사람 난소조직의 초자화 냉동보존과 면역결핍 동물에의 이식 (Xenografting of the Human Vitrified Ovarian Tissues into the Immune Deficient Animal)

  • Objective: The present study was conducted to evaluate the viability of germ cells from the adult and fetal ovarian tissues after vitrification followed by xenografting. Method: The human adult ovarian tissues were obtained from 33 years old patient, and the fetal ovarian tissues were obtained from 22 weeks and 25 weeks in gestation. Ovarian tissues were cryopreserved by vitrification with 5.5 M ethylene glycol (EG 5.5) and 1.0 M sucrose as cryoprotectants. Adult and fetal ovarian tissues were pre-equilibrated with EG 5.5 at room temperature for 10 and 5 minutes, respectively and plunged into liquid nitrogen immediately. Frozen-thawed tissues were xenografted into NOD-SCID mice to evaluate the viability and capacity for further growth of the primordial follicles. Grafts were recovered from the recipients 4 weeks after transplantation and histological analysis was accomplished. Result and Conclusion: Grafts recovered 4 weeks after transplantation contained less number of oocytes and primordial follicles compared to that of the fresh tissues. Survived follicles were mainly primordial and intermediary with larger diameter and more granulosa cells. It is confirmed that 1) the ovarian tissues were healthy and the germ cells were survived after vitrification, and 2) the survived fetal primordial follicles after vitrification resumed the growth in the xenografts.

초기배의 발달속도에 따른 후기배로의 배 발달율 (Effect of Cell Stage of Embryos at 48 Hours Post-Insemination On In Vitro Development of IVF Bovine Embryos)

  • This experiment was investigated the effect of cell stage of embryos at 48 hours post-insemination On in vitro development of IVF embryos. The ovaries of Korean native cows or heifers were obtained from an abattoir and kept on 25 to 28$^{\circ}C$ and transported to laboratorty within 2 hrs. The oocytes were matured in vitro(IVM) for 24 hrs. in TGM-199 supplemented with 35 $\mu$g/$m\ell$ FSH, 10 $\mu$g/$m\ell$ LH, 1 $\mu$g/$m\ell$ estradiol-17$\beta$ and granulosa cells at 39$^{\circ}C$ under 5% $CO_2$ in air. They were fertilized in vitro(IVF) by epididymal spermatozoa treated with heparin for 24 hrs. , and then the zygotes were co-cultured in vitro(IVC) with bovine oviductal epithelial cells for 7 to 9 days. At 48 hrs. post-insemination, the embryos were classfied into 5 to 8-cell, 3 to 4-cell or 2-cell stage and then were co-cultured in vitro(IVC) with bovine oviductal epithelial cells until the embyos reached blastocyst stage. Embryos developed to blastocyst stage were stained with Hoechst 33342 for cell counting. The embryos of 5 to 8-cell stage at 48 hrs. post-insemination with grade I oocytes were significantly (P<0.05) better developed to blastocysts(63.0%) than 3 to 4-cell(42.0%) and 2-cell stage(2.7%) embryos which delayed in the early cleavage, and those embryos cleaved faster in the very early stage seemed to develop to blastocysts earlier. These results indicate that the embryos cleaved faster at 48 hrs. post-insemination seemed to develop to blastocysts earlier.

북방산 개구리 여포의 스테로이드생성과정에 협막세포와 난구세포의 역할 (Roles of Theca and Granulosa Cells in Follicular Steroidogenesis in Rana dybowskii)

  • 본인 등은 참개구리를 이용하여 여포의 스테로이드 생성에 관한 two-cell type model을 제시한바 있다. 본 연구에서는 이 model이 북방산 개구리 (R. dybowskii)에도 적용 되는지와 협막세포층에 minor pathway(P 5$\longrightarrow$17$\alpha$-OHP$_4$)가 있는지의 여부를 조사하였다. 이를 위하여 북방산 개구리 난소로부터 intact follicles (IFs), granulosa cell enclosed-oocytes (GcEOs), theca/epithelium (THEP) layers 및 난구세포가 포함되어 있지 않은 순수한 theca/epithelium (P-THEP) layers를 미세해부기술로 분리해 내었다. 이들 여포 조직들은 전구 스테로이드나 개구리 뇌ㅏ수체추출물(FPH)이 포함되어 있는 배양액에서 6시간 배양한 후, 각 여포조직 의해 전환된 산물스테로이드의 양을 방사면역측정법으로 조사하였다.외부에서 첨가된 P 5와 P$_4$는 GcEOs와 IFs에 의하여 효율적으로 P$_4$혹은 17 $\alpha$-OHP$_4$로 전환되었으나 THEP에 의해서는 전환되지 않았다. 더욱이 순수한 협막층(P-THEP)에 의해서는 전환이 거의 이루어지지 않았다. 17 $\alpha$-OHP$_4$및 testosterone 역시 GCEOs와 IFs에 의해서 estradiol (E$_2$) 및 androstenedione (AD)으로 각각 전환되었으나 THEP에 위해서는 정환되지 않았다. 반면에, AD는 THEP과 IFs에 의해서만 T로 전환되어졌으며, AD를 제외한 다른 전구스테로이드들은 THEP 의해서도 T로 전환되지 못했다. 이러한 결과들은 P$_4$, 17 $\alpha$-OHP$_4$AD 및 E$_2$는 주로 난구세포에서 생성되고, T는 주로 협막세포에서 생성되며 T나 E$_2$의 효율적인 생성에 이들 두 세포의 협조가 필요하다는 것을 말해준다. 이는 참개구리에서 제시한 two-cell type model이 북방산개구리 여포의 스테로이드 생성과정에도 적용되며 협막 세포층에는 minor pathway ( P5 to 17$\alpha$-OHP$_4$)가 존재하지 않음을 또한 보여주고 있다.

6 MeV LINAC 방사선 조사에 의한 생쥐 난소조직의 미세구조 변화 (Fine Structural Modification of Mouse Ovarian Tissue by Irradiation of 6 MeV LINAC Radiation)

  • 본 연구는 6 MeV LINAC에서 발생한 X-선을 생쥐 생체에 조사한 후, 방사선 조사선량에 따른 난소 조직의 미세구조적 변화를 광학 및 전자현미경으로 관찰하였다. X-선을 200 cGy의 선량을 조사한 후 적출한 난소조직에서 성장난포의 과립층세포는 핵이 응축되어 나타났고, 난자를 둘러싸고 있는 투명층은 정상 대조군에서보다 경계가 불규칙하게 관찰되었다. 400 cGy로 조사된 난소조직에서 과립층세포는 대부분 응축된 핵을 가지고 있었으며, 피폭된 과립층세포의 사멸에 의한 세포질의 단백질 변성이 확인되었다. 600 cGy의 선량을 조사한 난소의 조직표본에서 성장난포는 위축되어 있었으며, 난포동에는 백혈구가 침윤되어 있었고, 난포액은 정상난포와 달리 불균질한 물질로 채워져 있었다. 투과전자현미경으로 관찰한 결과, 200 cGy의 방사선을 조사한 성장난포에서 과립층세포는 정상난포와는 달리 불규칙한 모양을 하고 있었다. 과립세포 사이의 난포동에는 많은 세포성분의 부스러기가 산재하여 있었으며, 이들 주변에 호중구와 대식세포가 관찰되어, 이 시기에 이미 방사선 피폭에 의한 세포사의 기작이 진행되고 있음이 확인되었다. X-선을 600 cGy로 조사한 난소조직의 과립층세포는 세포의 형태나 핵의 변형이 현저하여, 일부 세포는 핵의 분절화가 진행되는 등, apoptosis의 전형적인 특징이 관찰되었다. 특히 성장난포의 난포동에서는 염증반응에 관여하는 호중구가 잘 관찰되었으며, 주변에 산재된 세포 파쇄물 및 apoptotic body와 함께 대식세포의 출현도 확인되었다.

Polymorphism, Genetic Effect and Association with Egg Production Traits of Chicken Matrix Metalloproteinases 9 Promoter

  • Matrix metalloproteinases (MMP) are key enzymes involved in cell and tissue remodeling during ovarian follicle development and ovulation. The control of MMP9 transcription in ovarian follicles occurs through a core promoter region (-2,400 to -1,700 bp). The aim of this study was to screen genetic variations in the core promoter region and examine MMP9 transcription regulation and reproduction performance. A single cytosine deletion/insertion polymorphism was found at -1954 $C^+/C^-$. Genetic association analysis indicated significant correlation between the deletion genotype ($C^-$) with total egg numbers at 28 weeks (p = 0.031). Furthermore, luciferase-reporter assay showed the deletion genotype ($C^-$) had significantly lower promoter activity than the insertion genotype ($C^+$) in primary granulosa cells (p<0.01). Therefore, the identified polymorphism could be used for marker-assisted selection to improve chicken laying performance.

미성숙 생쥐에서 난포성숙호르몬 처리에 의한 난포의 퇴화 (Follicular Degeneration After Treatment of Follicle Stimulating Hormone in Prepubertal Mouse Ovary)

  • FSH는 미성숙 설치류의 난포성장을 촉진하며, 강소형성 난포의 퇴화비율을 감소시킨다. 본 연구는 미성숙 생쥐에 난포성숙호르몬을 투여한 후 유발되는 난포의 조직학적인 변화를 규명하기 위해 시행되었다. 3주령의 ICR생쥐에 10 i.u.의 재조합 난포자극호르몬을 복강주사한 후 1일, 2일, 3일에 좌측 난소의 조직학적 변화를 관찰하였다. 강소형성전 난포의경우 FSH처리 후 시간에 따라 퇴화난포의 비율이 증가하였으나 강소형성 난포의 경우에는 유의한 변화를 보이지 않았다. 퇴화되는 양상은 난포 내 세포자연사하는 과립세포의 증가, 대식세포 및 다형다핵백혈구의 증가 등이 관찰되었다. 이상의 결과로 보아, 과량의 FSH처리 후에 유발되는 난포의 퇴화는 과립세포의 세포자연사뿐 아니라 급성 염증반응을 수반하는 것으로 생각된다.

  • In order to study the growth and maturation of ovarian follicle, the localization and activity of alkaline phosphatase(ALPase) and adenosine triphosphatase(ATPase) of the granulosa cells and theca layer were examined according to the follicle size, the follicle state and the ovarian cyclic phase in pig. Theca interna of the small follicles was more heavyly localized with reaction product by the activites of ALPase and ATPase than that of the large follicles. It is assumed that, as the follicles proceed to growth and maturation, antrum formation is the result of the follicular fluid accumulation by means of active transport by the activities of ALPase and ATPase in theca interna. The activities of ALPase and ATPase in atretic follicles were higher than those of normal follicles. These results imply that the mechanisms of follicle maturation and atresia are different according to the phase of ovarian cycle.

  • Histological investigation of the number of follicles following gonadotropin treatments for superovulation was carried out in mature Sprague-Dwaley(SD) rats. Routinely serial sections of paraffin-embedded ovaries were stained with hematoxylin-eosin and evaluated with light microscope. During the study unusual cases of microscopic alterations were observed in the medulla of ovaries in two rats. Case one: An ovum and its follicular fluid outflowed in medulla of ovary. The follicular fluid was densly proteinuous. Corona raiata consisted of 2-6 layers thick cells in the periphery of the ovum. While the cortical side of the follicular wall was intact with normal granulosa cell layer the meullary side of it was ruptured. Case two: A large cyst was present in medulla of ovary hilus. The cyst occupied the entire medulla displacing the ovarian archetecture and enclosed by connective tissue and smooth muscle wall.

  • This study was carried out to investigate the ultrastructural changes of ooplasm and follicular membrane of oocytes, obtained from 150 of 3-year-old female rainbow trout(Oncorhynchus mykiss). All data were collected from March in 1989 to February in 1990, and from August to October in 1991. The size of the nucleoli and number of the yolk granules increased as the oocyte growed. Yolk granules were deposited in the oocyte as crystalline granules. Due to the presence of large early and late maturing oocytes, their ovaires were enlarged, transparent and granular. The lattice was broken down at hydration, leaving the egg transparent. As thepercentages of fish in LPO and EMO stage increased from September to October, Mean GSI values increased. Follicle cells such as granulosa cell and thecal cell change a squamous into cuboid shape in LPO and EMO stage. Seasonal changes in the microscopic appearance of the ovaries were well correlated with those in bothgonadosomatic index and macroscopic apearance. Under the natural conditions,t he ovarian follicle influences the histological development and periodical secretion of the hormones, sufficient for a oogenesis and gonadal steroid production. The electrophoretic pattern of major band in mature stage was much thicker(70∼110k dalton) than that in previtellogenic phase.

  • This study was carried out to investigate the histomorphological changes of ovaries obtained from 100 of 1-year-old female Korean loach(misgurnus anguillicaudatus). The light microscopic and ultrastructural changes ofooplasm and follicular membranes of oocytes, were observed by lightand transmission electron microscope during the reproductive cycle. All data were collected from November in 1991 to May in 1992. The results obtained in this study were as follows: The size of the nucleoli and number of the yolk granules increased as the oocytes grown. Yolk granules were loosely deposited in the oocytes as crystalline granules. Due to the presence of large early and late maturing oocytes, their ovaries were enlarged, transparent, granular and yellowish in color. The lattice was broken down at hydration, leaving the egg transparent. As the percentages of fish in LMO and RO stage increased from March to April, mean gonadosomatic index(GSI) values(18.49%) increased. Zona radiata change a squamous into cuboid shape in EMO stage. Processes from the granulosa cells and from the oocyte, microvilli grow and make contact with other in the pore canals of the zona radiata during vitellogenesis, but are withdrawn as the zona radiata becomes more compact and devoid of pore canals during oocyte maturation. Seasonal changes in the microscopic appearance of the ovaries were well correlated with those in both GSI and macroscopic appearance.
