Ultrastructural Changes of Oocyte in Korean Catfish, Silurus asotus (발달 단계에 따른 한국산 메기(Silurus asotus) 난모세포의 미세구조적 변화)

  • This study was carried out to investigate the histomorphological changes and the electrophoretic patterns of egg components, obtained from 100 of 1-year-old female catfish(Silurus asotus). Especially, the light microscopic and ultrastructural changes of ooplasm and follicular membranes of oocytes, were observed by light and transmission electron microscope. All data were collected from October in 1992 to May in 1993. The size of the nucleoli and number of the yolk granules increased as the oocyte grew. Yolk granules were deposited in the oocyte as fluid. Due to the presence of large early and late maturing oocytes, their ovaries were large, transparent, granular, and greenish in color. As the percentages of fish in LMO and RO stage increased from March to April, mean of GSI values(19.95%) increased. Follicle cells such as granulosa cell and thecal cell change a squamous into cuboid shape in LPO and EMO stage. Processes, microvilli, from the granulosa cells and from the oocyte grow and make contact with each other in the pore canals of the zona radiata during vitellogenesis, but are withdrawn as the zona radiate becomes more compact and devoid of pore canals during oocyte maturation. The electrophoretic pattern of major band in mature stage was much thicker(21k, 24k, 32k, 45k, 67∼110k, 170k dalton) than that in previtellogenic phase.

  • Ovarian follicles are faced with one of two fates, atresia or development. Up to 99% of follicles become degenerated rather than ovulated in female life span. Thus, atresia occurs at all stages of follicle development in mammalian ovaries. In the present experiment, the effect of ${\gamma}$-radiation on primordial follicles was morphologically analyzed in a mouse ovary. Thirty-seven percent of the primordial follicles in the non-irradiated control mice ovaries were abnormal. At day 8 post irradiation, most of primordial follicles became atretic. They lost their integrity of architecture in the follicular shape. Then, all the oocytes disappeared from the follicles. And only 3 to 4 granulosa cells lay down onto the basement membrane. Disappearance of granulosa cells or oocytes resulted from the radiation-induced apoptotic process. It is definitely clear that ${\gamma}$-radiation induces rapid apoptotic degeneration of the primordial follicles. The morphological degeneration induced by radiation in the primordial follicles can be used as an experimental model to draw out a deeper insight for radioprotectant researches.

Homing and Restorative Effects of Bone Marrow-Derived Mesenchymal Stem Cells on Cisplatin Injured Ovaries in Rats

  • Premature ovarian failure (POF) is a long-term adverse effect of chemotherapy treatment. However, current available treatment regimens are not optimal. Emerging evidence suggests that bone marrow-derived mesenchymal stem cells (BMSCs) could restore the structure and function of injured tissues, but the homing and restorative effects of BMSCs on chemotherapy injured ovaries are still not clear. In this study, we found that granulosa cell (GC) apoptosis induced by cisplatin was reduced when BMSCs were migrated to granulosa cells (GCs) in vitro. Chemotherapy-induced POF was induced by intraperitoneal injection of cisplatin in rats. BMSCs labeled with enhanced green fluorescent protein (EGFP) were injected into the rats via the tail vein to investigate the homing and distribution of BMSCs in vivo. The number of BMSCs in the ovarian hilum and medulla was greater than in the cortex, but no BMSCs were found in the follicles and corpus lutea. In addition, the BMSCs treatment group's antral follicle count and estradiol levels increased after 30 days, compared with the POF group. Hence, our study demonstrates that intravenously delivered BMSCs can home to the ovaries, and restore its structure and function in POF model rats.

  • Gangilosides are ubiquitous membrane components in mammalian cells and are suggested to play essential roles in cellular phenomena such as cell-cell interadion, differentiation, and signal transduction. Rat ovary contained GM3 as major gangiloside. Nn order to study GM3 distribution in the atretic follicles and its possible changes during follicular development, frozen sections were stained with spedfic monocional antibodies against eleven gangilo-series gangliosides including GM3. In the atretic follicles, Glf3 was expressed in a spatlo-temporally different manner during foilicular development, but GM1 and other gangliosides were not immunohistochemicaily detected. in atretic follicle from primary follicle stage, GM3 was expressed in all the theca cells and some granulosa cells adjacent to oocyte. In atretic follicle from secondary follicle stage, GM3 were expressed in all theca cells and granulosa cells. in atretic follicles from developing Graaflan follicle stages, GM3 was similarly expressed as in secondary follicle stage.

In Vitro Development of Nuclear Transplantation Bovine Embryos Using In Vitro Fertilized Embryos of Korean Native Heifers (한우 체외수정란을 이용한 핵 이식배의 체외발달에 관한 연구)

  • To improve nuclear transplantation(NT) efficiency and to produce a large scale genetically identical cloned calves, examined the in vitro development capacity after co-culture of bovine oviductal epithelial cells (BOEC) and granulosa cells in TCM-199 supplemented with 10% fetal calf serum (FCS) with early bovine embryos derived from in vitro matured fertilized(IVM-IVF) oocyte. In addition, the age dependence of IVM oocyte on electro-stimulation and the effective electric voltage on in ivtro development of bovine NT embryos were examined. The results obtained were summerized as follows; 1. The cleavage rates of IVM-IVF bovine embryos in co-culture with bovine oviductal epithelial cells and granulosa cells were not significantly different(P<0.05), but the developmental rate into morula and blastocyst stage were different showing 38.3 and 20.2%, respectively. 2. The activation (82.5%) and development in vitro(8.6%) into later embryo stages of the aging oocytes of 32 hours post-maturation (hpm) were significantly higher than those of 24 hpm at direct current (DC) voltage of 1.5kV/cm, 60$\mu$sec pulse duration and 1 pulse time. 3. The fusion rates of NT eggs of 32 hpm following to different DC voltages from range 0.75 to 1.5kV/cm were not differ, but the developmental rate into morula and blastocyst stages at DC voltages of 0.75 and 1.0kV/cm were higher(11.4 and 12.6%, respectively) than those of 1.5kV/cm(0%). From these results, it can be suggested the optimal culture system for in vitro culture of IVM-IVF bovine embryos is a co-culture system with BOEC in TCM-199 supplemented 10% FCS. The effective time and the DC voltage for activation, electrofusion and in vitro development of NT embryos derived from IVM-IVF bovine embryo are 32hpm and 0.75~1.0kV/cm. But to improve NT efficiency, the advanced research (cell cycle synchronization, micromanipulation, culture system, etc.) is needed.

An immunohistochemical study on distribution of proliferating cells in uterus and ovary of progesterone-treated rats (Progesterone이 rat 자궁과 난소의 증식세포 분포에 미치는 영향에 대한 면역조직화학적 연구)

  • The study was designed to investigate the effects of progesterone on the reproductive system. This investigation was performed by immunohistochemical methods using anti-bromodeoxyuridine-antibody following bromodeoxyuridine(Brdur) injection for labeling proliferating cells in the uterus and ovary of rats. Sixteen female rats(Wistar), weighing initially 300g, were randomly allotted into ovariectomized and unovariectomized large groups. These two large groups were subdivided into three subgroups of control, 3-day and 6-day groups, respectively. 3-days and 6-days group were injected with 1mg of progesterone/rat/day for 3 or 6 days, respectively. In gross findings, the uterus of ovariectomized groups markedly atrophied, and were not hypertrophied by progesterone injection for 3 days or 6 days and the uterus of unovariectomized groups also were not hypertrophied. Labeling index(LI, %) was measured by counting the number of Brdur-positive cells from 300 to 3,000 cells per layer in the uterus tissue. The average LI of the uterus in unovariectomized groups was higher than that of ovariectomized groups. The subgroups with higher LI in unovariectomized groups were ordered as 6-day group, 3-day group. So progesterone considerably effected to the proliferating of the cells in the uterus of unovariectomized groups. The layers with higher LI in the uterus wall were ordered as the functional zone of endometrium, epithelial layer of endometrium, basal zone of endometrium, myometrium and perimetrium. The cell types with higher LI in the uterus of unovariectomized groups were ordered as the surface epithelial cells, stromal cells, glandular epithelial cells and muscle cells. Growing follicles with proliferating cells from secondary and tertiary follicles in the ovary of unovariectomized groups appeared to be 37.66% in control group, 39.23% in 3-day groups, 39.47% in 6-day groups. Mature follicles in the ovary were more number in control group than those in 3-day groups but not appeared in 6-day groups. So progesterone not nearly effects to the number of the growing follicles but appeared to be related to suppression of the development and protrusion of the mid-tertiary and mature follicles on the ovary surface. The cell types with higher LI in the ovary of unovariectomized groups were respectively ordered as granulosa cells, theca interns cells in secondary follicles; theca interna cells, granulosa cells, theca externa cells in tertiary follicles; fibroblasts, theca in terns cells in atretic follicles; fibroblasts, luteal cells in corpus luteum.

MiR-26a promotes apoptosis of porcine granulosa cells by targeting the 3β-hydroxysteroid-Δ24-reductase gene

  • Objective: Apoptosis of ovarian granulosa cells (GCs) affects mammalian follicular development and fecundity. This study aimed to explore the regulatory relationship between microRNA-26a (miR-26a) and the 3β-hydroxysteroid-Δ24-reductase gene (DHCR24) gene in porcine follicular granular cells (pGCs), and to provide empirical data for the development of methods to improve the reproductive capacity of pigs. Methods: The pGCs were transfected with miR-26a mimic, miR-26a inhibitor and DHCR24-siRNA in vitro. The cell apoptosis rate of pGCs was detected by the flow cytometry. The secretion levels of estradiol (E2) and progesterone (P) in pGCs were detected by enzyme-linked immunosorbent assay. Double luciferase validation system was used to detect the binding sites between miR-26a and DHCR24 3'-UTR region. Qualitative real-time polymerase chain reaction and Western blotting were used to verify the DHCR24 mRNA and protein expression in pGCs, respectively, after transfecting with miR-26a mimic and miR-26a inhibitor. Results: Results showed that enhancement of miR-26a promoted apoptosis, and inhibited E2 and P secretion in pGCs. Meanwhile, inhibition of DHCR24 also upregulated the Caspase-3 expression, reduced the BCL-2 expression, promoted pGCs apoptosis, and inhibited E2 and P secretion in pGCs. There were the binding sites of miR-26a located within DHCR24 3'-UTR. Up-regulation of miR-26a inhibited DHCR24 mRNA and protein expression in pGCs. Conclusion: This study demonstrates that miR-26a can promote cell apoptosis and inhibit E2 and P secretion by inhibiting the expression of DHCR24 in pGCs.

Gonadotropin Regulation of Regulator of G Protein Signaling 2 (RGS-2) Expression in the Rat Ovary (백서 난소에서 성선자극호르몬에 의한 RGS-2의 발현 조절)

  • Objective: The purpose of the present study was to examine the hormonal regulation of RGS-2 in the rat ovary. Methods: Immature rats were injected with 10 IU of PMSG to induce multiple growth of preovulatory follicles and 10 IU of hCG to induce ovulation. Northern blot analysis performed for gene expression and in situ hybridization performed for mRNA localization. Results: Northern blot analysis revealed that pregnant mare's serum gonadotropin (PMSG) treatment did not affect RGS-2 mRNA levels. In contrast, human chorionic gonadotropin (hCG) treatment of PMSG-primed rats resulted in an increase in RGS-2 expression within $1{\sim}3\;h$. The major cell-types expressing RGS-2 mRNA were oocytes regardless of follicle size. Interestingly, hCG treatment caused the stimulation of RGS-2 gene expression in granulosa cells of preovulatory and growing follicles. In contrast, cell types expressing RGS-2 protein were theca cells regardless of hCG treatment. Like in vivo, treatment of preovulatory granulosa cells with LH in vitro stimulated RGS-2 levels within 1 h. Interestingly, GnRH antagonist II enhanced the stimulatory action of LH. Conclusion: The present study demonstrates the LH/hCG induction of RGS-2 in preovulatory granulosa cells and suggests a role of RGS-2 in Gq protein signaling pathway during ovulation.

miR-458b-5p regulates ovarian granulosa cells proliferation through Wnt/β-catenin signaling pathway by targeting catenin beta-1

  • Objective: Ovarian follicular development, which dependent on the proliferation and differentiation of granulosa cells (GCs), is a complex biological process in which miRNA plays an important role. Our previous study showed that miR-458b-5p is associated with ovarian follicular development in chicken. The detailed function and molecular mechanism of miR-458b-5p in GCs is unclear. Methods: The luciferase reporter assay was used to verify the targeting relationship between miR-458b-5p and catenin beta-1 (CTNNB1), which is an important transcriptional regulatory factor of the Wnt/β-catenin pathway. The cell counting kit-8 (CCK-8) assay, flow cytometry with propidium iodide (PI) and annexin V-fluorescein isothiocyanate (FITC) labeling were applied to explore the effect of miR-458b-5p on proliferation, cell cycle and apoptosis of chicken GCs. Quantitative real-time polymerase chain reaction and Western blot were used to detect the mRNA and protein expression levels. Results: We demonstrated that the expression of miR-458b-5p and CTNNB1 showed the opposite relationship in GCs and theca cells of hierarchical follicles. The luciferase reporter assay confirmed that CTNNB1 is the direct target of miR-458b-5p. Using CCK-8 assay and flow cytometry with PI and Annexin V-FITC labeling, we observed that transfection with the miR-458b-5p mimics significantly reduced proliferation and has no effects on apoptosis of chicken GCs. In addition, miR-458b-5p decreased the mRNA and protein expression of CD44 molecule and matrix metallopeptidase 7, which are the downstream effectors of CTNNB1 in Wnt/β-Catenin pathway and play functional roles in cell proliferation. Conclusion: Taken together, the data indicate that miR-458b-5p regulates ovarian GCs proliferation through Wnt/β-catenin signaling pathway by targeting CTNNB1, suggesting that miR-458b-5p and its target gene CTNNB1 may potentially play a role in chicken ovarian follicular development.
