천파석(穿破石)이 콜라겐유도 생쥐관절염에 미치는 억제효과 (Inhibitory Effect of Cudraniae Tricuspidatae Radix on Collagen-induced Arthritis in Mice)

  • Objectives : The present study was conducted to examine the effect of Cudraniae Tricuspidatae Radix(CTR), which is the radix part of Cudrania tricuspidata Bureau, on cytokine secretion from the joint cells and spleen cells of mice with arthritis induced by collagen and verify its efficacy against rheumatoid arthritis. Methods : Three kinds of extract were prepared from CTR through extraction with hot-water and ethanol. The levels of cytokine secreted from the cells were measured, after the mice knee joint cells were cultured with each extract. Results : The three kinds of extracts from CTR decreased the growth rate and levels of inflammatory cytokines such as IL-$1{\beta}$, IL-6, and TNF-${\alpha}$ from knee joint cells of mice. All of the organic-soluble fraction, such as hexane, ethyl ether, ethyl acetate, butanol, and residual water-soluble fraction decreased the levels of IL-$1{\beta}$, TNF-${\alpha}$, IL-6, and IL-10. Conclusion : These results suggest that CTR had some effects on rheumatitis, with fat and water-solubles in organic solvent considered as the effective element.

In vitro antimicrobial effect of the tissue conditioner containing silver nanoparticles

  • PURPOSE. The aim of this study was to identify in vitro antimicrobial activity of the tissue conditioner containing silver nanoparticles on microbial strains, Staphylococcus aureus, Streptococcus mutans and Candida albicans. MATERIALS AND METHODS. Experimental disc samples ($20.0{\times}3.0$ mm) of tissue conditioner (GC Soft-Liner, GC cooperation, Tokyo, Japan) containing 0.1 - 3.0% silver nanoparticles (0%: control) were fabricated. Samples were placed on separate culture plate dish and microbial suspensions (100 ${\mu}L$) of tested strains were inoculated then incubated at $37^{\circ}C$. Microbial growth was verified at 24 hrs and 72 hrs and the antimicrobial effects of samples were evaluated as a percentage of viable cells in withdrawn suspension (100 ${\mu}L$). Data were recorded as the mean of three colony forming unit (CFU) numerations and the borderline of the antimicrobial effect was determined at 0.1% viable cells. RESULTS. A 0.1% silver nanoparticles combined to tissue conditioner displayed minimal bactericidal effect against Staphylococcus aureus and Streptococcus mutans strains, a 0.5% for fungal strain. Control group did not show any microbial inhibitory effect and there were no statistical difference between 24 hrs and extended 72 hrs incubation time (P > .05). CONCLUSION. Within the limitation of this in vitro study, the results suggest that the tissue conditioner containing silver nanoparticles could be an antimicrobial dental material in denture plaque control. Further mechanical stability and toxicity studies are still required.

길항세균 Pseudomonas fluorescens의 Complemented Mutant에 대한 특성조사에 및 길항물질 유전자 Cloning (Characterization of Complemented Mutants in Pseudomonas fluorescens and Cloning of the DNA Region Related in Antibiotic Biosynthesis)

  • Pseudomonas fluorescens produces the antibiotic, 2,4-diacetylphloroglucinol (Phl), which promotes plant growth by inhibiting bacteria and fungi. Cosmids (genomic library) were mobilized into Phl-nonproducing mutants through the triparental matings with pRK2013 as the helper plasmid at the frequency of 8.37$\times$10-4. Complemented mutants that showed antibiotic activity were selected among about 2,000 transconjugants. The complemented mutants were confirmed by acquired drug resistances (kanamycin and tetracycline). The antibiotic substances of wild type and complemented mutants showed the most excellent anti-bacterial activity. Inhibitory effects of complemented P. fluorescens against phytopathogenic fungi were equal to the parental strain. Complemented mutant and wild type of P. fluorescens were causal microbes of fungal morphological abnormalities. Complemented mutants in potato dextrose agar supplemented with bromothymol blue also showed restoration of glucose utilization as wild type. Plasmids of complemented mutants were isolated from transconjugant sand transformed into competent cells of E. coli DH5$\alpha$. The plamid DNA was reisolated from transformed E. coli DH5$\alpha$.

Evaluation of the Antibacterial Activity of Rhapontigenin Produced from Rhapontin by Biotransformation Against Propionibacterium acnes

  • Biotransformation is often used to improve chemical activity. We evaluated the antimicrobial activity of rhapontigeuin, converted from rhapontin after treatment with Pectinex. Rhapontigenin showed 4-16 times higher antimicrobial activity than rhapontin. The activity was higher against Gram-positive strains than Gram-negative strains. Minimum inhibitory concentrations (MICs) of rhapontigenin, retinol, and five antibiotics were determined by the microbroth dilution method for antibiotic-sensitive and -resistant Propionibacterium acnes. We also investigated the in vitro antibacterial activity of rhapontigenin in combination with antibiotic against antibiotic-resistant P. acnes. The antibiotic combination effect against resistant P. acnes was studied by the checkerboard method. The combination formulations (rhapontigenin and clindamycin, retinol and clindamycin) showed synergistic effects on the inhibition of the growth of clindamycin-resistant P. acnes. It is predictable that the combination of antibiotics with rhapontigenin is helpful to treat acne caused by antibiotic-resistant P. acnes. The antibacterial activity of rhapontigenin was enhanced by biotransformation.

위암을 유도시킨 흰쥐에서 한국 전통 식품이 위암 발생에 미치는 영향 (Effects of Some Korean Traditional Foods on Gastric Cancer Induced by Carcinogen in Rats)

  • Gastric cancer was inducedd by N-nethyl-N'-nitro-N-nitrosoguanidine(MNNG) in Fisher 344 male rats. Freeze dried typical Koran feeds, aged kimchi, soybean paste and maejoo, and partially scorched barbecued bulgokee were fed to the rat in the diet, which were composing 10% of the total diet. Experimental desing was as follows ; (1) group C : Control, (2) group M : MNNG with control diet, (3) group MK : MNNG with 10% of kimchi, (4) group MS : MNNG with 10% of soybean paste and maejoo, (5) group MB : MNNG with 10% of barbecued bulagokee, (6) group MKSB ; MNNG with 10% of the mixture of kimchi, soybean paste and maejoo, and barbecued bulgokee. Each group was fed with isocaloric diet for 26 weeks. Comparing to control, the growth rate of the experimental group was decreased after administration of MNNG and experimental diet. The mortality rate of group MB was increased by 17% than the control group along with a significant decrease of body weight. The protein efficiency ratio and the food efficiency ratio of group MB were lower than the control. The incidence of gastric cancer in rats fed kimchi and barbecued bulgokee were 73% and 75%, respectively, while that of group M which fed MNNG remained only 56%. On the contrary, soybean paste and maejoo showed an inhibitory effect on the burden of gastric tumor. However, the combination of kimchi, soybean paste and maejoo, and barbecued bulogokee showed a synergistic effect of increasing tumorigenesis in rats. Pathological observations of the rat stomach represent that squamous cell type tumors occupied in most frequencies.

  • We have developed a new process for the production of new structure oligosaccharides using the mixed-culture fementation of Lipomyces starkeyi KSM22 and leuconostoc mesenteroides B-512FMCM.L.starkeyi KSM22 produces a novel DXAMase(an enzyme containing both dextranase and amylase activities). It hydrolyzes the soluble starch and dextran. The hydrolyzates were used as acceptors for dextransucrase of L.mesenteroides to synthesize the new oligosaccharides(NOS). In fermentation, as the concentration of sucrose was increased from 9%(w/v) to 15%(w/v), the yields of dextran(sum of dextran I, MW=66kD, and dextran II, MW=21kD) was increased from 12.7% to 42.5%, and NOS was increased from 3.9% to 5.2% of the theoretical, respectively. The NOS of dp(degree of polymerization) 5 and over was increased from 33.1% to 58.3% of the total NOS. The NOS showed heat resistant up to 12$0^{\circ}C$ and was stable at pHs ranged from 2 to 6. The NOS decreased the pH changes in the culture of S. mutans, and also showed inhibitory effects on the growth of S. aureus or S. typhimurium.

Evaluation of Two Biologically Active Compounds for Control of Wheat Root Rot and its Causal Pathogens

  • The main aim of this study is to evaluate the efficiency of two biologically active compounds(Strom and F-760) in control of wheat root rot disease and its causal organisms. Fusarium graminearum, F. oxysporum, F. solani and Bipolaris sorokiniana were used as target organisms. In vitro, the two compounds showed fungicidal effect on all investigated pathogens resulted in suppression of radial growth and mycelial dry weight of them. Under greenhouse conditions, treatment of wheat grains with either Strom or F-760 before cultivation significantly reduced the percent of disease distribution as well as the mean disease rating of plants in both seedling and flowering stages. Fresh and dry weights of plants as well as water maintenance capacity were increased as the result of applying these compounds as seed dressing. Also data showed that the membrane stability of plants was injured as a result of infection with all investigated organisms, while this injury was alleviated when F-760 and Strom were applied. The $K^+$ efflux and the leakage of UV absorbing metabolites was stimulated with fungal infection. However, F-760 and Storm treatment partially retarded the stimulatory effect on leakage of $K^+$ and UV-absorbing metabolites of fungal infected plants. On the other side, the fungal infection had inhibitory effects on pigment fractions(chlorophyll a, b, and carotenoids) biosynthesis in wheat leaves. This retarding effect was partially or completely alleviated as the grains were treated with the applied compounds.

Screening of Korean Traditional Medicines and Medicinal Herbs for Antimicrobial Activity Against Pathogenic Bacteria

  • This study screened 76 Korean traditional patent medicines and 58 medicinal herbs for antimicrobial activity against pathogenic bacteria. Among the prescriptions methanol and hot water extracts of Samsoum, Chujunghwang-yont'ang, and Sashinhwan showed antimicrobial activities against E. coli and Chujunghwang-yont'ang showed the strongest antimicrobial activity. Among the medicinal herbs, hot water and methanol extracts of Terminalia chebula, Caesalpinia sappan, and Coptis chinensis showed the strongest antimicrobial activities against Escherichia coli O157, Staphylococcus aureus, Shigella flexnery, Shigella sonnei and Salmonella typhimurium. An interesting characteristic of the traditional medicines such as Samsoum, Chujunghwang-yont'ang, and Sashinhwan was that both methanol and hot water extracts were active. Chujunghwang-yont'ang exhibited the strongest antimicrobial activities against the microorganisms tested. Water and methanol extracts of Terminalia chebula, Caesalpinia sappan, and Coptis chinensis showed the strong of inhibitory effects on the growth of all microorganisms tested.

홍화자약침(紅花子藥鍼)의 암전이 억제와 면역 활성화에 관한 연구 (The Study on Anti-Metastasis and Immune Activation of Carthami Semen Herbal Acupuncture)

  • Objectives : This study was guided to estimate inhibitory effects against hepatic metastasis and activation of immunocytes by Carthami Semen Herbal Acupuncture. Methods : Colon26-L5 carcinoma cells were injected through hepatic portal vein to induce hepatic metastatic cancer. Changes in weight, morphology of the cancer were estimated and cytokine level was analyzed to produce immunological changes. Results: The experimental group was significantly decreased metastatic size and proliferation compared with the control group. Observing immunocytes from the spleen of experimental group T-lymphocytes were significantly increased. As compared with control group, the differentiation of total T cell, helper T cell, cytotoxic T cell, Macrophage and NK cell was increased. Measuring the level of cytokine $IFN-{\gamma}$ which stimulates Th 1 was significantly increased in splenocyte with Con A stimuli. $IFN-{\gamma}$ and IL-12 were significantly increased in peritoneal exudate macrophage with LPS stimuli. Conclusion : These findings strongly indicate Carthami Semen Herbal Acupuncture improves immunity to inhibit the growth of cancer and metastasis.

Apoptosis of Colorectal Cancer UTC116 Cells Induced by Cantharidinate

  • Effects of Cantharidinate on apoptosis of human colorectal cancer UTC-116 cells were investigated by means of 3-(4, 5-dimethylthiazol-2-yl)-2, 5-diphenyltetrazolium bromide (MTT) assay, H and E staining, flow cytometry, and Raman Spectra analysis. The results showed Cantharidinate to exert inhibitory action on proliferation of human colorectal cancer UTC-116 cells, inducing apoptosis, arresting cells in G1 phase, with decline of S and G2 phases. In addition, the results of Raman spectrum showed significant changes in the UTC-116 cells chemical structure with stretching after the application of Cantharidinate. Taken together, these results suggest that the treatment of human colorectal cancer with Cantharidinate may be associated with multiple molecular mechanisms for apoptosis. Furthermore, similar to fluorouracil, Cantharidinate should be considered as novel assistant drug for controlling the growth of human colorectal cancer UTC-116 cells.
