The Effects of Vero Cell Co-culture on Mouse Embryo Development (Vero Cell과의 공동배양이 체외에서 생쥐 배아발생에 미치는 영향)

  • Embryos of most mammalian species grown in vitro would undergo developmental arrest at the approximate time of genomic activation. Stage-specific cell block and the resulting rapid loss of embryo viability in conventional culture media have limited the duration for which embryos may be cultured prior to transfer. As a result, embryos are usually transferred to the uterus at the 4-to 8-cell stage to avoid the loss of viability associated with long-term in vitro culture. Early transfer has led to asynchrony of the endometrium-trophectoderm interaction at the time of implantation and a resultant reduction in the rate of implantation. To overcome these problems, a variety of co-culture systems has been devised in which embryos can develop for a longer period prior to embryo transfer. Vero cells, derived from African green monkey kidney, share a common embryologic origin with cells from the genital tract. In addition, they are potentially safe to use, since they are highly controlled for viruses and other contaminants. Therefore, co-culture using Vero cells has been widely utilized to enhance embryo viability and development, although not without controversies. We thus designed a series of experiments to demonstrate whether Vero cells do indeed enhance mouse embryo development as well as to compare the efficacy of co-culturing mouse 1-cell embryos on Vero cell monolayer in both Ham's F-10 and human tubal fluid (HTF) culture media. 1-cell stage ICR mouse embryos were cultured either in the presence of Vero cells (Group A) or in conventional culture medium alone (Group B). In Ham's F-10 significantly more 3-to-8cell embryos developed in group A than group B (59.8 versus 10.0%; p<0.01). In contrast, there was no significant difference in embryonic development both group A and group B in HTF. However, significant differences were noted only in later embryonic stage (13 and 0%; p<0.05 of group A and B respectively, hatching or hatched). In Ham's F-10, we also could observe the beneficial effect of Vero cell on hatching process (70.7 and 42.1%; p<0.05 of group A and group B respectively).

  • In ornamental crops, the color and shape of flowers are one of the important traits. Generally, flower colors are determined by accumulating pigments such as carotenoids, flavonoids, and betalains. Among them, flavonoids are responsible for broad ranges of colors. Chrysanthemums are one of the most popular ornamental crops in the world, and there have been many efforts to change their flower color. In chrysanthemum flowers, cyanidin-based anthocyanin confers pink or red color, whereas terpenoid-based carotenoids are mainly responsible for yellow and green colors. However, blue colored chrysanthemums do not occur in nature. To date, there have been attempts to obtain blue or violet-colored chrysanthemum flowers through the introduction of a novel gene for accumulating delphinidin-based anthocyanins, while other studies have reported changing endogenous metabolites through the reconstruction of flavonoid biosynthesis. Since various transcription factors are involved in the regulation of flavonoid biosynthesis, it is important to understand not only the structural genes, but also the transcription factors required for the modification of flavonoid-based flower color. Therefore, in this paper, we describe the flavonoid biosynthetic pathway and its regulation, and review previous studies on the change in flower color through modification of flavonoid biosynthesis. This effort could be an important milestone in successfully achieving the modification of chrysanthemum flower color by means of plant biotechnology.

The Evaluation of Adaptability of Three Ever-green Perennial Herbaceous South Korea Native Plants under In-door Light Intensities

  • Though there is an on-going need for the diversity of ornamental plants for in-door environment, their growth and stress adaptability in comparatively low light intensity condition require further studies for implementation. Here investigates the growth and chlorophyll fluorescence parameters of Farfugium japonicum (L.) Kitam, Liriope muscari (Decne.) L.H.Bailey and Acorus gramineus Aiton under several light intensities which were based on common in-door environment. The growth measurement of the plants included the quantity, length, width and SPAD value of the leaves. Calculated values of Fm/Fo, Fv/Fm, Pi_Abs, ETo/RC and DIo/RC were used as the parameters of the chlorophyll fluorescence under 10, 50, 100 and 200 PPFD (μmol·m-2·s-1) light intensities. In-door plants group was put in a closed room allowing no other light sources for 10 weeks and the control group was put in glass-greenhouse for the same period. The overall in-door growth of L. muscari was not significantly different in all light intensities compared to the control group and even showed the higher SPAD values. Also, an increasing tendency of Pi_Abs value under 10 to 100 PPFD was observed implying that L.muscari could adapt well to in-door environment. Measurement of A. gramineus growth mostly showed the highest values in the control group especially in the number of the leaves. Nevertheless, chlorophyll fluorescence parameters showed no significant value difference between in-door and the control groups and thus, A. gramineus might have possibility of successful adaptation to in-door environment. F. japonicum showed deficient growth in plant height and leaf length compared to the control but, it seemed to be able to sustain ornamental value under in-door light intensities. Furthermore, Pi_Abs and DIo/RC values were increased under in-door light conditions suggesting potential adaptability of F. japonicum.

Sasa quelpaertensis Leaf Extracts Induce Apoptosis in Human Leukemia HL-60 Cells

  • In east Asia, the leaves of various Sasa species have been used in folk medicine for centuries. The effects of the methanolic extract and its subsequent fractions derived from the leaves of Sasa quelpaertensis Nakai on cell proliferation in human leukemia HL-60 cells were evaluated. The ethyl acetate fraction of this extract (ESQL) significantly reduced cell viability in a dose-dependent manner ($0-250\;{\mu}g/mL$). ESQL ($IC_{50}=24.8\;{\mu}g/mL$) exhibited growth inhibition comparable to the main constituent of green tea, epigallocatechin ($IC_{50}=26.2\;{\mu}g/mL$), which was used as a positive control. ESQL treatment induced apoptosis, which was confirmed by the presence of nuclear condensation and annexin V-staining. These results demonstrate that ESQL contains chemopreventive phytochemicals that may be useful in neutraceutical applications.

Protection of Green Leafy Vegetable Extracts Against Oxidation of Human Low Density Lipoprotein

  • Oxidation of low density lipoprotein (LDL) is regarded to play an important role in the development of atherosclerosis. In the present study, salad vegetables with a remarkable DPPH radical-scavenging activity were extracted with methanol, and the methanol extracts were evaluated for the inhibition of $Cu^{2+}$-induced oxidation of human LDL. Separately, the amount of total phenolics was determined colorimetrically using Folin-Ciocalteu reagent. The vegetable extracts, expressing a strong inhibition of LDL oxidation ($IC_{50}$ values, <$100\;{\mu}g/mL$), were from angelica, dandelion, mustard leaf, and water spinach, which contained relatively high level of polyphenol content. Noteworthy, a highly positive correlation was observed between inhibition of LDL oxidation and amount of total polyphenol (p<0.01). Based on these results, it is suggested that salad vegetables, especially angelica, dandelion, and mustard leaf, may be used as easily accessible sources of natural antioxidants, especially in anti-atherosclerosis.

  • Since 1970, concentrated development in Gang-nam caused serious differences with Gang-buk, such as housing environment, education, culture, industry, economy, and consumption behavior as well as Urban Infrastructure. In this result, the government has executed New Town project to solve these matters, but the residents' opinion wasn't reflected. The purpose of this study is to research New Town project district residents' consciousness and prefered image for housing and this research will be applied to basic study for housing environment improvement policy in future. The results of this study are summerized as follows : First, living satisfaction is revealed almost low level, and the most demands for improvement are to solve lacks of park and green yard. Second, image of Gang-nam, which of housing environment is better than Gang-buk, is expressed as the words, 'Complicated', 'Complex', 'Polished', 'Convenient', 'Rich'. Third, prefered types of housing are high-rise Residental Complex, Apartment mixed with detached housing, and tower-type apartment. And These are connected with there values

  • As a public transit playing an important role in the era of high gas prices, subway corresponds the national strategy 'Low carbon, Green growth'; accordingly, passengers who would use the transportation are steadily increasing. Subway operators are required to provide subway users with services based on higher-quality system, safety, convenience & high reliability level. Furthermore, as the legally allowed maximum life time of EMUs is extended, it is necessary to localize & standardize the parts of EMUs. This paper presents a modelling of systematic EMU(SR001) manufacturing technique which enables us to reflect passenger and subway operator's demands in the whole EMU manufacturing process from the initial design phase to its completion by making full use of the know-how based on subway operators' experiences. SR001 enabled SMRT's staffs to participate in the EMU design & its equipment manufacturing; consequently, it realized the innovative system which meets passengers' requirements. Moreover, the process lead time was reduced dramatically. The result of this research will be helpful in providing useful guidelines for railway industry policy and development making in Korea.

  • Since the "Low Carbon, Green Growth" as a social requirements is attracted public attention, the policies and investments of the railway which have been less estimated than road is re-considered. Since the starting the commercial service (2004. April), the KTX have been operated successfully while transport passengers over 100million for years. And also the project for development of KHST (Korea High Speed Train ; G7) had been accomplished with a good result, the first trainset of KTX-II which base on G7 project was manufactured and testing now. On the other hand, the world's technology of the high speed train is more faster, and changing to a distributed traction system. To catch up the world's leading technology, the HEMU(High-speed Electric Multiple Unit) project will secure new technology and aims to promote the technology of domestic high-speed train. The authors indicates the result of detail review such as a curve, slope of the track and electrical dead section of the catenary to test at 400kph and also the performance simulation of the developed rolling stock in this paper. As the result, the authors devise the test scenario to perform a maximum speed test (stability at max. speed, acceleration & deceleration etc) with the restricted conditions such as track length, track available time etc by considering above result.

  • Conventional railways are less competitive than other land transportation means in term of speed, and thus users preference and transportation share for rail system are relatively lower than others. For example, most of the conventional lines except the Seoul~Busan corridor run at an average speed of 70 km/h or less, which imposes certain constraints on roles and functions as the trunk lines. In this regard, the speed of the conventional lines should be improved up to 200 km/h to gain competitiveness, promote balanced regional development and lead the era of low carbon green growth. As track system is one of the most important elements for the speed-up, it is critical to come up with optimum technical solutions. Improvement of ballast track structure with efficient track installation can provide structural stability for higher speed and ensure operational safety with lower maintenance efforts. Thus, this study focuses on consequences followed by the speed-up including increase of load imposed on the track and impacts on track components, and provide solutions for track maintenance by analyzing impact on the track structure by speed. Also, it compares ballast and concrete tracks under designing and construction and considers how to meet needs for passengers comfort and environmental requirements as a strategic approach.

  • There has been a growing interest toward railroad conjunction with opening of high-speed rail in 2004 and concurrent implementation of nation's green growth policies. Meanwhile, users' expectation on service of public transportation, especially railroad has been growing than since ever due to heightened quality of life, entering aging society, voice of equity for minorities, and greater interest in environments of communities. In order to come up with the expectations, actual improvements for user's convenience/comfort and its assessment strategies in terms of service quality level are necessary. This study defines the index of this aspect of service level as 'Sensitivity Quality'. This is conceived as one of the important factors to define travel cost as do the travel time and fare, but hardly taken into consideration when assessing a transportation system or policy-making for a transportation project. The elements included for the assessment of the sensitivity quality in this study are feeling of a ride, conformability and convenience what users feel and recognize using a train. This paper suggests that establishing sensitivity quality elements would contribute in foundation of assessing service quality and provide improved public transportation services through their scientific and objective development and management.
