Evaluation of Microbial Load in Oropharyngeal Mucosa from Tannery Workers

  • Background: Animal skin provides an ideal medium for the propagation of microorganisms and it is used like raw material in the tannery and footware industry. The aim of this study was to evaluate and identify the microbial load in oropharyngeal mucosa of tannery employees. Methods: The health risk was estimated based on the identification of microorganisms found in the oropharyngeal mucosa samples. The study was conducted in a tanners group and a control group. Samples were taken from oropharyngeal mucosa and inoculated on plates with selective medium. In the samples, bacteria were identified by 16S ribosomal DNA analysis and the yeasts through a presumptive method. In addition, the sensitivity of these microorganisms to antibiotics/antifungals was evaluated. Results: The identified bacteria belonged to the families Enterobacteriaceae, Pseudomonadaceae, Neisseriaceae, Alcaligenaceae, Moraxellaceae, and Xanthomonadaceae, of which some species are considered as pathogenic or opportunistic microorganisms; these bacteria were not present in the control group. Forty-two percent of bacteria identified in the tanners group are correlated with respiratory diseases. Yeasts were also identified, including the following species: Candida glabrata, Candida tropicalis, Candida albicans, and Candida krusei. Regarding the sensitivity test of bacteria identified in the tanners group, 90% showed sensitivity to piperacillin/tazobactam, 87% showed sensitivity to ticarcillin/clavulanic acid, 74% showed sensitivity to ampicillin/sulbactam, and 58% showed sensitivity to amoxicillin/clavulanic acid. Conclusion: Several of the bacteria and yeast identified in the oropharyngeal mucosa of tanners have been correlated with infections in humans and have already been reported as airborne microorganisms in this working environment, representing a health risk for workers.

  • The intention of this research was to investigate the solubilization of primary sewage sludge using lactic acid bacteria cultured in a glucose and yeast extract medium. Glucose as the carbon source and yeast extract as the source of nitrogen were chosen as an economic medium with the potential for the mass production of lactic acid bacteria. The optimal concentrations of the medium were 3% (w/v) glucose and 2% (w/v) yeast extract. In this study, in order to improve field applications for the solubilization of sludge at sewage treatment plants, a powdered form of lactic acid bacteria was produced. The optimal inoculum of the powder for the maximum efficiency of solubilization was 1% (w/v). In that condition, the SCOD value increased from 8600 (mg/L) at the beginning of experiment to 10290 (mg/L) at 96 h, with the highest solubilization rate (20.6% DDCOD) and 11.2% (SCOD). Also, the TVFAs of the lactic acid bacteria inoculation group were produced more than that of the control group. In particular, acetic acid was produced 5 times more in the experimental group than in the control group. In this research, the potential of lactic acid bacteria in the pretreatment of primary sewage sludge as a solubilizer, and as an energy source producer for microbial fuel cells was revealed.

市販豆腐 成分 및 細菌汚染에 관한 硏究 (A Study on Ingredient and Bacterial Contamination of Bean Curd on the Market)

  • The purpose of this study was to analyze general ingredient of bean cured on the market and to examine its exposure to bacterial contamination. For this study, 17 samples (each 9 samples for general bean curd, soft bean curd, and uncurdled bean curd) were randomly collected from nine areas in Seoul from the beginning of April, 1983 to the beginning of June, 1983. The result of ingredient analysis of moisture, ash, and protein of bean curd was compared with the standard set by the Ministry of Health & Social Affairs. In order to find out exposure of bean curd on the market to bacterial contamination, total biological bacteria and coliform group were examined. Experimental results were shown as follows 1) Results of ingredient analysis of moisture, ash, and protein of general bean curd showed that total samples in both moisture and protein met the standard set by the Ministry of Health & Social Affairs but 44.4% of the samples in ash was below the above standard, indicating average 82.0%, 0.9% and 9.6% in moisture, ash, and protein order. 2) Experimental results of moisture, ash, and protein of soft bean curd demonstrated 90.2%, 0.5% and 4.3% respectively total samples in both moisture and protein satisfied the self-criteria set by the Soft Food Co-operative Association of Seoul City but 11.1% of the samples in ash didn't meet the self-criteria. 3) Total samples of uncurdled bean curd satisfied the self-criteria set by the above association, indicating average 92.0%, 0.4%, and 3.5% in moisture, ash, and protein order. 4) Total biological bacteria and coliform group detected in general bean curd showed that more than 10$^5$/g in total biological bacteria accounted for 88.8% of the samples and that 10$^4$/g or more in coliform group accounted for 77.7% of the samples. The result proves that general bean curd has been exposured to a severe bacterial contamination. 5) Result of total biological bacteria and coliform group detected in such packed bean curd as uncurdled bean curd and soft bean curd showed that 61.6% of the samples exceeded 10$^6$/g in total biological bacteria and 27.7% of the samples exceeded 10$^3$/g in coliform group. 6) According to the change with time and temperatures in total biological bacteria and coliform group of general bean curd, general bean curd began to decay around 72 hours at 4$\circ$C and around 48 hours at 23$\circ$C and around 24 hours at 37$\circ$C and, at that time, total biological bacteria approached 10$^6$/g while coliform group did 10$^6$/g. The result indicates that temperature has a great effect on bacteria counts and decay.

Photodynamic Therapy for Methicillin-resistant Staphylococcus aureus Using Various Photosensitizer

  • The aim of this study was to evaluate the photodynamic effect of various photosensitizing agents against methicillin-resistant Staphylococcus aureus (MRSA). MRSA was exposed to light from a 632 urn diode laser (15 J/$cm^2$) in the presence of various photosensitizer, such as photofrin, photogem, radachlorine and ALA. In vivo study was performed using ICR mice. Twenty eight mice had a standard wound ($100\;mm^2$) created on the dorsum, and MRSA was inoculated into the wound region. The four groups were classified as follows: (1) the untreated control group (bacteria alone), (2) the bacteria plus light group (15 J/$cm^2$), (3) the bacteria plus photofrin group (kept in the dark), and (4) the photodynamic therapy (PDT) group (bacteria, photofrin, and light). After photofrin (dose 1 mg/kg) injection, the experimental group was irradiated with 632 urn diode laser (15 J/$cm^2$) for 30 minutes after In vitro results of PDT showed the complete killing of MRSA at the photofrin, radachlorine, and photogem However, ALA-PDT was ineffective on MRSA viability. In vivo results showed that photofrin has therapeutic effect on the wound infection. These results demonstrate that selective lethal photosensitization of MRSA can be achieved using phofrin, photogem and radachlorin. Thus, PDT can inactivate MRSA survival.

안동식혜의 저장 안정성에 관한 연구 (Studies on the storage stability of Andong Sikhe)

  • This study was carried out to determine optimal conditions in storing Andong sikhe. We made Andong sikhe according to the traditional recipe, and fermentation it has been stored either with (group B) or without (group A) "Saengkiwon Songi" for 40 days at 7${\pm}$2$^{\circ}C$. The changes of pHs, sweetness, the number of total bacteria and lactic acid producing bacteria, the quantities of organic acids and free sugars produced had been measured regularly since the 3rd day after fermentation. The results are as follows: The pH of both groups were 5.63 immediately after fermentation, and then gradually decreased to 4.02∼4.05. The sweetness was higher in group A (4.0) until the 15th day of storage, and then the same (17.5) in both groups. The sample from the 6th day in group A, and the samples from the 3rd and the 9th day in group B obtained the highest scores in their sensory evaluation. The numbers of total bacteria and lactic acid producing bacteria showed maxium on the 3rd day in both groups. 8 kinds of organic acids were detected, lactic acid being the most of all. 3 kinds of free sugars - fructose, glucose and maltose - were detected ; glucose and maltose have gradually increased throughout the storage period but fructose was not been detected after the 6th day in both groups.

Bacillus subtilis from Soybean Food Shows Antimicrobial Activity for Multidrug-Resistant Acinetobacter baumannii by Affecting the adeS Gene

  • Exploring novel antibiotics is necessary for multidrug-resistant pathogenic bacteria. Because the probiotics in soybean food have antimicrobial activities, we investigated their effects on multidrug-resistant Acinetobacter baumannii. Nineteen multidrug-resistant A. baumannii strains were clinically isolated as an experimental group and 11 multidrug-sensitive strains as controls. The growth rates of all bacteria were determined by using the analysis for xCELLigence Real-Time Cell. The combination of antibiotics showed synergistic effects on the strains in the control group but no effect on the strains in the experimental group. Efflux pump gene adeS was absent in all the strains from the control group, whereas it exists in all the strains from the experimental group. Furthermore, all the strains lost multidrug resistance when an adeS inhibitor was used. One strain of probiotics isolated from soybean food showed high antimicrobial activity for multidrug-resistant A. baumannii. The isolated strain belongs to Bacillus subtilis according to 16S RNA analysis. Furthermore, E. coli showed multidrug resistance when it was transformed with the adeS gene from A. baumannii whereas the resistant bacteria could be inhibited completely by isolated Bacillus subtilis. Thus, probiotics from soybean food provide potential antibiotics against multidrug-resistant pathogenic bacteria.

  • The purpose of study was to confirm theory about the effectiveness of routine mental care on the reduction of catheter-associated urinary tract infection. The study was carried out at a university hospital from September 1,1987 to April 17, 1989 : 32 Patients with a foley -catheter were studied. The study compared the urinary tract infection rate of an experimental group with that of a control group and tested the antibiotic susceptibility of the isolated bacteria. The experimental group(16 patients) was given daily meatal care with 10% Betadine for periods ranging from 4 to 21 days. The control group(16 patients) was not given that care. The results obtained were as follows : 1. The urinary tract infection rate of the experimental group was 50.0 %, and that of the control group 43.8%. There was no significant difference between the groups. 2. Organisms isolated in the control group were bacteria 100%, and in the experimental group bacteria 50% and fungus 50%. The most common organisms of the 15 strains isolated in the total group were Staphylococcus coagulase negative (3 patients), and E-coli (3 patients). 3. Most of bacteria isolated in this study were sensitive to Norfloxacillin, but resistant to Ampicillin, Chloramphenicol, Kanamycin, Tetracycline, and Erythromycin. Hence the importance of controling catheter-associated urinary tract infections. Findings suggest the need to search for other sources of infection, further experimentation controling various sources of urinary tract infection and larger group of subjects.

단일 3차 의료기관에 내원한 당뇨병성 족부병변 환자의 창상 배양검사를 통한 세균 검출 현황 (The Current Status of Bacterial Identification by Wound Culture for Diabetic Foot Lesions in a Single Tertiary Hospital in South Korea)

  • Purpose: The present study aimed to develop guidelines regarding initial choice of antibiotics for diabetic foot ulcers (DFU) by investigating bacterial isolates. Materials and Methods: This study included 223 DFU patients that visited a single tertiary hospital and underwent bacterial culture between January 2016 and February 2020. The study was conducted in two parts: 1) to compare bacterial isolates and wound healing according to comorbidities such as chronic kidney disease (CKD) and peripheral artery disease (PAD), and 2) to compare bacterial isolates according to wound depth using the Wagner classification. Results: Of the 223 patients, 43 had CKD (group A), 56 had PAD (group B), 30 had CKD and PAD (group C), and 94 had none of these comorbidities (group D). The isolation rate for multidrug-resistant gram-negative bacteria (MRGNB) and gram-negative to gram-positive bacteria ratio were highest in group C (p=0.018, p=0.038), and the proportion that achieved wound healing was lowest in group C (p<0.001). In the second part of the study, subjects were classified into 5 grades by wound depth using the Wagner classification; 13 grade I, 62 grade II, 60 grade III, 70 grade IV, and 17 grade V. No significant difference was observed between these grades in terms of isolation rates or gram-negative to gram-positive bacteria ratios. Conclusion: This study suggests antibiotics that cover gram-negative bacteria including MRGNB produces better results in the presence of CKD and PAD and that initial antibiotic choice should be based on the presence of CKD and PAD rather than wound depth.

팔당호에서 Aggregates에 부착한 세균군집구조의 변화 (The Change of Attached Bacterial Community on Aggregates in Lake Paldang)

  • 팔당호에서 fluorescent in situ hybridization(FISH) 방법을 이용하여 aggregates에 부착한 세균군집의 변화를 조사하였다. 조사대상은 Eubacteria에 속하는 세균과 Class Proteobacteria에 속하는 세균중 $\alpha$-, $\beta$-, $\gamma$ -group과 Cytophaga-Flavobacterium group이었고, 환경요인의 변화를 파악하고자 영양염류와 엽록소 a를 측정하였다. Aggregate와 물시료의 조사항목을 비교하면, TN의 경우 5~15배, TP는 81~140배, 엽록소 a는 49~66배로 aggregate가 높게 나타났으며, 총 세균수 역시 물시료에서 전체적으로 1.0~$2.0{\times}10^6$cells.$ml^{-1}$이었고, aggregates 부착세균에서 0.2~~$3.6{\times}10^8$cells.$ml^{-1}$의 범위로 물시료보다는 aggregates에 부착한 세균의 밀도가 200배 높았다. 또 수심별로는 5m trap 보다 20m trap에서 더 많은 수가 측정되었다. 총 세균수에 대한 세균군집구조의 비율은 부유세균의 경우 $\alpha$-group이 4.5~8.3%, $\beta$-group이 2.2~8.0%, ${\gamma}$-group이 2.1~7.4%, Cytophaga-Flavobacterium group이 2.1~6.1% 'Other'group은 0.1~2.5로 매우 낮았으나 aggregate에 부착한 세균의 군집구조는 $\alpha$-, $\beta$-, $\gamma$-group과 Cytophaga-Flavobacterium group이 아닌 'Other'group이 약 10.2~32.1%로 우점하는 경향을 보였다. 이처럼 팔당호에서 aggregates에 부착한 세균의 군집구조는 부유세균과 비교해볼 때 독특한 군집구조를 나타내었다.

The Bacterial Community of Southern Lake Baikal in Winter

  • The bacterial abundance, proportion of respiring bacteria, and bacterial community of southern Lake Baikal were analyzed at 1 m and 400m depths during winter. The total bacterial numbers were 5.1${\times}$105 cells ml-1 at 1 m and 2.5${\times}$105 cells ml-1 at 400 m depth, which are about half and quarter of the numbers of other lakes. The proportion of respiring bacteria was as low as 2.5% at 1 m and 1.4% at 400 m depth. Considering the amount of organic carbon which need to be degraded and low proportion of respiring bacteria, the bacteria could be assumed to have high activities. The EUB/DAPI ratios were 77 and 89% at 1 m and 400 m depths, respectively. Of the bacterial community, the other group was dominant at both depths, and gamma group of protebacteria followed next. But the beta group of proteobacteria and Cytophaga-Flavobacterium groups occupied very small proportions.
