patio. The activation energy of seed growth is 16.16 KJ/mol and the rate equation of seed growth can be written as follows: $-\frac{d[Fe^{2+}]}{dt}=1.46{\times}10^4[P_{o2}]^{0.66}[OH^-]^{2.19}exp(-\frac{16.16}{dt})$.

  • Effects of some sources on the vegetative growth of Naematoloma sublateritium (Fr.) Karst. were investigated using liquid and solid culture media. Temperature, pH, carbohydrates as carbon sources, amino acids as nitrogen sources, the optimal carbon/nitrogen ratio, mineral element and organic acids were studied for good mycelial growth. We could improve a new semisynthetic medium for mycelial growth in N. sublateritium.

Effects of Dietary Supplemental $Megazone^{(R)}$ on Growth Performance, Nutrients Digestibility, Blood Characteristics, Meat Quality and Carcass Traits in Weaning-to-Finishing Pigs

  • This study was conducted to investigate the effects of $Megazone^{(R)}$ (a mineral mix) supplementation on growth performance, nutrients digestibility, blood characteristics, meat quality and carcass traits in weaning-to-finishing pigs. A total of 48 crossbred $(Landrace{\times}Yorkshire{\times}Duroc)$ pigs with initial body weight (BW) of $4.46{\pm}0.18kg$ were used in a 21 wks trial. Pigs were blocked by weight and allotted to two dietary treatments in a completly randomized design. There were 6 pens per treatment. Dietary treatments included: 1) Control (CON: basal diet) and 2) MT (basal diet+0.8% $Megazone^{(R)}$). Through the entire experimental period, there were no effects of dietary $Megazone^{(R)}$ supplementation on growth performance nutrients digestibility, blood characteristics and meat quality traits (p>0.05). Market weight and backfat thickness also had no differences between the two treatments (p>0.05). However, carcass weight and carcass ratio in MT treatment were improved significantly compared with CON treatment (p<0.05). In conclusion, supplmentation of $Megazone^{(R)}$ can increase carcass weight and carcass ratio in weaning-to-finishing pigs, however, it has no effects on growth performance, nutrients digestibility, blood characteristics and meat quality traits.

Fermentation Properties and in vitro Anticancer Effect of Young Radish Kimchi and Young Radish Watery Kimchi (열무김치 및 열무물김치의 발효특성과 in vitro 항암효과)

  • Fermentation properties and in vitro anticancer effect of young radish (YR) kimchi and young radish watery (YRW) kimchi were investigated during fermentation at 5℃. The fermentation of YR kimchi during 2-3 weeks led to the decrease of pH down to pH 4.3, increased acidity, and the highest Leuconostoc sp. counts. YR kimchi showed the acidity of 1.04-1.27% at the pH 4.3, when the kimchi was ripened properly. The fermentation of YRW kimchi during 9 days led to the decrease of pH down to pH 4.3 and the acidity of 0.20%. Inhibitory effects of the juices of YR, YR kimchi, and YRW kimchi on the growth of AGS human gastric adenocarcinoma cells in MTT assay were increased with the added concentration. The juice of YR kimchi had a higher inhibitory effect on the growth of AGS human gastric adenocarcinoma cells than that of YRW kimchi at same concentration. The juice of YR kimchi showed similar inhibitory effects on the growth of AGS human gastric and HT-29 human colon adenocarcinoma cells in MTT assay to baechu kimchi, which the inhibition rates are more than 50%.

Physiological Responses and Growth Performance of Alnus firma and Alnus hirsuta Seedlings to Artificial Soil Acidification (인위적인 토양산성화에 대한 사방오리나무와 물오리나무 묘목의 생리반응 및 생장특성)

  • To obtain basic information for evaluating resistance of soil acidification for growth, net photosynthesis, $N_2$ fixation rate and nutrient status of Alnus firma and Alnus hirsuta seedlings grown in brown forest soil acidified with $H_2SO_4$ or $HNO_3$ solution were investigated (control (pH=5.9), LN (pH=5.0; Low levels of Nitric acid), HN (pH=4.0; High levels of Nitric acid), LS (pH=5.0; Low levels of Sulfuric acid), and HS (pH=4.0; High levels of Sulfuric acid)). The shoot dry weight of Alnus firma and Alnus hirsuta and the root dry weight of Alnus hirsuta seedlings grown in the HN, LS and HS were significantly less than that of the seedlings grown in the control. The Chlorophyll a/b ratio in leaves of Alnus firma at LN, LS and HS was significantly lower than that of control. The concentration of N in leaves of Alnus hirsuta at HS was significantly higher than that of control. The net photosynthetic rate of Alnus firma and Alnus hirsuta seedlings at LN and HN was higher than that of control. Based on the results, we concluded that the negative effects of soil acidification due to sulfate deposition are greater than those of soil acidification due to nitrate deposition on growth, net photosynthesis and $N_2$ fixation rate of Alnus firma more than Alnus hirsuta.

Proton Effect on the Degradation of Phenolic Compound by Activated Sludge and Nocardia asteroides (활성슬러지 혼합미생물과 Nocardia asteroides에 의한 페놀화합물 분해시 양성자이온의 영향)

  • This study was investigated to evaluate the effect of the sodium ion and pH on toxicity of dinitrophenol at high concentrations (0.41 to 0.54 mM), over a sodium concentration range of 0.1 mM to 107 mM and over a pH range of 5 to 9. The concentration of sodium ions in the activated sludge mixed liquor seemed to have very little effect on dinitrophenol toxicity. However, lack of sodium in the growth media resulted in a reduction of the dinitrophenol degradation rate by bacterial isolate from the activated sludge culture, which has been identified as Nocardia asteroides. Dinitrophenol inhibition was found to be strongly dependent on mixed liquor pH. The dinitrophenol degradation rate was highest in the pH range of 6.95 to 7.84; at pH 5.94 degradation of 75 mg/L dinitrophenol was significantly inhibited; at pH < 5.77, dinitrophenol degradation was completely inhibited after approximately 30% of the dinitrophenol was degraded.
