Stability of the growth process at pulling large alkali halide single crystals

  • Principles of a novel pulse growing method are described. The method realized in the crystal growing on a seed from melts under raw melt feeding provided a more reliable control of the crystallization process when producing large alkali halide crystals. The slow natural convection of the melt in the crucible at a constant melt level is intensified by rotating the crucible, while the crystal rotation favors a more symmetrical distribution of thermal stresses over the crystal cross-section. Optimum rotation parameters for the crucible and crystal have been determined. The spatial position oi the solid/liquid phase interface relatively to the melt surface, heaters and the crucible elements are considered. Basing on that consideration, a novel criterion is stated, that is, the immersion extent of the crystallization front (CF) convex toward the melt. When the crystal grows at a <> CF immersion, the raised CF may tear off from the melt partially or completely due to its weight. This results in avoid formation in the crystal. Experimental data on the radial crystal growth speed are discussed. This speed defines the formation of a gas phase layer at the crystal surface. The layer thickness il a function of time a temperature at specific values of pressure in the furnace and the free melt surface dimensions in the gap between the crystal and crucible wall. Analytical expressions have been derived for the impurity component mass transfer at the steady-state growth stage describing two independent processes, the impurity mass transfer along the <> path and its transit along the <> one. The heater (and thus the melt) temperature variation is inherent in any control system. It has been shown that when random temperature changes occur causing its lowering at a rate exceeding $0.5^{\circ}C/min$, a kind of the CF decoration by foreign impurities or by gas bubbles takes place. Short-term temperature changes at one heater or both result in local (i.e., at the front) redistribution of the preset axial growth speed.

갈색거저리 유충 추출물의 항산화 활성 및 모발 성장 촉진 효과 (The Antioxidant Activities and Hair-growth Promotion Effects of Tenebrio molitor Larvae Extracts (TMEs))

  • 최근 들어 곤충을 식품 및 바이오 소재로 이용한 연구가 활발히 진행되고 있다. 그러나 곤충을 이용한 모발 성장 효과에 대한 연구는 아직 미흡한 실정이다. 따라서 본 연구에서는 탈모 예방 및 모발 성장 효과를 가진 새로운 천연물 소재 개발을 위해 갈색거저리 유충 추출물의 항산화 활성 및 모발 성장 촉진 효과를 연구하였다. 갈색거저리 유충 추출물의 항산화 활성 평가를 위해서 DPPH 라디칼 및 아질산염 소거능을 측정하였다. 모발 성장촉진 효과를 측정하기 위해서는 인간 모유두세포(human dermal papilla cell)와 섬유아세포(fibroblast, NIH3T3 cell)를 이용하였으며 MTS assay를 통해 세포생존율 및 세포증식률을 측정하였다. 모유두세포에서 dihydrotesteone (DHT)에 의한 세포사 억제 효과를 확인하였으며, 섬유아세포에서는 tolbutamide (TBM)의 potassium channel blocker 역할에 의한 세포사 억제 효과를 확인하였다. DPPH radical 및 아질산염 소거능 측정 결과 갈색거저리 유충 추출물은 항산화 역할이 뛰어난 것으로 보고된 블루베리와 유사하거나 높은 정도의 항산화능을 가지는 것으로 확인되었다. In vitro 상에서 갈색거저리 유충 추출물을 48시간 동안 처리한 경우, 모유두세포와 섬유아세포의 세포증식을 218% 및 116%까지 증가시켰다. 또한, 모유두세포에서 DHT 처리에 의한 세포사가 갈색거저리 유충 추출물에 의해 억제되는 것을 확인하였으며, 섬유아세포에서는 potassium channel blocker인 TBM에 의해 세포생존율이 감소하였으나 갈색거저리 유충 추출물 처리 시 세포생존율이 정상군과 비슷한 정도로 회복되는 것을 확인하였다. 이상의 결과로부터 갈색거저리 유충 추출물을 이용한 모발성장 및 탈모방지 기능성 소재 개발 가능성을 확인하였다.

Curcumol Induces Apoptosis in SPC-A-1 Human Lung Adenocarcinoma Cells and Displays Anti-neoplastic Effects in Tumor Bearing Mice

  • Curcumol is a sesquiterpene originally isolated from curcuma rhizomes, a component of herbal remedies commonly used in oriental medicine. Its beneficial pharmacological activities have attract significant interest recently. In this study, anti-cancer activity of curcumol was examined with both in vitro and in vivo models. It was found that curcumol exhibited time- and concentration-dependent anti-proliferative effects in SPC-A-1 human lung adenocarcinoma cells with cell cycle arrest in the G0/G1 phase while apoptosis-induction was also confirmed with flow cytometry and morphological analyses. Interestingly, curcumol did not display growth inhibition in MRC-5 human embryonic lung fibroblasts, suggesting the anti-proliferative effects of curcumol were specific to cancer cells. Anti-neoplastic effects of curcumol were also confirmed in tumor bearing mice. Curcumol (60 mg/ kg daily) significantly reduced tumor size without causing notable toxicity. In conclusion, curcumol appears a favorable anti-cancer candidate for further development.

느타리버섯 재배 토양으로부터 분리한 Ochrobactrum anthropi A-1의 식물생장촉진효과 (Plant Growth Promotion Effect of Ochrobactrum anthropi A-1 isolated from Soil of Oyster Mushroom Farmhouse)

  • An auxin-producing bacteria (A-1) was isolated from soils of Oyster mushroom farmhouse in Daejeon city, South Korea. The strain A-1 was classified as a novel strain of Ochrobactrum anthropi based on a chemotaxanomic and phylogenetic analyses. The isolate was confirmed to produce indole-3-acetic acid (IAA), one of auxin hormones, by TLC and HPLC analyses. The maximum concentration of IAA, $5.6mg\;L^{-1}$ was detected from the culture broth of O. anthropi A-1 incubated for 24 h at $35^{\circ}C$ in R2A broth containing 0.1% L-tryptophan. To investigate the growth-promoting effects to the crops, the culture broth of O. anthropi A-1 was inoculated to water cultures and seed pots of mung bean as well as lettuce. In consequence, the adventitious root induction and root growth of mung bean and lettuce were 2.7 and 1.4 times higher than those of the non-inoculated, respectively.

Probiotic bacteria의 생장에 대한 막걸리슬러지의 이용 (Utilization of Makgeolli sludge for growth of probiotic bacteria)

  • A number of health benefits have been claimed for probiotic bacteria such as Bifidobacterium (B) spp. Lactobacillus(L) acidophilus, and Lactococcus(Lc) cremoris. Viability of probiotic bacteria is important in order to provide health benefits. Only a limited culture media for the test purpose of probiotic bacteria are commercially available (MRS broth), but the media for large-scale propagation of viable cells which are able to be used as food additive are not available. The manufacture of a low priced and preferred novel medium for probiotic bacteria was therefore, attempted using whey protein concentrate(WPC) and Makgeolli sludge as a starting material. The effect of WPC and Makgeolli sludge on the growth of four strains (B. bifidum 15696, B. longum 15707, L. acidophilus CH-2, and Lc. cremoris 20076) was investigated. Medium prepared such as WPC, Makgeolli sludge, and WPC+Makgeolli sludge(WPCMs). It was observed that the growth of 4 strains (B. bifidum 15696, B. longum 15707, L. acidophilus CH-2, and Lc. cremoris 20076) was stimulated by Makgeolli sludge, WPC, WPCMs. Especially, Viable cell number of 4 strains in the WPCMs were higher than that of the single media. These result suggest the possibility that Makgeolli and WPC, acts as a growth factor for the growth of probiotic bacteria.

Degradation analysis of horizontal steam generator tube bundles through crack growth due to two-phase flow induced vibration

  • A correct understanding of vibration-based degradation is crucial from the standpoint of maintenance for Steam Generators (SG) as crucial mechanical equipment in nuclear power plants. This study has established a novel approach to developing a model for investigating tube bundle degradation according to crack growth caused by two-phase Flow-Induced Vibration (FIV). An important step in the approach is to calculate the two-phase flow field parameters between the SG tube bundles in various zones using the porous media model to determine the velocity and vapor volume fraction. Afterward, to determine the vibration properties of the tube bundles, the Fluid-Solid Interaction (FSI) analysis is performed in eighteen thermal-hydraulic zones. Tube bundle degradation based on crack growth using the sixteen most probable initial cracks and within each SG thermal-hydraulic zone is performed to calculate useful lifetime. Large Eddy Simulation (LES) model, Paris law, and Wiener process model are considered to model the turbulent crossflow around the tube bundles, simulation of elliptical crack growth due to the vibration characteristics, and estimation of SG tube bundles degradation, respectively. The analysis shows that the tube deforms most noticeably in the zone with the highest velocity. As a result, cracks propagate more quickly in the tube with a higher height. In all simulations based on different initial crack sizes, it was observed that zone 16 experiences the greatest deformation and, subsequently, the fastest degradation, with a velocity and vapor volume fraction of 0.5 m/s and 0.4, respectively.

Low-Temperature Si and SiGe Epitaxial Growth by Ultrahigh Vacuum Electron Cyclotron Resonance Chemical Vapor Deposition (UHV-ECRCVD)

  • Low-temperature epitaxial growth of Si and SiGe layers of Si is one of the important processes for the fabrication of the high-speed Si-based heterostructure devices such as heterojunction bipolar transistors. Low-temperature growth ensures the abrupt compositional and doping concentration profiles for future novel devices. Especially in SiGe epitaxy, low-temperature growth is a prerequisite for two-dimensional growth mode for the growth of thin, uniform layers. UHV-ECRCVD is a new growth technique for Si and SiGe epilayers and it is possible to grow epilayers at even lower temperatures than conventional CVD's. SiH and GeH and dopant gases are dissociated by an ECR plasma in an ultrahigh vacuum growth chamber. In situ hydrogen plasma cleaning of the Si native oxide before the epitaxial growth is successfully developed in UHV-ECRCVD. Structural quality of the epilayers are examined by reflection high energy electron diffraction, transmission electron microscopy, Nomarski microscope and atomic force microscope. Device-quality Si and SiGe epilayers are successfully grown at temperatures lower than 600℃ after proper optimization of process parameters such as temperature, total pressure, partial pressures of input gases, plasma power, and substrate dc bias. Dopant incorporation and activation for B in Si and SiGe are studied by secondary ion mass spectrometry and spreading resistance profilometry. Silicon p-n homojunction diodes are fabricated from in situ doped Si layers. I-V characteristics of the diodes shows that the ideality factor is 1.2, implying that the low-temperature silicon epilayers grown by UHV-ECRCVD is truly of device-quality.

Comparative Genome-Scale Expression Analysis of Growth Phase-dependent Genes in Wild Type and rpoS Mutant of Escherichia coli

  • Numerous genes of Escherichia coli have been shown to growth phase-dependent expression throughout growth. The global patterns of growth phase-dependent gene expression of E. coli throughout growth using oligonucleotide microarrays containing a nearly complete set of 4,289 annotated open reading frames. To determine the change of gene expression throughout growth, we compared RNAs taken from timecourses with common reference RNA, which is combined with equal amount of RNA pooled from each time point. The hierarchical clustering of the conditions in accordance with timecourse expression revealed that growth phases were clustered into four classes, consistent with known physiological growth status. We analyzed the differences of expression levels at genome level in both exponential and stationary growth phase cultures. Statistical analysis showed that 213 genes are shown to, growth phase-dependent expression. We also analyzed the expression of 256 known operons and 208 regulatory genes. To assess the global impact of RpoS, we identified 193 genes coregulated with rpoS and their expression levels were examined in the isogenic rpoS mutant. The results revealed that 99 of 193 were novel RpoS-dependent stationary phase-induced genes and the majority of those are functionally unknown. Our data provide that global changes and adjustments of gene expression are coordinately regulated by growth transition in E. coli.

Characterization of dental phenotypes and treatment modalities in Korean patients with Parry-Romberg syndrome

  • Objective: To investigate the dental phenotypes and treatment modalities (Tx-Mod) in Korean patients with Parry-Romberg syndrome (PRS) using longitudinal data. Methods: The samples consisted of 10 PRS patients, who were treated and/or followed-up at Seoul National University Dental Hospital between 1998 and 2019. Using a novel PRS severity index based on the numbers of the atrophy-involved area and asymmetry-involved item, we classified them into mild (n = 3), moderate (n = 2), and severe (n = 5). Dental phenotypes, including congenitally missing tooth (Con-Missing-Tooth), microdontia, tooth with short root (Short-Root), tooth with dilacerated root, and delayed eruption/impacted tooth, were investigated along with Tx-Mod. Results: The side of occurrence of all dental phenotypes showed 100% concordance with the side of PRS involvement. The most two common dental phenotypes were Con-Missing-Tooth and Short-Root (n = 29 and n = 17 in six patients). The sums of the average number of Con-Missing-Tooth and Short-Root increased from mild PRS to moderate PRS and severe PRS cases (1.0, 6.0, and 6.2). In terms of Tx-Mod, growth observation due to mild atrophy, fixed orthodontic treatment, and grafting were used for mild PRS cases. Tx-Mod for moderate PRS cases involved growth observation for surgery due to an early age at the initial visit. For severe PRS cases, diverse Tx-Mod combinations including unilateral functional appliance, fixed orthodontic treatment, growth observation, grafting, and orthognathic surgery were used. Conclusions: The novel PRS severity index may be useful to provide primary data for individualized diagnosis and treatment planning for PRS patients.

Pig large tumor suppressor 2 (Lats2), a novel gene that may regulate the fat reduction in adipocyte

  • Clenbuterol, a $\beta_2$-adrenoceptor agonist, has been proven to be a powerful repartition agent that can decrease fat deposition. Based on results from our previous cDNA microarray experiment of pig clenbuterol administration, a novel up-regulated EST was full-length cloned (4859 bp encoding 1041 amino acids) and found to be the pig homolog of large tumor suppressor 2 (Lats2). We mapped pig Lats2 to chromosome 11p13-14 by using FISH, and western blotting demonstrated that pig Lats2 protein was most abundant in adipose. In Drosophila, Lats2 ortholog was reported as a key component of the Hippo pathway which regulates cell differentiation and growth. Here, we show that pig Lats2 exhibit inverted expression to YAP1, another member of the Hippo pathway which positively regulates cell growth and proliferation, during the differentiation of 3T3-L1 preadipocytes. Our results suggested that Lats2 may involve in Hippo pathway regulating the fat reduction by inhibiting adipocyte differentiation and growth.
