The ultrastructure and histochemical characteristics of the submandibular gland was examined in the big white-toothed shrew, Crocidura lasiura. A submandibular gland of Crocidura lasiura was a mixed gland composed of serous and mucous acinar cells. Secretory granules from the acini were discharged through the intercalated duct, the granular duct and the striated duct into the oral cavity. Serous and mucous acinar cells and granular duct cells had large amount of rough endoplasmic reticulum, free ribosome and prominent Golgi apparatus at the basal cytoplasm of the cell, and many granules at the apical cytoplasm. Oval type serous granules had a homogeneously pale round shape of bead at the center. Mucous granules were distinct from those of the other mammalian species having variety patterns with several dense bands into homogeneous pale matrix. A serous-like secretory granules and myelin-like body were observed in the cytoplasm and the lumen of granular duct cells. The myelin-like body is a characteristic structure only reported in the salivary glands of two shrews, Suncus murinus and C. dsinezumi. Striated duct cell had numerous well-developed mitochondria but secretory granule was not shown at all.
Neural cell survival is an essential concern in the aging brain and many diseases of the central nervous system. Neural transplantation of the stem cells are already applied to clinical trials for many degenerative neurological diseases, including Huntington's disease, Parkinson's disease, and strokes. A critical problem of the neural transplantation is how to reduce their apoptosis and improve cell survival. Neurotrophic factors generally contribute as extrinsic cues to promote cell survival of specific neurons in the developing mammalian brains, but the survival factor for neural stem cell is poorly defined. To understand the mechanism controlling stem cell death and improve cell survival of the transplanted stem cells, we investigated the effect of plausible neurotrophic factors on stem cell survival. The neural stem cell, HiB5, when treated with PDGF prior to transplantation, survived better than cells without PDGF. The resulting survival rate was two fold for four weeks and up to three fold for twelve weeks. When transplanted into dorsal hippocampus, they migrated along hippocampal alveus and integrated into pyramidal cell layers and dentate granule cell layers in an inside out sequence, which is perhaps the endogenous pathway that is similar to that in embryonic neurogenesis. Promotion of the long term-survival and differentiation of the transplanted neural precursors by PDGF may facilitate regeneration in the aging adult brain and probably in the injury sites of the brain.
The primary neuroendocrine carcinomas[NEC] of the lung are relatively rare. There are classified histologically into the following groups. 1] bronchial carcinoid, 2] well differentiated neuroendocrine carcinoma[WDNC], 3] intermediate cell neuroendocrine carci-noma[ICNC], 4] small cell neuroendocrie carcinoma[SCNC]. The NE cells can be found throughout the mucosa of the bronchial tree by the electron microscopy. By the immunocytochemistry, Neuron-specific enolase[NSE], serotonin, born-csin, and calcitonin are found in normal solitary NE cells and neuroepithelial bodies. The immunohistochemistry has the advantage that it can be applied on routine pathology specimens. The NSE was localized in all of the NE granule bearing tumors but also present in 57 per cent of the non NE carcinomas. Together the combination of chromogranin A, bombesin and NSE should provide an exellent signature for tumor cells expressing NE features, such as carcinoids and other NE carcinomas. We have experienced two cases of primary pulmonary NEC in 59 and 29 years old men. The former was admitted due to exertional dyspnea and left chest pain, the latter, newly developed mass on chest X-ray. They were treated by Lt pneumonectomy with LN dissection and RLL resection with LN dissection The postoperative diagnoses were could be confirmed by NSE study as NEC which were SCNEC and INEC in subgroups. The postoperative courses were uneventful.
The cutaneous xanthophore differentiation from larvae to adult in Bombina orientalis-the Korean fire bellied toad-is studied with light and electron microscopes. General structure of adult xanthophore which is composed of many pterinosomes and a small quantity of carotenoid vesicles is forming chromatophore complex with other dermal pigment cells. And the cytoplasmic process of xanthophore is distributed just beneath the basement membrane. The first differentiated xanthophore is originated from both rER rich cells and Golgi complex rich cells before and after the feeding larval stage. Formation of the pigment granule is proceeded gradually along the sequental metamorphic stages. After the metamorphosis, rapid multiplication of pterinosome is observed and enlargement of carotenoid vesicle is appeared after hybernation. These pigment granules are seen several structures by the differentiated level.
Ultrastructure of the cutaneous granular glands and production of their secretory granules in the water toad, Bufo steinegeri Schmidt, are studied with light and electron microscopes. Cutaneous granular glands of the water toad have gland cavity in dermis and gland duct in epidermis. Each gland cavity of the granular glands is consisted of 3 types of cells which are inner glandular epithelial cells, outermost myoepithelial cells and another kind of epithelial cells near the gland duct. Characteristically, cytoplasms of the glandular epitelial cells appeared multinucleated masses without differentiation into cells. Poisonous secretory graules excreated by the merocrine secretion are basicafly composed of 2 kinds of granules which are electron dense granules and electron lucent granules. These granules are fused each other and forming compounded structures. According to the granular size and differentiated levels, they are subdivided into 4 types of granules. Synthesis of these secretory granules is occurred at the smooth endoplasmic reticulums of the glandular epithelial cells and limiting membranes of these granules are also originated from these cell organelles.
In this study, we investigated primary biocompatibility and osteogenic gene expression of porous granular BCP bone substitutes with or without strontium (Sr) doping. In vitro biocompatibility was investigated on fibroblasts like L929 cells and osteoblasts like MG-63 cells using a cell viability assay (MTT) and one cell morphological observation by SEM, respectively. MTT results showed a cell viability percent of L929 fibroblasts, which was higher in Sr-BCP granules (98-101%) than in the non-doped granules (92-96%, p < 0.05). Osteoblasts like MG-63 cells were also found to proliferate better on Sr-doped BCP granules (01-111%) than on the non-doped ones (92-99%, p < 0.05) using an MTT assay. As compared with pure BCP granules, SEM images of MG-63 cells grown on sample surfaces confirmed that cellular spreading, adhesion and proliferation were facilitated by Sr doping on BCP. Active filopodial growth of MG-63 cells was also observed on Sr-doped BCP granules. The cells on Sr-doped BCP granules were well attached and spread out. Gene expression of osteonectin, osteopontin and osteoprotegrin were also evaluated using reverse transcriptase polymerase chain reaction (RT-PCR), which showed that the mRNA phenotypes of these genes were well maintained and expressed in Sr-doped BCP granules. These results suggest that Sr doping in a porous BCP granule can potentially enhance the biocompatibility and bone ingrowth capability of BCP biomaterials.
Preparative isolation of lipid granules from Fhodotorula glutinis, which has been studied for long time to produce edible lipids, was carried out by flotation method in Ficoll-Linear density gradient. When the isolated lipid granules were suspended in a series of solutions containing varying concentration of osmotic stabilizer (sorbitoal and mannitol) ranging from 0.8M to 0M, the lipid granules appeared to be disrupted at a concentration between 0.8M and 0.7, and again at a concentration below 0.1M, suggesting that lipid granules have a membraneous structure and that at least two types of lipid granules are present. Compositional analysis of lipids from lipid granules revealed that lipids are composed mainly of neutral lipids (87.8% of total lipids), predominantly as triacylglycerols (71.89%). Marked differences were observed inphospholipids between lipids of lipid granules and those of whole cells . The major components of phospholipids in lipid granules and inwhole cells are phosphatidylcholine(38.6%) and phosphatidylserine(42.8%), respectively. In addition, significant differences were also observed in the fatty acid composition of phospholipids. As phospholipids are important structural components of membranes, these differences lead to the suggesting that the membrane of lipid granules may be distinct functionally and structurally from other membranes of yeast cells. The major fatty acid components of neutral lipidss of whole cells and lipid granules are palmitic , oleic and linoleic acid. However , degreeof fatty acid unsaturation of neutal lipids of lipid granules was much lower than that of neutral lipids of whole cells.
The effect of aromatic amino acids such as phenylalaine, tryptophan and tyrosine on somitogenesis at the early stage of chick embryo has been investigated morphologically using light and electron microscopy. Micrographs of aromatic amino acid injected chick embryo showed that an incomplete somite segmentation occurred and some decremental effect on the nervous system were observed. Somites were poorly developed and their size were variable. Electron micrograph of somatic cells from aromatic amino acid injected chick embryo showed that chromatins were coagulated, some of mitochondria were damaged, and nucleus were transformed considerably in some cases. The protein and nucleic acid levels and some enzyme activities of 15-day chick embryo which received the injection of 1mg of aromatic amino acid in 0.05 ml of saline 24 hours after the incubation were analyzed. Protein, DNA and RNA levels of the test group were not lowered significantly but the activities of enzymes for basic metabolism, such as lactate dehydrogenase, succinate dehydrogenase, malate dehydrogenase and glucose 6-phosphate dehydrogenase were considerably lowered as compared with those of control. From the present expeerimental results, it was tentatively suggested that the administration of amino acid might slow down the yolk granule degradation probably by feed back mechanism resulting in the disturbance of amino acid balance in the cell, which might give rise to impair normal metabolic pattern leading to abnormal somitogenesis to chick embryo at very early stage of development.

The study was conducted to know the cyclic changes in gonadal maturation and to investigate the developmental stages of oocytes and testicular germ cells of an air-breathing fish, Channa striata. Fish were sampled monthly from lentic and lotic environments of three geographical locations of Bangladesh from December to November and the histological analysis of their gonad was done to evaluate the objectives. The highest mean GSI was $5.95{\pm}0.20$ for female in July and $0.14{\pm}0.01$ for male also in July showing that the gonadal development reached its peak during this month. The highest mean oocyte diameter was $1257.50{\pm}24.17{\mu}m$ observed in July implying that the oocyte reached maturity in this month. Histological study of ovary revealed the evidence of early yolk granule stage and late yolk granule stage from April to July. In case of male four stages of spermatogenesis were distinguished and spermatozoa were highly abundant in June and July. So the monthly pooled values of GSI and the analysis of gonadal histology indicated that the peak breeding season of C. striata occurred in July in the lentic and lotic environments. Samples collected from lentic and lotic habitats are suggestive of no difference in the development of the gonad. The results of the present study will be useful for selective breeding programme, conservation and sustainable fishery management of C. striata in its natural habitat.
These studies were designed to examine the differential effect of nitric oxide (NO) and cGMP on glutamate neurotransmission. In primary cultures of rat cerebellar granule cells, the glutamate receptor agonist N-methyl-D-aspartate (NMDA) stimulates the elevation of intracellular calcium concentration ($[Ca^{2+}]_i$), the release of glutamate, the synthesis of NO and an increase of cGMP. Although NO has been shown to stimulate guanylyl cyclase, it is unclear yet whether NO alters the NMDA-induced glutamate release and ${[Ca^{2+}]}_i$ elevation. We showed that the NO synthase inhibitor, NG-monomethyl-L-arginine (NMMA), partially prevented the NMDA-induced release of glutamate and elevation of ${[Ca^{2+}]}_i$ and completely blocked the elevation of cGMP. These effects of NO on glutamate release and [Ca2+]i elevation were unlikely to be secondary to cGMP as the cGMP analogue, dibutyryl cGMP (dBcGMP), did not suppress the effects of NMDA. Rather, dBcGMP slightly augmented the NMDA-induced elevation of ${[Ca^{2+}]}_i$ with no change in the basal level of glutamate or ${[Ca^{2+}]}_i$. The extracellular NO scavenger hydroxocobalamine prevented the NMDA-induced release of glutamate providing indirect evidence that the effect of NO may act on the NMDA receptor. These results suggest that low concentration of NO has a role in maintaining the NMDA receptor activation in a cGMP-independent manner.
