To produce bioactive compound enriched yeast using medicinal Gugiga (Lycium chinensis Mill), several edible Saccharomyces species were cultured in Gugija extracts added yeast extract, peptone and dextrose medium (GE - YEPD medium) at $30^{\circ}C$ for 24 hr, and their growth were determined. Growth of Saccharomyces cerevisiae K-7 and Sacchromyces cerevisiae ACTC 7904 were better than those of the other yeasts. Two yeasts were selected and then determined their some physiological functionalities after cultivated the yeasts in the GE - YEPD medium and compared those grown on YEPD medium. Antihypertensive angiotensin I-converting enzyme (ACE) inhibitory activity of S. cerevisiae K-7 grown on GE - YEPD medium was about 20% higher than that grown on YEPD medium. Superoxide dismutase-like activity of S. cerevisiae ACTC 7904 was also about 12% more high. However, the other physiological functionalities were almost same or lower. Optimal addition concentration of Gugija extract was 10%, and maximally growth and ACE inhibitory activity of S. cerevisiae K-7 were shown when the strain was cultured in 10% Gugija extracts containing YEPD medium at $30^{\circ}C$ for 12 hr.

An attempt was made to verify the antibacterial activities of the soluble extracts of Gardenia jasminoides(SEGJ) against mycobacteria other than tubercle bacilli (MOTT); M. kansasii, M. scrofulaceum, M. simiae, M. szulgai, M.xenopi, M. avium-intracellulare complex, M. gordonae and M. flarescens. In addition, the effect of the SEGJ on the yellow pigmentation and violet pigmentation (crocin reaction) of the colonies of the MOTT grown on7H10-crocin agar plates was observed. The SEGJ gave a growth stimulatory activity against most species of mycobacteria except for M. szulgai at low concentration of the SEGJ in terms of crocin pigment OD=0.02 or less, but exerted an inhibitory activity at high concentration of OD=0.04 or more. The growth-inhibitory activity of the SEGJ was dose-dependent but the yellow pigmentation of the scotochromogens was not affected by the dose of the SEGJ that was growth-inhibitory. Crocin positive reaction was observed only with M. kansasii.

The antimicrobial effect of a novel flavonoid (7-O-butyl naringenin) on Helicobacter pylori ATCC 26695 and its inhibitory effects on the urease activity of the strain were evaluated by comparing with quercetin and naringenin. H. pylori was cultured with brain heart infusion supplemented with 5% horse serum at $37^{\circ}C$ under 10% $CO_2$ atmosphere and the inhibitory effects of flavonoids against the strain were detected using micro-plate methods. During 12 hr of incubation time, the optical densities of phenol red reduced (pink color) in the urea broth by producing ammonia were detected at 560 nm with a spectrophotometer. The results indicated that both quercetin and 7-O-butyl naringenin were effective against the growth of H. pylori. Moreover, inhibitory effect of 7-O-butyl naringenin on the growth of H. pylori was about two-fold higher than quercetin at the same concentration. With regard to H. pylori urease activity, 7-O-butyl naringenin had a greater inhibitory effect than did naringenin or quercetin at the same concentration.

Helicobacter pylori were found to be resistant to azide but sensitive to vanadate, suggesting that defect in the P-type ATPase activity rather than F-type ATPase would be lethal to cell survival or growth. To elucidate the relationship between this enzyme inhibition and H. pylori death, we determined the effect of omeprazole (OMP) plus vanadate on enzyme activity and cell growth. The minimum inhibitory concentration (MIC; ca. 0.8$\mu$mol/disk) of vanadate for H. pylori growth was lowered over l0-fold with the aid of OMP, whereby its inhibitory potential toward the P-type ATPase activity was diametrically increased. Alternatively, we found that this enzyme activity was essential for active transport in H. pylori. From these observations, we strongly suggest that the immediate cause of the growth inhibition of H. pylori cells with OMP and/or vanadate might be defective in the cell's active transport due to the lack of P-type ATPase activity. From the spectral data with circular dichroism (CD) spectroscopy, we found that activated OMP (OAS) at concentration below MIC did not disrupt helical structures of membrane proteins. Separately, we determined the cytopathic effect of OAS by SDS-PAGE, indicating the change in the production of cytoplasmic protein but not cell membrane.
The insecticidal and antibacterial activity of new synthesized carbamate derivatives(8-Hydroxy-5-chloroquinolyl-N-methylcarbamate(I), 8-Hydroxy-5-chloroquinolyl-N-ethylcarbamate(II)) was examined using 0.2 w/v% acetone solutiolas and 50 r/ml~1000 r/m N,N'-dimethylformamide-$H_2O$(2:3) solutions of each compounds, respectively. 1) Two carbamates exerted insecticidal effects on Sogata furcifera HORVATH, Delphacodes Striatella FAUEN, Whereas no significant effects were observed on the Nilaparvate lugens STAL, Inazuma dorasails MOISCHIULSKY and Nephateffix apicalis Cincticeps UHLER. 2) These compounds exhibited growth-inhibitory activity against Staphylococcus aureus, Salmonella paratyphi A, Shigella dysenteriae 1a, Escherichia coli NL 1401,at the concentration range of 100~500 r/ml in general.
This study was designed to identify the effects of Reynoutria Japonica on antibacterial activity aganist Bordetella pertussis ATCC 9797 which is cause of whooping cough. The ethanol- and water-extracts of more than 80 oriental herbal medicine were Investigated by Kirby -Bauer method to determine their inhibitory effects on growth of B. pertussis ATCC 9797 in vitro. For that Reynoutria Japonica was selected. The ethanol-soluble extract of Reynoutria Japonica showed relatively high antivacterial activity against B. pertussis ATCC 9797. However, the water-soluble extract of Reynoutria Japonica showed no antibacterial activity. The ethanolic extract was further fractionated with organic solvents such as hexane, chloroform, and ethyl acetate, in that order. Among the fraction tested, the chloroform fraction showed the highest antibacterial activity when the ethanol-soluble extract of Reynoutria Japonica minial inhibitory concentration(MIC) was $25{\mu}g/m{\ell}l$. Results support a role far Reynoutria Japonica in inhibiting the cell growth of B. pertussis ATCC 9797, but further experimentation is required.

Resveratrol (3,5,4’-trihydroxy-trans-stilbene), a naturally occurring phytoallexin abundant in grapes and several plants, has been shown to be active in inhibiting proliferation and inducing apoptosis in several human cancer cell lines. On the line of the biological activity of resveratrol, a variety of resveratrol analogs were synthesized and evaluated for their growth inhibitory effects against several human cancer cell lines. In the present study, we found that one of the resveratrol analogs, 3,4,5-trimethoxy-4’-bromo-cis-stilbene, markedly suppressed human colon cancer cell proliferation (EC$_{50}$ = 0.01 ${\mu}$g/ml), and the inhibitory activity was superior to its corresponding trans-isomer (EC$_{50}$ = 1.6 ${\mu}$g/ml) and resveratrol (EC$_{50}$ = 18.7 ${\mu}$g/ml). Prompted by the strong growth inhibitory activity in cultured human colon cancer cells (Col2), we investigated its mechanism of action. 3,4,5-Trimethoxy-4’-bromo-cis-stilbene induced arrest of cell cycle progression at G2/M phase and increased at sub-G1 phase DNA contents of the cell cycle in a time- and dose-dependent manner. Colony formation was also inhibited in a dose-dependent manner, indicating the inhibitory activity of the compound on cell proliferation. Moreover, the morphological changes and condensation of the cellular DNA by the treatment of the compound were well correlated with the induction of apoptosis. These data suggest the potential of 3,4,5-trimethoxy-4’-bromo-cis-stilbene might serve as a cancer chemotherapeutic or chemopreventive agent by virtue of arresting the cell cycle and inducing apoptosis for the human colon cancer cells.
This study verifies the polyphenol and flavonoid contents of a dried extract, as well as its antioxidant effect and growth inhibitory effect on cancer cells to investigate the potential of yellow cherry tomatoes as a physiologically active food material. The polyphenol and flavonoid contents were determined as 10.96 ± 1.57 and 4.12 ± 0.41 mg/g, respectively. The antioxidant activity was confirmed by measuring DPPH and ABTS radical scavenging ability, and RC50-the concentration that reduces free radicals by 50%-were determined as 490.83 ± 17.35 ㎍/mL and 355.90 ± 0.79 ㎍/mL, respectively. The dried extract showed no cytotoxicity with respect to normal hepatocytes (Chang) and no growth inhibitory activity with respect to A549 lung cancer cells, whereas dried extract showed growth inhibitory activities of 15.2% and 18.4% with respect to human cervical adenocarcinoma (HeLa) and human hepatocellular carcinoma (HepG2) cells, respectively, when treated with a concentration at 100㎍/mL. The results of this study confirm the potential of yellow cherry tomatoes as a physiologically active food material by verifying their antioxidant activity and their growth inhibitory activity with respect to cervical and liver cancer cells.
